AI Technical Summary
Technical Problem
There is a need for compositions and methods that allow for controlled release of glycan precursors in live cell settings, particularly for regulating glycosylation pathways in a spatiotemporal manner.
Method used
The development of controllably-releasable glycan precursors, which include a photocage or chemocage component covalently bonded to a glycan precursor component, enabling controlled release upon exposure to light or specific bioorthogonal chemical reactions.
Benefits of technology
This approach provides spatiotemporal control over glycosylation pathways, allowing for real-time chemical control of sugar-driven phenotypes in living cells without interfering with cellular processes.

Abstract
Compositions are provided according to aspects of the present disclosure which include: a controllably-releasable glycan precursor, the controllably-releasable glycan precursor having a photocage component or a chemocage component covalently bonded to a glycan precursor component. Methods of controlled metabolic engineering in a cell are provided which include introducing a controllably-releasable glycan precursor of the present disclosure into a cell; and exposing the controllably-releasable glycan precursor in the cell to a stimulus effective to release the glycan precursor from the photocage component or chemocage component, thereby providing the glycan precursor to the cell, and providing controlled metabolic engineering in the cell.
