Abstract

Provided in the present invention are a series of substrate binding pocket-modified firefly luciferases having high enzyme activity and high thermal stability, a preparation method therefor and the use thereof. Through the analysis of the spatial structure of the substrate binding pocket of the firefly luciferase, and on the basis of considering a large number of amino acid sites related to enzyme activity and thermal stability of firefly luciferase, the present inventors design, construct and detect a series of firefly luciferase mutants. The firefly luciferase has a high enzyme activity and thermal stability.
