Abstract
Description
[Technical field]
[0001] The present invention relates to an oral pouched product according to claim 1, an oral pouched product comprising according to claim 69 and a method for producing an oral pouched product according to claim 71. [Background technology]
[0002] Delivery of nicotine through smoking has many well-known drawbacks, particularly health-related problems such as the inclusion of carcinogens. However, tobacco substitutes also have drawbacks, such as insufficient relief of cravings in users. Summary of the Invention [Problem to be solved by the invention]
[0003] It is an object of one embodiment of the present invention to provide a nicotine-containing pouch, for example as a tobacco substitute, which can solve the above problems. Pouched products for oral delivery, such as nicotine, have proven to be attractive, user-friendly and attractive vehicles for the delivery of certain types of substances, such as active ingredients, sugars or sweeteners, etc. However, a challenge associated with such vehicles is that the substance to be delivered must be released from the pouch, at least in part, as a result of salivation in the user's mouth. If such salivation is inhibited or reduced in any way, the intended delivery profile may be unintentionally altered. For example, when releasing nicotine, complete drying of the pouch contents may affect the contact area of the oral cavity.
[0004] The above problems are addressed in several aspects of the present invention. [Means for solving the problem]
[0005] The present invention relates in a first aspect to an oral pouched product, said oral pouched product comprising: Saliva-permeable pouches and pouch compositions, Including, The pouch composition comprises: A powdered composition; Water, A liquid flavoring composition; wherein the liquid flavor composition is added to the powdered composition after the water.
[0006] The advantage of the present invention can be that a highly desirable flavor release can be obtained. By using the liquid flavor composition according to the present invention, the user experiences an initial noticeable flavor burst. The liquid flavor composition distributes over a wide surface of the provided mixture containing sugar alcohol and water, thereby promoting effective contact with saliva, which also advantageously promotes flavor release.
[0007] The invention, on the other hand, counteracts the drying of the mucosa in order to stimulate saliva secretion, especially in the initial phase of use of the pouch. The invention provides an initial burst of flavor, which also promotes saliva secretion, which affects the release, and at the same time makes it possible to obtain an impressive flavor profile that is indeed very well perceived by the user.
[0008] In a further aspect, it is further noted that the synergy between the use of liquid flavors and water (as a result of wetting the pouch) brings taste perception to a new level while also affecting and dissolving sweeteners such as sugar alcohols, thereby providing additional saliva production.
[0009] The rapid and effective contact of the liquid flavoring with saliva promotes increased saliva secretion, which promotes faster wetting of the pouch, thereby providing the user with an improved wetness of the pouch. This increased saliva secretion can also increase the dissolution of saliva-soluble ingredients, such as sugar alcohols or any active ingredients, providing the user with a stronger initial sweetness sensation.
[0010] An important advantage of the present invention is that it does not require drying of the composition after addition of the liquid flavor composition, so water is added followed by the liquid flavor composition. In an advantageous embodiment of the present invention, the pouch composition further comprises a powdered flavour composition.
[0011] By combining a liquid flavorant composition with a powdered flavorant composition in accordance with the present invention, the user experiences both an initial noticeable burst of flavor and a sustained release of flavor. The liquid flavoring agent is distributed over a large surface area of the provided mixture, which may include, for example, sugar alcohol and water, thereby facilitating effective contact with saliva, which also advantageously promotes flavoring agent release, while the powdered flavoring composition promotes flavoring agent release over a longer period of time, i.e., sustained flavoring agent release.
[0012] A powdered flavoring composition refers to a flavoring composition provided as a solid or contained in a solid matrix. A powdered flavoring composition can be obtained by adding a liquid flavoring to a solid matrix composition, such as a sugar alcohol, to obtain a solid powdered flavoring composition. A powdered flavoring material can be obtained, for example, by a spray drying method.
[0013] Therefore, according to the present invention, the liquid flavor composition is added to the powdered composition after the water, i.e., after the water is added to the powdered composition, the liquid flavor composition is added to the resulting mixture of the powdered composition and water.
[0014] In an advantageous embodiment of the invention, the pouch composition comprises the powdered flavour composition in an amount of at least 1% by weight of the pouch composition, such as at least 3% by weight of the pouch composition, such as at least 5% by weight of the pouch composition.

[0029] In an advantageous embodiment of the invention, the at least one sugar alcohol is chosen from xylitol, maltitol, mannitol, erythritol, isomalt, sorbitol, lactitol, and mixtures thereof.
[0030] In an advantageous embodiment of the invention, the at least one sugar alcohol is chosen from xylitol, maltitol, mannitol, erythritol, isomalt, lactitol, and mixtures thereof.
[0031] In one embodiment of the invention, the at least one sugar alcohol comprises xylitol and / or erythritol. In an advantageous embodiment of the invention, the pouched composition comprises at least two alcohols.
[0032] In one embodiment of the invention, the at least two sugar alcohols are selected from xylitol, maltitol, mannitol, erythritol and isomalt. [0034] In one embodiment of the invention, the pouch composition comprises at least one sugar alcohol in an amount of 5-40% by weight of the composition, such as 5-30% by weight of the composition. In an advantageous embodiment of the invention, the at least one sugar alcohol comprises a DC (directly compressible) grade sugar alcohol.
[0035] In an advantageous embodiment of the invention, at least 50% by weight of the at least one sugar alcohol is a DC (directly compressible) grade sugar alcohol. In one embodiment of the invention, the at least one sugar alcohol comprises a DC (directly compressible) grade sugar alcohol in an amount of 50-100% by weight of the at least one sugar alcohol, such as 60-80% by weight of the at least one sugar alcohol.
[0036] In one embodiment of the invention, the at least one sugar alcohol comprises a non-DC (non-directly compressible) grade sugar alcohol. In an advantageous embodiment of the invention, the powdered composition comprises water-insoluble fibers.
[0037] In an advantageous embodiment of the invention, the powdered composition comprises a sugar alcohol and a water-insoluble fiber. The advantage of using a combination of sugar alcohol, water and water insoluble fiber is that a very appealing soft, moist, moldable texture and mouthfeel is obtained. While the desired texture and mouthfeel is obtained, the produced pouches can still be stored adjacent to each other, for example in a can, without sticking too much to each other which would result in the pouch bursting when removed.
