Description

[Technical field]

[0001] The present invention relates to a method for producing an antibody-drug conjugate, and also to a mutant enzyme of endo-β-N-acetylglucosaminidase used in the production method. [Background technology]

[0002] Antibody drug conjugates (hereinafter referred to as ADCs) are antibodies that specifically bind to a target and are conjugated with a drug. For example, an ADC in which a payload with a strong cell-killing effect is conjugated with an antibody that targets cancer cells can selectively deliver a drug (payload) to cancer cells, thereby significantly reducing the systemic toxicity that is a side effect, and effectively killing cancer cells, compared to the use of a single payload. A representative ADC is T-DM1 (Kadcyla (registered trademark)), which is an anti-HER2 antibody trastuzumab conjugated with a tubulin inhibitor, and has been approved as a drug (Non-Patent Documents 1, 2, and 3). Since T-DM1 randomly binds the payload to the Lys residue on the antibody, it is produced as a mixture with heterogeneous binding positions and numbers of bindings to the antibody of the payload. Similar ADCs include gemtuzumab ozogamicin (generic name: Mylotarg) (Non-Patent Document 4), brentuximab vedotin (generic name: Adcetris) (Non-Patent Document 5), etc.

[0003] In recent years, it has been reported that the homogeneity of ADCs affects pharmacokinetics, drug release rate, and efficacy (Non-Patent Document 6). From the viewpoint of ADC quality control, site-selective payload binding methods are being actively developed with the aim of producing highly homogeneous ADCs. One such method is a method of binding to antibody glycans using an enzyme. The ADC production process using this method that has been reported so far is as follows: (1) homogenization of glycans by enzymatic cleavage of heterogeneous antibody glycans (hydrolysis process), (2) enzymatic binding of a glycosyl homogenized antibody to another glycan (transglycosylation process), and (3) binding of the azide group of another glycan to a payload (chemical reaction process).

[0004] When the purpose of this method is to convert sugar chains, an enzyme family called endo-β-N-acetylglucosaminidase is used in the hydrolysis step and the sugar transfer step (Non-Patent Document 7). Endo-β-N-acetylglucosaminidase is isolated from various species and used depending on the type of antibody sugar chain. Representative endo-β-N-acetylglucosaminidases used in the hydrolysis step include EndoA, EndoD, EndoM, EndoH, EndoF2, EndoF3, EndoE, EndoS, EndoS2, etc.

[0005] Among these, it has been reported that hydrolysis ability is suppressed in EndoS D233Q, in which Asp at position 233 of EndoS is substituted with Gln (Patent Document 1, Non-Patent Document 8), EndoS2 D184M or EndoS2 D184Q, in which Asp at position 184 of EndoS2 is substituted with Met or Gln (Patent Document 2, Non-Patent Document 9), and EndoF3 D165A or EndoF3 D165Q, in which Asp at position 165 of EndoF3 is substituted with Ala or Gln (Patent Document 3, Non-Patent Document 10). It has also been reported that these mutant enzymes promote glycosylation between a glycosylated antibody (GlcNAc antibody) and a substrate in which the reducing end of the glycan is oxazolinated.

[0006] Furthermore, it has been reported that a homogeneous ADC was produced by using EndoS D233Q to transfer a GlcNAc antibody and a reducing end of a glycan having an azide group to an oxazolinated substrate, and then chemically binding the azide group of the resulting conjugate to MMAE, a type of payload (Non-Patent Documents 11 and 12). [0009] The manufacturing process of ADC using glycosylated antibodies and enzymes that has been reported so far requires three steps: hydrolysis of the antibody glycan, glycotransfer to the glycosylated antibody, and linking of the payload to the antibody-glycan. Considering the purification of ADC at each step, it is expected that the cost of ADC production will increase and the performance and function of the antibody will decrease due to purification, so a simpler manufacturing method is desired.

[0010] In general, enzyme reactions are substrate-specific. Therefore, if a substrate glycan modified with a payload is to be conjugated to a glycosylated antibody in one step using a glycosyltransferase, the reaction efficiency of the enzyme is expected to be significantly reduced. In fact, the present inventors have attempted to conjugate a glycan modified with a peptide or drug to an antibody using various glycosyltransferases, but the reaction efficiency was significantly reduced compared to the case of linking a simple glycan to an antibody.

