[0112] (Example 1) Synthesis of 5-bromo-2-(4-cyclopropyl-2-fluoroanilino)-3,4-difluorobenzamide A reaction vessel containing a 24% lithium bis(trimethylsilyl)amide THF solution (94.7 kg, 136 mol) was cooled to an external temperature of -15°C, and a solution of 4-cyclopropyl-2-fluoroaniline (8.1 kg, 53.5 mol) in THF (18.7 kg) was added dropwise at an internal temperature of 5°C or below. The reaction vessel was then rinsed with THF (4.7 kg), and a solution of 5-bromo-2,3,4-trifluorobenzoic acid (10.5 kg, 41.2 mol) in THF (65.3 kg) was added dropwise while maintaining the internal temperature at 5°C or below, followed by stirring for 30 minutes. 5M hydrochloric acid (88.7 kg) was added to the reaction mixture, which was then warmed to room temperature and extracted with isopropyl acetate (45.9 kg). The organic layer was washed once with 2M hydrochloric acid (42.8 kg) and twice with 15% aqueous sodium chloride solution (52.5 kg), and then concentrated under reduced pressure. THF (74.8 kg) was added to the resulting concentrate, and the mixture was concentrated until the total volume reached 75 L. THF (74.8 kg) was again added to the concentrate, and the mixture was concentrated until the total volume reached 75 L. Acetonitrile (100 kg) and THF (31.6 kg) were then sequentially added to the concentrate. 1,1'-carbonyldiimidazole (10.0 kg, 48.8 mmol) was added to the resulting solution, and the mixture was stirred at room temperature for 1 hour. 28% aqueous ammonia (14.5 kg) was added to the reaction mixture, and the mixture was stirred at room temperature for 1.5 hours. The resulting suspension was heated to 60°C. After confirming dissolution of the suspended matter, the solution was cooled to 25°C, and seed crystals (80 g) of 5-bromo-2-(4-cyclopropyl-2-fluoroanilino)-3,4-difluorobenzamide and water (191 kg) were sequentially added. The precipitated crystals were collected by filtration, and the filter cake was washed twice with a mixed liquid of acetonitrile (18.7 kg) and water (23.9 kg) and then dried under reduced pressure at an external temperature of 40°C to obtain the title compound (13.84 kg, yield 87.3%).
[0113] The seed crystals were produced according to the method described in Synthesis Example 2 (2-2) below.
[0114] Example 2: Preparation of sodium salt of 2-(4-cyclopropyl-2-fluoroanilino)-3,4-difluoro-5-[[3-fluoro-2-(methylsulfamoylamino)pyridin-4-yl]methyl]benzamide (Compound 3)
[0115] (2-1) Effect of Various Additives 5-[(2-amino-3-fluoropyridin-4-yl)methyl]-2-(4-cyclopropyl-2-fluoroanilino)-3,4-difluorobenzamide (Compound 2) (1.00 g, 2.32 mmol) and THF (10.0 mL) were added to a reaction vessel, and the reaction vessel was degassed under reduced pressure while stirring and purged with nitrogen. Pyridine (0.564 mL, 6.97 mmol) was added to the resulting solution, and the mixture was cooled to a temperature of -20°C to 0°C. N-methylsulfamoyl chloride was then added stepwise while the reaction rate was monitored by HPLC (analysis condition 1). After confirming that the reaction rate had reached 95% or higher, 10% aqueous sodium chloride solution (5.00 g) was added while maintaining the temperature of the reaction solution below 10°C. The organic layer obtained by layer separation was washed sequentially with 10% aqueous sodium chloride solution (5.0 g) and 20% aqueous sodium chloride solution (5.00 g). While maintaining the resulting solution at 10°C or below, acetone (3.7 mL) was added, and THF was added to the mixture to adjust the volume ratio of acetone to THF in the solution to 3:8. The resulting suspension was then filtered to obtain a clear solution. Water was added to adjust the water content in the solution to approximately 5% by mass, and 2 M sodium tert-butoxide (1.11 mL, 2.21 mmol) was added dropwise with or without an additive (acetaminophen, BHT, phenol, or m-cresol) while maintaining the temperature of the reaction solution in the range of 20°C to 40°C. Subsequently, a suspension of seed crystals of Compound 3 (12 mg) in acetone (0.24 mL) was added at 40°C. Acetone (2.2 mL) was added over 30 minutes, followed by stirring for 1 hour. Acetone (6.1 mL) was then added over 20 minutes, followed by stirring for 30 minutes. TBME (6.1 mL) was then added over 20 minutes, followed by stirring for an additional 30 minutes. The resulting suspension was cooled to 25°C over 30 minutes and stirred for an additional 30 minutes. The precipitated solid was collected by filtration, washed twice with a mixed solvent of acetone (3.7 mL) and TBME (3.7 mL), and then dried under reduced pressure to obtain compound 3. A sample of the resulting compound 3 (hereinafter referred to as "sample 3a") was subjected to HPLC analysis using analytical condition 1.