[0038] In an advantageous embodiment of the invention, the water-insoluble fibre is a vegetable fibre. In an advantageous embodiment of the invention, the water insoluble fibre is selected from wheat fibre, pea fibre, rice fibre, maize fibre, oat fibre, tomato fibre, barley fibre, rye fibre, sugar beet fibre, buckwheat fibre, potato fibre, cellulose fibre, apple fibre, cocoa fibre, cellulose fibre, powdered cellulose, bran fibre, bamboo fibre and any combination thereof.
[0039] Powdered cellulose within the scope of the present invention is understood to be cellulose prepared by processing α-cellulose obtained as a pulp from several types of fibrous plant material, such as wood pulp.
[0040] In an advantageous embodiment of the invention, the water-insoluble fibre comprises or consists of a water-insoluble fibre selected from wheat fibre, oat fibre, pea fibre, powdered cellulose, or combinations thereof.
[0041] An advantage of the above embodiment may be that it can promote a relatively efficient release of flavouring. When including a further substance to be released, such as nicotine, efficient release may be promoted. In one embodiment of the invention, the water-insoluble composition comprises or consists of a water-insoluble fiber selected from wheat fiber, oat fiber, pea fiber, or combinations thereof.
[0042] In one embodiment of the invention, the water-insoluble composition comprises or consists of a water-insoluble fiber selected from wheat fiber, oat fiber, or a combination thereof. Non-limiting examples of water insoluble fibers that may be used include Vitacel® WF 600, Vitacel® HF 600, Vitacel® P95, Vitacel® WF 200, Vitacel® L00, Vitacel® Erbsenfaser EF 150, Vitacel® bamboo fiberbaf 90, Vitacel® HF 600, Vitacel® Cellulose L700G, Vitacel® PF200, Vitacel® potatoofiber KF200, Vitacel® bamboo fiberhaf BAF40, Vitacel® Haferfaser / oat fiber HF-401-30 US.
[0043] Non-limiting examples of powdered cellulose that can be used include Vitacel® L00, Vitacel® Cellulose L700G, Vitacel® LC1000, Vitacel® L600-20, Vitacel® L600.
[0044] In one embodiment, the powdered cellulose is not chemically modified, and therefore the powdered cellulose can be a chemically unmodified cellulose fiber, e.g., free of microcrystalline cellulose (MCC).
[0045] In an advantageous embodiment of the invention, the water-insoluble fibres have a water binding capacity of at least 200%, such as at least 300%, such as at least 400%.
[0046] An advantage of the above embodiment is that the high water binding capacity allows for a pouch composition with a high moisture content. Additionally, it has been found that pouch compositions having a high moisture content provide the desired texture and mouthfeel while still allowing the produced pouches to be stored adjacent to one another, for example in a can, without sticking too much to one another which would result in the pouch bursting when removed.
[0047] In one embodiment of the present invention, the water-insoluble fiber has a water binding capacity of 200-1500%, such as 300-1300%, such as 200-800%, such as 300-800%, such as 400-600%.
[0048] In one embodiment of the present invention, the water-insoluble fiber has a water binding capacity of 200 to 1500%, such as 300 to 1300%, such as 300 to 900%, such as 300 to 700%, such as 400 to 700%.
[0049] In one embodiment of the present invention, the water-insoluble fiber has a water binding capacity of 200-1500%, such as 400-1500%, such as 500-1500%, such as 500-1200%, such as 500-1000%.
[0050] In one embodiment of the present invention, the water-insoluble fiber has a water binding capacity of 300-1500%, such as 400-1300%. In one embodiment of the invention, the water insoluble fibre has a density of 50-500 grams per litre, such as 100-400 grams per litre, such as 200-300 grams per litre.
[0051] In one embodiment of the present invention, the water-insoluble fiber has a swelling capacity of at least 5.0 mL / g, such as 5.0 to 20 mL / g. The advantage of the above embodiment is that the amount of water-insoluble fiber can be reduced without compromising the mouthfeel during use: when a certain amount of water-insoluble fiber is substituted for the water-soluble ingredient, the water-insoluble fiber swells during use and prevents the water-soluble ingredient from dissolving, so that the user will not experience a loss of pouch content during use.
[0052] In an advantageous embodiment of the invention, the water-insoluble composition comprises or consists of water-insoluble fibres in an amount between 5 and 50% by weight of the pouch composition and a water content between 15 and 70% by weight of said pouch composition.
[0053] In an advantageous embodiment of the invention, the pouch composition comprises water in an amount of at least 5% by weight of the composition, such as at least 8% by weight of the composition, such as at least 10% by weight of the composition, such as at least 20% by weight of the composition.
[0054] In an advantageous embodiment of the invention, the pouch composition comprises water in an amount of less than 65% by weight of the pouch composition, such as less than 60% by weight of the pouch composition, such as less than 50% by weight of the pouch composition, such as less than 40% by weight of said composition.
[0055] In an advantageous embodiment of the invention, the pouch composition comprises water in an amount of 8-60% by weight of the composition, such as 8-50% by weight of the composition, such as 8-40% by weight of the composition, such as 20-40% by weight of the composition.
[0056] In one embodiment of the invention, the pouch composition comprises water in an amount of 8-65% by weight of the composition, such as 10-65% by weight of the composition, such as 15-65% by weight of the composition, such as 20-65% by weight of the composition.
[0057] In one embodiment of the present invention, the pouch composition has a moisture content of 15-70% by weight of said pouch composition, such as 15-50% by weight of said pouch composition, such as 15-40% by weight of said pouch composition, such as 15-30% by weight of said pouch composition, such as 15-25% by weight of said pouch composition.
[0058] In one embodiment of the invention, the pouch composition comprises water in an amount of 8-60% by weight of the composition, such as 8-50% by weight of the composition, such as 8-40% by weight of the composition, such as 8-30% by weight of the composition.
[0059] In an advantageous embodiment of the invention, the pouch composition comprises water in an amount of 20-65% by weight of the composition, such as 20-60% by weight of the composition, such as 20-50% by weight of the composition, such as 20-40% by weight of the composition.
