Description

[Technical field]

[0001] The present invention relates to an improved process for the preparation of 2-(3,5-dichlorophenyl)-6-benzoxazole carboxylic acid 1-deoxy-1-methylamino-D-glucitol, also known as 6-carboxy-2-(3,5-dichlorophenyl)-benzoxazole meglumine or tafamidis meglumine. The process of the present invention is particularly suitable for industrial scale production of tafamidis meglumine salt with high purity and high yield.

[0002] The present invention also relates to the potassium salt of 2-(3,5-dichlorophenyl)benzoxazole-6-carboxylic acid, which is useful for the preparation of 6-carboxy-2-(3,5-dichlorophenyl)-benzoxazole meglumine in high yield and purity.

[0003] The present invention also describes an improved process for the preparation of methyl 4-(3,5-dichlorobenzamido)-3-hydroxybenzoate, a key intermediate suitable for industrial scale production of the purified product in high yield and purity. [Background technology]

[0004] The compound of formula I below, 6-carboxy-2-(3,5-dichlorophenyl)-benzoxazole meglumine (tafamidis meglumine), is an orphan drug used to slow nerve damage caused by transthyretin amyloidosis, a genetic disease in which fibers called amyloid build up in the tissues of the body, including around the nerves. Tafamidis meglumine is marketed under the trademark Vyndaqel by Pfizer. [ka]

[0005] 6-Carboxy-2-(3,5-dichlorophenyl)-benzoxazole (tafamidis free acid) was first disclosed in European Patent EP1587821B1 in the name of Scripps Research Institute. EP1587821B1 describes in Example 5 a synthetic approach for the preparation of tafamidis as shown in Scheme 1. Condensation of 4-amino-3-hydroxybenzoic acid with 3,5-dichlorobenzoyl chloride in the presence of pyridine in refluxing tetrahydrofuran gives 4-(3,5-dichlorobenzamido)-3-hydroxybenzoic acid, which, without isolation, is cyclized in the presence of p-toluenesulfonic acid monohydrate in refluxing xylene and then methylated with Me3SiCHN2 in benzene / methanol to give methyl 2-(3,5-dichlorophenyl)benzo[d]oxazole-6-carboxylate, which is finally hydrolyzed with LiOH·H2O in tetrahydrofuran / methanol / H2O to give the free acid tafamidis as a white solid. [ka]

[0006] The method described in the aforementioned patent EP1587821B1 provides tafamidis free acid in low yields of 8%-27% over four steps. In addition, toxic reagents and solvents such as pyridine and benzene and also dangerous reagents such as the diazomethane derivative Me3SiCHN2 are used. The purity is not disclosed.

[0007] Razavi et al. [Angew. Chem. Int. Ed. 2003, 42, 2758-2761] and WO2013 / 168014A1 disclose the preparation of tafamidis free acid, which includes the same four steps as shown in Scheme 1. In particular, the intermediate methyl 2-(3,5-dichlorophenyl)-benzoxazole-6-carboxylate and tafamidis free acid are purified by column chromatography. The yield and purity are not disclosed. This method has several drawbacks, such as purification by column chromatography, which is a method unsuitable for application on an industrial scale.

[0008] Alternatively, Bulawa et. al. [Proceedings of the National Academy of Sciences of the USA, Early Edition, 2012, vol. 109, n° 24, 9629-9634] disclose the preparation of tafamidis free acid and tafamidis meglumine salt via the key intermediate methyl 4-(3,5-dichlorobenzamido)-3-hydroxybenzoate, as shown in Scheme 2. Methyl 4-amino-3-hydroxybenzoate hydrochloride is reacted with 3,5-dichlorobenzoyl chloride in the presence of pyridine in dichloromethane to give methyl 4-(3,5-dichlorobenzamido)-3-hydroxybenzoate which is further purified from acetone and water, isolated by filtration, and then cyclized in the presence of p-toluenesulfonic acid monohydrate to give methyl 2-(3,5-dichlorophenyl)benzo[d]oxazole-6-carboxylate which is further isolated by filtration and then reacted with LiOH in tetrahydrofuran / HO (1:1 v / v) at 40-45°C followed by pH adjustment with hydrochloric acid to give the free acid tafamidis. This compound is further converted to the meglumine salt by reacting it with N-methyl-D-glucamine (also 1-deoxy-1-methylamino-D-glucitol) in a mixture of isopropyl alcohol / HO at 65°C-70°C, which is then isolated by filtration. [ka]

[0009] An alternative method described by Bulawa et al. avoids the use of dangerous diazomethane derivatives such as Me3SiCHN2, but still uses toxic reagents such as pyridine. Yields and purity are not disclosed.

