Description
Technical Field
[0001] The present invention relates to novel compositions for cosmetic or dermatological treatment of the skin. Background Art
[0002] Many compounds of animal or plant origin have been shown to have beneficial properties for humans or animals.
[0003] For example, Jean Soon Park et al., Astaxanthin Reduces Oxidative Stress and Inflammation and Enhances Immune Response in Humans, Nutr Metab (Lond), 2010, 7:18, revealed that dietary intake of astaxanthin increased both humoral and cell-mediated immune responses in young healthy women.
[0004] The literature by PP Ward et al., Multifunctional Roles of Lactoferrin: A Critical Review, Cell. Mol. Life Sci., 2005, Vol. 62, pp. 2540-2548, describes the different functions of lactoferrin, such as iron capture in the intestine, antibacterial effects, anti-inflammatory functions, anticancer effects, and a role in bone morphogenesis.
[0005] The literature by Didier Levieux et al., Le colostrum, a milk component particularly rich in various components: is it possible to detect their presence in milk transport?, Lait, 1999, vol. 75, pp. 465-488 (Colostrum, a milk component particularly rich in various components: is it possible to detect their presence in milk transport?), describes the composition of bovine colostrum, and the literature by Sylvie F. Gauthier et al., Growth factors in bovine milk and colostrum: composition, extraction and biological activity, Lait, 2006, vol. 86, pp. 99-125, lists the growth factors contained in colostrum and milk and describes their properties.
[0006] However, the beneficial properties of these ingredients when ingested as part of a meal are generally known.

[0042] Now, the present invention is described in more detail in the following description in a non-limiting manner. In the present specification, unless otherwise expressly stated, all indicated percentages (%) are weight percentages.
[0043] In the present application, the term "iron-binding glycoprotein" is to be understood as "one or more iron-binding glycoproteins", and "casein hydrolysate" is to be understood as "one or more casein hydrolysates". The same applies to the other compounds in the composition.
[0044] Composition
[0045] The present invention relates to a composition comprising an iron-binding glycoprotein (compound A) and a casein hydrolysate (compound B).
[0046] Preferably, the iron-binding glycoprotein is lactoferrin. Lactoferrin is a protein contained in whey (e.g., human milk or cow's milk). Lactoferrin is a multifunctional glycoprotein that has the effect of protecting cells from intrinsic attack. It affects the proliferation, maturation and activation of a variety of immune cells, and plays a role in the regulation, maturation and activation of neutrophils and macrophages, and the maturation and function of lymphocytes. Lactoferrin can also regulate immune function by promoting the action of cytokines. When colostrum is contained in the composition, lactoferrin enhances its effect.
[0047] Preferably, the casein hydrolysate is α s1 A hydrolyzate of casein. More preferably, the casein hydrolyzate is α-casozepine or a fragment thereof (having a size greater than or equal to 5 amino acid residues). α-casozepine is a decapeptide of SEQ ID NO.1. Examples of fragments of α-casozepine that can be used in the present invention are a nonapeptide of SEQ ID NO.2, an octapeptide of SEQ ID NO.3, a heptapeptide of SEQ ID NO.4, a hexapeptide of SEQ ID NO.5, and a pentapeptide of SEQ ID NO.6. α-casozepine and the above-mentioned fragments have a sedative effect. They increase the activity of gamma-aminobutyric acid (GABA), which is a neurotransmitter that has an anxiety-inhibiting effect and a relaxing property that promotes a decrease in blood pressure.
[0048] The casein hydrolysate preferably has a molecular weight of 500 to 2000 Da, preferably 1000 to 1500 Da.
[0049] The composition advantageously comprises from 5% to 50% by weight, preferably from 10% to 30% by weight, more preferably from 15% to 20% by weight of iron-binding glycoprotein, preferably lactoferrin, relative to the total weight of the composition.
[0050] The composition may comprise 2% to 30% by weight, preferably 3% to 20% by weight, more preferably 5% to 10% by weight of casein hydrolysate, preferably α-casozepine or a fragment thereof, relative to the total weight of the composition.
