AI Technical Summary
When measuring antigen content, existing chemiluminescence measurement systems are difficult to maintain linearity within the high concentration range, and cannot effectively expand the upper limit measurement value while maintaining the sensitivity of low concentration measurements.
Chemiluminescence reactants containing enhancer compounds with pyridine backbones are employed, which are able to provide excellent linear relationships (high slopes) and increase chemiluminescence intensity in high concentration ranges.
Excellent linear measurements in high concentration ranges are achieved, and the intensity and time-holding of the chemiluminescence reaction are enhanced, and the problems of limited measurement range and poor linearity in the prior art are solved.

Abstract
Description
Chemiluminescence reaction enhancer reagents and compounds
[0001] The present invention relates to a chemiluminescent reaction enhancer reagent that can enhance the amount of chemiluminescence when measuring a substance to be measured using chemiluminescence. The present invention further relates to a compound useful as a chemiluminescent reaction enhancer reagent.
[0002] In a chemiluminescence measurement system using luminol or the like, it is known that PPP (4-phenylphenol) and TP (thiazolephenol) are used as enhancers to increase the amount of chemiluminescence.
[0003] Patent Document 1 describes TP (thiazole phenol) as an enhancer, and Patent Document 2 describes PPP, but neither mentions the measurement range on the high concentration side. As an effect, they describe an improvement in sensitivity in the low concentration range.
[0004] Patent No. 03030043 Publication Special Publication No. 03-005539
[0005] For example, in the measurement of human chorionic gonadotropin (hCG) or estradiol (E2), it is necessary to measure samples with a wide range of antigen concentrations, from low to high. However, through studies by the present inventors, it has been found that measurements using TP or PPP have the problem of being unable to measure samples with a wide range of antigen concentrations, i.e., unable to extend the upper limit of measurement while maintaining the lower limit of measurement, and unable to maintain linearity over a wide range.
[0006] An object of the present invention is to provide an enhancer reagent for enhancing the amount of chemiluminescence, which can achieve excellent linearity in the high concentration range (hereinafter referred to as high-value slope) in the measurement of a substance to be measured. Another object of the present invention is to provide a compound that can enhance the amount of chemiluminescence.
[0007] The present inventors have conducted extensive research to solve the above problems and, as a result of screening various compounds, have found that enhancer reagents having a pyridine skeleton can achieve an excellent high slope. The present invention was completed based on the above findings. Specifically, the present invention provides the following:

[0042] In one example, the substance to be measured to which peroxidase is bound via a binding substance that recognizes the substance to be measured may be obtained by reacting the substance to be measured with a first binding substance that recognizes the substance to be measured to form a complex, and then reacting the complex with a second binding substance to which peroxidase is bound that recognizes the substance to be measured.
[0043] In one example, the analyte to which peroxidase is bound via a binding substance that recognizes a complex of the analyte and a first binding substance that recognizes the analyte may be obtained by reacting the analyte with the first binding substance that recognizes the analyte to form a complex, and then reacting the complex with a "third binding substance that recognizes the complex of the analyte to which peroxidase is bound and the first binding substance that recognizes the analyte."
[0044] For example, a substance to be measured in a sample can be measured using a solid phase carrier such as magnetic silica particles on the surface of which a first binding substance that recognizes the substance to be measured is immobilized.
[0045] Examples of binding substances that recognize the analyte (first binding substances that recognize the analyte, or second binding substances that recognize the analyte) include substances that bind to the analyte through interactions such as an "antigen"-"antibody" reaction, a "sugar chain"-"protein" reaction, a "sugar chain"-"lectin" reaction, a "protein"-"peptide chain" reaction, a "protein"-"protein" reaction, and a "protein"-"nucleotide chain" reaction.
[0046] Examples of binding substances that recognize the above complexes include those that bind to the above complexes through interactions such as an "antigen"-"antibody" reaction, a "sugar chain"-"protein" reaction, a "sugar chain"-"lectin" reaction, a "protein"-"peptide chain" reaction, a "protein"-"protein" reaction, and a "protein"-"nucleotide chain" reaction.
[0047] The measurement may be carried out in accordance with the sandwich method described in literature commonly referred to in the art [for example, Enzyme Immunoassay, 2nd Edition (edited by Ishikawa Eiji et al., Igaku Shoin), 1982].
