[0002] Dandruff is an issue that affects many consumers globally; and it not only negatively impacts the aesthetics, but also causes different scalp problems, such as itchiness and seborrheic dermatitis. Especially in recent years, with the speed up of life pace and the increase of stress, more and more people suffer from dandruff.
[0003] Current anti-dandruff hair care products available in the market, which focus on the anti-malassezia efficacy, provide good efficacy of instant dandruff removal. However, after using them, the dandruff is easily recurrent; and they always bring other scalp problems, and in particular, they negatively affect scalp health, scalp sensory properties, and a hydrating and smoothing efficacy of scalp.

[0035] Piroctone olamine or 1-Hydroxy-4-methyl-6- (2, 4, 4-trimethylpentyl) -2- (1 H) pyridinone, 2-aminoethanol salt or octopirox is a heterocyclic compound and ethanolamine salt of the hydroxamic acid derivative piroctone. It is often used in anti-dandruff shampoo as a replacement for the commonly used compound zinc pyrithione. It is a white or slightly yellow crystalline powder. It is soluble in solution containing 10%ethanol in water, soluble in solution containing surfactants in water or in 1%-10%ethanol. Piroctone olamine is an anti-dandruff active ingredient, soothes inflamed scalp and reduces flaking. It has special functions such as thickening, preservation and elimination of body offensive odour.

[0156] To evaluate the effect of the component (A) on the long-lasting anti-dandruff efficacy, for the composition of Ex. 2, a clinical test, which is a human efficacy test, was carried out, in which 32 Chinese female subjects aged 18 to 40 completed this test. In order to obtain the initial ASFS values of the 32 subjects, according to the Adherent Scalp Flaking Score (ASFS) grading method as shown in Fig. 1, the scalps of the 32 subjects were evaluated as follows: each of the scalps was divided into 8 sections and graded by professional dermatologist according to the Grading scale as shown in Fig. 1, the grades from the 8 sections on the scalp were summed up to obtain the ASFS value, and the higher ASFS value means the heavier dandruff problem. All of the initial ASFS values of the 32 subjects were equal to or higher than 24.

In order to obtain the initial dandruff score of the 20 subjects, the scalps of the 20 subjects were divided into 5 grades from 1 to 5 by professional dermatologist, in which 1 means slight dandruff, and 5 means severe dandruff. All of the initial dandruff scores of the 20 subjects were higher than 3.

[0175] In particular, 323 subjects, including 44 normal ones (VAS ≤ 2) and 132 itchy ones (VAS > 2) , were tested, in which the VAS refers to Visual Analogue Scale which is used for measuring the severity of itch with a numerical score of 1 to 10; and in particular, the VAS is a 10-cm long line, on which individuals indicated the intensity of pruritus by crossing the line at the point that corresponded to their pruritus severity, and individuals have been informed that the beginning of the scale refers to no pruritus (0 points) and the end to the most severe pruritus they can imagine (10 points) .

[0178] Thirdly, microorganism collection areas were marked on the scalps of all the subjects. To collect as many microorganisms as possible in the microorganism collection areas, each of the sites of the normal subjects (hereinafter referred to as “normal sample” ) and the itchy sites of the itchy subjects (hereinafter referred to as “itchy sample” ) , whose area was 6 cm x 0.5 cm, was scraped with 5 sterile cotton swabs dipped in sterile saline for at least ten times. In total, 41 normal samples and 123 itchy samples were collected. The bacterial DNAs of all the samples were extracted according to slightly modified Guidelines for DNeasy Blood &Tissue Kit (Qiagen, #69506) , and all the samples were subjected to a high-throughput sequencing in Shanghai Personalbio Technology Co., Ltd with the Illumina Novaseq sequencing platform and PE250 strategy. Then, all the samples were used for droplet digital PCR quantification, and all the microorganism data obtained from the above were analyzed using USEARCH (version 11.0.667) and QIIME2 platform (version 2021.04, https: / / qiime2. org) .

[0189] To evaluate the effect of the combination of the components (A) , (B) and (C) on the performances of scalp repair, long-lasting anti-dandruff efficacy and scalp health, for the composition of Ex. 1, a clinical test, which is a human efficacy test, was carried out, in which Chinese female subjects aged 18 to 60 completed this test, their dandruff grades varied from light to serious (ASFS values of from 24 to 80, which were measured according to the above ASFS grading method) , and the test group included sensitive scalps.

