AI Technical Summary

Technical Problem

In the current technology, there is limited research on FGF5 in tumor angiogenesis, and there is a lack of effective inhibition methods, which makes it difficult to control tumor angiogenesis and growth.

Method used

Anti-tumor angiogenesis drugs were prepared by using siRNA targeting the gene encoding fibroblast growth factor 5 to inhibit the expression or function of FGF5, thereby reducing the migration and invasion capabilities of tumor vascular endothelial cells.

Benefits of technology

Effectively inhibiting tumor angiogenesis and slowing tumor formation and growth, by knocking down or silencing FGF5 gene expression, significantly reducing the migration and tubule formation ability of tumor endothelial cells, verifying the role of FGF5 in promoting tumor growth.

Abstract

The invention provides application of an FGF5 gene inhibitor in preparation of anti-tumor angiogenesis drugs, and belongs to the technical field of biological medicines. The invention provides application of a reagent for inhibiting expression of an FGF5 coding gene or a reagent for inhibiting expression or functions of FGF5 in preparation of anti-tumor angiogenesis drugs. In the invention, tumor angiogenesis can be inhibited by inhibiting expression of FGF5. Results of the embodiment of the invention show that overexpression of the FGF5 protein and the coding gene thereof can effectively promote migration and invasion of human umbilical veins and tubule formation, and knock-down of expression of the FGF5 inhibits migration and invasion ability of the human umbilical veins and tubule formation ability. A subcutaneous tumor generation experiment further verifies that the FGF5 can promote the growth of a subcutaneous tumor HGC27 of a nude mouse, and promote tumor angiogenesis and tumor endothelial cell proliferation and migration.
