Abstract

A method for improving the signal-to-noise ratio in a DNA methylation partition assay is provided. [Solution] The present disclosure provides a method for determining the methylation status, comprising the steps of: providing a biological sample of nucleic acid molecules; distributing at least one subset of nucleic acid molecules in the biological sample into a plurality of distribution sets based on the methylation status of the nucleic acid molecules; digesting at least one subset of one or more of the plurality of distribution sets with at least one methylation-sensitive restriction enzyme; enriching at least one subset of nucleic acid molecules in the plurality of distribution sets with respect to a genomic region of interest, wherein at least one subset of nucleic acid molecules includes nucleic acid molecules digested in one or more distribution sets; and determining the methylation status of nucleic acid molecules in at least one of the distribution sets at one or more loci.