[0060] In an advantageous embodiment of the invention, the pouch composition comprises water and water insoluble fibre in a weight ratio of 3.0 or less, such as 2.5 or less, such as 2.0 or less, such as 1.5 or less, such as 1.0 or less.
[0061] In one embodiment, the pouch composition comprises water and water insoluble fiber in a weight ratio of 3.0 to 0.2, such as 2 to 0.2, such as 1.5 to 0.5. Thus, the above weight ratio has in the numerator the water content, in weight %, of the pouch composition, and in the denominator the water insoluble fiber content, in weight %, of the pouch composition.
[0062] Having a moisture content within the range of the present invention may promote fast release within an initial fast release period, such as within the first 120 seconds, such as within the first 5 minutes, since the pouch is already wetted or partially wetted with water from the start of use.
[0063] On the other hand, the moisture content should not be too high. Having too high moisture content may affect the liquid diffusion into and out of the pouch. A fully wetted pouch may have lower liquid diffusion into and out of the pouch during use, while a partially wetted pouch may have higher liquid diffusion into and out of the pouch. Therefore, a pouch with low liquid diffusion may have lower release, such as a low initial release of ingredients such as flavors or any active ingredients.
[0064] In one embodiment of the invention, the pouch composition has a moisture content of 60% or less by weight of the pouch composition, such as 50% or less by weight of the pouch composition, such as 40% or less by weight of the pouch composition, such as 30% or less by weight of the pouch composition.
[0065] In one embodiment of the present invention, the pouch composition comprises a total amount of flavor compositions of 10% or less by weight of the pouch composition, such as 8% or less by weight of the pouch composition, such as 5% or less by weight of the pouch composition, the total amount of flavor compositions being the sum of the liquid flavor composition and the powdered flavor composition.
[0066] In an advantageous embodiment of the invention, the pouch composition comprises a water insoluble fiber selected from pea fiber, powdered cellulose and combinations thereof, and a flavoring composition in an amount of up to 10% by weight of the pouch composition.
[0067] An advantage of the above embodiment may be that a relatively efficient release of the flavouring composition may be promoted by the inclusion of pea fibre and / or powdered cellulose. In one embodiment of the present invention, the pouch composition comprises a water insoluble fiber selected from pea fiber, powdered cellulose, and combinations thereof, and a total amount of a flavoring composition in an amount of 0.01-10% by weight of the pouch composition.
[0068] In one embodiment of the invention, the soluble fiber comprises or consists of cereal fiber. In one embodiment of the invention, the soluble fibre comprises or consists of fruit fibre and / or vegetable fibre.
[0069] In an advantageous embodiment of the invention, the pouch composition further comprises nicotine. The use of liquid flavourings according to the provisions of the present invention in combination with nicotine has proven to be very attractive from the user's point of view, for example with respect to liquid menthol or liquid mint flavourings, particularly in those embodiments in which the flavouring partially encapsulates or coats the nicotine / nicotine-containing particles.
[0070] In an advantageous embodiment of the invention, the nicotine is selected from the group consisting of nicotine salts, nicotine free base, nicotine bound to an ion exchanger such as an ion exchange resin such as nicotine polacrilex resin, nicotine inclusion complexes or any non-covalently bound nicotine, nicotine bound to zeolites, nicotine bound to cellulose such as microcrystalline cellulose, or starch microspheres, and mixtures thereof.
[0071] One example of a combination of different types of nicotine is a combination of free base nicotine mixed with polacrilex resin, where some nicotine binds to the ion exchange resin and some nicotine remains unbound.
[0072] Free base nicotine includes nicotine mixed with sugar alcohols, modified calcium carbonate, water soluble fiber, ion exchange resins, and combinations thereof. Nicotine bound to modified calcium carbonate is described in WO 2010 / 121619, which is incorporated herein by reference.
[0073] In an advantageous embodiment of the invention, the nicotine comprises non-salt nicotine. In an advantageous embodiment of the invention, the nicotine comprises free base nicotine.
[0074] A very important advantage of the above embodiment would be that it provides a long shelf life for the pouched product, providing a long life taste and texture. By providing nicotine in free base form, it is easier to achieve a high pH in the pouch composition without the need for significant alkaline pH control agents.
[0075] Therefore, in the above embodiments, the amount of alkaline pH control agent can be reduced without compromising shelf life and long life taste and texture. In an advantageous embodiment of the invention, the nicotine comprises nicotine mixed with an ion exchange resin.
[0076] In one embodiment of the invention, the nicotine comprises nicotine mixed with an ion exchange resin, such as a polacrilex resin, and the nicotine pouch composition further comprises nicotine bound to the ion exchange resin, i.e., a nicotine ion exchange resin complex. Thus, the nicotine can be nicotine mixed with a polacrilex resin, where some nicotine is bound to the ion exchange resin, but some nicotine remains unbound as free base nicotine.
[0077] In an advantageous embodiment of the invention, the nicotine comprises free base nicotine mixed with an ion exchange resin in a weight ratio between the free base nicotine and the ion exchange resin of from 0.1 to 2.0, preferably from 0.5 to 2.0, most preferably from about 0.67 to 1.0.
[0078] In one embodiment of the invention, the nicotine comprises free base nicotine mixed with ion exchange resin in a weight ratio between the free base nicotine and the ion exchange resin of from 1:1 to about 1:10, preferably from 1:2 to 1:6, and most preferably from about 1:4 to 1:5.
[0079] Here, the weight ratio means the ratio of the mass of the first component divided by the mass of the second component. The term mixing ratio may also be used. Thus, in the above embodiments, the nicotine comprises free base nicotine mixed with ion exchange resin in a weight ratio between the free base nicotine and the ion exchange resin of from 0.1 to about 1, preferably from 0.17 to 0.5, and most preferably from about 0.2 to 0.25.
[0080] In one embodiment, the pouch composition comprises water and water insoluble fiber in a weight ratio of 0.2 to 0.8. In an advantageous embodiment of the invention, the nicotine comprises a nicotine salt.