[0010] WO2016 / 038500A1 discloses crystalline solid forms of tafamidis free acid (6-carboxy-2-(3,5-dichlorophenyl)-6-benzoxazole) designated as Forms 1, 2, 4 and 6, as well as methods for their preparation. According to WO2016 / 038500A1, Form 1 is a non-hygroscopic anhydrous crystalline solid obtained by recrystallization from 2-propanol; Form 2 is a crystalline tetrahydrofuran solvate; Form 4 is another non-hygroscopic anhydrous crystalline solid form obtained from a solvent (tetrahydrofuran) / anti-solvent (toluene) crystallization method at low temperatures (-10°C to 25°C) for 12 hours; Form 6 is another non-hygroscopic anhydrous crystalline solid form obtained from a tetrahydrofuran / acetamide / dichloromethane mixture at low temperatures (about 0°C). In general, these crystallization methods carried out at low temperatures are unsuitable for use on an industrial scale. Purity is not disclosed.

[0011] Furthermore, according to WO2016 / 038500A1, the tafamidis free acid obtained by reproducing the procedure of Razavi et al. (i.e. also described in EP1587821B1 and WO2013 / 168014A1) is essentially amorphous and physically unstable and therefore unsuitable for use on an industrial scale and, most importantly, for use in pharmaceutical manufacturing.

[0012] As already explained, the prior art has not provided an efficient method for obtaining tafamidis meglumine salt with high purity and high yield. The lack of an efficient manufacturing method increases the cost of the final tafamidis meglumine salt and pharmaceutical compositions containing it, which are already expensive pharmaceutical products. Therefore, in view of the pharmacological value of the compound of formula I, it is desirable to develop an efficient and safe manufacturing method for tafamidis meglumine salt with high purity and high yield, which can be easily applied to industrial scale with low energy demand and cost. Summary of the Invention

[0013] The present invention provides an efficient and environmentally friendly process for the preparation of 2-(3,5-dichlorophenyl)-6-benzoxazole carboxylic acid 1-deoxy-1-methylamino-D-glucitol (tafamidis meglumine salt) which allows the production in high yield and high purity and is applicable on an industrial scale. The process of the present invention makes it possible to obtain tafamidis meglumine salt without the need for laborious and unfeasible purification steps and to obtain a salt product of high purity complying with pharmaceutical standards.

[0014] In a first aspect, the present invention provides an improved process for preparing a compound of formula I, 2-(3,5-dichlorophenyl)-6-benzoxazole carboxylic acid 1-deoxy-1-methylamino-D-glucitol, comprising the steps of: a) converting a compound of formula II, wherein R is C1-C4 alkyl; and b) converting 6-carboxy-2-(3,5-dichlorophenyl)-6-benzoxazole of formula III into 2-(3,5-dichlorophenyl)-6-benzoxazole carboxylic acid 1-deoxy-1-methylamino-D-glucitol of formula I, wherein step a) comprises the steps of: a1) converting a compound of formula I, wherein R is C1-C4 alkyl; and b) converting 6-carboxy-2-(3,5-dichlorophenyl)-6-benzoxazole of formula III into 2-(3,5-dichlorophenyl)-6-benzoxazole carboxylic acid 1-deoxy-1-methylamino-D-glucitol of formula I, The method comprises the substep of treating the compound of formula I with a first inorganic base selected from the group consisting of sodium hydroxide and potassium hydroxide in the presence of a first solvent or a first solvent mixture to obtain a compound of formula III, where M is a cation selected from the group consisting of sodium and potassium, and the substep of a2) treating the compound of formula IIIa obtained in substep a1) with an acid, preferably a hydroacid such as hydrochloric acid, hydrobromic acid or hydroiodic acid, more preferably hydrochloric acid, to obtain 6-carboxy-2-(3,5-dichlorophenyl)-6-benzoxazole of formula III, as shown in scheme 3. The use of said base makes it possible to carry out the method without isolating the compound of formula III, tafamidis free acid (i.e. the method of the first embodiment is carried out in one reaction vessel). Advantageously, the method of the present invention provides the compound of formula I, 2-(3,5-dichlorophenyl)-6-benzoxazole carboxylic acid 1-deoxy-1-methylamino-D-glucitol, in high yield and high purity. [ka]

