[0001] KRAS G12D Mutation Targeting Compounds

[0002] TECHNICAL FIELD

[0003] The present disclosure is directed towards a new series of KRAS G12D mutation targeting compounds as defined herein, pharmaceutical compositions containing these KRAS G12D mutation targeting compounds, and methods of treating cancer associated with the KRAS G12D mutation using said targeting compounds.

[0004] BACKGROUND

[0005] The KRAS gene provides instructions for the production of the KRAS protein, which relays signals that promote cell division and growth (proliferation).

[0006] The KRAS protein is a GTPase, binding to guanosine triphosphate (GTP) and converting it to guanosine diphosphate (GDP) through hydrolysis. The gene oscillates between two forms, an ‘active’ form in which it is bound to GTP and able to transmit signals to promote proliferation, and an ‘inactive’ form in which it is bound to GDP, following GTP to GDP conversion, and unable to transmit signals. The KRAS protein typically maintains a balance between inactive and active states so that cell proliferation is appropriately regulated.

[0007] When this KRAS protein is damaged, a KRAS mutation may occur, causing the gene to mutate and change form. The most common KRAS mutations are known as KRAS G12C, G12D and G12V mutations. The ‘G12X’ nomenclature relates to the amino acid position (12) on the KRAS protein at which the mutation occurs, as well as the amino acid substitution involved in the mutation (X). Typically, these mutated (changed) forms of the KRAS protein (KRAS mutations) mean that the normal function of the gene is altered. The reversible nature (GTP to GDP activity) of the KRAS protein is impaired such that it remains predominantly in the active state and directs cells to proliferate in an uncontrolled manner.

[0008] KRAS mutations are associated with a number of cancers, including non-small cell lung cancer, colorectal cancer, and pancreatic cancer. It is one of the most frequently mutated gene families in human cancers. The conformational changes to the KRAS protein discussed above result in the protein directing cells to proliferate in an uncontrolled manner, leading to tumour formation and the spread of cancer.

[0009] KRAS mutated proteins were initially considered as ‘undruggable’ due to the failure to identify specific and potent molecules that could directly target the KRAS protein. However, in recent years, KRAS G12C inhibitors have been identified. These inhibitors typically work by covalently ‘tethering’ to a nucleophilic cysteine at position 12, i.e., G12C, of the active site of the KRAS protein whilst it is in the ‘inactive’ state, irreversibly stabilising this state and preventing the KRAS protein with the KRAS G12C mutation from instructing the excessive proliferation of cells.

[0010] However, a selective inhibitor has yet to be approved that can target the more prevalent and oncogenic mutation of the KRAS protein, KRAS G12D.

[0011] Identified KRAS G12C inhibitors cannot be utilised, as the point of mutation and thus target of the inhibitor differs greatly in terms of chemistry between KRAS G12C (cysteine) and KRAS G12D (aspartic acid). The configurations of other regions of the complex structure of the KRAS protein must also be considered. In addition, when compared to KRAS G12C mutations, selective inhibition of KRAS G12D mutations presents a significant challenge due to the requirement of the inhibitors to selectively bind with high enough affinity typically without the benefit of covalent interaction (aspartic acid binding is typically considered to require a non-covalent interaction).

[0012] There is therefore a need for new KRAS G12D mutation targeting compound capable of targeting the KRAS G12D mutation to enable treatment of diseases associated with the KRAS G12D mutation, such as cancer.

[0013] It is an object of the present disclosure to provide a solution to the above-mentioned or other problems.