[0081] In one embodiment of the invention, the nicotine salt is selected from nicotine ascorbate, nicotine aspartate, nicotine benzoate, nicotine monotartrate, nicotine bitartrate, nicotine chloride (e.g., nicotine hydrochloride, nicotine dihydrochloride), nicotine citrate, nicotine fumarate, nicotine gensitate, nicotine lactate, nicotine mucate, nicotine laurate, nicotine levulinate, nicotine malate, nicotine perchlorate, nicotine pyruvate, nicotine salicylate, nicotine sorbate, nicotine succinate, nicotine zinc chloride, nicotine sulfate, nicotine tosylate, and hydrates thereof (e.g., nicotine zinc chloride monohydrate).
[0082] In one embodiment of the invention, the nicotine salt comprises or consists of nicotine bitartrate. In an advantageous embodiment of the invention, the nicotine comprises nicotine bound to an ion exchange resin.
[0083] In one embodiment of the invention, the ion exchange resin is a polacrilex resin. In one embodiment of the present invention, the polacrilex resin is Amberlite® IRP64.
[0084] In an advantageous embodiment of the invention, the nicotine comprises synthetic nicotine. In an advantageous embodiment of the invention, the nicotine comprises nicotine isolated from tobacco.
[0085] In an advantageous embodiment of the invention, the pouch composition comprises nicotine in an amount of at least 0.1% by weight of the pouch composition, such as at least 0.2% by weight. In an advantageous embodiment of the invention, the pouch composition comprises nicotine in an amount of 0.1 to 5.0% by weight of the pouch composition, such as 0.2 to 4.0% by weight of the pouch composition, such as 1.0 to 2.0% by weight of the pouch composition.
[0086] In one embodiment of the invention, the pouch composition is substantially homogeneous. The homogeneity of a pouch composition can be assessed by evaluating the distribution of the single components of the composition among the individual pouches.
[0087] For example, the standard deviation of the nicotine content, i.e., the uniformity of the nicotine content (CU), is related to the homogeneity of the pouch composition: pouches prepared from the same pouch composition with a low standard deviation of the nicotine content will have a high homogeneity of the pouch composition, whereas pouches prepared from the same pouch composition with a high standard deviation of the nicotine content will have a low homogeneity of the pouch composition.
[0088] In an advantageous embodiment of the invention, the nicotine content among a series of at least 10 oral pouches of said pouch composition maintains a relative standard deviation (RSD) of not more than 10%, preferably not more than 8%, more preferably not more than 6%, even more preferably not more than 4%, and most preferably not more than 2%.
[0089] In one embodiment of the present invention, the nicotine content among a series of at least 10 oral pouches comprising said pouch composition maintains a relative standard deviation (RSD) of 0.1-10%, preferably 0.1-8%, more preferably 0.1-6%, even more preferably 0.1-4%, and most preferably 0.1-2%.
[0090] By adding the water to the powdered composition followed by the liquid flavor composition, a more homogeneous pouch composition is obtained. In an advantageous embodiment of the invention, the pouch composition is free of tobacco, tobacco fibre and tobacco-derived fibre.
[0091] In some alternative embodiments, the pouch composition may include a trace amount of tobacco. For example, nicotine provided as part of a tobacco such as powdered tobacco is in addition to the free base nicotine. Such tobacco may be included, for example, to provide a tobacco flavor.
[0092] In one embodiment, the pouch composition may include tobacco, tobacco fiber, or tobacco-derived fiber in an amount of 0.1-5.0% by weight of the pouch composition, such as 0.1-3.0% by weight of the pouch composition. Thus, the pouch composition may, in some embodiments, include small amounts of tobacco, but this is in addition to the free base nicotine, and thus the pouch composition is not tobacco-based.
[0093] In one embodiment of the invention, the pouch composition comprises, for example, no tobacco, such as less than 0.1% by weight of the pouch composition, such as less than 0.5% by weight of the pouch composition, such as less than 1.0% by weight of the pouch composition, such as less than 3.0% by weight of the pouch composition, or less than 5.0% by weight of tobacco.
[0094] In one embodiment of the invention, the pouch composition does not include tobacco, tobacco fiber, or tobacco-derived fiber, and therefore in this embodiment, the water-insoluble fiber is a non-tobacco fiber, i.e., does not include tobacco, tobacco fiber, or tobacco-derived fiber.
[0095] In one embodiment of the invention, the pouch composition is free of microcrystalline cellulose (MCC), such as being cellulose-free. In one embodiment of the invention, the pouch composition comprises cellulose and does not comprise microcrystalline cellulose (MCC).
[0096] In an advantageous embodiment of the present invention, the liquid flavouring composition at least partially covers the surface of the powdered composition. In an advantageous embodiment of the present invention, the liquid flavouring composition is at least partially adsorbed onto the surface of the powdered composition.
[0097] In an advantageous embodiment of the invention, the pouch composition has a bulk density of at most 0.8 g / cm3, such as at most 0.7 g / cm3, such as at most 0.6 g / cm3, such as at most 0.5 g / cm3.
[0098] An advantage of the above described embodiment may be that it results in a low density composition. Surprisingly, the combination of water and sugar alcohol did not result in a very dense and compacted unworkable pouch composition, but rather allowed for a relatively light weight and low density composition.
[0099] In an advantageous embodiment of the invention, the pouch composition has a bulk density of 0.2 g / cm3 to 0.8 g / cm3, such as 0.3 g / cm3 to 0.7 g / cm3, such as 0.3 g / cm3 to 0.6 g / cm3, such as 0.4 to 0.5 g / cm3.
[0100] In one embodiment of the present invention, the composition has a bulk density of 0.2 g / cm3 to 0.8 g / cm3, such as 0.2 g / cm3 to 0.7 g / cm3, such as 0.2 g / cm3 to 0.6 g / cm3, such as 0.2 to 0.5 g / cm3.
[0101] In one embodiment of the present invention, the composition has a bulk density of 0.2 g / cm3 to 0.8 g / cm3, such as 0.3 g / cm3 to 0.8 g / cm3, such as 0.4 g / cm3 to 0.8 g / cm3, such as 0.5 to 0.8 g / cm3.
[0102] The density of the pouch composition can be influenced by many parameters, especially the type and amount of sugar alcohol, the type and amount of fiber, the water content, and the mixing time of the following process. For example, varying the amount of water also affects the mobility, such that too much water can lead to a dense and dense pouch formulation, but by selecting a fiber with a high water binding capacity, a higher water content can be at least partially offset. It has also been observed that excessive mixing can lead to a very dense and dense pouch composition.