[0116] The above compound 2 was produced from 5-bromo-2-(4-cyclopropyl-2-fluoroanilino)-3,4-difluorobenzamide obtained in the above Example 1 according to the method described in Synthesis Example 3 (3-1) below.
[0117] The seed crystals were produced from 5-bromo-2-(4-cyclopropyl-2-fluoroanilino)-3,4-difluorobenzamide obtained in Example 1 above according to the methods described in Synthesis Examples 3 (3-1) and 4 (4-1) below.
[0118] The reaction rate was calculated according to the following formula: Reaction rate (%) = [(Peak area of compound 1 + Peak area of compound X + Peak area of compound Y) / (Peak area of compound 1 + Peak area of compound 2 + Peak area of compound X + Peak area of compound Y)] × 100 (%)
[0119] The HPLC retention times were about 3.26 minutes for compound 1, 1.02 minutes for compound 2, 3.54 minutes for compound X, and 4.05 minutes for compound Y.
[0120] The structural formulae of Compound X and Compound Y are as follows:
[0121] The results of the HPLC analysis of Sample 3a are shown in Table 2. Table 2 shows the content of Compound 3 in Sample 3a, the HPLC purity of Compound 3 in Sample 3a (the percentage of the peak area of Compound 3 relative to the sum of the peak areas of all substances detected in the HPLC analysis of Sample 3a), and the amount of the impurity 2-(4-cyclopropyl-2-fluoroanilino)-3,4-difluoro-5-[[3-fluoro-2-(methylsulfamoylamino)pyridin-4-yl]methyl]benzoic acid (Compound 4) in Sample 3a (relative amount to Compound 3).
[0122] The content of Compound 3 in Sample 3a was calculated from the content of Compound 3 in a standard sample of Compound 3 (hereinafter referred to as "Standard 3") according to the following formula: Content of Compound 3 in Sample 3a (% by mass) = [mass (mg) of Standard 3 subjected to HPLC analysis / mass (mg) of Sample 3a subjected to HPLC analysis] × [peak area of Compound 3 obtained by HPLC analysis of Sample 3a / peak area of Compound 3 obtained by HPLC analysis of Standard 3] × content of Compound 3 in Standard 3 (% by mass).
[0123] Sample 3 was obtained by jet milling a sample produced from 5-bromo-2-(4-cyclopropyl-2-fluoroanilino)-3,4-difluorobenzamide obtained in Example 1 according to the methods described in Synthesis Examples 3(3-1) and 4(4-1) below. HPLC analysis of Sample 3 was carried out under Analysis Condition 1.
[0124] The content of Compound 3 in Sample 3 was determined by quantitative NMR using dimethyl sulfone as an internal standard. The quantitative NMR measurement was carried out under the following conditions: Deuterated solvent: dimethyl sulfoxide-d 6 and heavy water (D 2 O) were used in a volume ratio of 9:1. 1 H Pulse angle: 90° Delay time: 60 seconds Digital resolution: 0.25Hz Spinning: Off Number of accumulations: 32
[0125] The HPLC purity of Compound 3 in Sample 3a was calculated according to the following formula: HPLC purity of Compound 3 in Sample 3a (%) = 100 (%) - [total peak areas of all impurities detected in HPLC analysis of Sample 3a / total peak areas of all substances detected in HPLC analysis of Sample 3a] × 100 (%)
[0126] The amount of compound 4 in sample 3a (relative to compound 3) was calculated according to the following formula: Amount of compound 4 in sample 3a (%) = [peak area of compound 4 obtained by HPLC analysis of sample 3a / peak area of compound 3 obtained by HPLC analysis of sample 3a] × 100 (%)
[0127] The structural formula of compound 4 is as follows:
[0128] Table 2 also shows the pKa of the additive, the solubility of the sodium salt of the additive, and the solubility of the additive.
[0129] The pKa values shown in Table 2 are pKa values in water described in the following documents. The pKa values of acetaminophen, phenol, and m-cresol are pKa values in water at 25°C. Acetaminophen: Org. Biomol. Chem., 2009, 7, 5103. BHT: IUPAC Chemical Data Series, No. 23; Pergamon Press: Oxford, UK, 1979. Phenol: Handbook of Biochemistry and Selected Data for Molecular Biology, 2nd ed.; CRC: Cleveland, OH, 1970. m-Cresol: Handbook of Chemistry and Physics, 85th ed. ;CRC: Boca Raton, FL, 2004-2005.