[0028] DEFINITIONS

[0029] Where used herein and unless specifically indicated otherwise, the following terms are intended to have the following meanings in addition to any broader (or narrower) meanings the terms might enjoy in the art:

[0030] As used herein, the term "KRAS G12D mutation" refers to mutation at the 12th amino acid glycine of the KRAS protein. The 12th amino acid glycine is changed to aspartic acid. Position of the mutation may be in reference to the wild-type KRAS protein, for example Uniprot accession number P01116 (SEQ ID NO: 1). The mutation is an altered form of the KRAS protein, and normal function is impaired. The mutated protein may be referred to as a "KRAS G12D mutated protein". As discussed above, the reversible nature (GTP to GDP activity) of the KRAS protein is impaired such that it remains predominantly in the active state and directs cells to proliferate in an uncontrollable manner. The term is well known in the art. As used herein, the term "disease associated with the KRAS G12D mutation" or "cancer associated with the KRAS G12D mutation" refers to a disease or cancer in which the KRAS G12D mutation has been diagnosed or detected, in any amount. This is typically done by PCR based KRAS sequencing of clinical samples.

[0031] As used herein, the term “wild-type” refers to the most common form, appearance or strain of a KRAS protein existing in nature. This is the normal, non-mutation version of the KRAS protein common in nature. This means that there is no mutation of the 12th amino acid of the KRAS protein. The sequence of the wild type KRAS protein can be found at Uniprot accession number P01116:

[0032] MTEYKLVWGAGGVGKSALTIQLIQNHFVDEYDPTIEDSYRKQWIDGETCLLDILDTAGQEE YSAMRDQYMRTGEGFLCVFAINNTKSFEDIHHYREQIKRVKDSEDVPMVLVGNKCDLPSRT VDTKQAQDLARSYGIPFIETSAKTRQRVEDAFYTLVREIRQYRLKKISKEEKTPGCVKIKKCII M (SEQ ID NO:1 ).

[0033] As used herein, the term “disease” is used to define any abnormal condition that impairs physiological function and is associated with specific symptoms. The term is used broadly to encompass any disorder, illness, abnormality, pathology, sickness, condition or syndrome in which physiological function is impaired irrespective of the nature of the aetiology (or indeed whether the aetiological basis for the disease is established). It therefore encompasses conditions arising from trauma, injury, surgery, radiological ablation, poisoning or nutritional deficiencies.

[0034] As used herein, the term "treatment" or "treating" includes an intervention (e.g. the administration of a compound of formula (I) according to the first aspect of the present disclosure, or a pharmaceutically acceptable salt, hydrate, solvate, prodrug or isotope thereof, to a subject) which ameliorates or lessens the proliferation of cells and formation of tumours and the spread of a disease such as cancer through removal of (or lessening the impact of) its cause(s) (for example, through degradation of the KRAS G12D mutated protein of the KRAS protein). In this case, the term is used synonymously with the term “therapy”. Thus, the treatment of a disease, such as cancer, associated with the KRAS G12D mutation according to the disclosure may be characterized by the (direct or indirect) action of the compound of formula (I) according to the first aspect of the present disclosure, or a pharmaceutically acceptable salt, hydrate, solvate, prodrug or isotope thereof. Thus, the compounds of formula (I) according to the first aspect of the present disclosure, or a pharmaceutically acceptable salt, hydrate, solvate, prodrug or isotope thereof, find application in methods of degrading the KRAS G12D mutation, preventing cell proliferation, decreasing tumour formation (or even shrinking tumour size), and preventing or reducing the spread of disease such as cancer.

[0035] Additionally, the term "treatment" or "treating" includes an intervention (e.g. the administration of a compound of formula (I) according to the first aspect of the present disclosure, or a pharmaceutically acceptable salt, hydrate, solvate, prodrug or isotope thereof, to a subject) which prevents or delays the onset or progression of a disease associated with the KRAS G12D mutation such as cancer or reduces (or eradicates) its incidence within a treated population. In this case, the term treatment is used synonymously with the term “prophylaxis” or “prevention”.

