PREPARATION METHOD OF A BRUTON’S TYROSINE KINASE DEGRADER THEREOF1. FIELD

[0001] Provided herein are processes generally pertaining to the field of chemical synthesis, and more specifically to methods of synthesis of certain compounds of formula (II) and formula (III) used as intermediates for preparing (R) -3- (tert-butyl) -N- (1- (4- (6- (6- (4- ( (1- (4- (2, 4-dioxotetrahydropyrimidin-1 (2H) -yl) phenyl) piperidin-4-yl) methyl) piperazin-1-yl) pyridin-3-yl) -7H-pyrrolo [2, 3-d] pyrimidin-4-yl) -2-methylphenyl) ethyl) -1, 2, 4-oxadiazole-5-carboxamide, a Bruton’s tyrosine kinase (BTK) degrader.2. BACKGROUND

[0002] Bruton’s tyrosine kinase (BTK) belongs to the Tec tyrosine kinase family primarily expressed in most hematopoietic cells such as B cells. BTK plays an important role in B-cell receptor (BCR) and FcR signaling pathways, which involve in B cell development and differentiation (Khan, Immunol. Res. 23: 147, 2001) . Inhibition of BTK has been shown to affect cancer development (B cell malignancies) . Inhibition of BTK has also been reported via alternative strategies, such as through degradation of BTK. International publication WO2021219070 disclosed BTK degraders.
