Abstract

Provided are a capture oligonucleotide, composition or kit for nucleic acid amplification, and a method. Specifically, the capture oligonucleotide comprises, from the 5' end to the 3' end, a universal sequence (3s) and a binding capture sequence (1s); wherein (1), the binding capture sequence (1s) is at least partially identical to the 5' end sequence of a target molecule or the binding capture sequence (1s) is at least partially identical to the 3' end sequence of a specific modification site on the target molecule; (2), the capture oligonucleotide further comprises a nucleic acid extension-blocking modification located at the 3' end of the binding capture sequence (1s); and optionally (3), the 3' end of the binding capture sequence (1s) has a nucleic acid analogue modification. Also provided are a composition or kit used for nucleic acid amplification detection, a detection system, and a method. The product and method can achieve nucleic acid detection with good specificity and high sensitivity.