[0051] The weight ratio of iron-binding glycoprotein to casein hydrolysate is advantageously 0.1 to 6, preferably 0.5 to 3, more preferably 2. In some embodiments, the weight ratio of iron-binding glycoprotein to casein hydrolysate is 0.1 to 0.2, or 0.2 to 0.3, or 0.3 to 0.4, or 0.4 to 0.5, or 0.5 to 0.6, or 0.6 to 0.7, or 0.7 to 0.8, or 0.8 to 0.9, or 0.9 to 1, or 1 to 2, or 2 to 3, or 3 to 4, or 4 to 5, or 5 to 6.
[0052] The composition may further comprise colostrum (compound C), such as bovine, ovine, equine or porcine colostrum. Preferably, colostrum is bovine colostrum, which is the milk secreted by female mammals just after giving birth. It contains many components, including immunoglobulins IgG1, IGg2, IgM, IgA, lactoferrin, serum albumin, transferrin and α1-acid glycoprotein. Colostrum has the property of stimulating vitality. It can make cells resist internal and external attacks and can maintain their vitality for a longer time. It strengthens cell membranes.
[0053] For example, the composition may comprise 15% to 70% by weight, preferably 20% to 50% by weight, more preferably 25% to 40% by weight of colostrum dry matter relative to the total weight of the composition.
[0054] The weight ratio of colostrum dry matter relative to casein hydrolysate is advantageously from 0.5 to 10, preferably from 1 to 6, more preferably from 4. In some embodiments, the weight ratio of colostrum relative to casein hydrolysate is from 0.5 to 0.6, or from 0.6 to 0.7, or from 0.7 to 0.8, or from 0.8 to 0.9, or from 0.9 to 1, or from 1 to 2, or from 2 to 3, or from 3 to 4, or from 4 to 5, or from 5 to 6, or from 6 to 7, or from 7 to 8, or from 8 to 9, or from 9 to 10.
[0055] It is to be understood that the above-mentioned weights and ratios of colostrum dry matter relate to the total weight of the compounds of the colostrum dry matter, excluding those compounds also included in the composition by specific additions. Therefore, the weight of colostrum dry matter does not take into account the weight of lactoferrin contributed by colostrum (calculated separately). However, in the composition of the present invention, the amount of lactoferrin contributed by colostrum is generally negligible compared to the total amount of colostrum dry matter.
[0056] In some embodiments, the composition further comprises a nitric oxide precursor (compound D), preferably L-arginine. Nitric oxide is a signaling molecule with the function of dilating blood vessels. Nitric oxide precursors promote the migration of other compounds in the composition to the skin. In particular, when colostrum is contained in the composition, the nitric oxide precursor allows the colostrum compounds to easily penetrate into the skin. In addition, arginine can stimulate cells to produce more proteins, such as collagen and elastin, and can repair damaged genetic inheritance.
[0057] The composition may comprise from 10% to 50% by weight, preferably from 15% to 40% by weight, more preferably from 20% to 30% by weight of the nitric oxide precursor relative to the total weight of the composition.
[0058] The weight ratio of nitric oxide precursor to casein hydrolysate is advantageously 0.1 to 10, preferably 1 to 5, more preferably 3. In some embodiments, the weight ratio of nitric oxide precursor to casein hydrolysate is 0.1 to 0.2, or 0.2 to 0.3, or 0.3 to 0.4, or 0.4 to 0.5, or 0.5 to 0.6, or 0.6 to 0.7, or 0.7 to 0.8, or 0.8 to 0.9, or 0.9 to 1, or 1 to 2, or 2 to 3, or 3 to 4, or 4 to 5, or 5 to 6, or 6 to 7, or 7 to 8, or 8 to 9, or 9 to 10.