[0015] A second aspect of the present invention provides a compound of formula IIIa, wherein M is selected from the group consisting of sodium and potassium, preferably M is sodium. [ka]

[0016] A third aspect of the present invention relates to the use of compound IIIa, wherein M is selected from the group consisting of sodium and potassium, preferably M is sodium, as an intermediate for the preparation of compound of formula I (tafamidis meglumine salt).

[0017] A fifth aspect of the present invention relates to a method for preparing a pharmaceutical composition comprising a compound of formula I 2-(3,5-dichlorophenyl)-6-benzoxazole carboxylic acid 1-deoxy-1-methylamino-D-glucitol, said method comprising obtaining 2-(3,5-dichlorophenyl)-6-benzoxazole carboxylic acid 1-deoxy-1-methylamino-D-glucitol of formula I by the method defined in the first aspect, combining said compound of formula I with at least one pharma- ceutically acceptable carrier or excipient, optionally wherein said composition is adapted for use in the treatment of transthyretin amyloidosis.

[0018] definition The term "one reaction vessel reaction" or "one reaction vessel process" is generally known in the art and refers to a chemical reaction in which the starting materials are converted into the final product of the reaction in a single reaction vessel, i.e., there are no intermediate reaction products to be isolated, removed or purified. In its broadest sense, a "one reaction vessel reaction" still allows for the formation of intermediate products, which are further converted into the final product by the addition of further reactants (in situ generation of intermediates). A one reaction vessel reaction also encompasses a reaction in a single reaction vessel in which the starting product is converted into the final product by the formation of one or more intermediate products that are formed in sequence without further addition of reagents ("multi-step" reactions). Thus, a "one reaction vessel process" is characterized by at least two reaction steps that are carried out without isolation and / or purification of intermediates or products, preferably carried out in a single reaction vessel / vessel. It will be understood by those skilled in the art that although the intermediate products are not isolated and / or purified, the simple transfer of the entire reaction mass at an intermediate stage is still a "one-reaction vessel process" according to the present invention, since such a process would still achieve the technical advantages associated with the one-reaction vessel process, not least in that the intermediates formed in situ do not need to be isolated and / or purified.

[0019] The term "leaving group," as used herein, refers to a molecular fragment that leaves with an electron pair in a heterolytic bond cleavage and is selected from the group consisting of acyls such as acetyl, sulfonate esters such as tosylates and mesylates, and halogen atoms such as fluorine, chlorine, bromine, and iodine, among others. Preferably, the "leaving group" is a halogen, such as Br, I, or Cl, more preferably Cl.

[0020] The term "hydroacid" (also known as binary acid), as used herein, refers to an inorganic acid in which hydrogen is combined with a second non-metallic element, such as S, F, Cl, Br, or I. More preferably, the "hydroacid" is selected from the group consisting of hydrochloric acid, hydrobromic acid, and hydroiodic acid.

Preferably, the alkyl group is a C1-C4 alkyl, more preferably, the alkyl is methyl and ethyl.

[0025] The term "halogen," as used herein, refers to an atom selected from the group consisting of fluorine, chlorine, bromine and iodine.

[0026] The term "inorganic base" as used herein refers to a substance that tends to accept a proton. Inorganic bases contain metal cations and do not contain an organic moiety, in contrast to organic bases, which are substances that contain an organic moiety. Inorganic bases suitable for the present invention are hydroxides, carbonates or bicarbonates of alkali metals such as lithium, sodium, potassium and cesium, or alkaline earth metals such as calcium and magnesium.