[0130] The solubility of the additive or its sodium salt was measured as follows. A mixed solvent of tetrahydrofuran, acetone, tert-butyl methyl ether, and water (the volume ratio of tetrahydrofuran, acetone, and tert-butyl methyl ether was 8:10:5, and the water content was 3 mass% relative to the mass of the mixed solvent) was prepared. The temperature of the solvent was brought to 25°C, and an additive (acetaminophen, BHT, phenol, or m-cresol) or its sodium salt was added, and the resulting suspension was stirred for 6 hours or more. The supernatant was subjected to HPLC analysis using analysis condition 1.
[0131]
[0132] As is evident from the above results, the addition of BHT, phenol or m-cresol results in a product with a higher content of Compound 3 compared to the product without additive or the product with the addition of acetaminophen.
[0133] When acetaminophen was added, residual impurities were confirmed in Sample 3a, and when phenol was added, the peak of Compound 4 was not detected.
[0134] (2-2) Investigation of inorganic salt removal method (1) Inorganic salt removal by concentration 5-[(2-amino-3-fluoropyridin-4-yl)methyl]-2-(4-cyclopropyl-2-fluoroanilino)-3,4-difluorobenzamide (compound 2) (1.00 g, 2.32 mmol) and THF (10.0 mL) were added to a reaction vessel, and the reaction vessel was degassed under reduced pressure while stirring and purged with nitrogen. Pyridine (0.564 mL, 6.97 mmol) was added to the resulting solution, and the mixture was cooled to a temperature of -20°C to 0°C. N-methylsulfamoyl chloride was then added stepwise while the reaction rate was monitored by HPLC (analysis condition 2). After confirming that the reaction rate had reached 95% or higher, 10% aqueous sodium chloride solution (5.00 g) was added while maintaining the temperature of the reaction solution at 10°C or below. The organic layer obtained by layer separation was washed sequentially with a 10% aqueous sodium chloride solution (5.0 g) and a 20% aqueous sodium chloride solution (5.00 g). THF (15.0 mL) was added and the mixture was concentrated to a total volume of 5 mL twice at 40°C. The resulting suspension was filtered to obtain a clear solution. Acetone (3.7 mL) and THF were added to adjust the volume ratio of acetone to THF in the solution to 3:8. Water was added to adjust the water content in the solution to approximately 5% by mass, followed by the addition of m-cresol, and 2 M sodium tert-butoxide (1.10 mL, 2.21 mmol) dropwise while maintaining the temperature of the reaction solution between 20°C and 40°C. Subsequently, a suspension of seed crystals (12 mg) of Compound 3 in acetone (0.20 mL) was added at 40°C. Acetone (2.5 mL) was added over 30 minutes, followed by stirring for 1 hour. Further, acetone (6.0 mL) was added over 20 minutes, followed by stirring for 30 minutes. Thereafter, TBME (6.0 mL) was added over 20 minutes, followed by stirring for an additional 30 minutes. [0137] (2) Removal of inorganic salts by adding solvent 5-[(2-amino-3-fluoropyridin-4-yl)methyl]-2-(4-cyclopropyl-2-fluoroanilino)-3,4-difluorobenzamide (Compound 2) (1.00 g, 2.32 mmol) and THF (10.0 mL) were added to a reaction vessel, and the reaction vessel was degassed under reduced pressure while stirring and purged with nitrogen. While maintaining the resulting solution at 10°C or below, methyl ethyl ketone (3.7 mL) or 2-MeTHF (3.7 mL) was added, and THF was added to the mixture to adjust the volume ratio of methyl ethyl ketone or 2-MeTHF to THF in the solution to 3:8. The resulting suspension was then filtered to obtain a clear solution. Water was added to adjust the water content in the solution to approximately 5% by mass, followed by the addition of m-cresol, and 2 M sodium tert-butoxide (1.10 mL, 2.21 mmol) dropwise while maintaining the temperature of the reaction solution in the range of 20°C to 40°C. Subsequently, a suspension of seed crystals of Compound 3 (12 mg) in acetone (0.20 mL) was added at 40°C. Acetone (2.5 mL) was added over 30 minutes, followed by stirring for 1 hour. Furthermore, acetone (6.0 mL) was added over 20 minutes, followed by stirring for 30 minutes. Thereafter, TBME (6.0 mL) was added over 20 minutes, and the mixture was stirred for an additional 30 minutes. The resulting suspension was cooled to 25°C over 30 minutes, and the mixture was stirred for an additional 30 minutes. The precipitated solid was collected by filtration, and the residue was washed twice with a mixed solvent of acetone (3.5 mL) and TBME (3.5 mL), and then dried under reduced pressure to obtain compound 3. A sample of the resulting compound 3 (hereinafter referred to as "sample 3d") was subjected to HPLC analysis using analytical condition 2.