[0036] The term "subject" (which is to be read to include "individual", "animal", "patient" or "mammal" where context permits) defines any subject, particularly a mammalian subject, for whom treatment is indicated. Mammalian subjects include, but are not limited to, humans, domestic animals, farm animals, zoo animals, sport animals, pet animals such as dogs, cats, guinea pigs, rabbits, rats, mice, horses, cattle, cows; primates such as apes, monkeys, orangutans, and chimpanzees; canids such as dogs and wolves; felids such as cats, lions, and tigers; equids such as horses, donkeys, and zebras; food animals such as cows, pigs, and sheep; ungulates such as deer and giraffes; rodents such as mice, rats, hamsters and guinea pigs; and so on. Preferably, in the context of the present disclosure, the subject is a human.

[0037] The term ‘degrades’, ‘degrading’, or ‘degradation’, or like terms as used herein, in relation to the KRAS G12D mutation is meant the partial or complete physical breakdown of the unfavourable KRAS G12D protein. The KRAS G12D mutated protein is depleted and no longer directs cells to proliferate in an uncontrollable manner.

[0038] As used herein, an "effective amount" or a "therapeutically effective amount" of a compound of formula (I) according to the first aspect of the present disclosure, or a pharmaceutically acceptable salt, hydrate, solvate, prodrug or isotope thereof, defines an amount that can be administered to a subject without excessive toxicity, irritation, allergic response, or other problem or complication, commensurate with a reasonable benefit / risk ratio, but one that is sufficient to provide the desired effect, e.g. the treatment or prophylaxis manifested by a permanent or temporary improvement in the subject's condition. The amount will vary from subject to subject, depending on the age and general condition of the individual, mode of administration and other factors. Thus, while it is not possible to specify an exact effective amount, those skilled in the art will be able to determine an appropriate "effective" amount in any individual case using routine experimentation and background general knowledge. A therapeutic result in this context includes eradication or lessening of symptoms, reduced pain or discomfort, prolonged survival, improved mobility and other markers of clinical improvement. A therapeutic result need not be a complete cure alone and can be used in combination with other agents.

[0039] The term "pharmaceutically acceptable salt" as applied to the compounds of formula (I) according to the first aspect of the disclosure defines any organic or inorganic acid addition salt of the free base which are suitable for use in contact with the tissues of humans and lower animals without undue toxicity, irritation, allergic response and which are commensurate with a reasonable benefit / risk ratio. Suitable pharmaceutically acceptable salts are well known in the art. Examples are the salts with inorganic acids (for example hydrochloric, hydrobromic, sulphuric and phosphoric acids), organic carboxylic acids (for example acetic, propionic, glycolic, lactic, pyruvic, malonic, succinic, fumaric, malic, tartaric, citric, ascorbic, maleic, hydroxymaleic, dihydroxymaleic, benzoic, phenylacetic, 4- aminobenzoic, 4-hydroxybenzoic, anthranilic, cinnamic, salicylic, 2-phenoxybenzoic, 2- acetoxybenzoic and mandelic acid) and organic sulfonic acids (for example methanesulfonic acid and p-toluenesulfonic acid).

[0040] The term “solvate” as applied to the compounds of formula (I) according to the first aspect of the disclosure means a physical association of the compound with one or more solvent molecules, whether organic or inorganic. This physical association can include hydrogen bonding. In certain instances, the solvate will be capable of isolation, for example when one or more solvent molecules are incorporated in the crystal lattice of the crystalline solid. The solvate may comprise either a stoichiometric or nonstoichiometric amount of the solvent molecules. For example, a solvate with a nonstoichiometric amount of solvent molecules may result from partial loss of solvent from the solvate. “Solvate” encompasses both solutionphase and isolable solvates. Exemplary solvates include hydrates, ethanolates, methanolates, isopropanolates and the like. Methods of solvation are generally known in the art.