[0027] The term "conventional isolation techniques" or "purification" as used herein refers to methods by which a product can be rendered free of extraneous elements to obtain a purified product. The term "industrial purification" refers to purification that can be carried out on an industrial scale, such as solvent extraction, filtration, slurrying, washing, phase separation, evaporation, centrifugation, or crystallization. [0037] Detailed Description of the Invention According to a first aspect of the present invention, there is provided 2-(3,5-dichlorophenyl)-6-benzoxazole carboxylic acid 1-deoxy-1-methylamino-D-glucitol (tafamidis meglumine, compound of formula I): [ka] 1. An improved process for producing a) Formula II: [ka] (Wherein, R is C1-C4 alkyl.) with a compound of formula III, 6-carboxy-2-(3,5-dichlorophenyl)-6-benzoxazole: [ka] converting the Here, step a) is a1) treating a compound of formula II with a first inorganic base selected from the group consisting of sodium hydroxide and potassium hydroxide in the presence of a first solvent or a first mixture of solvents to obtain a compound of formula IIIa: [ka] where M is a cation selected from the group consisting of sodium and potassium. and a2) The compound of formula IIIa obtained in substep a1) is treated with an acid, preferably a hydroacid such as hydrochloric acid, hydrobromic acid or hydroiodic acid, more preferably hydrochloric acid, to give the compound of formula III: [ka] Sub-step of obtaining 6-carboxy-2-(3,5-dichlorophenyl)-6-benzoxazole and b) converting the compound of formula III obtained in step a) into the compound of formula I, 2-(3,5-dichlorophenyl)-6-benzoxazole carboxylic acid 1-deoxy-1-methylamino-D-glucitol; A method is described that includes:

[0038] The use of sodium or potassium hydroxide as the first inorganic base allows for the process to be carried out without isolation of the compound of formula III.

[0039] The inventors have realized that when reproducing the method as described by Bulawa et al., tafamidis free acid, a compound of formula III, precipitates in the reaction medium immediately after the addition of aqueous hydrochloric acid. Thus, the obtained tafamidis free acid is isolated from the reaction medium by filtration and further converted to meglumine salt. On the other hand, the inventors have surprisingly found that the compound of formula I can be prepared by steps a) and b) according to the method of the first aspect in one single reaction vessel. Importantly, the tafamidis free acid, a compound of formula III, prepared according to the first aspect is not isolated because it does not precipitate in the reaction medium. Advantageously, the method of the invention according to the first aspect provides the compound of formula I, 2-(3,5-dichlorophenyl)-6-benzoxazole carboxylic acid 1-deoxy-1-methylamino-D-glucitol, in high yield and high purity.

[0040] Advantageously, the process of the present invention avoids the isolation of advanced intermediates, i.e., the compound of formula IIIa and the compound of formula III, and thus reduces the number of steps to provide the compound of formula I, 2-(3,5-dichlorophenyl)-6-benzoxazole carboxylic acid 1-deoxy-1-methylamino-D-glucitol, which constitutes a preferred embodiment of the present invention.

[0041] In another embodiment of the first aspect of the invention, step a) further comprises an additional sub-step a3) of adding water to the resulting mixture after sub-step a2). The amount of water (volume in milliliters) can be from 1:1 (v / w) to 10:1 (v / w), preferably from 1:1 (v / w) to 5:1 (v / w) relative to the weight (grams) of the compound of formula II treated in step a1).

[0042] In another embodiment of the first aspect of the invention, step a) comprises sub-step a3) as described in the preceding paragraph followed by an additional sub-step a4) of decanting the mixture resulting in sub-step a3) to separate the aqueous phase (containing mainly inorganic salts) from the desired organic phase (containing mainly the compound of formula III) and then discarding the aqueous phase.

[0043] Advantageously, the separation of the by-product impurities inorganic salts from the compound of formula III obtained in substep a2) allows the following step b) of the first aspect of the invention, the formation of the meglumine salt, since the competition for salt formation is significantly reduced, thus increasing the purity of the compound of formula I as well as the overall yield of the process.

[0044] In another embodiment of the first aspect, the first solvent or the first solvent mixture used in substep a1) is a mixture of an organic solvent and water. Suitable organic solvents may be selected from the group consisting of ethers, alcohols and mixtures thereof. [0045] In another embodiment of the first aspect, the ratio (by volume) of tetrahydrofuran to water used in substep a1) ranges from 2:1 (v / v) to 15:1 (v / v), preferably from 5:1 (v / v) to 10:1 (v / v), more preferably from 6:1 (v / v) to 8:1 (v / v).

