Abstract
Description
Technical Field
[0001] The invention relates to a traditional Chinese medicine preparation for treating varicella-zoster virus infection, belonging to the technical field of traditional Chinese medicines. Background Art
[0002] Herpes zoster (shingles) is a common dermatological disease caused by the varicella-zoster virus. The global incidence of herpes zoster is 3-5 per 1,000 people per year in the general population and 3-10 per 1,000 people per year in the Asia-Pacific region, with an annual increase of 2.5% to 5.0%. The typical symptom is the development of vesicles along sensory nerves in the corresponding innervated segments. Clinically, the rash is characterized by clusters of erythematous papules and blisters. Most patients experience severe pain during the acute phase and are prone to postherpetic neuralgia. Some patients even experience anxiety and depression, which can severely impact their daily lives. Current Western medical treatments primarily focus on antiviral, analgesic, and immune-boosting therapies. Antiviral drugs primarily include the commonly used open-ring nucleoside drugs (valacyclovir, acyclovir, famciclovir, etc.), interferons, and non-nucleoside drugs (sodium foscarnet, adenosine monophosphate, etc.). These drugs work by binding to the DNA polymerase of the varicella-zoster virus, reducing viral activity and exerting an antiviral effect. However, their clinical efficacy is less than satisfactory and they are associated with certain side effects. Traditional Chinese medicine (TCM) has some experience in treating herpes zoster. The current mainstream consensus in TCM circles is that the primary etiology of herpes zoster is damp-heat in the liver and gallbladder, spleen dysfunction, blocked collaterals, liver depression and qi stagnation, emotional disturbances, and dietary irregularities. However, a widely used and effective TCM preparation remains lacking in terms of therapeutic efficacy. Summary of the Invention

DETAILED DESCRIPTION
[0028] The traditional Chinese medicine syndrome differentiation of herpes zoster mainly includes liver-gallbladder damp-heat type, spleen dysfunction type, qi stagnation and blood stasis type, and the treatment strategy focuses on clearing away heat and toxic substances, invigorating the spleen and removing dampness, promoting blood circulation and removing blood stasis, promoting qi circulation and removing stagnation, and relieving pain. The present invention considers that herpes zoster is common in middle-aged and elderly people and people with physical weakness. Such patients are liver-kidney insufficiency, old and weak, and lack of vital energy, which leads to the internal release of latent pathogens, poor circulation of qi and blood, and internal generation of blood stasis, which blocks the meridians and affects the normal distribution and movement of body fluids, causing body fluids to accumulate, thereby generating phlegm-dampness, further blood stasis and phlegm-dampness blocking the meridians, and the obstruction of the meridians, so the disease occurs. In view of this cause of disease, the present invention uses ginseng, mistletoe, schisandra chinensis, and gastrodia elata as the main medicines to strengthen the body constitution, nourish the spleen and benefit the lungs, and strengthen the body and eliminate evil. Among them, ginseng can greatly replenish vital energy and has the effect of replenishing qi and strengthening deficiency. The classics record that gastrodia elata can "be taken for a long time to improve qi and strength, grow yin, be fat and healthy, lighten the body, and increase longevity". Schisandra chinensis nourishes the lungs and kidneys, while mistletoe nourishes the liver and kidneys and strengthens the tendons and bones. The auxiliary herbs cinnamon twig, saposhnikovia divaricata, and processed Evodia rutaecarpa nourish the liver and kidney yang, warm the meridians, dispel cold, and relieve pain. The adjuvant herbs Chuanxiong, Salvia miltiorrhiza, Angelica sinensis, Cortex moutan, Paeonia lactiflora, Scorpio, and Scolopendra subspinipes promote blood circulation, dissipate stasis, resolve toxins, and resolve stagnation, unblocking the meridians and relieving spasms. This unclogs the meridians and relieves pain. Tangerine peel, Pinellia ternata, and Herba scutellariae are also added to regulate qi and balance the middle qi, dissipate dampness, and resolve phlegm. Pinellia ternata is particularly important for treating phlegm-dampness. Roasted Licorice root tonifies qi and strengthens the spleen, harmonizing the other herbs. Together, these herbs strengthen the body's vital energy, dispel pathogenic factors, dispel cold and relieve spasms, unblock the meridians and relieve pain, and detoxify and resolve dampness.

[0042] ① Criteria for determining efficacy Cured: After treatment, the rash subsides, pain disappears, and scabs form completely, with no PHN. Markedly effective: After treatment, the number of rashes decreases significantly, pain is significantly alleviated, and most rashes scab over, but mild PHN symptoms may occur occasionally. Effective: After treatment, the number of rashes decreases, pain improves somewhat, and rashes partially scab over, although some patients may develop PHN. Ineffective: The rash does not decrease or may even increase, pain persists, the rash does not scab over, and new rashes may appear repeatedly. Some patients may develop PHN. The total effective rate = cure rate + marked effective rate + effective rate.
[0043] ②Evaluate indicators such as pain relief and complete scab formation time ③Comparison of pain levels between the two groups ④Comparison of the incidence of PHN between the two groups (5) Statistical methods Data were collated and statistically analyzed using R (version 4.5.0). Continuous data were first subjected to the Shapiro-Wilk normality test; if normal distribution was satisfied, the t-test was used; if not, the Wilcoxon rank-sum test was used. For incidence ratios, the Fisher test was used to determine the association between variables in a 2×2 contingency table. For enumeration data, the chi-square test was used based on theoretical frequency and sample size, and two-sided or one-sided tests were used based on the study objectives. P < 0.05 was considered statistically significant.
[0044] 2. Treatment Results (1) Comparison of clinical efficacy between the two groups of patients Comparison of clinical efficacy between the two groups showed that the total effective rate of the observation group was 97.50% (39 / 40), which was significantly higher than that of the control group (80.00% (32 / 40). The difference was statistically significant ( P <0.05). See Table 2 for details.