[0059] The composition may also contain an antioxidant (compound E), preferably a carotenoid. In a particularly preferred manner, the antioxidant is astaxanthin. Astaxanthin has anti-inflammatory and antioxidant effects, and protects the mitochondrial double membrane by improving the function of the mitochondrial double membrane, thereby improving the energy production capacity of the mitochondria. It can be distributed throughout the body, providing antioxidant and anti-inflammatory effects to all organs and tissues. It also strengthens cell membranes.
[0060] Alternatively, or in combination with a carotenoid (eg astaxanthin), the antioxidant may be a retinoid, such as retinol, vitamin C (ascorbic acid and its salts) and / or vitamin E.
[0061] Preferably, the content of the antioxidant is from 2% to 30% by weight, preferably from 3% to 20% by weight, more preferably from 5% to 10% by weight, relative to the total weight of the composition.
[0062] The weight ratio of antioxidant to casein hydrolysate is advantageously 0.1 to 5, preferably 0.5 to 3, more preferably 1. In some embodiments, the weight ratio of antioxidant to casein hydrolysate is 0.1 to 0.2, or 0.2 to 0.3, or 0.3 to 0.4, or 0.4 to 0.5, or 0.5 to 0.6, or 0.6 to 0.7, or 0.7 to 0.8, or 0.8 to 0.9, or 0.9 to 1, or 1 to 2, or 2 to 3, or 3 to 4, or 4 to 5.
[0063] The composition may further comprise serotonin or one of its precursors (compound F). Preferably, the composition comprises a serotonin precursor, more preferably tryptophan, and further preferably L-tryptophan. Serotonin and its precursors, including tryptophan, have a sedative effect and play a role in depression, anxiety, mood and appetite control. The use of serotonin precursors such as tryptophan is particularly advantageous because serotonin cannot pass the blood-brain barrier, while precursors such as tryptophan have this ability. In addition, tryptophan can enhance the effect of casein hydrolysate. It participates in the formation of active lymphocytes in the body's defense system and acts on cell stress.
[0064] The composition may comprise 2% to 30% by weight, preferably 3% to 20% by weight, more preferably 5% to 10% by weight of serotonin or one of its precursors relative to the total weight of the composition.
[0065] The weight ratio of serotonin or one of its precursors to casein hydrolysate is advantageously 0.1 to 5, preferably 0.5 to 3, more preferably 1. In some embodiments, the weight ratio of serotonin or one of its precursors to casein hydrolysate is 0.1 to 0.2, or 0.2 to 0.3, or 0.3 to 0.4, or 0.4 to 0.5, or 0.5 to 0.6, or 0.6 to 0.7, or 0.7 to 0.8, or 0.8 to 0.9, or 0.9 to 1, or 1 to 2, or 2 to 3, or 3 to 4, or 4 to 5.
[0066] In some embodiments, the composition further comprises at least one mineral filler, preferably clay, more preferably selected from illite, montmorillonite, kaolinite or mixtures thereof. In a particularly preferred manner, the composition comprises a mixture of illite, montmorillonite and kaolinite.
[0067] When the composition comprises a mineral filler, it is advantageously contained in an amount ranging from 5% to 80% by weight and preferably from 10% to 50% by weight relative to the total weight of the composition.
[0068] The weight ratio of mineral filler to casein hydrolysate is advantageously from 0.1 to 10, preferably from 0.5 to 5. In some embodiments, the weight ratio of mineral filler to casein hydrolysate is from 0.1 to 0.2, or from 0.2 to 0.3, or from 0.3 to 0.4, or from 0.4 to 0.5, or from 0.5 to 0.6, or from 0.6 to 0.7, or from 0.7 to 0.8, or from 0.8 to 0.9, or from 0.9 to 1, or from 1 to 2, or from 2 to 3, or from 3 to 4, or from 4 to 5, or from 5 to 6, or from 6 to 7, or from 7 to 8, or from 8 to 9, or from 9 to 10.

[0101] Example
[0102] The following are non-limiting examples that illustrate the invention.
