Method for preparing protein-drug conjugate

Abstract

Provided is a method for preparing a protein-drug conjugate (in particular, an antibody-drug conjugate). In the method, an Fc-containing protein (such as an antibody) prior to purification is used, for example, a cell culture supernatant or an affinity chromatography eluate thereof is directly used, to carry out site-specific glycan remodeling under the action of an endoglycosidase and a glycosyltransferase together with a substrate thereof; and then a conventional purification step is carried out to obtain a purified site-specific glycan remodeling product, which is then conjugated with a small‑molecule drug. The method does not rely on a purified protein (such as an antibody) to carry out site-specific glycan remodeling, thereby saving considerable time, reagents and costs consumed in protein purification, and thus significantly shortening the preparation time of the antibody-drug conjugate and greatly reducing the cost.
