Abstract

The invention discloses a method for improving the editing efficiency of a guide editing system and application thereof. The invention further discloses an editing guiding system PE-At2. Experiments prove that compared with other guide editing systems, the T1-generation average PE efficiency of the guide editing systems PE-At2.1 and PE-At2.2 is relatively high, and the T1-generation average PE efficiency of the guide editing system PE-At2.1 is the highest. In addition, more importantly, the guide editing system PE-At2.1 is more suitable for being used for creating target editing plant mutants which can be stably inherited and do not contain transgenic ingredients. The invention provides a more accurate and powerful tool for the research of plant gene functions, especially the research of dicotyledon gene functions.