[0103] Example 1
[0104] Preparation of composition
[0105] The composition (Product A) was prepared by mixing the following components in powder form:
[0106]
[0107] A composition (Product P2) was then prepared by diluting 15% of Product A in carboxymethylcellulose gel (CMC gel).
[0108] Explant preparation and treatment
[0109] Nine explants with a diameter of 12 mm (± 1 mm) were prepared from a 50-year-old Caucasian female during abdominoplasty. The explants were placed in BEM medium (BIO-EC explant medium) and kept at 37°C, 5% CO 2 survive in humid environments.
[0110] The explants were divided into 3 batches as follows:
[0111] batch deal with Number of explants Number of days to stop the experiment T0 (plasty control) none 3 D0 T (untreated control) none 3 D12 LP2 Product P2 3 D12
[0112] D0 represents the start date of the experiment, and Dn represents the nth day after the start of the experiment.
[0113] At D0, D2, D3, D6, D8 and D10, a spatula was used to apply 2 mg / cm 2 Product P2 was topically applied to the explants corresponding to the LP2 batch in an amount of 1 mL. The explants of the control batches (T0 and T) did not receive any treatment except for the renewal of the medium. At D2, D3, D6, D8 and D10, half of the medium was renewed (1 mL BEM medium per well).
[0114] At D0, three explants from batch T0 were removed and cut in half. One half was fixed in formalin buffer solution and the other half was frozen at -80°C. At D12, explants from each batch T and LP2 were collected and processed in the same way.
[0115] Explant characterization methods
[0116] - Histological processing and cell viability control
[0117] After 24 h in buffered formalin, the samples were dehydrated and paraffin impregnated using a Leica TP 1020 automated tissue processor. They were paraffin embedded in a Leica EG 1160 embedding station. 5 μm sections were obtained using a Minot-type microtome, Leica RM 2125, and mounted on Histological slides. Microscopic observations were performed under light microscopy using a DMLB or Olympus BX43 Leica microscope. Images were captured using an Olympus DP72 camera and Cell^D software.
[0118] Cell viability was observed on paraffin sections after staining with Masson's trichrome Goldner variant. Evaluation was performed by microscopic examination.
[0119] -Measurement of epidermal thickness by image analysis.
[0120] The images obtained after staining with Masson's trichrome stain were analyzed to measure the thickness of the epidermis. Three target areas were measured per image, i.e. 27 measurements per batch.
[0121] result
[0122] At D12, the epidermal cell viability of control batch T was quite good, and the dermal cell viability was good.
[0123] In explants treated with product P2, an increase in epidermal thickness due to hyperplastic acanthosis was observed (increased epidermal thickness due to increased size of keratinocytes). The cells became larger, indicating an increased intracellular nutrient absorption.
[0124] The following table summarizes the results of the epidermal thickness measurements:
[0125] batch T0 T LP2 Measurement days D0 D12 D12 Average thickness of epidermis (μm) 28.5 50.3 66.1 Standard Deviation 4.6 7.3 6.9
[0126] The application of product P2 significantly increased epidermal thickness by 31% compared to untreated control epidermis (p<0.01).
[0127] The cells of explants treated with product P2 were larger than those of untreated control explants. They were in good condition and able to withstand various biological and environmental stresses. In addition, the cell lifespan was improved.
[0128] Example 2
[0129] Preparation of the product.
[0130] The following compositions were prepared:
[0131] - Product P1': Lactoferrin (powder) diluted in carboxymethylcellulose gel (CMC gel) to a final concentration of 15% by weight;
[0132] - Product P2': α-casozepine (powder) diluted in CMC gel to a final concentration of 15 wt.%;
[0133] - Product P3': a mixture of lactoferrin and α-casozepine (powder) diluted in CMC gel to a final concentration of 15% by weight;
[0134] - Product P4': Product A (powder) as described above in Example 1 was diluted in CMC gel to a final concentration of 15% by weight.
[0135] Products P3' and P4' are compositions of the invention, products P1' and P2' are comparative examples.
