Abstract

Methods and assays for determining the activation level of the plasma kallikrein (pKal) system and their use to assess the activity of pKal modulators on the pKal system are provided. [Solution] The present invention features an ex vivo activation method, which includes (i) incubating a plasma sample obtained from a subject with an activator of the plasma kallikrein (pKal) system (e.g., factor XIIa (FXIIa)), (ii) measuring the concentration of intact high molecular weight kininogen (HMWK), cleaved HMWK, or both, in the plasma sample before and after incubation, and (iii) determining the reduction of intact HMWK in the sample after activation.
