Abstract
Description
Novel ionized lipid and lipid nanoparticle composition comprising the same
[0001] The present invention relates to a novel ionizable lipid and a lipid nanoparticle composition comprising the same.
[0002] This invention was researched as a project (S3368433) for "Development of a novel biodegradable ionized lipid material as a core carrier for lipid nanoparticle technology for gene delivery" of the Regional Specialized Industry Promotion+ (R&D) project hosted by the Small and Medium Business Technology Information Promotion Agency of the Ministry of SMEs and Startups.
[0003] Nucleic acid-based medicines are being used not only as therapeutic agents but also as preventative agents that protect against diseases by injecting genes capable of expressing antigens for specific diseases. Among these, gene-based vaccines are categorized as DNA vaccines, RNA vaccines, and viral vector vaccines. RNA vaccines, which inject mRNA encoding an antigen into the body, trigger antigen expression and antibody production. RNA vaccines do not have the potential risks of infection associated with viral vector-based vaccines or genetic mutation associated with DNA vaccines. Furthermore, they offer the advantage of rapid development, drawing attention as an effective response to the COVID-19 outbreak in 2019.
[0004] However, genes are easily broken down by nucleases in the human body and, as negatively charged macromolecules, are difficult to deliver into cells. Therefore, a stable and efficient method for delivering them to the desired location is needed. Various delivery methods based on materials such as lipids, polymers, dendrimers, and inorganic metals have been reported as gene delivery systems. Among these, lipid nanoparticles, as lipid delivery vehicles, were used in the development of patisiran, the first siRNA drug approved by the FDA in 2018, as well as the mRNA vaccine for COVID-19, which received emergency use authorization in 2020. Since then, lipid nanoparticle technology has been recognized for its technological value as a clinically useful delivery system for nucleic acid drugs such as siRNA and mRNA.
[0005] Current lipid nanoparticles are generally used in a form in which four components are mixed in a certain ratio: ionized lipid, phospholipid (helper lipid), cholesterol (structural maintenance lipid), and PEG-lipid. However, as the development of gene therapy for various indications is in full swing, unmet needs have been identified in various aspects of lipid nanoparticle-based carriers to date, such as enhanced gene delivery ability, maintenance of gene delivery ability in specific diseases such as liver disease, enhanced stability, improved targeting characteristics, ensuring ease of storage and distribution, mitigation of side effects, cost reduction, and response to breakthrough infections. Therefore, there is a continuous demand for the development of lipid nanoparticles using new ionized lipid carriers.

[0150] <Example 2> Manufacturing of lipid nanoparticles
[0151] Lipid nanoparticles encapsulating Firefly Luciferase mRNA (RNA Gene, Korea) were prepared using the compound (ionized lipid) prepared in Example 1 above, dioleoylphosphatidylethanolamine (DOPE) or distearoylphosphatidylcholine (DSPC) (neutral lipid), cholesterol (steroid), and PEG 2000-DMG (polymerized lipid).
[0152] Ionized lipids, DOPE / DSPC, cholesterol, and PEG 2000-DMG were mixed in a molar ratio of 49:10:38.5:2.5 and dissolved in ethanol at a concentration of 4.8 mg / mL to prepare a lipid solution. Separately, Firefly Luciferase mRNA was dissolved in a citric acid buffer solution (pH 4.0±1.0) or an acetic acid buffer solution (pH 5.0±1.0) at a weight ratio of 1:16 for mRNA: ionized lipid to prepare an active ingredient solution. Then, lipid nanoparticles were prepared by passing the mixture through a microfluidic mixing device (Benchtop Nanoassemblr, Precision Nanosystems) at a flow rate of approximately 15 mL / min so that the lipid solution: active ingredient solution had a volume ratio of 1:3. The manufactured lipid nanoparticles were diluted with Tris buffer containing 8.7% sucrose and dialyzed using a dialysis centrifuge tube to ensure that the ethanol content was less than 1%, and the final concentration was manufactured to be 0.2 mg / mL based on mRNA.
[0153] For comparison, lipid nanoparticles encapsulating Firefly Luciferase mRNA (RNA Gene, Korea) were prepared using MC3 ionized lipid, SM-102 ionized lipid, distearoylphosphatidylcholine (DSPC) (neutral lipid), cholesterol (steroid), and DMG-PEG2000 (polymer polymerized lipid). MC3, DSPC, cholesterol, and DMG-PEG2000 or SM-102, DSPC, cholesterol, and DMG-PEG2000 were prepared in a molar % ratio of 50:10:38.5:1.5 and used as lipid nanoparticles for comparison.

