AI Technical Summary

Technical Problem

Existing digital PCR systems are complex, costly, and require skilled personnel due to the need for multiple devices for droplet generation, amplification, and fluorescence signal reading, which hinders the speed of target sequence detection.

Method used

A compact, cost-effective, and integrated digital PCR system that consolidates all procedures into a microfluidic chip and a detection instrument with a small footprint, featuring a simplified design with a flow induction unit, optical detector, and signal processor for digital PCR analysis.

Benefits of technology

The system achieves absolute quantification of nucleic acids, allows for specific detection of rare gene mutations, and provides high precision in distinguishing allelic copy number variations, while reducing operational complexity and increasing speed of target sequence detection.

Abstract

A digital PCR system includes (a) a microfluidic chip having a sample inlet, an oil inlet, a nozzle, in fluid communication with the sample inlet and the oil inlet to mix the sample and the oil phase to generate a water-in-oil microdroplet; a microdroplet reaction region for performing thermal cycling with the microdroplet to form a reacted microdroplet; an optical detection region, configured to allow detection of a fluorescence signal from the reacted microdroplet; and (b) a chip processing instrument having an optical detection module for detecting a fluorescent signal, and a temperature control module for thermally regulating temperature of the microdroplet reaction region.