[0048] In the sandwich method, a first binding substance that recognizes the analyte is immobilized on the surface of a solid-phase carrier, such as magnetic silica particles. A sample containing the analyte is mixed with the solid-phase carrier and a second binding substance labeled with peroxidase that recognizes the analyte. The immobilized first binding substance, the analyte in the sample, and the second binding substance are brought into contact with each other. This results in the formation of a peroxidase-labeled complex, which is a complex of the immobilized first binding substance, the analyte in the sample, and the second binding substance labeled with peroxidase. The solid-phase carrier carrying the peroxidase-labeled complex is then subjected to B / F separation to measure the amount of peroxidase in the peroxidase-labeled complex, and the amount of the analyte in the sample is determined based on the amount of peroxidase in the peroxidase-labeled complex.
[0049] Alternatively, for example, a first binding substance that recognizes the analyte is immobilized on the surface of a solid-phase carrier such as magnetic silica particles. A sample containing the analyte is mixed with the solid-phase carrier and a peroxidase-labeled "third binding substance that recognizes the complex between the analyte and the first binding substance that recognizes the analyte." The immobilized first binding substance is then contacted with the analyte in the sample and the third binding substance. This results in the formation of a peroxidase-labeled complex, which is a complex between the immobilized first binding substance, the analyte in the sample, and the peroxidase-labeled third binding substance. The solid-phase carrier carrying the peroxidase-labeled complex is then subjected to B / F separation to measure the amount of peroxidase in the peroxidase-labeled complex, and the amount of the analyte in the sample is measured based on the amount of peroxidase in the peroxidase-labeled complex.
[0050] The solid phase carrier can be any support (particularly an insoluble support) used in conventional immunoassays, and examples thereof include organic substances such as polystyrene, polyacrylic acid, polymethacrylic acid, polymethyl methacrylate, polyacrylamide, polyglycidyl methacrylate, polypropylene, polyolefin, polyimide, polyurethane, polyester, polyvinyl chloride, polyethylene, polychlorocarbonate, silicone resin, silicone rubber, agarose, dextran, and ethylene-maleic anhydride copolymer; inorganic substances such as glass, silicon oxide, diatomaceous earth, porous glass, ground glass, alumina, silica gel, and metal oxides; magnetic substances such as iron, cobalt, nickel, magnetite, and chromite; and substances prepared from alloys of these magnetic substances.
[0051] Immobilization can be performed on a solid phase (for example, any surface such as beads, magnetic beads, membranes, plates, etc.). Various commercially available magnetic beads can be used. For example, the beads disclosed in WO2012 / 173002A may be used as magnetic beads.
[0052] Specifically, for example, a target substance in a sample is contacted with a first binding substance immobilized on the surface of a solid-phase carrier such as magnetic silica particles, which recognizes the target substance, to form a complex between the target substance in the sample and the first binding substance immobilized on the surface of the solid-phase carrier, which recognizes the target substance. Next, the complex is contacted with a complex of a second binding substance labeled with peroxidase, to form a complex (peroxidase-labeled complex) between the first binding substance immobilized on the solid-phase carrier, which recognizes the target substance in the sample, and the second binding substance labeled with peroxidase. The peroxidase-labeled complex is then subjected to B / F separation, the amount of peroxidase in the peroxidase-labeled complex is measured, and the amount of the target substance in the sample can be determined based on the amount of peroxidase in the peroxidase-labeled complex.
[0053] In the above method, the substance to be measured in the sample is reacted with a first binding substance that recognizes the immobilized substance to be measured, and then a second binding substance labeled with peroxidase is reacted. However, it is also possible to react the second binding substance labeled with peroxidase with the substance to be measured in the sample, and then react with the first binding substance that recognizes the immobilized substance to be measured, or these three reactions may be performed simultaneously.
[0054] The B / F separation (bond / free separation) in the sandwich method refers to the separation of a substance supported on a solid phase carrier from other substances. That is, the B / F separation in the present invention includes: (1) the separation of a "complex of a first binding substance that recognizes a substance to be measured immobilized on the surface of a solid phase carrier and the substance to be measured in a sample" from the substance to be measured or other substances in the reaction system that were not involved in the formation of the complex; and (2) the separation of a peroxidase-labeled complex from a second binding substance labeled with peroxidase or other substances in the reaction system that were not involved in the formation of the peroxidase-labeled complex.