[0138] The compound 2 and the seed crystals were prepared in the same manner as in (2-1) above. The reaction rate was calculated in the same manner as in (2-1) above.
[0139] The content of compound 3 in sample 3d was as follows (shown together with the solvent initially added in the inorganic salt removal step). The content of compound 3 in sample 3d was calculated in the same manner as the content of compound 3 in sample 3a. Methyl ethyl ketone: 99.4% by mass 2-MeTHF: 97.4% by mass
[0140] (3) Discussion It was shown that whether inorganic salts were removed by concentration or by adding a solvent, a product with a high content of Compound 3 could be obtained. Furthermore, when inorganic salts were removed by adding a solvent, it was shown that acetone or methyl ethyl ketone, for example, was particularly excellent as the solvent (see Table 2 (m-cresol) above for acetone).
[0141] Although inorganic salts can be removed by any of the above methods, the method of adding a solvent is superior in terms of simplicity.
[0142] Since removal of inorganic salts by concentration is carried out at a relatively high temperature (e.g., 40°C), if the concentration takes a long time, there is a possibility that Compound 1 may be decomposed during the concentration process. The method using solvent addition is advantageous in that it allows removal of inorganic salts at a low temperature (e.g., 10°C or lower).
[0143] (2-3) Preparation of Compound 3 Under a nitrogen atmosphere, THF (21.32 kg) was added to a reaction vessel, followed by the addition of 5-[(2-amino-3-fluoropyridin-4-yl)methyl]-2-(4-cyclopropyl-2-fluoroanilino)-3,4-difluorobenzamide (Compound 2) (3.00 kg, 6.97 mol). While rinsing the weighing vessel, THF (5.34 kg) was added to the reaction vessel, followed by the dropwise addition of pyridine (1.65 kg, 20.9 mol). The solution obtained after stirring was cooled to a temperature of -20°C to -10°C, and N-methylsulfamoyl chloride (total 0.942 kg, 7.27 mol) was added stepwise while maintaining the solution temperature within this range. While maintaining the temperature of the reaction solution below 10°C, a 10% aqueous sodium chloride solution (15.0 kg) was added. While maintaining the temperature of the organic layer obtained by layer separation in the range of -5°C to 5°C, the organic layer was washed successively with a 10% aqueous sodium chloride solution (15.0 kg), a 20% aqueous sodium chloride solution (15.1 kg), and a 20% aqueous sodium chloride solution (15.0 kg). While maintaining the temperature of the organic layer after washing in the range of -5°C to 5°C, acetone (8.63 kg) and THF (8.59 kg) were added successively to the organic layer. The resulting suspension was filtered to obtain a clear solution. The reaction vessel was washed and filtered with THF (3.24 kg), and the obtained filtrate was combined with the above-mentioned clear solution and cooled to a temperature of -5°C to 5°C in a crystallization vessel. Water was added to this solution to adjust the water content in the solution to approximately 5% by mass, and then, while maintaining the temperature of the solution in the range of -5°C to 5°C, 2 M sodium tert-butoxide THF solution (2.89 kg, 6.66 mol) and m-cresol (0.38 kg, 3.51 mol) (which had been mixed in advance under a nitrogen atmosphere) were added dropwise. The vessel used to mix the 2 M sodium tert-butoxide THF solution and m-cresol was washed with THF (8.28 kg), and the resulting mixture was added to a crystallization vessel, and the resulting mixture was heated to a temperature of 25°C to 35°C. Subsequently, seed crystals of Compound 3 (18.0 g) were suspended in acetone (0.57 kg) and added to the crystallization vessel. The resulting suspension was heated to a temperature of 30°C to 40°C and stirred for 30 minutes. The suspension was heated to a temperature of 35°C to 45°C and stirred for 30 minutes, after which acetone (5.2 kg) was added dropwise to the crystallization vessel over 1 hour and the mixture was stirred for another 1 hour.Acetone (14.4 kg) was added dropwise over 30 minutes or more, and the mixture was stirred for an additional 30 minutes. Thereafter, TBME (13.5 kg) was added over 30 minutes or more, and the mixture was stirred for an additional 30 minutes. The resulting suspension was cooled to a temperature of 20°C to 30°C, and the mixture was stirred for an additional 30 minutes. The precipitated solid was filtered, and the filter cake was washed twice with a mixture of acetone (8.64 kg) and TBME (8.10 kg), and then dried under reduced pressure to obtain compound 3 (3.10 kg, yield 81.1%). A sample of the resulting compound 3 (hereinafter referred to as "sample 3b") was subjected to HPLC analysis using analytical condition 2.