[0103] In an advantageous embodiment of the invention, the pouch composition further comprises a pH controlling agent, such as a basic pH controlling agent, such as a basic buffering agent. In one embodiment of the invention, the pouch composition further comprises a combination of at least two pH control agents, such as a combination of at least two basic buffering agents, such as a basic buffer pair, such as a combination of at least two basic pH control agents.
[0104] An advantage of the above embodiment may be that more efficient nicotine uptake may be obtained, especially when a basic (alkaline) pH control agent is used. Another advantage of the above embodiment may be that a desirable mouthfeel may be obtained during use.
[0105] In embodiments, by avoiding the use of nicotine salts such as nicotine bitartrate, a lower amount of pH control agent can be applied, but it may still be desirable to further increase the pH by adding a pH control agent.
[0106] In an advantageous embodiment of the invention, the pouch composition comprises an alkaline buffer. As used herein, the term alkaline buffer is used interchangeably with basic buffer, i.e., alkaline is used in the sense of "basic" as opposed to acidic.
[0107] In an advantageous embodiment of the invention, the pouch composition comprises a pH controlling agent in an amount of less than 6% by weight of the pouch composition, such as less than 5% by weight of the pouch composition, such as less than 4% by weight of the pouch composition, such as less than 2% by weight of the pouch composition, such as less than 1% by weight of the pouch composition, such as no pH controlling agent.
[0108] In one embodiment of the present invention, the pouch composition comprises a pH control agent in an amount of 0-6% by weight of the pouch composition, such as 0-5% by weight of the pouch composition, such as 0-4% by weight of the pouch composition, such as 0-3% by weight of the pouch composition, such as 0-2% by weight of the pouch composition, such as 0-1% by weight of the pouch composition.
[0109] In one embodiment of the present invention, the pouch composition comprises a pH control agent in an amount of 0.1 to 6 wt% of the pouch composition, such as 0.1 to 5 wt% of the pouch composition, such as 0.5 to 5 wt% of the pouch composition, such as 0.5 to 4 wt% of the pouch composition, such as 0.1 to 3 wt% of the pouch composition, such as 0.1 to 2 wt% of the pouch composition, such as 0.1 to 1 wt% of the pouch composition.
[0110] In one embodiment of the present invention, the pouch composition includes a pH control agent, for example in an amount of 0.01 to 15% by weight of the pouch composition. In one embodiment of the present invention, the pouch composition comprises a pH control agent in an amount of 0.01 to 15% by weight of the pouch composition, such as 0.5 to 10% by weight of the pouch composition, such as 1 to 10% by weight of the pouch composition, such as 5 to 10% by weight of the pouch composition.
[0111] In an advantageous embodiment of the invention, the pouch composition is adapted to provide a pH of at least 8.0, such as a pH of at least 8.2, such as a pH of at least 8.5, such as a pH of at least 8.7, such as a pH of at least 9.0, when 2.0 grams of the pouch composition is added to 20 mL of 0.02 M potassium dihydrogen phosphate buffer (pH adjusted to 7.4).
[0112] An advantage of the above embodiment may be that relatively efficient nicotine uptake is promoted due to the high pH value obtained. A further advantage of the above embodiments may be that the need for preservatives may be reduced or even eliminated, and that such preservatives may be used in smaller amounts, if not absent.
[0113] The resulting high pH value may also advantageously impart a tingling sensation in the mouth that may be perceived as a desirable mouthfeel, for example, similar to tobacco-based pouch products.
[0114] In an advantageous embodiment of the invention, the pH control agent is selected from the group consisting of acetic acid, adipic acid, citric acid, fumaric acid, glucono-δ-lactone, gluconic acid, lactic acid, malic acid, maleic acid, tartaric acid, succinic acid, propionic acid, ascorbic acid, phosphoric acid, sodium orthophosphate, potassium orthophosphate, calcium orthophosphate, sodium diphosphate, potassium diphosphate, calcium diphosphate, pentasodium triphosphate, pentaposodium triphosphate, sodium polyphosphate, potassium polyphosphate, carbonic acid, sodium carbonate, sodium bicarbonate, potassium carbonate, calcium carbonate, magnesium carbonate, magnesium oxide, or any combination thereof.

[0116] In one embodiment of the invention, the pouch composition further comprises a humectant. The humectant may attract and retain water in the oral cavity during use, however, the humectant may also further moderate the release of an active ingredient, such as nicotine, for example to facilitate sustained release.
[0117] In one embodiment of the present invention, the pouch composition further comprises a water retention agent in an amount of 0.5-10% by weight of the pouch composition, such as 0.5-5% by weight of the pouch composition, such as 1-3% by weight of the pouch composition.

In one embodiment of the invention, the pouch composition further comprises a powdered flavoring composition comprising water insoluble fiber and at least one sugar alcohol, such as at least one sugar alcohol selected from xylitol, maltitol, mannitol, erythritol, isomalt, lactitol, and mixtures thereof, the pouch composition further comprises nicotine, and the liquid flavoring is at least partially adsorbed on the surface of the powdered composition.
[0137] In a second aspect, the present invention relates to an oral pouched product comprising a pouch, a powdered composition and a liquid flavouring composition, the liquid flavouring composition covering at least a portion of the surface of the powdered composition.
[0138] Thus, in one embodiment, the liquid flavourant is at least partially adsorbed onto the surface of the powdered composition. In one embodiment of the invention, an oral pouched product of the second aspect is provided according to the oral pouched product of the first aspect of the invention or any of its embodiments, provided that the limitations of claim 1 are not observed.
[0139] In one embodiment of the invention, the oral pouched product of the second aspect is provided according to the oral pouched product of the first aspect of the invention or any of its embodiments. In one embodiment of the invention, the pouch is a pouch membrane.
[0140] In one embodiment of the invention, the pouch is a saliva-permeable membrane. Typically, the pouch membrane includes an opening with characteristic opening dimensions adapted to characteristic dimensions of the pouch composition to retain the pouch composition inside the pouch prior to use and / or to retain a portion of the pouch composition, such as a water-insoluble composition, inside the pouch during use.
