AI Technical Summary
Existing pharmaceutical tablets containing axitinib exhibit rapid elution of the active ingredient, leading to undesirable side effects due to variations in solubility caused by differences in crystal polymorphism, necessitating a solution to control and manage dissolution properties.
Incorporation of polyvinyl acetate into the pharmaceutical tablet formulation, along with other excipients, to suppress the elution of axitinib, particularly in acidic conditions, by adjusting the dissolution rate through the use of water-soluble polymers, poorly water-soluble excipients, and disintegrants.
The formulation effectively controls the dissolution of axitinib, ensuring stable pharmacokinetics by suppressing initial elution at pH 1.2 and maintaining desired elution properties, as confirmed by dissolution tests.

Abstract
Description
[Technical field]
[0001] The present invention relates to a pharmaceutical tablet that contains axitinib as an active ingredient and can inhibit the dissolution of axitinib. [Background technology]
[0002] Axitinib is a compound having the structure represented by formula (1) and the chemical name N-methyl-2-({3-[(1E)-2-(pyridin-2-yl)ethen-1-yl]-1H-indazol-6-yl}sulfanyl)benzamide.
[0003] [ka]
[0004] Axitinib is a selective kinase inhibitor targeting vascular endothelial growth factor receptors (VEGFR-1, -2 and -3). VEGFR is a major receptor tyrosine kinase that regulates angiogenesis and lymphangiogenesis and is involved in tumor growth and metastasis. Axitinib is available under the trade name Inlyta® tablets. It is known that axitinib has multiple types of crystalline polymorphism. Patent Document 1 discloses axitinib polymorphs I, II, III, IV, VI, VII and VIII. Patent Document 2 also describes polymorphism XLI, crystalline polymorphisms including multiple types of solvates, and amorphous forms. Different crystalline polymorphs have different physical properties such as water solubility, and therefore different dissolution and absorption properties in pharmaceuticals. Among these, existing axitinib formulations contain an anhydrous XLI crystalline polymorph as an active ingredient and are provided as film-coated pharmaceutical tablets. Non-Patent Document 1 describes a pharmaceutical tablet obtained by dry granulating a mixture of axitinib (XLI), lactose hydrate, crystalline cellulose and croscarmellose sodium, adding magnesium stearate to the mixture and tableting the mixture, and film-coating the pharmaceutical tablet using a coating agent containing titanium oxide (Opadry (registered trademark) Red). [0007] In pharmaceutical tablets, rapid dissolution of the active ingredient causes side effects, so it is desirable that the active ingredient be appropriately controlled and managed. The present inventors have found that the solubility varies greatly depending on the crystal polymorphism of the axitinib drug substance, and that the dissolution varies greatly when pharmaceutical tablets are prepared using different crystal polymorphic drug substances. The object of the present invention is to provide a pharmaceutical tablet containing axitinib as an active ingredient, which can suppress dissolution. In particular, the object of the present invention is to provide a pharmaceutical tablet in which the dissolution is suppressed and controlled regardless of the crystal polymorphism of the axitinib drug substance. [Means for solving the problem]

In the second step, the composition is compressed to prepare a granulated product. The method for preparing the granulated product is a procedure in which the composition is compressed to granulate, the granules are sized, and then, if necessary, the granulated product is obtained by appropriately screening the granulated product. In the third step, the granulated material is molded into a pharmaceutical tablet, i.e., the composition is mixed with any other additives and molded into a pharmaceutical tablet. The pharmaceutical tablet can be prepared by molding using rotary tableting, single shot tableting, etc. The pharmaceutical tablet can be prepared in any form such as a plain tablet, a film-coated tablet, a dispersible tablet, an orally disintegrating tablet, a chewable tablet, or an effervescent tablet, etc.

The dry granules were mixed to a ratio of 173.1 mg to 0.9 mg croscarmellose sodium. The mixture was mixed to a ratio of 174 mg to 1.8 mg magnesium stearate, and tableted to prepare the tablet of Example 11.
[0044] [Comparative Example 1] The contents per tablet were 5 mg axitinib (IV type), 83 mg lactose hydrate, 83 mg crystalline cellulose, and 9.1 mg croscarmellose sodium, which were mixed and dry granulated, and the resulting granules were sized. The dry granules were mixed and compressed to a ratio of 180.1 mg to 1.8 mg magnesium stearate, to prepare the tablet according to Comparative Example 1.
[0045] [Test Example 1] The dissolution rates were evaluated by Japanese Pharmacopoeia Dissolution Test Method 2 (paddle method) using the tablets of Examples 1 to 11 and Comparative Example 1, as well as Inlyta (registered trademark) tablets, an existing axitinib formulation, and a test solution of pH 1.2 prepared by the method described in the Japanese Pharmacopoeia. The dissolution rate was evaluated using a dissolution tester (NTR-6600A, Toyama Sangyo Co., Ltd.) and an ultraviolet-visible spectrometer (UV-1900, Shimadzu Corporation) with a test liquid volume of 900 mL, a test liquid temperature of 37±0.5°C, and a paddle rotation speed of 50 rpm. The dissolution rate at 15 minutes after the start of the test was used to judge similarity, and a dissolution rate within ±15% of the Inlyta (registered trademark) tablet was judged to be similar. The axitinib dissolution rates of each tablet sample are summarized in Table 1.
[0046] [Table 1]
[0047] The pharmaceutical tablet of the present invention had a dissolution rate of 75% or less at the initial dissolution time of 5 minutes, 90% or less at 10 minutes, and 90% or less at 15 minutes, and was able to suppressively control the dissolution of axitinib. In addition, the pharmaceutical tablet of the present invention was judged to be similar to Inlyta (registered trademark) tablets at 15 minutes after the start of the test. On the other hand, Comparative Example 1 was unable to suppressively control the initial dissolution, and showed a dissolution rate of 90% or more at 5 minutes and 95% or more at 10 minutes. That is, the present invention can provide an axitinib tablet in which the initial dissolution rate is particularly suppressively controlled, and even when a crystalline polymorphic drug substance different from that of the existing formulation, Inlyta (registered trademark) tablets, is used, it is possible to provide a pharmaceutical tablet that shows a dissolution behavior similar to that of the existing formulation.
