Description

[0001] This application is a divisional application of the invention patent application filed on February 7, 2018, with application number 201880000825.X and titled “Aminopyrimidine compounds suitable for use as SSAO inhibitors”. Technical Field

[0002] The present invention relates to aminopyrimidine compounds, pharmaceutically acceptable salts of said compounds and therapeutic uses of said compounds and their salts. Background Art

[0003] Semicarbazide-sensitive aminooxidase / vascular adhesion protein-1 (SSAO / VAP-1) exists in two forms: a membrane-bound isoform and a plasma-soluble isoform. It is mainly expressed from the surface of endothelial cells, vascular smooth muscle, and adipocytes. SSAO / VAP-1 is involved in many cellular processes, including glucose disposal, inflammatory responses and related pain, and leukocyte recruitment. High activity levels of this enzyme are associated with diabetes, atherosclerosis, stroke, chronic kidney disease, and Alzheimer's disease, among other conditions. SSAO / VAP-1 has been implicated in the pathogenesis of liver diseases, such as fatty liver. (Weston CJ et al., J. Neural. Transm., 2011, 118, 1055.) Fatty liver disease (FLD) covers a range of disease conditions characterized by excessive accumulation of fat in the liver, often accompanied by inflammation. FLD can cause non-alcoholic fatty liver disease (NAFLD), which is characterized by insulin resistance. If left untreated, NAFLD can progress to persistent inflammation or non-alcoholic steatohepatitis (NASH), progressive liver fibrosis, and ultimately cirrhosis. There is a need to provide alternative treatments for liver diseases such as NAFLD and / or NASH.

[0004] It is believed that SSAO / VAP-1 inhibitors will reduce liver inflammation and fibrosis and thus provide treatment for liver disease, especially for the treatment of NAFLD and / or NASH. In addition, because the activation of SSAO / VAP-1 is involved in inflammation and related pain, the inhibition of SSAA / VAP-1 enzymes may be useful for the treatment of pain and pain associated with osteoarthritis in particular. (Luis M. et al., J of Pharm and Experimental Therapeutics, 2005, 315, 553).

[0005] US Patent No. 8,426,587 discloses haloallylamine compounds useful as SSAO / VAP1 inhibitors.

[0006] Currently, there are no approved drugs for the treatment of NASH; the standard of care for NASH consists of dietary control and / or lifestyle changes. In addition, the current standard of care for pain is dominated by nonsteroidal anti-inflammatory drugs (NSAIDS) and opiates. Both drug classes are recommended for short-term use only. It is desirable to have more treatment options to control pain, especially chronic pain. The present invention provides compounds that inhibit the SSAO / VAP-1 enzyme and can address one or more of these needs. Summary of the invention

[0037] Process 1

[0038]

[0039] Scheme 1 depicts a general synthesis of compounds of formula 1, wherein "PG" is a protecting group for the hydroxyl group. Specifically, in step 1, sub-step 1, the chlorine of compound A can be replaced by the nitrogen of R1 substituted with a cyclic amine to obtain compound B. In step 1, sub-step 2, deprotection of the hydroxyl group can be achieved by a variety of methods depending on the specific protecting group. In step 2, sub-step 1, the hydroxyl group of the resulting compound B can be alkylated with a suitably protected amine 2-bromo-3-fluoro-propyl-2-ene-amine to obtain a compound of formula 1. Alkylation can generally be achieved under alkaline conditions. The protected amine can be deprotected to obtain a compound of formula 1.

The compound of Example 1 was administered to rats at a dose of 15, 3, 0.6, 0.12, 0.025, 0.005 mg / kg. The same volume (2 ml / kg) of dosing vehicle (hydroxyethyl cellulose 1% w / v, 0.25% Tween 80) was administered to the control group. The plasma and liver after 2 or 24 hours of compound treatment were collected and stored at -78°C until analysis. Tissue lysates were prepared by homogenization in lysis buffer (20 mM HEPES, pH 7.4; 150 mM NaCl, 1 mM EDTA, 1 mM EGTA, 1% Triton X-100 and 1× Roche Complete protease inhibitor tablet). Tissue pellets were removed by centrifugation at 12,000 rpm for 30 min at 4°C. 40 μl of plasma or liver lysate was incubated with clorgyline (10 μM) and pargyline (10 μM) at room temperature for 20 minutes, followed by the addition of luminescent substrate (50 μM) for 60 min. The products produced were quantified according to the manufacturer's procedure.