[0041] The term “hydrate” as applied to a compound of formula (I) according to the first aspect of the disclosure refers to the compound when it is associated with water in the molecular form, i.e., in which the H — OH bond is not split, and may be represented, for example, by the formula R H2O, where R is a compound of formula (I) according to the first aspect of the disclosure. A given compound may form more than one hydrate including, for example, monohydrates (R H2O) or polyhydrates (R nH2O wherein n is an integer >1) including, for example, dihydrates (R 2H2O), trihydrates (R-3H2O), and the like, or hemihydrates, such as, for example, R n / 2H2O, R n / 3H2O, R n / 4H2O and the like wherein n is an integer. As used herein, the term "prodrug" as applied to a compound of formula (I) according to the first aspect of the present disclosure means the pharmacologically acceptable derivatives such as esters, amides and phosphates, such that the resulting in vivo biotransformation product of the derivative is the active drug as defined in the compounds of formula (I) according to the first aspect of the present disclosure. The reference by Goodman and Gilman (The Pharmaco- logical Basis of Therapeutics, 8 ed, McGraw-HiM, Int. Ed. 1992, "Biotransformation of Drugs", p 13-15) describing prodrugs generally is hereby incorporated. Prodrugs of a compound of formula (I) according to the first aspect of the present disclosure are prepared by modifying functional groups present in the compound in such a way that the modifications are cleaved, either in routine manipulation or in vivo, to the parent compound. Prodrugs of the compounds of formula (I) according to the first aspect of the present disclosure include those compounds wherein the amino group(s) of the compound of formula (I) according to the first aspect of the present disclosure is bonded to any group that, when the prodrug is administered to a subject, cleaves to form a free amino. The term includes all regioisomers thereof.

[0042] In its broadest aspect, the present disclosure contemplates all optical isomers of the compounds of formula (I) according to the first aspect of the present disclosure, or pharmaceutically acceptable salts, hydrates, solvates, prodrugs, or isotopes thereof.

The heteroaryl ring may contain up to 4 heteroatoms, such as up to 3 heteroatoms, or up to 2, for example a single heteroatom. The said heteroaromatic ring may be attached to the rest of the molecule through either a heteratom or a carbon atom of the ring system. Examples of heteroaryl groups include but are not limited to furyl, pyrrolyl, thienyl, oxazolyl, isoxazolyl, oxadiazolyl, imidazolyl, oxatriazoly, thiazolyl, isothiazolyl, tetrazolyl, pyrazolyl, pyridinyl, pyrimidinyl, pyridazinyl, pyrazinyl, triazinyl and thiadiazolyl. [0052] DETAILED DESCRIPTION

[0053] The present disclosure provides the solution to a previously unmet need for a KRAS G12D mutation targeting compound for the treatment of KRAS G12D mutation, including cancer associated with the KRAS G12D mutation.

[0054] The compounds of formula (I) according to the first aspect of the present disclosure, or a pharmaceutically acceptable salt, hydrate, solvate, prodrug or isotope thereof, function as a proteolysis targeting chimera (PROTAC) composed of two protein-binding molecules (left- and right-hand sides of the compound as depicted in formula (I)) joined by a linker. The linker includes Li shown in formula (I).

[0055] The compound of formula (I) according to the first aspect of the present disclosure, or a pharmaceutically acceptable salt, hydrate, solvate, prodrug or isotope thereof, functions by inducing selective intracellular proteolysis. As depicted in formula (I), the left-hand side of the compound of formula (I) according to the first aspect of the present disclosure, or a pharmaceutically acceptable salt, hydrate, solvate, prodrug or isotope thereof, binds to the unfavourable KRAS G12D mutation, and is connected to an E3 ubiquitin recognition unit (the right-hand side of the compound of formula (I)) via a central linker (including Li, shown in formula (I)).