[0011] The present invention aims to provide an enzyme that can efficiently conjugate a glycan modified with a payload to a glycosylated antibody, and to provide a novel method for producing an ADC that uses this enzyme to conjugate an antibody and a payload in a single step, whereas the conventional method required two steps. [0015] The present invention relates to a method for producing antibody-drug conjugates (hereinafter referred to as "ADC"), comprising the steps of: The production method of the present invention comprises a step of conjugating a glycosylated antibody with a glycosylated drug. The step of conjugating a glycosylated antibody with a glycosylated drug can be carried out by a synthetic method or an enzymatic method.

[0016] [enzyme] When the conjugation step is carried out by an enzymatic method, an endo-β-N-acetylglucosaminidase mutant enzyme can be used. In the present invention, the term "endo-β-N-acetylglucosaminidase mutant enzyme" refers to an endo-β-N-acetylglucosaminidase mutant enzyme with suppressed hydrolysis ability. Representative examples of endo-β-N-acetylglucosaminidase include EndoA, EndoD, EndoM, EndoH, EndoF2, EndoF3, EndoE, EndoS, and EndoS2.

[0017] In a preferred embodiment, among the endo-β-N-acetylglucosaminidase mutant enzymes, a mutant enzyme in which asparagine (D) at position 233 of EndoS is substituted, a mutant enzyme in which asparagine (D) at position 184 of EndoS2 is substituted, or a mutant enzyme in which asparagine (D) at position 165 of EndoF3 is substituted, preferably a mutant enzyme in which asparagine (D) at position 233 of EndoS is substituted, or a mutant enzyme in which asparagine (D) at position 165 of EndoF3 is substituted.

[0018] More preferred examples of the endo-β-N-acetylglucosaminidase mutant enzymes include mutant enzymes consisting of any one of the following proteins: (1) a protein consisting of the amino acid sequence of SEQ ID NO: 1, or the amino acid sequence of SEQ ID NO: 1 in which the 165th position is substituted with an amino acid other than aspartic acid (D); (2) A protein having the amino acid sequence of the protein according to (1) in which one or more amino acids have been substituted, deleted, inserted, and / or added, with the proviso that the 165th amino acid is an amino acid other than D, and having conjugation activity; (3) A protein having an amino acid sequence having at least 90% identity to the amino acid sequence of the protein according to (1), with the proviso that the 165th amino acid is an amino acid other than D, and having a conjugation activity; (4) a protein consisting of the amino acid sequence of SEQ ID NO: 11, or the amino acid sequence of SEQ ID NO: 11 in which the 233rd position is substituted with an amino acid other than D; (5) A protein having the amino acid sequence of the protein according to (4) in which one or more amino acids are substituted, deleted, inserted, and / or added, with the proviso that the 233rd position is an amino acid other than D, and having conjugation activity; (6) A protein having an amino acid sequence having at least 90% identity to the amino acid sequence of the protein according to (4), with the proviso that the 233rd amino acid is an amino acid other than D, and having a conjugation activity; (7) A protein consisting of the amino acid sequence of SEQ ID NO: 24, or the amino acid sequence of SEQ ID NO: 24 in which the 165th position is replaced with an amino acid other than aspartic acid (D).

[0019] Another example of an endo-β-N-acetylglucosaminidase mutant enzyme is a mutant enzyme consisting of any one of the following proteins: (1) a protein consisting of the amino acid sequence of SEQ ID NO: 1, or the amino acid sequence of SEQ ID NO: 1 in which the 165th glutamine (Q) is changed to alanine (A); (2) A protein having the amino acid sequence of the protein according to (1) in which one or more amino acids are substituted, deleted, inserted, and / or added, with the proviso that the 165th amino acid is Q or A, and having a conjugation activity; (3) A protein consisting of an amino acid sequence having identity to the amino acid sequence of the protein according to (1), with the proviso that the 165th amino acid is Q or A, and having a conjugation activity; (4) a protein consisting of the amino acid sequence of SEQ ID NO: 11; (5) A protein having the amino acid sequence of the protein according to (4) in which one or more amino acids are substituted, deleted, inserted, and / or added, with the proviso that the 233rd position is Q, and having a conjugation activity; (6) A protein consisting of an amino acid sequence having identity to the amino acid sequence of the protein according to (4), with the proviso that the 233rd position is Q, and having a conjugation activity; (7) A protein consisting of the amino acid sequence of SEQ ID NO: 24, or the amino acid sequence of SEQ ID NO: 24 in which the 165th position is replaced with an amino acid other than aspartic acid (D).