[0141] To obtain a pouch membrane with a suitable opening size in view of the pouch composition to be used, the materials of the pouch membrane, including, for example, woven and / or nonwoven fabrics, may be selected accordingly.
[0142] In other words, according to various embodiments, the pouch membrane allows the passage of saliva and prevents or blocks the passage of undissolved composition and water-insoluble fibers. The pouch membrane can be any suitable material, such as woven or nonwoven fabrics (e.g., cotton, fleece, etc.), heat-sealable nonwoven cellulose such as long fiber paper, or other polymeric materials such as synthetic, semi-synthetic, or natural polymeric materials. One example of a suitable material for the pouch membrane is paper made of pulp and a small amount of a wet enhancer. The material suitable for use must provide a semi-permeable membrane layer to prevent the powder or composition from exiting the bag or pouch during use. The suitable material is also one that does not have a significant effect on the release of nicotine from the pouch.
[0143] More specifically, in terms of material, the pouch membrane can be a natural, synthetic, semi-synthetic hydrophilic or hydrophobic membrane. It can be made of one or more biocompatible and physiologically acceptable polymeric materials. Examples of suitable materials for the pouch membrane are cellulose acetate and its derivatives, carboxymethylcellulose, polycellulose esters, other cellulose derivatives including ethylcellulose, propylcellulose, polyethylene, polypropylene, polystyrene, polyvinyl chloride, polyvinyl acetate, methacrylate and acrylate polymers, natural rubber, polycarbonate, polyethylene terephthalate, polyester, polyamide, and nylon. Other suitable materials have been mentioned earlier in this specification.
[0144] Rayon fibers, such as viscose rayon fibers (i.e., regenerated cellulose), may also be used, for example, in combination with an acrylic polymer to act as a binder in the nonwoven material and provide a heat seal for the pouch membrane during its manufacture. Other binders, such as one or more copolymers of vinyl acetate and acrylic acid esters, may also be used.
[0145] Suitable pouch membranes are available under the trade names "Taboka", CatchDry, Ettan, General, Granit, Goteborgs Rape, GrovSnus White, Metropol Kaktus, Mocha Anis, Mocha Mint, Mocha Wintergreen, Kicks, Probe, Prince, Skruf, TreAnkrare, Camel Snus Original, Camel Snus Frost, and Camel Snus Spice. The pouch membrane provides a liquid permeable container of a type that may be considered similar in properties to the mesh-type materials used in the construction of tea bags The desired component of the nicotine composition to be released diffuses through the pouch membrane and into the user's mouth.
[0146] The pouch membrane material may have the form of a net, a sieve, a paper with holes, a permeable cloth, and the like. For example, a pouch material made from rice paper in a net form, or straw paper with holes, may dissolve in the mouth of the user. In some exemplary embodiments, the pouch membrane material may be made using water-dispersible film-forming materials (e.g., binders such as alginate, carboxymethylcellulose, xanthan gum, pullulan, and the like), as well as those materials in combination with materials such as ground cellulose derivatives (e.g., fine grain wood pulp). The preferred pouch material, while water-dispersible or soluble, may be designed and manufactured such that a significant amount of the nicotine content will permeate the pouch material under normal use conditions before the time at which the pouch loses its physical integrity. If desired, flavoring ingredients, disintegration aids, and other desired ingredients may be incorporated into or applied to the pouch material.
[0147] Examples of various types of pouch membrane materials are described in U.S. Patent No. 5,167,244 to Kjerstad. Fleece materials for use as pouch membranes are described, for example, in WO 2008 / 152469, GB 673,587, and EP 2 692 254.
[0148] In one embodiment of the invention, the membrane comprises water-insoluble fiber of a different origin than the water-insoluble fiber contained in the pouched product. [0161] As used herein, the term "pouch composition" refers to a pouched product, i.e. a composition for use in a pouch for oral use.
[0162] As used herein, the term "pouch" is intended to mean a container typically formed by a web of fibrous material surrounding a cavity. The pouch may be designed for administering a compound such as nicotine into the oral cavity. In any case, it is suitable for use in the oral cavity and is non-toxic and non-water soluble. The fibrous material may form, for example, a woven or non-woven web or fabric. The pouch may be formed, for example, by sealing two corresponding parts of a web or fabric together along their edges to form a cavity for the pouch composition. To release the flavorant (i.e., the flavorant compound and any further ingredients to be released), the pouch is water permeable so that saliva from the oral cavity penetrates the pouch and enters the cavity, where it comes into contact with the flavorant composition, thereby releasing the flavorant from the oral pouch.
[0163] Other active ingredients to be applied in the pouch composition within the scope of the present invention include, but are not limited to, the group consisting of phytochemicals such as resveratrol and anthocyanins; herbs such as green tea and thyme; antioxidants such as polyphenols; micronutrients; mouth moisteners such as acids; throat soothing ingredients; appetite suppressants; breath fresheners such as zinc compounds or copper compounds; dietary supplements; cold preventatives; cough suppressants; vitamins such as vitamin A, vitamin C or vitamin E; minerals such as chromium; metal ions; alkaline materials such as carbonates; salts; dental treatments such as herbs, remineralizing agents, antibacterial agents, anti-caries agents, plaque acid buffers, teeth whitening agents, stain removers or desensitizing agents; and combinations thereof.
[0164] In one embodiment of the invention, the active ingredient is selected from the group consisting of a dipeptide, a tripeptide, an oligopeptide, a decapeptide, a decapeptide KSL, a decapeptide KSL-W, an amino acid, a protein, or any combination thereof.
[0165] In one embodiment of the invention, the active ingredient comprises probiotic bacteria such as Lactobacillus, Bifidobacterium, Lactococcus, Streptococcus, Leuconostoccus, Pediococcus or Enterococcus.
[0166] As used herein, the term "powder composition" refers to a composition in the form of a powder, i.e. as particulate matter having a relatively small particle size, for example between 1 and 1200 micrometers. In particular, powder composition does not refer to powdered tobacco.