[0055] The sandwich method can also be performed using a microfluidic chip (micro-total analysis systems; μTAS). The microfluidic chip includes a flow channel with a sample inlet. For example, a first binding substance bound to a polycation or polyanion (e.g., DNA) that recognizes the analyte in the sample is contacted at the sample inlet to form a complex between the first binding substance that recognizes the analyte and the analyte. Next, an electric field is applied to move the complex, and when the complex comes into contact with a second binding substance labeled with peroxidase present in the flow channel, a complex (peroxidase-labeled complex) is formed between the first binding substance, the analyte, and the second binding substance labeled with peroxidase. The electric field further moves the peroxidase-labeled complex through the flow channel, where it undergoes B / F separation and reaches a detection unit in the flow channel. In the detection section, the amount of peroxidase in the peroxidase-labeled complex is measured, and the amount of the substance to be measured in the sample is determined based on the amount of peroxidase in the peroxidase-labeled complex.
[0056] In the measurement method of the present invention, the method for contacting the analyte in the sample with the first binding substance that recognizes the analyte immobilized on the surface of the solid phase carrier may be carried out by a conventional treatment such as stirring, mixing, etc. The reaction time may be appropriately set depending on the analyte and the first binding substance, but is usually 1 minute to 24 hours, preferably 1 minute to 1 hour, more preferably 1 to 10 minutes, and particularly preferably 1 to 5 minutes.
[0057] B / F separation in the measurement method of the present invention can be achieved, for example, by utilizing the magnetic properties of the solid carriers, collecting the solid carriers from the outside of the reaction vessel using a magnet or the like, discharging the reaction solution, adding a washing solution, removing the magnet, mixing and dispersing the solid carriers, and washing them. The above procedure may be repeated 1 to 3 times. The washing solution is not particularly limited as long as it is one commonly used in this field.
[0058] As the peroxidase, horseradish peroxidase (HRP) or microperoxidase can be used.
[0059] To bind peroxidase to the second or third binding substance, a method commonly used in this field, for example, a known labeling method commonly used in known EIA [e.g., Medical Chemistry Experiment Lectures, Vol. 8, edited by Yamamura Yuichi, 1st Edition, Nakayama Shoten, 1971; Illustrated Fluorescent Antibodies, by Kawao Akira, 1st Edition, Soft Science Co., Ltd., 1983; Enzyme Immunoassay, edited by Ishikawa Eiji, Kawai Tadashi, and Miyai Kiyoshi, 2nd Edition, Igaku Shoin, 1982, etc.] may be used.
[0060] The amount of peroxidase used is preferably such that the second or third binding substance and peroxidase are used in a molar ratio of, for example, typically 1:0.5 to 20, preferably 1:1 to 15, and more preferably 1:1 to 10. The binding substance that recognizes the peroxidase-labeled analyte may be contained in a buffer solution commonly used in this field, such as Tris buffer, phosphate buffer, veronal buffer, borate buffer, or Good's buffer (e.g., MES (2-morpholinoethanesulfonic acid) buffer). The concentration of the peroxidase bound to the second binding substance or the third binding substance is not particularly limited, but is preferably 0.1 nmol / L or more and 5000 nmol / L or less, more preferably 1 nmol / L or more and 500 nmol / L or less, even more preferably 3 nmol / L or more and 300 nmol / L or less, and particularly preferably 5 nmol / L or more and 150 nmol / L or less.
[0061] When an antibody is used as the binding substance, the pH of the buffer solution may be within a range that does not inhibit the antigen-antibody reaction, and is typically between pH 5 and 9. The buffer solution may also contain stabilizers such as albumin, globulin, water-soluble gelatin, and polyethylene glycol, surfactants, and sugars, as long as they do not inhibit the antigen-antibody reaction. In this specification, pH refers to the value measured in accordance with JIS K0400-12-10:2000 (measurement temperature: 25°C).
[0062] <Measurement Kit> The present invention can provide a measurement kit comprising a peroxidase bound to a binding substance that recognizes a substance to be measured, a chemiluminescent substance, an oxidizing agent, and the chemiluminescent reaction enhancer reagent of the present invention. The present invention further provides a measurement kit comprising a peroxidase bound to a binding substance that recognizes a complex of the substance to be measured and the binding substance that recognizes the substance to be measured, a chemiluminescent substance, an oxidizing agent, and the chemiluminescent reaction enhancer reagent of the present invention.
[0063] Preferred embodiments and specific examples of the peroxidase bound to a binding substance that recognizes the analyte, the peroxidase bound to a binding substance that recognizes a complex between the analyte and a binding substance that recognizes the analyte, the chemiluminescent substance, the oxidizing agent, and the chemiluminescent reaction enhancer reagent of the present invention are as described above in this specification.
