AI Technical Summary

Technical Problem

Existing prostate cancer treatment methods are prone to recurrence in patients with local diseases. Androgen depletion therapy can only temporarily control the condition and lack effective targeted treatment methods.

Method used

Chimeric antigen receptor (CAR) targeting human kallirein-2 (hK2) was developed, which contains extracellular domains, transmembrane domains and intracellular signaling domains, to modify T cells to specifically recognize and kill prostate cancer cells.

Benefits of technology

It achieves specific killing of prostate cancer cells, enhances tumor immune response, and provides a new targeted therapeutic method for the treatment of prostate cancer.

2. An isolated lymphocyte that expresses the CAR according to claim 1.

3. The isolated lymphocyte according to claim 2, wherein the lymphocyte is a T lymphocyte.

4. The isolated lymphocyte according to claim 2, wherein the lymphocyte is a natural killer (NK) cell.

5. An isolated nucleic acid molecule that encodes the CAR according to claim 1.

6. A vector that comprises the nucleic acid molecule according to claim 5.

7. A cell that expresses the nucleic acid molecule according to claim 5.

8. A pharmaceutical composition that comprises an effective amount of the lymphocyte according to any one of claims 2 to 4.

9. A pharmaceutical composition that comprises an effective amount of the lymphocyte according to any one of claims 2 to 4 and a pharmaceutically acceptable excipient.

10. Use of the pharmaceutical composition according to claim 8 or 9 in the preparation of a medicament for use in treating a subject having prostate cancer or breast cancer expressing an androgen receptor.

11. Use of the pharmaceutical composition according to claim 8 or 9 in the preparation of a medicament for use in treating a subject having prostate cancer or breast cancer expressing an androgen receptor, wherein the lymphocyte induces killing of prostate cancer cells or breast cancer cells expressing an androgen receptor in the subject.

12. Use of the pharmaceutical composition in the preparation of a medicament for detecting the presence of prostate cancer or breast cancer expressing an androgen receptor in a subject, wherein the pharmaceutical composition comprises the CAR according to claim 1.
