Method for improving expression of immunomodulatory factor in mesenchymal stem cells and use thereof in mesenchymal stem cell culture

By treating mesenchymal stem cells with a combination of interferon-γ and budesonide, and possibly with the addition of valproic acid, the problems of unstable efficacy and cell damage in existing culture methods have been solved. This approach enhances the immunomodulatory capacity and cell yield of mesenchymal stem cells, resulting in higher expression of immunomodulatory factors and lower risk of damage.

AI Technical Summary

Technical Problem

Existing mesenchymal stem cell culture methods suffer from problems such as residual xenogeneic components, unstable efficacy, low cell yield, complex operation, and high risk of cell damage, making it difficult to improve their immunomodulatory capacity and clinical efficacy.

Method used

Using a combination of interferon-γ and budesonide, possibly with the addition of valproic acid, to treat mesenchymal stem cells to enhance the expression of their immunomodulatory factors, and using a mesh-structured carrier during culture to avoid the use of collagenase to harvest cells.

🎯Benefits of technology

It significantly enhanced the expression of immunomodulatory factors in mesenchymal stem cells, especially TNFR1, TSG6 and PGE2, thereby improving cell survival and immunomodulatory capacity and reducing the risk of cell damage.

Abstract

The present invention provides a method for improving the expression of an immunomodulatory factor in mesenchymal stem cells, comprising contacting the mesenchymal stem cells with a composition comprising interferon-γ and budesonide, wherein the immunomodulatory factor is at least one of the following: tumor necrosis factor receptor 1, tumor necrosis factor-stimulated gene 6, and prostaglandin E2. Also provided are a method for culturing a mesenchymal stem cell highly expressing an immunomodulatory factor and a cell obtained by the method, wherein the method comprises the following steps: (1) culturing mesenchymal stem cells in a first culture medium; (2) contacting the mesenchymal stem cells provided in step (1) with interferon-γ and budesonide for 18-36 hours; and (3) collecting the cells from step (2) using a cell detachment enzyme.
