[0003] Currently, only the content of individual characteristic components has been determined, and there is no literature report on the compositional qualitative analysis and comprehensive quality evaluation of QCBP. Summary of the invention
[0004] The invention aims to provide a quality evaluation method for Qingxin Chenxiang Bawei Pills.
[0005] The present invention uses HPLC-Q-Exactive-MS technology combined with literature data to identify and analyze the chemical components of commercially available QCBP, establishes fingerprints of 13 batches of commercially available QCBP, and conducts comprehensive quality evaluation of QCBP through cluster analysis (CA), principal component analysis (PCA), orthogonal partial least squares discriminant analysis (OPLS-DA) and variance analysis, and quantitatively analyzes 4 differential components, providing a basis and reference for quality control of commercially available QCBP.
[0006] The purpose of the present invention is implemented by the following technical scheme: a method for evaluating the quality of Qingxin Chenxiang Bawei Pills, comprising the following steps: (1) establishing fingerprints of each QCBP test solution and calibrating common peaks; (2) performing multi-dimensional quality difference evaluation and analysis on a number of QCBP test samples; (3) if there is a quality difference, screening out the key factors that cause the large differences between the QCBP test samples from the calibrated common peaks of the QCBP test solutions; (4) determining the content of each component that causes the sample quality difference; wherein, (1) Establish fingerprints of each test solution and calibrate common peaks: Take several QCBP samples, prepare them into QCBP test solutions, perform chromatographic analysis on them, record the chromatograms, convert the collected chromatograms into CDF format files and import them into the Chinese medicine chromatographic fingerprint similarity evaluation system, select the chromatogram of any QCBP test solution and set it as the reference spectrum for fingerprint analysis, after multi-point correction and Marker peak matching, obtain the chromatogram overlay and reference fingerprint of each QCBP test solution, and calibrate several common peaks; after comparing with the spectrum of each single reference stock solution, multiple chromatographic peaks are identified; (2) Conduct multi-dimensional quality difference evaluation and analysis on several QCBP test products: conduct fingerprint similarity evaluation on the fingerprints established in step (1); conduct cluster analysis with several common peak areas as indicators; conduct principal component analysis with several common peak areas as variables; use orthogonal partial least squares discriminant analysis and variance analysis to analyze the differences in several common peak areas; and determine whether there are quality differences among several QCBP test products based on the above analysis results; (3) If there are quality differences, the key factors that cause the large differences between the QCBP test samples are screened out from the common peaks of the calibrated QCBP test sample solutions: evaluation method based on variable weight values [25-26] Each QCBP test product was analyzed. When the VIP value was > 1, it indicated that the component was the key factor causing the large differences among the QCBP test products; (4) Determination of the content of each component that causes the difference in sample quality: Weigh the QCBP test solution separately, and use the HPLC-MS quantitative analysis method to calculate the content of each component that causes the difference in sample quality in each sample, and determine the content difference of each component that causes the difference in sample quality in the QCBP test solution.
[0007] Specifically, in the step (1), 13 batches of commercially available QCBP samples from three manufacturers were taken; 20 common peaks were calibrated, of which 6 chromatographic peaks were identified, namely: peak 1 was gallic acid, peak 3 was protocatechuic acid, peak 5 was hydroxysafflor yellow A, peak 6 was agarwood tetraol, peak 7 was ferulic acid, and peak 10 was ellagic acid; the single reference substance stock solutions were ferulic acid stock solution, agarwood tetraol stock solution, gallic acid stock solution, hydroxysafflor yellow A stock solution, protocatechuic acid stock solution, quinic acid stock solution, kaempferol stock solution, costunolide stock solution, and rutin stock solution; In step (2), there were quality differences among 13 batches of commercially available QCBP samples from three manufacturers; In the step (3), there are 14 common peaks with VIP values greater than 1, which are arranged from large to small: peak 2, peak 16, peak 15, peak 17, peak 11, ferulic acid, agarwood tetraol, peak 20, peak 18, gallic acid, peak 19, hydroxysafflor yellow A, peak 12, and peak 9; that is, these 14 chromatographic peaks are the key factors causing the quality differences of the 13 batches of samples; In the step (4), the contents of ferulic acid, agarwood tetraol, gallic acid and hydroxysafflor yellow A, which are key factors leading to sample quality differences, are measured to determine the content differences of the four components in the test solution.