[0046] In another embodiment of the first aspect, sub-step a1) may be carried out at a temperature range of 40°C to 70°C, preferably 45°C to 65°C, more preferably 50°C to 60°C.

[0047] In another embodiment of the first aspect, the acid used in sub-step a2) according to the first aspect of the invention is an inorganic or organic acid. Suitable acids are those having a pKa (vs. water) less than 5, preferably less than 3, more preferably less than 1. Preferably, the acid has a pKa (vs. water) of -10 to 5, preferably -10 to 3, more preferably -10 to 1. Suitable acids include, but are not limited to, hydrofluoric acid, hydrochloric acid, hydrobromic acid, phosphoric acid, sulfuric acid, formic acid, acetic acid, dichloroacetic acid, methanesulfonic acid, p-toluenesulfonic acid and camphorsulfonic acid. Preferably, the acid is a hydroacid such as hydrochloric acid, hydrobromic acid or hydroiodic acid, which may exist as a gas or aqueous solution or may be generated in situ, for example from an alkylsilyl halide, in the presence of a protic solvent.

[0048] In a preferred embodiment of the first aspect, the acid used in substep a2) is hydrochloric acid. The molar ratio of the compound of formula IIIa to the acid is 1:1 to 1:3. Preferably, the molar ratio of the compound of formula IIIa to the acid is 1:1 to 1:1.5.

[0049] In another preferred embodiment of the first aspect, sub-step a2) may be carried out at a temperature range of 35°C to 70°C, preferably 40°C to 60°C, more preferably 45°C to 55°C.

[0050] In another preferred embodiment of the first aspect, sub-step a3) may be carried out at a temperature range of 35°C to 70°C, preferably 40°C to 60°C, more preferably 45°C to 55°C.

[0051] In another preferred embodiment of the first aspect, sub-step a4) may be carried out at a temperature range of 35°C to 70°C, preferably 40°C to 60°C, more preferably 45°C to 55°C.

[0052] In another embodiment of the first aspect of the present invention, R is selected from the group consisting of methyl and ethyl, preferably, R is methyl.

[0053] In another embodiment of the first aspect, the cation M is sodium. Thus, the alkali metal hydroxide used in sub-step a1) is sodium hydroxide.

[0054] In a first preferred embodiment, the process for preparing a compound of formula I comprises the following steps and substeps: a1) reacting a compound of formula II, wherein R is methyl, with an alkali metal hydroxide selected from the group consisting of sodium hydroxide and potassium hydroxide in the presence of a first solvent or a first solvent mixture to obtain a compound of formula IIIa: [ka] wherein M is selected from the group consisting of sodium and potassium. a sub-step of obtaining a compound of formula (I) a2) adding hydrochloric acid to the compound of formula IIIa obtained in substep a1) to obtain a compound of formula III: [ka] a sub-step of obtaining tafamidis free acid, which is a compound of formula (I); a3) adding water to the mixture resulting from substep a2), a4) decanting the mixture resulting from substep a3) and then discarding the aqueous phase obtained; and b) converting the compound of formula III to the compound of formula I, 2-(3,5-dichlorophenyl)-6-benzoxazole carboxylic acid 1-deoxy-1-methylamino-D-glucitol wherein the compound of formula IIIa and the 6-carboxy-2-(3,5-dichlorophenyl)-6-benzoxazole of formula III are not isolated (i.e., the process is carried out in one reaction vessel).

[0055] In another embodiment of the first aspect, step b) comprises converting the compound of formula III obtained in step a) into the compound of formula I, 2-(3,5-dichlorophenyl)-6-benzoxazole carboxylic acid 1-deoxy-1-methylamino-D-glucitol, in the presence of a second solvent or a second solvent mixture.