[0167] As used herein, the term "flavor" is understood to have its ordinary meaning within the art. Flavor is provided by flavor compounds and may be included in liquid or powdered flavor compositions. Thus, flavor does not include, by definition, sweeteners (such as sugars, sugar alcohols, and high-intensity sweeteners), acids that impart pure acidity / sourness, or compounds that impart pure saltiness (e.g., NaCl) or pure bitterness. Flavor enhancers include substances that impart only saltiness, bitterness, or sourness. When flavor enhancers are included in the pouch composition, they can be added as part of the flavor composition or separately. Thus, flavor enhancers include, for example, NaCl, citric acid, ammonium chloride, and the like.
[0168] As used herein, the term "powdered flavor composition" refers to a flavor composition in powder form. It includes flavors that are solid in nature, or flavors immobilized in or contained within a powdered carrier, such as a water-soluble or water-insoluble carrier. Thus, a powdered flavor composition may include a solid carrier and / or a flavor enhancer, but always includes at least one flavor compound. Powdered flavor compositions include, for example, encapsulated flavors. Some powdered flavor compositions include several flavor ingredients to obtain a desired flavor profile, while other powdered flavor compositions include only a single flavor ingredient.
[0169] As used herein, the term "liquid flavor composition" refers to a flavor composition in liquid form. It includes flavors that are essentially in liquid form and flavor ingredients that are liquid because of the liquid carrier. As such, liquid flavor compositions do not include encapsulated flavors. Liquid flavor compositions may include liquid carriers and / or flavor enhancers, but always include at least one flavor compound. Some liquid flavor compositions include several flavor ingredients to obtain a desired flavor profile, while other liquid flavor compositions include only a single flavor ingredient.
[0170] As used herein, the term "humectant" is understood as a moisturizing agent used to attract moisture or water in the form of saliva. Humectants typically include compositions that are suitably hygroscopic. In some cases, humectants may also be referred to as moistening agents, since they act to attract moisture. Examples of humectants include glycerol, propylene glycol, alginate, modified starch, hydroxypropyl cellulose, triacetin, polyethylene glycol (PEG), pectin, xanthan gum, and the like.
[0171] As used herein, the term "water soluble" refers to a relatively high water solubility, e.g., a water solubility of a water soluble composition or substance greater than 5 grams per 100 mL of water, measured at 25 degrees Celsius and a pH of 7.0. When referring to a "soluble" composition or substance, water soluble is meant unless otherwise specified.
[0172] As used herein, the term "water-insoluble" refers to a relatively low water solubility, for example, a water soluble composition or substance having a water solubility of less than 0.1 grams per 100 mL of water, measured at 25 degrees Celsius and a pH of 7.0. When referring to "insoluble", water insolubility is meant unless otherwise specified.
[0173] As used herein, the term "nicotine" refers to nicotine used as a purified / isolated substance. In particular, nicotine does not refer to tobacco materials that contain nicotine.
[0174] As used herein, the term "free base nicotine" refers to nicotine in unprotonated form, and therefore does not include nicotine salt and nicotine provided as a complex of nicotine and ion exchange resin. However, free base nicotine can be mixed with an amount of ion exchange resin or water-soluble composition such as sugar alcohol or water-soluble fiber.Free base nicotine includes both free base nicotine extracted from tobacco and synthetically produced free base nicotine, but free base nicotine is not provided in the form of tobacco or powdered tobacco.Typically, free base nicotine is provided as a liquid.
[0175] As used herein, the term "initial" can refer to the first few minutes of use, such as the first 5 minutes of use. Generally, the pouch has an opening whose characteristic opening dimensions match the characteristic dimensions of the pouch composition to retain the pouch composition within the pouch prior to use and / or to retain a portion of the pouch composition, such as a water-insoluble composition, within the pouch during use.
[0176] The materials of the pouch membrane, including, for example, woven and / or nonwoven fabrics, may be appropriately selected to obtain a pouch membrane with a suitable opening size in view of the pouch composition to be used. In other words, according to various embodiments, the pouch membrane allows the passage of saliva and prevents or blocks the passage of undissolved pouch composition. The pouch membrane can be any suitable material, such as woven or nonwoven fabric (e.g., cotton, fleece, etc.), heat-sealable nonwoven cellulose, or other polymeric materials, such as synthetic, semi-synthetic, or natural polymeric materials. One example of a suitable material for the pouch is paper made of pulp and a small amount of wet enhancer. The material suitable for use must provide a semi-permeable membrane layer to prevent the powder or composition from exiting the bag or pouch during use. The suitable material is also one that does not have a significant effect on the release of flavor from the pouch.
[0177] The pouch composition is filled into the pouch and maintained within the pouch by sealing. An ideal pouch would be chemically and physically stable, pharma- ceutically acceptable, insoluble in water, capable of being filled with powder and sealed, providing a semipermeable membrane layer to prevent the powder from exiting the bag, but allowing saliva and ingredients from the pouch composition within the pouch, such as flavorings, dissolved therein or suspended in small enough amounts, to pass through the pouch.
[0178] The pouch can be placed in the mouth by a user, after which saliva enters the pouch and flavorings and other ingredients that are soluble in the saliva begin to dissolve and are carried along with the saliva out of the pouch and into the mouth where the nicotine can be absorbed.
[0179] According to one embodiment of the present invention, the pouch composition includes one or more pH controlling agents, such as a buffering agent. In one embodiment of the invention, the pH control agent is selected from the group consisting of acetic acid, adipic acid, citric acid, fumaric acid, glucono-δ-lactone, gluconic acid, lactic acid, malic acid, maleic acid, tartaric acid, succinic acid, propionic acid, ascorbic acid, phosphoric acid, sodium orthophosphate, potassium orthophosphate, calcium orthophosphate, sodium diphosphate, potassium diphosphate, calcium diphosphate, pentasodium triphosphate, pentaposodium triphosphate, sodium polyphosphate, potassium polyphosphate, carbonic acid, sodium carbonate, sodium bicarbonate, potassium carbonate, calcium carbonate, magnesium carbonate, magnesium oxide, or any combination thereof.
[0180] According to various embodiments of the present invention, one or more sugar alcohols may be included in the pouch as part of the pouch composition, for example, as a carrier or part thereof, as a humectant, or as a sweetener.Suitable sugar alcohols include sugar alcohols selected from the group of sorbitol, erythritol, xylitol, lactitol, maltitol, mannitol, hydrogenated starch hydrolysates, isomalt, or any combination thereof.