[0008] Specifically, the preparation method of the QCBP test solution is as follows: weigh the QCBP samples respectively, grind them into powder, sieve them, add a methanol solution with a mass percentage concentration of 70%-80% to dissolve them, seal them, and ultrasonicate them for 30min-50min, with an ultrasonic power of 60W-80W and an ultrasonic frequency of 50kHz-90kHz. After cooling to room temperature, add a methanol solution with a mass percentage concentration of 70%-80%, shake them well, and filter them to obtain each QCBP test solution.
[0009] Specifically, the mass volume ratio of the sieved commercially available QCBP sample powder to the methanol solution with a mass percentage concentration of 70%-80% is 0.1 g:1 mL~0.2 g:1 ml.
[0010] Specifically, in the step (1), the chromatographic analysis conditions are as follows: the mobile phase is methanol (A)-0.2% phosphoric acid aqueous solution (B), the flow rate is 1.0 mL / min, the column temperature is 35°C, the injection volume is 10 μL, and the detection wavelength is 254 nm; the gradient elution program is: 0-8 min, 15%→30% A; 8-17 min, 30%→35% A; 17-20 min, 35%→35% A; 20-25 min, 35%→40% A; 25-30 min, 40%~45% A; 30-50 min, 45%~60% A; 50-55 min, 60%→65% A; 55-65 min, 65%→65% A; 65-70 min, 65%~80% A; 70-80 min, 80%~15% A; 80-90 min, 15%~15% A.
[0011] Specifically, in the step (4), the HPLC-MS quantitative analysis method, the chromatographic column quantitative analysis conditions are: the mobile phase is methanol (A)-water (containing 0.1% formic acid) (B), the gradient elution program is: 0.01~0.5min, 15%~20%A; 0.5~1.0min, 20%~35%A; 1~3 min, 35%~60%A; 3.0~5.0min, 65%~75%A; 5.0~5.5min, 75%~95%A; 5.5~7.0min, 95%A; 7.0~7.01min, 95%~10%A; 7.01~9.5min, 10%A; volume flow rate 0.25mL / min; column temperature 35℃; injection volume 3μL.
[0012] Specifically, in the step (4), the HPLC-MS quantitative analysis method and the mass spectrometry quantitative analysis conditions are: using an electrospray ion source (ESI); positive and negative ion scanning; multiple reaction monitoring (MRM) mode; spray voltage 3.80 kV (+), 3.50 kV (-), nebulizing gas volume flow rate 3 L / min; heating gas volume flow rate 10 L / min; interface temperature 300°C; desolvation temperature 526°C; heating block temperature 400°C.
[0013] Advantages of the present invention: (1) The present invention is based on the component analysis of HPLC-Q-Exactive-MS technology combined with the establishment of HPLC fingerprint and the quantitative evaluation of HPLC-MS differential components, which provides a reference for the quality evaluation system of commercially available QCBP.
[0014] (2) The present invention investigated the characterization effects of samples with different ultrasonic extraction times, different mobile phases and different concentrations. The results showed that when the ultrasonic extraction time was 40 min, the mobile phase was methanol and 0.1% formic acid water as the mobile phase for gradient elution, and the sample was diluted 1000 times, the separation effect of each component, mass spectrum abundance and peak shape were better than those under other conditions.
[0015] (3) To ensure that the detection conditions are stable and feasible, and the HPLC fingerprint characterization information is comprehensive, the mobile phase, chromatographic column, and detection wavelength were investigated on the basis of HPLC-Q-Exactive-MS conditions. The experiment found that the detection wavelength was significantly higher when methanol and 0.2% phosphoric acid water were used as the mobile phase and the chromatographic column was Agilent Eclipse XDB-C 18 The fingerprint spectrum with a detection wavelength of 254 nm and a diameter of 4.6 mm × 250 mm (5 μm) can make each chromatographic peak achieve the most ideal peak shape and response value, and can more intuitively reflect the information of multi-component chromatographic peaks. Peak 3 (gallic acid) is used as the reference peak. The results show that the relative retention time RSD of the 20 common peaks in the sample is less than 0.10%, and the relative peak area RSD is less than 1.72%, indicating that the method has good repeatability.