[0156] The size and surface charge of lipid nanoparticles prepared according to the ionized lipid in Example 1 were measured. To measure the size of lipid nanoparticles, each lipid nanoparticle was diluted with PBS to a concentration of 1 μg / mL, and the diameter and polydispersity index (PDI) of LNPs were measured using dynamic light scattering (DLS) on a Malvern Zetasizer Nano (Malvern Instruments, UK).
[0157] As a result, as shown in Table 1 below, all lipid nanoparticles exhibited a size of approximately 120 nm or less and a good polydispersity of less than 0.2.

[0160] <Example 4> Confirmation of gene inclusion rate
[0161] To determine the gene encapsulation rate of lipid nanoparticles, Quant-iTTM RibogreenTM RNA Reagent and Kit were used. 5 μL of ribosomal RNA standard (100 μg / mL in TE buffer) was diluted with 245 μL TE buffer or 0.4% Triton-TE buffer to prepare a stock solution for calibration curve. 2, 5, 10, 25, and 50 μL of the stock solution were diluted with TE buffer or 0.4% Triton-TE buffer to make a total volume of 100 μL. 100 μL of Quant-iTTM RibogreenTM RNA Reagent was added, mixed, and the fluorescence intensity (excitation 475 nm and emission 500–550 nm) was measured to prepare a calibration curve.
[0162] 5 μL of the nanoparticle solution prepared in Example 2 was diluted with 245 μL of TE buffer (10 mM Tris-HCl, 1 mM EDTA, pH 7.5 in DEPC-treated water). 50 μL of the diluted nanoparticle solution was added with 50 μL of TE buffer or 2% Triton-TE buffer, and incubated at 37°C for approximately 10 minutes. 40 μL of each culture medium was then placed in a microplate, followed by sequential addition of 60 μL of TE buffer and 100 μL of Quant-iT™ Ribogreen™ RNA Reagent, mixing, and measuring the fluorescence intensity. The measured fluorescence intensity was applied to the calibration curve to calculate the gene content (TO) of the TE buffer and the gene content (T2) of the 2% Triton-TE buffer, and then the gene inclusion rate was calculated using the following mathematical equation 1. The gene inclusion rate was confirmed and is shown in Table 2. As a result, all showed good gene inclusion rates.

[0170] To confirm the in vivo gene delivery efficiency of lipid nanoparticles, a single intravenous injection of 1 mg / kg based on the luciferase gene encapsulated in lipid nanoparticles was performed into 7-week-old C57BL / 6 mice, and the mice were sacrificed after 4 hours and their livers were removed.
[0171] To determine the level of luciferase expression in tissue samples, the weight of the liver tissue slices was measured and transferred to a tube containing 3 mm metal beads. 500 μl of Glo-Lysis Buffer (Promega, WI, USA) per 50 mg was added, and the tissue was homogenized using a bead homogenizer. The supernatant of the centrifuged homogenate was then diluted with Glo-Lysis Buffer at an appropriate ratio. 50 μl of the diluted tissue homogenate and Steady-Glo Luciferase Assay solution were each transferred to a 96-well white plate at a 1:1 ratio and incubated at room temperature. After 5 minutes, the luminescence value was detected using a GloMax Discover microplate reader (Promega). A calibration curve was generated using recombinant luciferase (Promega) as a standard, and the level of luciferase expression in the sample (ng / g tissue) was derived.
[0172] As a result, as shown in Table 3, it was confirmed that the lipid nanoparticles using the ionized lipid of the present invention exhibited an excellent gene transfer effect through systemic administration by showing a good level of luciferase expression in liver tissue when injected intravenously.

