Abstract

This method for producing natural killer T (NKT) cells comprises: differentiating induced pluripotent stem cells (iPS cells) into hematopoietic stem progenitor cells by primary differentiation induction; and differentiating the hematopoietic stem progenitor cells into NKT cells by secondary differentiation induction. The timing of switching from the primary induction to the secondary induction is determined by the ratio of CD41a expression negative cells in the hematopoietic stem progenitor cells.

Claims

1. A method for producing NKT cells, comprising differentiating induced pluripotent stem cells (iPS cells) into hematopoietic stem progenitor cells by primary differentiation induction, and differentiating the hematopoietic stem progenitor cells into natural killer T (NKT) cells by secondary differentiation induction, wherein the timing of switching from primary differentiation induction to secondary differentiation induction is determined by the proportion of CD41a-negative cells in the hematopoietic stem progenitor cells.

2. The method for producing NKT cells according to claim 1, wherein the proportion of CD41a-negative cells is 50% or more.

3. The method for producing NKT cells according to claim 1, wherein the timing is further determined by the proportion of CD34-positive and CD43-positive cells in the hematopoietic stem progenitor cells.

4. The method for producing NKT cells according to claim 3, wherein the proportion of CD34-expression-positive and CD43-expression-positive cells is 80% or more.

5. The method for producing NKT cells according to any one of claims 1 to 4, wherein the iPS cells include a gene encoding a therapeutic gene product.

6. An NKT cell preparation manufacturing kit containing hematopoietic stem progenitor cells in which the proportion of CD41a-negative cells is 50% or more, and the proportion of CD34-positive and CD43-positive cells is 80% or more.

7. The NKT cell preparation manufacturing kit according to claim 6, wherein the hematopoietic stem progenitor cells include a gene encoding a therapeutic gene product.
