AI Technical Summary
Technical Problem
Generating multispecific antibodies that exhibit desired activity and minimize off-target activity is challenging due to a lack of knowledge about the spatial location of epitopes on cell surfaces relative to therapeutic targets.
Method used
A novel multispecific antibody format is developed by linking antigen-binding domains via linkers of different lengths and sequences, optimizing antigen binding and minimizing off-target activity.
Benefits of technology
The novel format allows for fine-tuning of multispecificity, enhancing on-target activity while reducing off-target effects, thereby delivering effective and safe therapeutics.

Abstract
The present invention provides a novel multispecific antigen-binding protein capable of binding to multiple targets, a pharmaceutical composition containing the multispecific antigen-binding protein, and a method for producing these. [Solution] A multispecific antibody construct comprising: a) a first polypeptide comprising an antibody Fc region including a first hinge region, a first CH2 region, and a first CH3 region; b) a second polypeptide comprising an antibody heavy chain construct, wherein the antibody heavy chain construct comprises i) scFv and ii) an antibody heavy chain, wherein the scFv is bound at its C-terminus to the N-terminus of a second VH region of the antibody heavy chain; and c) a third polypeptide comprising an antibody light chain including a second VL and CL, wherein the second VH of the antibody heavy chain and the second VL of the antibody light chain are bound to form a second antigen-binding domain.