[0056] The left-hand side of the compound of formula (I), or a pharmaceutically acceptable salt, hydrate, solvate, prodrug or isotope thereof, as shown below:

[0057] The right-hand side of the compound of formula (I), or a pharmaceutically acceptable salt, hydrate, solvate, prodrug or isotope thereof, as shown below:

[0058] The left-hand side of the compound of formula (I) according to the first aspect of the present disclosure, or a pharmaceutically acceptable salt, hydrate, solvate, prodrug or isotope thereof, interacts with the protein of interest, in this case, the KRAS G12D protein when it is a KRAS G12D mutated protein. The right-hand side of the compound of formula (I), is an E3 ubiquitin recognition unit, which is able to interact with an E3 ubiquitin ligase protein. The left- and right-hand sides are joined by a central linker (including Li shown in formula (I)).

[0059] Ultimately, the structure and configuration of the compound of formula (I) according to the first aspect of the present disclosure means that the E3 ubiquitin recognition unit (right-hand side of the compound of formula (I) as depicted) can interact with an E3 ubiquitin ligase protein and facilitate the transfer of ubiquitin from a ubiquitin carrier to the target KRAS G12D mutation, advantageously resulting in selective degradation of the KRAS G12D mutation. The compound of formula (I) according to the first aspect of the present disclosure, or pharmaceutically acceptable salt, hydrate, solvate, prodrug or isotope thereof, brings the unfavourable KRAS G12D mutated protein into close proximity with the E3 ubiquitin ligase protein to catalyse the degradation thereof. This advantageously reduces the instance of the unfavourable KRAS G12D mutated protein and lessens the proliferation of cells and formation of tumours and the spread of cancer through removal of (or lessening the impact of) its cause(s).

[0060] Advantageously, the compounds of formula (I) according to the first aspect of the present disclosure, or pharmaceutically acceptable salts, hydrates, solvates, prodrugs or isotopes thereof, are selective for the degradation of the KRAS G12D mutation, in particular, and advantageously, over KRAS G12C mutations and KRAS wild type.

[0061] The compounds of formula (I) according to the first aspect of the present disclosure, or pharmaceutically acceptable salts, hydrates, solvates, prodrugs or isotopes thereof, demonstrate potent effects on the degradation of the KRAS G12D mutated protein.

[0062] The advantageous effects of the compounds of formula (I) according to the first aspect of the present disclosure, or pharmaceutically acceptable salts, hydrates, solvates, prodrugs or isotopes thereof, with respect to potency and selectivity for the degradation of the KRAS G12D mutated protein are attributed by the present inventors to the structure and configuration of the compound of formula (I). The present inventors consider that the structure and configuration of the linker between the left- and right-hand sides of the compound of formula (I), or pharmaceutically acceptable salt, hydrate, solvate, prodrug or isotope thereof, as depicted, plays a significant role in this. In particular, the structure and configuration of the linker, including Li as described herein.

[0063] The linker of the compound of formula (I) according to the present disclosure, or a pharmaceutically acceptable salt, hydrate, solvate, prodrug or isotope thereof, is shown below:

[0064] The linker of the compound of formula (I) according to the present disclosure, or pharmaceutically acceptable salts, hydrates, solvates, prodrugs or isotopes thereof, begins on the left-hand side of the compound, and includes, the cyclopropyl (m is 1 ) or cyclobutyl (m is 2) aliphatic group. The linker links the left-hand side of the compound to the right-hand side of the compound as discussed above. The linker ends at the right-hand side of the molecule and includes, the (CH2)nand CO moiety. The structure of this linker is important to achieving the advantages of the disclosure as set out above. As described in more detail below, it is the structural significance of an aliphatic group linked to a tertiary amine linkage (part of Li), preferably connected to an oxygen atom (part of Li), and to a carbon chain (CH2)n which means the linker is of sufficient length and forms a structural configuration optimal to interact with the E3 ubiquitin ligase protein and form a productive ternary complex.

[0065] The structure of the compound of formula (I) according to the first aspect of the present disclosure, or pharmaceutically acceptable salts, hydrates, solvates, prodrugs or isotopes thereof, ensures that the compound binds securely and strongly to the target KRAS G12D mutation and operable to bring an E3 ubiquitin ligase protein into connection therewith to selectively, and potently, degrade the KRAS G12D mutated protein.