[0136] Explant preparation and treatment
[0137] Eighteen explants with a diameter of 12 mm (± 1 mm) were prepared from a 45-year-old Caucasian female undergoing abdominoplasty. The explants were placed in a humidified atmosphere at 37°C with 5% CO. 2 The cells were placed in BEM survival medium (explant medium for BIO-EC) in a moist environment.
[0138] Explants were divided into 6 batches as follows:
[0139] batch deal with Number of explants Number of days to stop the experiment T0'(plasty control) none 3 D0 T'(untreated control) none 3 D18 LP1' Product P1' 3 D18 LP2' Product P2' 3 D18 LP3' Product P3' 3 D18 LP4' Product P4' 3 D18
[0140] D0 represents the start date of the experiment, and Dn represents the nth day after the start of the experiment.
[0141] At D0, D2, D3, D6, D8, D10, D13, D15 and D17, products P1', P2', P3' and P4' were topically applied at 2 μL per explant and spread with a spatula on the explants of batches LP1', LP2', LP3' and LP4', respectively. The explants of the control batches (T0' and T') did not receive any treatment except renewal of the medium. At D2, D3, D6, D8, D10, D13, D15 and D17, half of the medium was renewed (1 mL BEM medium per well).
[0142] At D0, three explants from batch T0' were removed and cut in half. One half was fixed in buffered formalin solution and the other half was frozen and stored at -80°C. At D18, explants from each batch T', LP1', LP2', LP3' and LP4' were removed and processed in a similar manner.
[0143] Explant characterization methods
[0144] Cell viability control and measurement of epidermal thickness were performed as described in Example 1.
[0145] General morphology was examined in the same manner as cell viability, ie, assessed on paraffin sections after staining with Masson's trichrome Goldner variant under a microscope.
[0146] result
[0147] At D0, epidermal and dermal cell viability was good on the control plasty batch T0'. The dermal-epidermal junction (DEJ) zone was clear and the dermal density was evident.
[0148] At D18, on the untreated control batch T', the cell viability of the epidermis was significantly impaired and that of the dermis was slightly impaired. The dermal-epidermal junction (DEJ) zone was moderate and the dermis was thin.
[0149] After further application of products P1', P2', P3' and P4' the following effects on the general morphology of the explants were observed at D18 compared to the control batch T' at D18.
[0150] Application of product P1' resulted in a slight improvement in cell viability in the epidermis and a very slight improvement in cell viability in the dermis. In addition, it caused a slight increase in epidermal thickness caused by hyperplastic acanthosis. Finally, it caused a slight increase in dermal density.
[0151] Application of product P2' resulted in a very slight improvement in cell viability in the dermis, but no changes in the epidermis. In addition, it caused a slight increase in epidermal thickness caused by hyperplastic acanthosis. Finally, it caused a slight increase in dermal density.
[0152] Application of product P3' resulted in a slight improvement in epidermal cell viability and a very slight improvement in dermal cell viability. In addition, it caused a moderate increase in epidermal thickness caused by hyperplastic acanthosis. Finally, it caused a slight increase in dermal density.
[0153] Application of product P4' resulted in a moderate improvement in epidermal cell viability and a very slight improvement in dermal cell viability. In addition, it caused a moderate increase in epidermal thickness caused by hyperplastic acanthosis. Finally, it caused a moderate increase in dermal density.
[0154] The following table summarizes the results of the epidermal thickness measurements:
[0155]
[0156] * = significant value, p < 0.01
[0157] Application of product P3' resulted in an increase in epidermal thickness that was significantly greater than that obtained using products P1' and P2'. Therefore, product P3' has a better anti-aging effect than products P1' and P2'. In addition, product P3' increased cell viability compared to product P2'.
[0158] Application of product P4' also resulted in an increase in epidermal thickness, which was significantly greater than that obtained with products P1' and P2'. Therefore, product P4' has a better anti-aging effect than products P1' and P2'. In addition, application of product P4' increased cell viability and dermal density compared to products P1', P2' and P3'.