[0020] In the sequence listing, the amino acid sequence of EndoF3 D165Q, in which the 165th amino acid of EndoF3 is replaced with glutamine (Q), is shown as SEQ ID NO: 1. Also, the amino acid sequence of EndoS D233Q, in which the 233rd amino acid of EndoS is replaced with glutamine (Q), is shown as SEQ ID NO: 11. Note that, in the following, the present invention may be described using as an example the case where EndoF3 D165Q or EndoS D233Q is used as the endo-β-N-acetylglucosaminidase mutant enzyme, but those skilled in the art can understand the description by appropriately applying it to the case where other mutant enzymes are used.

[0021] The endo-β-N-acetylglucosaminidase mutant enzyme used for conjugation may be one in which a point mutation has been introduced into the above mutant enzyme. Examples of such mutant enzymes include a mutant enzyme consisting of a protein having an amino acid sequence of SEQ ID NO: 1, or an amino acid sequence in which the 165th glutamine (Q) in the amino acid sequence of SEQ ID NO: 1 is alanine (A), and having at least one mutation selected from the group consisting of K81T, M97W, Y, F, V, G, S, or Q, V129G, Q250I, N270R, L, or T, and Y282W. [0022] In the present invention, the term conjugation activity in relation to an endo-β-N-acetylglucosaminidase mutant enzyme means, unless otherwise specified, that when a conjugation reaction is carried out between a glycosylated antibody and a glycosylated drug, at least 20% of the antibody (adduct) to which the drug is attached can be produced (100(%) - residual rate (%) is 20% or more). The rate (%) here can be calculated based on the peak area in the HPLC chromatogram of the conjugation reaction solution. When multiple types of adducts are produced, for example when a 1-adduct and a 2-adduct are produced, all of the adducts are taken into consideration. Specifically, it can be calculated based on the following formula.

[0023] Residual rate (%) = [peak area value of unreacted antibody] / [(peak area value of unreacted antibody) + (peak area value of adduct: if multiple types of adducts are formed, the sum of the peak areas of each adduct) × 100

[0024] 1-adduct conversion rate (%) = [peak area value of 1-adduct] / [(peak area value of unreacted antibody)+(peak area value of 1-adduct)+(peak area value of 2-adduct)]×100

[0025] 2-adduct conversion rate (%) = [peak area value of 2-adduct] / [(peak area value of unreacted antibody)+(peak area value of 1-adduct)+(peak area value of 2-adduct)]×100

[0051] [ka]

[0052] In a particularly preferred embodiment, the process of the present invention can be carried out as follows. (1) The glycan of an anti-HER2 antibody, which originally has a heterogeneous glycan structure, is cleaved. The glycan can be cleaved using endo-β-N-acetylglucosaminidase (EndoS). EndoS cleaves the N-type complex glycan, which is bound to asparagine (Asn) 297 located in the CH domain of the Fc region of the antibody, leaving one N-acetylglucosamine (GlcNAc) residue at the reducing end. The N-acetylglucosamine at the reducing end may or may not be bound to fucose. The antibody with the cleaved glycan may then be purified using chromatography. Examples of chromatography include gel filtration chromatography, affinity chromatography, ion exchange chromatography, and hydrophobic chromatography. (2) A glycan-binding drug is conjugated to the glycan of the antibody-GlcNAc obtained by (1). Conjugation can be performed using an endo-β-N-acetylglucosaminidase mutant enzyme. N-acetylglucosamine (GlcNAc) residue at the reducing end of the N-complex glycan bound to asparagine (Asn) at position 297 located in the CH domain of the Fc region of the antibody-GlcNAc is bound to an oxazolinyl N-acetylglucosamine at the reducing end of the glycan-binding drug. Thereafter, the antibody with the cleaved glycan can be purified by chromatography. Examples of chromatography include gel filtration chromatography, affinity chromatography, ion exchange chromatography, and hydrophobic chromatography.

[0053] [Term etc.] In the present invention, when a protein or amino acid sequence is referred to as an "amino acid sequence in which one or more amino acids have been substituted, deleted, inserted, and / or added," the number of amino acids to be substituted or otherwise modified is not particularly limited in any protein, unless otherwise specified, so long as the protein consisting of that amino acid sequence has the desired function, but is about 1-750, 1-500, 1-250, 1-200, 1-150, 1-100, 1-50, 1-40, 1-30, 1-20, 1-15, 1-9, or 1-4 amino acids, or a larger number of substitutions, etc., may be possible as long as the substitutions are with amino acids of similar properties. Means for preparing polynucleotides or proteins relating to such amino acid sequences are well known to those skilled in the art.