[0066] For compounds of formula (I) according to the first aspect of the present disclosure, or pharmaceutically acceptable salts, hydrates, solvates, prodrugs or isotopes thereof, Ri , R2, Li, m and n, may be as described below. Each of Ri , R2, Li , m and n, as described below, is applicable to all embodiments of the first aspect of the present disclosure.

[0067] Ri

[0068] Ri is selected from C=CH or Ci-4alkyl.

[0069] Preferably, Ri is C=CH, methyl or ethyl. More preferably, Ri is C=CH or ethyl. More preferably, Ri is C=CH.

[0070] R2

[0071] R2is selected from a phenyl or a monocyclic 5- or 6-membered heteroaryl, wherein the phenyl and 5- or 6-membered heteroaryl are optionally substituted with one or more substituents selected from Ci-4alkyl and halogen. Preferably, R2is selected from a phenyl optionally substituted with one or more halogen substituents, such as fluorine, and a 5- or 6-membered heteroaryl optionally substituted with one or more Ci .4alkyl substituents.

[0072] Preferably, R2is selected from a phenyl or a monocyclic 5-membered heteroaryl, wherein the phenyl and 5-membered heteroaryl are optionally substituted with one or more substituents selected from Ci-4alky I and halogen. More preferably, R2is selected from a phenyl optionally substituted with one or more halogen substituents, such as fluorine, and a 5-membered heteroaryl optionally substituted with one or more Ci .4alkyl substituents.

[0073] More preferably, R2is selected from a phenyl or thiazole optionally substituted with one or more substituents selected from Ci4alkyl and halogen.

[0074] More preferably, R2is a phenyl optionally substituted with one or more halogen substituents, such as fluorine, or a thiazole optionally substituted with one or more Ci -4alkyl substituents.

[0075] For all descriptions of R2above, the phenyl or 5- or 6-membered heteroaryl is preferably substituted with the stated one or more substituents.

[0076] More preferably, R2is a phenyl substituted with one or more halogen substituents, such as fluorine, or a thiazole substituted with one or more Ci4al kyl substituents.

[0077] For all descriptions of R2above, the halogen is preferably fluorine.

[0078] More preferably, R2is selected from a compound of formula (Illa) or (II I b):

[0079] More preferably, R2is a compound of formula (Illa). n n is from 1 to 6.

[0080] Preferably, n is from 1 to 4. More preferably, n is 1 to 3. Most preferably, n is 2.

[0081] Hl m is 1 or 2. Preferably, m is 1 . Li

[0082] Li is selected from the following (Ila), (lib) and (lie): where R4and R6are each independently selected from H or F; and

[0083] R7is Ci-4alkyl.

[0084] It will be appreciated that the carbon atom to which R4or Re is attached in (Ila) and (lib) also has a H attached thereto.

[0085] Preferably, R7is methyl or ethyl. More preferably, R7is methyl.

[0086] (Ila), (lib) and (lie) are advantageous as they contain a tertiary amine. It is this tertiary amine that is protonated at physiological pH (the pH typically prevailing in the human body, around 7.4) and interacts with E62 of KRAS more strongly.

[0087] Preferably, Li is of formula (Ila) or (lib), more preferably formula (lib).

[0088] Preferably, when Li is of formula (Ila), R4is F.

[0089] Preferably, when Li is formula (lib), either or both of R4and R6are F. More preferably, both of R4and Re are F. Most preferably, Re is H, and R4is F. It will be appreciated that when R4and Re are not selected to be F, they are H.