[0056] In a further embodiment of the first aspect, step b) comprises b1) a substep of adding N-methyl-D-glucamine (1-deoxy-1-methylamino-D-glucitol) to the compound of formula III obtained in step a), b2) removing the solvent of the mixture resulting from substep b1), preferably by distillation; and b3) a substep of treating the mixture resulting from substep b2) with a second solvent or a second solvent mixture. [0068] A second embodiment provides a compound of formula IIIa, wherein M is selected from the group consisting of sodium and potassium, preferably M is potassium. Advantageously, the potassium salt of compound IIIa (potassium salt of 2-(3,5-dichlorophenyl)benzoxazole-6-carboxylic acid) is particularly useful for the preparation of compound of formula I, 2-(3,5-dichlorophenyl)-6-benzoxazole carboxylic acid 1-deoxy-1-methylamino-D-glucitol, in high yield and purity. [ka]

[0069] The inventors have recognized that the compound of formula IIIa, in which M is a cation selected from lithium, competes with the meglumine salt in the corresponding salt formation process, precipitating the lithium salt of compound IIIa instead of the meglumine salt. Surprisingly, the compound of formula IIIa, in which M is potassium or sodium, i.e. potassium 2-(3,5-dichlorophenyl)benzoxazole-6-carboxylate or sodium 2-(3,5-dichlorophenyl)benzoxazole-6-carboxylate, remains soluble in the reaction medium and does not compete in precipitating the meglumine salt. Thus, the compound IIIa, in which M is potassium or sodium, preferably the compound in which M is sodium, is particularly useful for the preparation of the compound of formula I, 2-(3,5-dichlorophenyl)-6-benzoxazole carboxylate 1-deoxy-1-methylamino-D-glucitol, in high yield and high purity, due to its high solubility in the reaction medium when compared with the corresponding known lithium salt of compound IIIa.

[0070] A third aspect of the present invention relates to the use of compound IIIa, wherein M is selected from the group consisting of sodium and potassium, as an intermediate for the preparation of compound of formula I (tafamidis meglumine salt).

[0071] A fourth aspect of the present invention relates to a compound of formula II: [ka] 1. A process for the preparation of a compound of the formula i) Formula V: [ka] wherein R is C1-C4 alkyl. with a compound of formula VI: [ka] wherein X is a leaving group, preferably a halogen, more preferably X is chloride. in a third solvent or a third solvent mixture in the presence of a second inorganic base to produce a compound of formula IV: [ka] where R is as defined above. obtaining a compound of formula (I), ii) converting the compound of formula IV to a compound of formula II. The present invention provides a method for producing the composition comprising the steps of:

[0072] In one embodiment of the fourth aspect, R is selected from the group consisting of methyl and ethyl, preferably methyl.

[0073] In another embodiment of the fourth aspect, the second inorganic base is an alkali metal carbonate or bicarbonate, preferably selected from the group consisting of sodium carbonate, sodium bicarbonate, potassium carbonate and potassium bicarbonate, more preferably sodium bicarbonate.

[0074] In another embodiment of the fourth aspect, the third solvent is tetrahydrofuran or the third solvent mixture comprises tetrahydrofuran, preferably the third solvent is tetrahydrofuran.

[0075] In another embodiment of the fourth aspect, the method further comprises isolating the compound of formula IV, preferably by conventional isolation techniques, more preferably by filtration.

[0076] In another embodiment of the fourth aspect, the method further comprises purifying the compound of formula IV, preferably by conventional purification techniques, more preferably by crystallization.

[0077] The compound of formula IV can be converted to the compound of formula II as described in the prior art (e.g., Bulawa et. al. [Proceedings of the National Academy of Sciences of the USA, Early Edition, 2012, vol. 109, n° 24, 9629-9634). In particular, compound IV can be cyclized in the presence of p-toluenesulfonic acid in toluene (preferably at reflux temperature) and then, optionally, treated with charcoal to obtain the corresponding 2-(3,5-dichlorophenyl)-6-benzoxazole carboxylate of formula II. In a preferred embodiment, the compound of formula II thus obtained is purified in a mixture of acetone and water.

[0078] A fifth aspect of the present invention relates to a method for preparing a pharmaceutical composition comprising tafamidis or a pharma- ceutically acceptable salt thereof, preferably tafamidis meglumine, which method comprises obtaining 2-(3,5-dichlorophenyl)-6-benzoxazole carboxylic acid 1-deoxy-1-methylamino-D-glucitol of formula I according to the method defined in the first aspect, and further combining said tafamidis or a pharma- ceutically acceptable salt thereof with at least one pharma- ceutically acceptable carrier or excipient, optionally wherein said pharmaceutical composition is adapted for use in the treatment of transthyretin amyloidosis.