[0057] 2.7.4 Fingerprint establishment Thirteen batches of commercially available QCBP from three manufacturers were used to prepare test sample solutions of each batch according to the method in "2.2". Samples were injected and analyzed according to the chromatographic conditions in "2.4". Chromatograms were recorded. The collected chromatograms were converted into CDF format files and imported into the Chinese Medicine Chromatographic Fingerprint Similarity Evaluation System (2012 Edition). Sample S1 was set to perform fingerprint analysis with reference to the chromatogram (the method was the median method and the time window was set to 0.1 min). After multi-point calibration and Marker peak matching, the QCBP chromatogram overlay and reference fingerprint were obtained, as shown in Figure 2. Fig. 9 As shown in the figure, the calibration results showed that there were 20 peaks in total. After comparison with the spectra of the single reference stock solutions prepared in 2.1, Fig.10 As shown, six chromatographic peaks were identified, namely gallic acid (peak 1), protocatechuic acid (peak 3), hydroxysafflor yellow A (peak 5), agalate tetraol (peak 6), ferulic acid (peak 7), and ellagic acid (peak 10).
[0058] 2.7.5 Similarity Analysis The data of 13 batches of commercially available QCBP were imported into the Traditional Chinese Medicine Fingerprint Similarity Evaluation System (2012 Edition) for fingerprint similarity evaluation. The similarity results of the fingerprints of 13 batches of commercially available QCBP and the generated reference fingerprints are shown in Table 2. The similarity of the 13 batches of commercially available QCBP was between 0.769 and 0.960, indicating that there were certain differences in the quality of the 13 batches of QCBP from three manufacturers. Taking peak 3 (protocatechuic acid) as the reference peak, the relative peak area RSD of 20 common peaks of 13 batches of QCBP was 28.0%~96.0%, indicating that there were large differences in the content of certain components of different batches of QCBP.
[0059] Table 2: Similarity evaluation results of 13 batches of commercially available QCBP
[0060] 2.8 Chemical pattern recognition analysis 2.8.1 Cluster Analysis (CA) The Weishengxin online website platform was used to analyze the 20 common peak areas of 13 batches of commercially available QCBP. The clustering method was the completion method, and the distance metric was set to euclidean.
[22] , the results are as follows Fig.11 As shown in the figure, the 13 batches of QCBP can be clustered into two categories, S1~S6, S10~S13 are clustered into one category (Manufacturer A, C), and S7~S9 are clustered into one category (Manufacturer B), which is consistent with the similarity results.
[0061] 2.8.2 Principal Component Analysis (PCA) The 20 common peak areas of 13 batches of QCBP were used as variables and the principal component analysis was performed using SIMCA 14.1 software. The results are as follows: Fig.12 , manufacturers A and C are clearly separated from manufacturer B, and the results are consistent with those of CA.
[0062] 2.8.3 Orthogonal Partial Least Squares-Discriminant Analysis (OPLS-DA) SIMCA 14.1 software was used to select supervised OPLS-DA to analyze the differences in 20 common peak areas among 13 batches of QCBP products. The cumulative explanatory power parameters of the constructed supervised model were ( R 2 X , R 2 Y ) were 0.783 and 0.903 respectively, and the prediction ability parameters Q 2 (cum) is 0.805, and all three parameters are greater than 0.5, indicating that the OPLS-DA model has good explanatory power and stability. [23-24] Based on the above, the score diagram is shown in Fig.13 , the samples from the three manufacturers were clustered into one category, and the classification results were more obvious, which further indicated that there were quality differences among the QCBP produced by different manufacturers.
[0063] In order to screen out the components that cause large inter-batch differences, 13 batches of samples were analyzed using the variable importance in projection (VIP) evaluation method. When the VIP value is greater than 1, it indicates that the component is the key factor in the inter-group classification. [25-26] .Depend on Fig.14 It can be seen that there are 14 common peaks with VIP values >1, which are arranged from large to small: peak 2 (VIP=1.367), peak 16 (VIP=1.100), peak 15 (VIP=1.096), peak 17 (VIP=1.095), peak 11 (VIP=1.090), ferulic acid (VIP=1.082), agarwood tetraol (VIP=1.064), peak 20 (VIP=1.063), peak 18 (VIP=1.053), gallic acid (VIP=1.044), peak 19 (VIP=1.039), hydroxysafflor yellow A (VIP=1.033), peak 12 (VIP=1.030), peak 9 (VIP=1.012). That is, these 14 chromatographic peaks are the key factors causing the quality differences of 13 batches of samples.