The 10Q Irritate Total Scalp is a questionnaire which had 10 questions regarding 10 factors and situations including: sun, heat, air dryness, wind, humidity, exercises, stress, alcohol, hot beverage, and spices. Within each question, the subjects were asked whether they had been influenced by this factor, and to describe their perception of the onset of itching, prickling, tightness, pain, or burning feelings. This method was named as 10Q (10 questions) , and counted 1 score for each symptom of each question; thus, the highest score of each question was 5, and the maximum total score would be 50.

[0196] As shown in Fig. 4, by the treatment with the shampoo composition of Ex. 1 comprising the specific combination of the components (A) , (B) and (C) , from T0 to T4W (i.e., from the starting point to the end point of the treatment phase) , the scalp hydration improved significantly as compared to T0. As shown in Fig. 5, by the treatment with the shampoo composition of Ex. 1 comprising the specific combination of the components (A) , (B) and (C) , from T0 to T4W (i.e., from the starting point to the end point of the treatment phase) , the TEWL was reduced significantly as compared to T0, which means the scalp was repaired.
[0197] As shown in Fig. 6, by the treatment with the shampoo composition of Ex. 1 comprising the specific combination of the components (A) , (B) and (C) , in both Treatment and Regression phases, as compared to T0 (i.e., the starting point of the treatment phase) , the dandruff level was significantly reduced, which means the anti-dandruff efficacy can last for up to 8 weeks; and by the treatment with the shampoo composition of Ex. 1 comprising the specific combination of the components (A) , (B) and (C) , as compared to T4 (i.e., the end point of the treatment phase) , there was no significant rising trend, which means in Regression phases (from the end of the treatment phase to the four weeks after that) , the dandruff level did not recur.
[0198] As shown in Figs. 7 and 8, by the treatment with the shampoo composition of Ex. 1 comprising the specific combination of the components (A) , (B) and (C) , the irritation was eliminated significantly, which means that the scalp health maintenance performance was significantly improved.
[0199] In view of the above, it can be seen that the anti-dandruff composition according to the present invention, especially the specific combination of the components (A) to (C) according to the present invention, provides a long-lasting anti-dandruff efficacy, and improves scalp health, scalp sensory properties, and a hydrating and smoothing efficacy of scalp.

Claims
1.An anti-dandruff composition, comprising:(A) piroctone olamine,(B) at least one substance selected from the group consisting of salicylic acid and its derivatives, and(C) decylene glycol.2.The anti-dandruff composition according to claim 1, wherein the amount of the component (A) is in the range of from 0.1 to 1.5 wt. %, preferably in the range of from 0.3 to 1.0 wt. %, more preferably in the range of from 0.5 to 0.9 wt. %, and most preferably 0.8 wt. %, based on the total weight of the anti-dandruff composition.3.The anti-dandruff composition according to claim 1 or 2, wherein the amount of the component (B) is in the range of from 0.1 to 3.0 wt. %, preferably in the range of from 0.2 to 2.0 wt. %, more preferably in the range of from 0.3 to 0.8 wt. %, and most preferably 0.5 wt. %, based on the total weight of the anti-dandruff composition.4.The anti-dandruff composition according to any one of claims 1 to 3, wherein the amount of the component (C) is in the range of from 0.001 to 3.0 wt. %, preferably in the range of from 0.005 to 2.0 wt. %, more preferably in the range of from 0.008 to 0.1 wt. %, and most preferably 0.01 wt. %, based on the total weight of the anti-dandruff composition.5.The anti-dandruff composition according to any one of claims 1 to 4, wherein the component (B) is selected from the group consisting of salicylic acid, sodium salicylate, potassium salicylate, magnesium salicylate, calcium salicylate, MEA-salicylate, TEA-salicylate, capryloyl salicylic acid, chitosan salicylate, benzyl salicylate, and combinations thereof; and preferably, the component (B) is salicylic acid.6.The anti-dandruff composition according to any one of claims 1 to 5, further comprising at least one additive selected from the group consisting of a surfactant, a conditioning agent, a pH adjuster, a thickening agent, a preservative, a fragrance, water and combinations thereof.7.The anti-dandruff composition according to claim 6, wherein the composition further comprises a surfactant selected from the group consisting of an anionic surfactant, a zwitterion surfactant, a non-ionic surfactant, a cationic surfactant, and combinations thereof.8.The anti-dandruff composition according to any one of claims 1 to 7, which is a skin or hair care product.9.The anti-dandruff composition according to claim 8, which is a shampoo, a hair conditioner, a hair oil, a scalp essence, a scalp care solution or a hair mask.10.A method for removing dandruff comprising a step of applying the anti-dandruff composition according to any one of claims 1 to 9 to a desired scalp surface.
