BISPECIFIC ANTIBODY AGAINST IL-6R AND TL1A AND USE THEREOFFIELD OF THE INVENTION

[0001] The present disclosure relates to a bispecific antibody against IL-6R and TL1A and use thereof.BACKGROUND OF THE INVENTION

[0002] Interleukin-6 (IL-6) is a multifunctional cytokine discovered in the 1980s and is produced by various types of cells such as T-cells, B-cells, monocytes, fibroblasts, osteoblasts, keratin-forming cells, endothelial cells, mesangial cells and some tumor cells. IL-6 transmits different biological signals to various tissues and cells by interacting with different IL-6 receptors on the cell surface and then via downstream signaling pathways.

[0003] Interleukin-6 (IL-6) binds to two receptors, one is the specific receptor IL-6R (80 kDa type I transmembrane protein) and the other is gp130, which is a common receptor subunit for members of the IL-6 family of cytokines. The gp130 can be expressed in all cells, but the expression of interleukin-6 receptor (IL-6R) is more restricted and is found mainly in hepatocytes, neutrophils, monocytes and CD4+ T cells. IL-6 plays an important role in immune regulation, hematopoiesis, inflammation and tumor formation. IL-6 is involved in the treatment of a disease or condition and has been the target of drug discovery for the treatment of that disease or condition.

[0004] TL1A, also known as TNFSF15 and VEGI, is a member of the tumor necrosis factor superfamily. It was first identified in 2002 and is expressed in a variety of cells. TL1A regulates different immune cells to produce inflammatory cytokines and chemokines. TL1A activates the TRADD pathway by binding to the receptor DR3. TRADD can promote PI3K, MAPKs, NF-κB, and other pathways by regulating TRAF2 and RIP1, thereby exerting pro-inflammatory effects. It also participates in apoptosis and necrotic cell death through activating the FADD, RIP3, and Caspase-8 / 3 / 7 pathways.

[0005] TL1A is associated with a variety of autoimmune diseases, including rheumatoid arthritis (RA) , inflammatory bowel disease (IBD) , psoriasis (PSA) , primary biliary cirrhosis (PBC) , systemic lupus erythematosus (SLE) , and ankylosing spondylitis (AS) , and TL1A shows abnormal expression in all these diseases.

[0006] Diseases such as ulcerative colitis and Crohn's disease are very prevalent and the number of available therapies is limited, and there are some patients who do not respond to initial treatment or whose response disappears over time. Currently, the only therapy for patients who do not respond to first-line treatment is surgery, that is stricturoplasty (shaping of the bowel) or resection (removal of the bowel) . Surgical treatment of IBD is invasive, and some patients who undergo surgery experience postoperative risks such as anastomotic fistula, infection, and bleeding. The pathogenesis of IBD is thought to involve an uncontrolled immune response that may be triggered by certain environmental factors in genetically susceptible hosts. The heterogeneity of disease pathogenesis and clinical course, combined with the different responses to treatment and its associated side effects, suggests that a targeted therapeutic approach to treating these diseases is an ideal therapeutic strategy. Therefore, novel therapeutic approaches that specifically target the pathogenesis of IBD, drugs that inhibit TL1A activity, have become popular targets for the development of IBD therapies. There exists a huge clinical demand for IBD with broad market potential.

[0008] In order to address one of the technical problems existing in the prior art, the present disclosure provides a bispecific antibody, which can better maintain the activity of each individual monoclonal antibody, and can simultaneously and specifically bind to both interleukin-6 receptor (IL-6R) and TNF-like ligand 1a (TL1A) targets. When binding to two different cell surface antigens, it potentially increases binding specificity and reduces side effects caused by off-target effects. Compared to combination therapy with monoclonal antibodies, it can effectively reduce the cost of treatment.