[0175] 5-2. Confirmation of in vivo gene transfer efficiency 2
[0176] To determine the in vivo gene transfer efficiency of repeated administration of lipid nanoparticles, lipid nanoparticles loaded with EPO mRNA (TriLink BioTechnologies, CA, USA) were intravenously administered to 7-week-old C57BL / 6 mice at a dose of 0.5 mg / kg per gene. To determine the in vivo EPO expression level, blood samples were collected 6 hours after administration, and the collected blood was anticoagulated to separate plasma.
[0177] To determine the concentration of EPO protein in plasma, an EPO enzyme-linked immunosorbent assay (ELISA) kit (R&D Systems, MN, USA) was used, and the analysis was performed according to the manufacturer's method. More specifically, the plasma sample was diluted 1:3000 using the specimen diluent included in the kit, and 100 μL of each (1:1 ratio) was added to the microwell and incubated at room temperature for 2 hours. After removing all wells, 200 μL of EPO conjugate was added and incubated for another 2 hours at room temperature. After washing four times with 300 μL of washing buffer, 200 μL of substrate solution was added and incubated for 20 minutes. To terminate the reaction, 100 μL of stop solution was added, and the absorbance was measured at wavelengths of 450 nm and 600 nm using a microplate (Promega). A calibration curve was derived using standard materials, and the EPO protein concentration (ng / mL) of each sample was derived within the defined range of the standard materials.
[0178] As a result, as shown in Table 4, the lipid nanoparticles using the ionized lipid of the present invention showed a good level of EPO expression in the blood compared to the lipid nanoparticles using the ionized lipids MC3 and SM-102 used for comparison when injected intravenously, confirming that they exhibit an excellent gene transfer effect through systemic administration.

[0181] 5-3. Confirmation of in vivo gene transfer efficiency 3
[0182] To confirm the gene delivery ability of lipid nanoparticles in liver damage disease, liver damage was induced in 6-week-old C57BL / 6 mice by intraperitoneal administration of CCl4 diluted to 20% in olive oil at 2 mL / kg three times a week for 2 weeks. The plasma ALT and AST levels were checked the day after the last CCl4 administration. Compared to the plasma ALT of 2,592 U / L and AST of 5,652 U / L in the olive oil vehicle group, liver damage was confirmed to have been induced. Mice with induced liver damage (liver damage model) and mice administered olive oil as a vehicle (normal model) were intravenously injected once at a dose of 1 mg / kg based on the loaded luciferase gene of each lipid nanoparticle. Six hours after administration, the mice were sacrificed and their livers were removed.
[0183] To confirm the luciferase expression level in tissue samples, the weight of the liver tissue slices was measured and transferred to a tube containing 3 mm metal beads. 500 μL of Glo-Lysis Buffer (Promega, WI, USA) was added per 50 mg, and the tissue was homogenized using a bead homogenizer. The supernatant of the centrifuged homogenate was then diluted with Glo-Lysis Buffer at an appropriate ratio. 50 μL of each of the diluted tissue homogenate and Steady-Glo Luciferase Assay solution were transferred to a 96-well white plate at a 1:1 ratio and incubated at room temperature. After 5 minutes, the luminescence value was detected using a GloMax Discover microplate reader (Promega). A calibration curve was generated using recombinant luciferase (Promega) as a standard, and the luciferase expression level (ng / g tissue) of the sample was derived.
[0184] As a result, as shown in Table 5, the lipid nanoparticles using the ionized lipid of the present invention exhibited the characteristic of maintaining excellent gene transfer ability without difference from a normal animal model when intravenously injected into a liver injury animal model. In contrast, the lipid nanoparticles using SM-102, used as a comparison, exhibited the characteristic of reduced gene transfer ability in a liver injury animal model.
