Abstract

This invention relates to an additive for NK cell induction and expansion culture, a culture medium, and their applications. The additive comprises tanshinone I and berberine. Specifically, PBMCs are isolated from human peripheral blood. On the third day of NK cell activation culture, tanshinone I (final concentration 0.1-10 µM) and berberine (final concentration 10-100 µM) are added to the culture system. Both tanshinone I and berberine are natural products with wide availability. Their combined use can significantly increase the proportion of memory NK cells, while exhibiting high viability, high purity, high expansion rate, and high cytotoxic activity. Flow cytometry analysis showed a significant increase in the positive expression rates of some NK cell markers, including CD62L, CD127, and Eomes.