[0090] Li is thus preferably of formula (lid):

[0091] The presence of an electron withdrawing group (F) on the piperidine ring of formula (Ila) or (lib) advantageously reduces the pKa of the tertiary amine (piperidine nitrogen). Preferably, the electron withdrawing group (F) reduces the pKa of the tertiary amine (piperidine nitrogen) to less than 10, such as less than 8. However, this does not reduce potency of the compound of formula (I), or pharmaceutically acceptable salt, hydrate, solvate, prodrug or isotope thereof, according to the first aspect of the present disclosure. Cellular activity and KRAS G12D selectivity of the compound of formula (I) according to the first aspect of the present disclosure is improved through one or both of R4and R6being independently selected to be F.

[0092] When one or both of R4and Re being independently selected to be F, such as, for example, when Li is of formula (lid) shown below, the compound of formula (I), according to the first aspect of the present disclosure , or a pharmaceutically acceptable salt, hydrate, solvate, prodrug or isotope thereof, may be a mixture of diastereomers - cis and trans, or may be a mixture of enantiomers, e.g. a mixture of cis enantiomers or mixture of trans enantiomers, or may be a cis or trans enantiomer. Preferably, the compound of formula (I), according to the first aspect of the present disclosure, or a pharmaceutically acceptable salt, hydrate, solvate, prodrug or isotope thereof, may be a mixture of cis enantiomers or a cis enantiomer. The presence of the substituent F introduces chirality into the compound of formula (I) according to the first aspect of the present disclosure, or pharmaceutically acceptable salt, hydrate, solvate, prodrug or isotope thereof. The present disclosure thus encompasses this mixture of diasteroeisomers, or mixture of enantiomers as detailed above, for all compounds of formula (I), or pharmaceutically acceptable salts, hydrates, solvates, prodrugs or isotopes thereof, one or both of R4and Re being independently selected to be F.

[0093] Accordingly, the following embodiments of the first aspect of the present disclosure are contemplated.

[0113]

[0114]

[0115] More preferably, the compound of formula (I), or a pharmaceutically acceptable salt, hydrate, solvate, prodrug, or isotope thereof, according to the first aspect of the present disclosure is selected from:

[0116] A compound of formula (I), or pharmaceutically acceptable salts, hydrates, solvates, prodrugs, or isotopes thereof, or a pharmaceutical composition according to the second aspect of the present disclosure may be used in the treatment of disease associated with the KRAS G12D mutation, such as cancers associated with the KRAS G12D mutation. Thus, the present disclosure contemplates the compounds of formula (I) according to the first aspect of the present disclosure, or pharmaceutically acceptable salts, hydrates, solvates, prodrugs, or isotopes thereof, as described herein, for use in medicine (e.g. for use in treatment or prophylaxis / prevention), methods of medical treatment or prophylaxis involving the administration of the compounds of formula (I) according to the first aspect of the present disclosure, or pharmaceutically acceptable salts, hydrates, solvates, prodrugs, or isotopes thereof, as described herein. The present disclosure also contemplates the administration of pharmaceutical compositions comprising the compounds of formula (I) according to the first aspect of the present disclosure, or a pharmaceutically acceptable salt, hydrate, solvate, prodrug, or isotope thereof.

[0117] It will be appreciated by a skilled person that the compounds of formula (I) according to the first aspect of the present disclosure, or pharmaceutically acceptable salts, hydrates, solvates, prodrugs, or isotopes thereof, are KRAS G12D mutation targeting compounds. They selectively degrade the KRAS G12D mutated protein. They provide treatment for diseases associated with the KRAS G12D mutation, such as cancer associated with the KRAS G12D mutation, by binding to and degrading the KRAS proteins displaying this KRAS G12D mutation (the KRAS G12D mutated proteins). Degradation of the KRAS G12D mutations means that proliferation of cells is prevented or significantly reduced, such that, for example, the growth of a tumour decreases (or even shrinks in size), and the spread of disease such as cancer is reduced. The compounds of formula (I) according to the first aspect of the present disclosure, or pharmaceutically acceptable salts, hydrates, solvates, prodrugs, or isotopes thereof, can therefore be considered to be anti-tumour agents.