[0182] In one embodiment of the invention, the pouch composition comprises sugar and / or non-sugar sweeteners, such as sugar alcohols. In one embodiment of the present invention, the pouch composition comprises sugar and / or sugarless sweeteners in an amount of 1.0 to about 80% by weight of the pouch composition, more typically comprising 5 to about 70% by weight of the pouch composition, and more commonly comprising 10 to 30% by weight of the pouch composition or 5 to 25% by weight of the pouch composition. In some other embodiments, the sugar and / or sugarless sweeteners comprise 10 to 60% by weight of the pouch composition, or 10 to 50% by weight of the pouch composition. The sugar and / or sugarless sweeteners may function not only as sweeteners but also as humectants. In some embodiments, the inclusion of certain ingredients can further limit the about amounts of sugar and / or sugarless sweeteners. In some embodiments, the content of sugar and / or non-sugar sweeteners in the pouch composition is 20% or less by weight of the pouch composition, such as 15% or less by weight of the pouch composition, such as 10% or less by weight of the pouch composition, such as 5% or less by weight of the pouch composition.
[0183] Sweeteners can often support the flavor profile of a pouch composition. Sugar sweeteners generally include, but are not limited to, sugar-containing ingredients commonly known in the pouch art, such as sucrose, dextrose, maltose, saccharose, lactose, sorbose, dextrin, trehalose, D-tagatose, dry invert sugar, fructose, levulose, galactose, corn syrup solids, glucose syrup, hydrogenated glucose syrup, etc., either alone or in combination. These sugar sweeteners may also be included as humectants.

[0390] Pouches PPC101 to PPC101 indicate pouches with different water contents and water-insoluble fiber contents. Example 4 L-Content uniformity measurement Content uniformity (CU) of pouch samples was determined by analysis of 10 replicate subsamples. Nicotine content was measured for each subsample of approximately 500 mg using standard HPLC techniques. Nicotine content of the subsamples was expressed as a percentage of the nominal nicotine content in the sample (i.e., % label claim). For example, if a pouch sample with a nominal nicotine content of 20 mg / g was analyzed to find an actual content of 19 mg / g, the label claim nicotine content would be 95%.
[0391] The content uniformity of the samples is then determined as the relative standard deviation (RSD) of the individual analyses of relative nicotine content in the 10 replicates. Homogeneity was evaluated on two batches of PPC46 produced in different ways: PPC46 was produced according to claim 1, i.e. the liquid flavouring was added after the water, but PPC46 * was similarly prepared by adding a liquid flavoring composition to a powdered composition, and then adding water. * When preparing the liquid flavor composition, the liquid flavor composition is added before the water is added.
[0392] PPC46 * It was visually apparent that PPC46 was significantly less homogeneous than PPC46. This was confirmed by nicotine CU analysis, as can be observed from Table 19 below. Furthermore, PPC46 * The powder composition contained a variety of sized agglomerates and was visually confirmed to be unevenly distributed. The relative standard deviation was measured and the PPC46 * was found to be less homogeneous than PPC46.
[0393] Both pouch compositions were sieved through a 1400 micron sieve and the RSD of nicotine was then measured again. * was found to be less uniform than PPC46.
[0394] [Table 24]
[0395] Example 5 - Evaluation The manufactured pouches of the present invention were evaluated and found to have a very appealing flavor release. This was observed in pouches with and without nicotine.
[0396] Pouched products PPC1 and PNN1 were compared with comparative pouches P1 and P2 (Comp. P1 and P2) with regard to the perception of flavour release. Evaluation of perceived flavor release is performed as described below.
[0397] Perceived flavor release was evaluated by a test panel of four trained judges, each judge evaluating all samples in duplicate. An average rating was estimated. Pouched products PNN1 and PPC1 were rated by all four judges as having a significantly faster onset of flavor release and a more pronounced initial burst of flavor compared to comparisons P1 and P2.
[0398] Pouch product PPC1 was evaluated for the perceived effects of nicotine and for the burning sensation. The perceived effects of nicotine and the burning sensation were assessed as follows:
[0399] The cognitive effects of nicotine and the burning sensation were evaluated by a test panel of four trained judges. Each judge evaluated all samples in duplicate. The average rating was estimated.
[0400] Pouch product PPC1 was rated by all four reviewers as having a fast onset of action and a high nicotine cognitive effect, and pouch product PPC1 was rated by all four reviewers as having a high burning sensation.
[0401] Similarly, the pouch product PPC1 was evaluated for nicotine-induced cognitive effects in the same manner as above. The pouch product PPC1 was rated as having a high nicotine-induced cognitive effect by all four judges.
[0402] Additionally, a burst of flavor was rated as desirable in combination with a burning (tingling) sensation. Example 6 - User evaluation Pouches PNN30 and Comparative P1 were prepared according to Example 2C. Comparative P1 (COMP.P1) was prepared with 8% by weight blackcurrant powdered flavor and no liquid flavor. PNN30 was prepared with 5% by weight blackcurrant powdered flavor and 1.5% by weight blackcurrant liquid flavor.
[0403] The amounts of liquid and optionally powdered flavor compositions were adjusted taking into consideration the potency of the flavor applied, the concentration / loading of the flavor compounds, and the desired flavor intensity. Pouches PPC28 and comparative pouch P2 (COMP.P2) were prepared according to Example 4A.
[0404] The pouches were evaluated by a test panel of four trained judges, with each judge evaluating all samples in duplicate, over a 30 minute period. The test panel evaluated the pouches on four different parameters: moisture sensation, flavor intensity, sweetness intensity, and overall taste.
[0405] [Table 25]
[0406] The test panel found that the pouches containing liquid flavor exhibited higher scores in all parameters during the initial use time, i.e., the first 5 minutes, compared to the pouches not containing liquid flavor.
[0407] PNN30 improved moisturizing sensation during initial use. PNN30 exhibited higher flavor intensity and sweetness intensity both at the initial use time and during the use period, indicating that the pouch promotes not only an initial rapid release of flavor and sweetener, but also a sustained release during use.
[0408] Similarly, overall taste intensity was found to be superior for PNN30 both initially and over the course of use. The above test was repeated with pouch PPC28 and comparative P2 pouch, confirming that the inclusion of liquid flavor resulted in higher flavor intensity.