Regression analysis was performed with the injection volume of the mixed standard solution as the horizontal axis (X) and the peak area as the vertical axis (Y). The results are shown in Table 4, which shows that the linear relationship of each component is good within its respective range.
[0065] Table 3: Mass spectrometry parameters of each component

The samples were injected and analyzed under the conditions of "2.3". The RSDs of the peak areas of ferulic acid, agarwood tetraol, gallic acid and hydroxysafflor yellow A were 4.46%, 1.63%, 1.61% and 4.06%, respectively, indicating that the method has good repeatability.
[0069] 2.9.4 Stability test Take QCBP sample S7, prepare the test solution according to item "2.2", and analyze the samples at 0, 2, 4, 8, 12, and 24 h under the conditions of item "2.3". The RSDs of the peak areas of ferulic acid, agarwood tetraol, gallic acid, and hydroxysafflor yellow A were 0.76%, 1.86%, 1.11%, and 1.22%, respectively, indicating that the sample solution has good stability within 24 h.

[0074] 3 Discussion High performance liquid chromatography-quadrupole / electrostatic field orbital trap high resolution mass spectrometry (HPLC-Q-Exactive-MS) has the characteristics of high resolution (resolution greater than 10000 (FWHM)), high sensitivity, and high selectivity. [27-28] The present invention investigated the characterization effects of samples with different ultrasonic extraction times, different mobile phases and different concentrations. The results showed that when the ultrasonic extraction time was 40 min, the mobile phase was methanol and 0.1% formic acid water as the mobile phase for gradient elution, and the sample was diluted 1000 times, the separation effect of each component, mass spectrum abundance and peak shape were better than other conditions. In order to make the detection conditions stable and feasible, the HPLC fingerprint characterization information was comprehensive. Based on the HPLC-Q-Exactive-MS conditions, the mobile phase, chromatographic column and detection wavelength were investigated by single factors. The experiment found that when methanol and 0.2% phosphoric acid water were used as the mobile phase and the chromatographic column was Agilent Eclipse XDB-C 18 The chromatogram with a diameter of 4.6 mm × 250 mm, 5 μm and a detection wavelength of 254 nm can make each chromatographic peak achieve the most ideal peak shape and response value, and can more intuitively reflect the information of multi-component chromatographic peaks.
[0075] The main drug in QCBP is agarwood, of which agarwood tetraol is listed as an indicator component of agarwood in the 2020 edition of the Chinese Pharmacopoeia.
[29] Agarwood tetraol is a chromone component, which has antioxidant
[30] , improve the body's immunity
[31] and inflammation regulation
[32] , there are 8 chromone components in the chemical composition analysis results of QCBP based on HPLC-Q-Exactive-MS, which can provide a material basis for subsequent research. Through HPLC fingerprint similarity evaluation combined with CA, PCA and OPLS-DA analysis, 13 batches of QCBP on the market were clustered into two categories (S1~S6, S10~S13 were clustered into one category (A, C manufacturers), S7~S9 were clustered into one category (B manufacturer)), and the clustering in S1~S6 of manufacturer A was relatively scattered, indicating that the intra-group differences were large. The reasons for the intra-group differences may be related to the origin, processing, and harvesting of the medicinal materials used. The 13 batches of samples were analyzed by the variable weight value evaluation method, and 14 common peaks had VIP values greater than 1. Among them, ferulic acid, agarwood tetraol, gallic acid, and hydroxysafflor yellow A were determined by HPLC-MS. The results showed that the contents of these four components were quite different.
[0076] In summary, the present invention provides a reference for the quality evaluation system of commercially available QCBP based on the component analysis of HPLC-Q-Exactive-MS technology combined with the establishment of HPLC fingerprint and the quantitative evaluation study of HPLC-MS differential components.