[0118] The compounds of formula (I) according to the first aspect of the present disclosure, or pharmaceutically acceptable salts, hydrates, solvates, prodrugs, or isotopes thereof, or the pharmaceutical composition according to the second aspect of the present disclosure, may target any disease associated with the KRAS G12D mutation, including any cancer associated with the KRAS G12D mutation.

[0120] Various types of malignancies may display the KRAS G12D mutation. Cancers typically associated with the KRAS G12D mutation include lung cancer, non-small cell lung cancer, colorectal cancer, pancreatic cancer, invasive ductal carcinoma, biliary tract malignancies, gastric cancer, endometrial cancer, cervical cancer, bladder cancer, liver cancer, myeloid leukaemia and breast cancer, ovarian cancer, and other gastrointestinal cancers such as oesophageal, and gastroesophageal junction adenocarcinoma. Accordingly, the compounds of formula (I) according to the first aspect of the present disclosure, or pharmaceutically acceptable salts, hydrates, solvates, prodrugs, or isotopes thereof, or the pharmaceutical compositions according to the second aspect of the present disclosure are particularly effective at treating cancer associated with the KRAS G12D mutation, in particular lung cancer, non-small cell lung cancer, colorectal cancer, pancreatic cancer, invasive ductal carcinoma, biliary tract malignancies, gastric cancer, endometrial cancer, cervical cancer, bladder cancer, liver cancer, myeloid leukaemia and breast cancer, ovarian cancer, and other gastrointestinal cancers such as oesophageal, and gastroesophageal junction adenocarcinoma, associated with the KRAS G12D mutation.

[0121] Preferably, the compounds of formula (I) according to the first aspect of the present disclosure, or pharmaceutically acceptable salts, hydrates, solvates, prodrugs, or isotopes thereof, or the pharmaceutical compositions according to the second aspect of the present disclosure, are used to treat any disease associated with the KRAS G12D mutation, such as any cancer associated with the KRAS G12D mutation, such as lung cancer, non-small cell lung cancer, colorectal cancer, pancreatic cancer, invasive ductal carcinoma, biliary tract malignancies, gastric cancer, endometrial cancer, cervical cancer, bladder cancer, liver cancer, myeloid leukaemia and breast cancer, ovarian cancer, and other gastrointestinal cancers such as oesophageal, and gastroesophageal junction adenocarcinoma, associated with the KRAS G12D mutation.

[0122] Preferably, the compounds of formula (I) according to the first aspect of the present disclosure, or pharmaceutically acceptable salts, hydrates, solvates, prodrugs, or isotopes thereof, or the pharmaceutical compositions according to the second aspect of the present disclosure, are used in the treatment of disease associated with KRAS G12D mutation, such as cancer associated with KRAS G12D mutations, in the human body. However, they may also be used in the treatment of disease associated with the KRAS G12D mutations, such as cancer having KRAS G12D mutations, in the animal body. In particular, the compounds of formula (I) according to the first aspect of the present disclosure, or pharmaceutically acceptable salts, hydrates, solvates, prodrugs, or isotopes thereof, or the pharmaceutical compositions according to the second aspect of the present disclosure, can be used to treat commercial animals such as livestock. Alternatively, the compounds of formula (I) according to the first aspect of the present disclosure, or pharmaceutically acceptable salts, hydrates, solvates, prodrugs, or isotopes thereof, or the pharmaceutical composition according to the second aspect of the present disclosure can be used to treat companion animals such as cats, dogs, etc. For the compounds of formula (I) according to the first aspect of the present disclosure, or pharmaceutically acceptable salts, hydrates, solvates, prodrugs, or isotopes thereof, or the pharmaceutical compositions according to the second aspect of the present disclosure, the dosage of the compound of formula (I) administered will vary with, for example, the mode of administration, the treatment desired, the type of disease such as cancer, and the combinational regimen (such as in combination with chemotherapy or radiation.
