[0002] Gene therapy technology is a hot topic of research in modern biomedical field, and the nucleic acid drugs can prevent and treat cancer, bacterial and viral infections, and treat diseases with inherited pathogenic factor, etc. Since nucleic acid drugs are easily degradable and difficult to enter cells, they usually need to be encapsulated and delivered to target cells with the help of carriers, therefore, the development of safe and efficient delivery carriers is a precondition for the clinical application of gene therapy.

[0555] Cholesterol in the blood is mainly synthesized by the liver, which is also the main organ for breaking down excess cholesterol. The presence of an LDL receptor (LDLR) on the surface of the liver that binds to cholesterol circulating back to the liver, which in turn breaks it down into bile acids that are excreted out of the body via the intestine; PCSK9 is a liver-synthesized protease that binds to LDL receptors and promotes LDL receptor entry into hepatocytes, leading to LDL receptor degradation by lysosomes and a decrease in the number of LDL receptors; thus,  inhibition of the activity of PSCK9, can increase the number of LDLR, thereby enhancing the uptake and decomposition of cholesterol. Basic and clinical studies have shown that the PCSK9 gene is an effective target for the treatment of hyperlipidemia and atherosclerosis. Figure 1 illustrates the changes in the number of LDL receptors and the eventual changes in cholesterol metabolism that result before and after site-specific editing of the PCSK9 gene.

[0576] 3.1 Lipid nanoparticles encapsulating mRNA and sgRNA encoding the base editor ABE8e prepared in step 2 and containing dilinoleylmethyl-4-dimethylaminobutyrate (DLin-MC3-DMA, usually abbreviated to MC3) , the compound of the present invention (see Table 2) , were injected via tail vein at a dose of 0.1 mg / kg into 6-7 week old C57BL / 6 female mice (purchased from Gempharmatech Co., Ltd) , respectively. Lipid nanoparticles containing dilinoleylmethyl-4-dimethylaminobutyrate (DLin-MC3-DMA, usually abbreviated to MC3) encapsulating mRNA and sgRNA of base editor ABE8e were administered to mice of comparable week age and sex groups as positive controls. In addition, PBS buffer was also injected caudally intravenously in a similar manner into mice of comparable week age and sex groups as a negative control.