[0054] In the description of the present invention, with regard to amino acids or amino acid residues, unless otherwise specified, A is alanine, C is cysteine, D is aspartic acid, E is glutamic acid, F is phenylalanine, G is glycine, H is histidine, I isoleucine, K is lysine, L is leucine, M is methionine, N is asparagine, P is proline, Q is glutamine, R is arginine, S is serine, T is threonine, U is selenocysteine (3), V is valine, W is tryptophan, and Y is tyrosine.

[0055] In the present invention, when the term "identity" is used with respect to a base sequence (sometimes called a nucleotide sequence) or an amino acid sequence, it means the percentage of the number of identical nucleotides or amino acids shared between two sequences when the two sequences are aligned in an optimal manner, unless otherwise specified. That is, identity can be calculated as follows: identity = (number of identical positions / total number of positions) x 100, and can be calculated using a commercially available algorithm. In addition, such an algorithm is incorporated in the NBLAST and XBLAST programs described in Altschul et al., J. Mol. Biol. 215 (1990) 403-410. More specifically, searches and analyses regarding the identity of base sequences or amino acid sequences can be performed using algorithms or programs (e.g., BLASTN, BLASTP, BLASTX, ClustalW) well known to those skilled in the art. When using a program, parameters can be appropriately set by those skilled in the art, and the default parameters of each program may be used. Specific techniques for these analysis methods are also well known to those skilled in the art. The identity may be calculated using genetic information processing software Genetyx (registered trademark) (Genetyx Corporation). If the subject sequence for which the identity percentage is to be determined has an additional sequence at the end that is not present in the sequence to be compared, such as a tag sequence, the additional sequence portion is not included in the identity percentage calculation.

[0056] In the present invention, when referring to identity with respect to a base sequence or amino acid sequence, unless otherwise specified, it refers to sequence identity of at least 50%, for example 60% or more, 70% or more, preferably 80% or more, more preferably 85% or more, even more preferably 90% or more, even more preferably 95% or more, even more preferably 97.5% or more, and even more preferably 99% or more.

[0079] [Table 1]

[0080] Analysis time: 23 min Flow rate: 0.50 mL / min, column temperature: 25℃, detection wavelength: 214 nm, 280 nm, injection volume: 10 μL.

[0081] For each HPLC chromatogram, the conjugation with daunorubicin-attached glycan by the EndoF3 mutant enzyme is shown in Figure 5, and the conjugation with MMAE-attached glycan by the EndoF3 mutant enzyme is shown in Figure 6. [0082] Antibody-GlcNAc remaining rate (%) = [peak area value of antibody-GlcNAc] / [(peak area value of antibody-GlcNAc)+(peak area value of monoadduct)+(peak area value of diadduct)]×100

[0083] 1-adduct conversion rate (%) = [peak area value of 1-adduct] / [(peak area value of antibody-GlcNAc) + (peak area value of 1-adduct) + (peak area value of 2-adduct)] × 100

[0084] 2-adduct conversion rate (%)=[peak area value of 2-adduct] / [(peak area value of antibody-GlcNAc)+(peak area value of 1-adduct)+(peak area value of 2-adduct)]×100

[0085] In addition, intact mass analysis was performed for each conjugate. The obtained MS chromatograms were deconvoluted. The results of conjugation with daunorubicin-linked glycans using the EndoF3 mutant enzyme are shown in Figure 7, and the results of conjugation with MMAE-linked glycans using the EndoF3 mutant enzyme are shown in Figure 8.

[0086] The activity of the EndoS mutant enzyme was measured in the same manner as that of the EndoF3 mutant enzyme.

[0087] The activity of each mutant enzyme in the case of daunorubicin-linked glycan is shown in the table below.

[0088] [Table 2]

[0089] Among the EndoF3 mutant enzymes, D165Q / M97G showed the lowest survival rate. This mutant enzyme is expected to show similar high activity even when the glycan structure is different.

[0090] The activity of each mutant enzyme in the case of daunorubicin-linked glycan is shown in the table below.

[0091] [Table 3]

[0092] High conversion rates were obtained when the EndoS mutant enzyme was used.

[0093] [Table 4]

[0094] Although the EndoS2 mutant enzyme was also able to perform conversion to a certain extent, the EndoF3 mutant enzyme and the EndoS mutant enzyme had higher conversion rates under the experimental conditions. In addition, it was predicted that the conversion rate would be lower when conjugating a glycan with a payload added than when conjugating a glycan with a linker, but contrary to expectations, it was found that the conversion rate was higher when conjugating a daunorubicin- or MMAE-glycan.
