Description

FIELD OF THE INVENTION

[0001] The present invention relates to compounds derived from 7-azaindole useful as AXL kinase inhibitors for the treatment of infections. Their preparation process is also described. TECHNOLOGICAL BACKGROUND

[0002] AXL is a Receptor Tyrosine Kinase (RTK) belonging to the TAM family composed of TYRO-3, AXL and MER. Initially discovered and demonstrated in patients with chronic myeloid leukemia (CML), the involvement of the AXL receptor in viral infections such as dengue, Zika virus, Chikungunya or Ebola fever has been described in the literature (Chen, J. et al. Nature Microbiology, 2018, 3, pp.302-309; Fedeli, C. et al. Journal of Virology, 2018, 92:e01613-17; Hastings, AK et al., iScience, 2019, 13 pp.339-350; Meertens, L. et al., Cell Reports, 2017, 18, 3, pp.324-333). This receptor, by interacting via the Gas6 adaptor with the phosphatidylserines present in viral envelopes, promotes the attachment of enveloped viruses to their cellular target. AXL thus stimulates viral endocytosis.AXL engagement during this interaction stimulates phosphorylation of the intracytoplasmic domain of AXL and activates associated signaling pathways. These signals neutralize interferon production and associated antiviral responses, promoting the escape of these viruses from host immune control.

[0003] To date, several kinase inhibitors active against AXL exist (Myers et al., J. Med. Chem. (2015), 59(8): 3593-3608), but none are truly selective or specific for this kinase. In the vast majority of cases, activity against AXL is secondary to the primary activity sought against MET or MER due to the sequence similarity between these RTKs. This is notably the case for Bosutinib or Cabozantinib, which are multi-target kinase inhibitors or MTKIs ( MultiTargeted Kinase Inhibitors) already on the market, or for BMS777607 (currently in clinical phase 2). This is also the case for the 7-azaindole derivative NPS-1034, which has a relatively broad inhibition profile, inhibiting a wide panel of kinases such as AXL / DDR1 / FLT3 / KIT / MEK / MET / ROS1 and TIE1. Bosutinib

[0004] Cabozantinib

[0005] BMS-777607

[0006] NPS-1034

[0007] Another AXL kinase inhibitor is Bemcentinib (also known as R428 or BGB324), currently in clinical trials. This compound, which is structurally very different from currently available kinase inhibitors, has also been shown to be active against other kinases such as ABL / KIT / JAK2-3 / LCK / PDGFRB / TIE2. R428 / BGB324

[0008] US 2003 / 207910 describes 6-aza-indoles as antiviral compounds. KR 2012 0061011 describes 7-aza-indoles as inhibitors of Phospho Inositide 3 Kinase.

[0009] There is therefore a need for new AXL inhibitory compounds that are more selective and effective against viral infections.

[0010] The present invention thus provides novel 7-azaindole derivatives that strongly inhibit AXL kinase, for use as antiviral agents.

[0011] Due to this very specific inhibition profile, unique to date for this type of structure, these compounds can be used in therapy in the treatment of infections caused by viruses using the AXL receptor to multiply. In order to combat viral infections, by inhibiting the phosphorylation of AXL, these compounds will act on the one hand by blocking the endocytosis of enveloped viruses in their cellular target, and on the other hand by neutralizing the intracellular signals controlled by AXL known to inhibit the production of interferons and antiviral responses of the host. The inhibitors of the present invention can thus be used for the treatment of diseases in which AXL is involved, in particular viral infections and in particular viral entry into cells. [0133] The compounds of the invention were evaluated for their Axl kinase inhibitory activity, selectively. 1- Inhibition of Axl kinase phosphorylation in cellulo

[0134] A549 cells were preincubated for 1 h with 2.5 µM of compound 1, 2 or 3. They were then stimulated for 5 min with 1 mM pervanadate, a compound known to activate Axl kinase by phosphorylation.

[0135] Inhibition of phosphorylation of residue Y779 of the intracytoplasmic domain of Axl is visualized by immunoblotting using the R&D Systems antibody (AF2228).

[0136] The results are illustrated in Figure 1 They show a much lower or even no activation of the Y779 residue of AxL in the presence of compounds 1, 2 and 3 compared to the control (DMSO). 2- Selective inhibition of Axl kinase

[0137] The inhibitory activity of the compounds on a panel of kinases including AXL and FLT3 was evaluated by Thermo Fisher Scientific using Z'-LYTE ®< and ADAPTA ®< Select Screen technologies.

[0138] The fluorescence-based biochemical assay uses coupled enzymes and is based on the difference in sensitivity of phosphorylated and non-phosphorylated peptides to proteolytic cleavage. The peptide substrate is labeled with two fluorophores—one at each end—enabling fluorescence resonance transfer (FRET).

[0139] Compounds 1, 2 and 3 are tested in wells at the final concentration of 100 nM in 1% DMSO.

[0140] The results are presented in the following table:

[0141] Percentage of kinase inhibition by the compounds of the invention. Columns from left to right: the kinases tested - compound 1- compound 2- compound 3.

[0142] The results show that each of the compounds of the invention shows excellent activity (95%, 95% and 94% inhibition) on Axl kinase when, with respect to other kinases, the results are disparate from one compound to another.

[0143] To calculate the IC 50 values for inhibition of Axl and FLT3 by the compounds of the invention, 10 concentrations of each compound 1, 2 and 3 were prepared by serial dilutions of one-third from the starting concentration. Axl or FLT3 kinases were diluted to a 2X concentration in kinase buffer. All ATP solutions were diluted to a 4X working concentration (50 mM HEPES pH 7.5, 0.01% BRIJ-35, 10 mM MgCl2, 1 mM EGTA).

[0144] Each compound is incubated at 10 concentrations ranging from 100 nM to 0.00495 nM to calculate the IC 50 .

[0145] The results are illustrated in figure 2 and summarized in the table below Compound IC 50 Axl IC 50 FLT3 1 5.36 nM 2.5 nM 2 2.36 nM 2.33 nM 3 6.3 nM 48.2 nM

[0146] Compounds 1, 2 and 3 show excellent activity against both Axl and FLT3 kinases. Compound 3 even shows selectivity for Axl over FLT3, with Axl playing a critical role in FLT3 activation. B- Antiviral evaluation

[0147] The compounds of the invention were evaluated for their antiviral activity in vitro, by testing the inhibitory effect of the molecules on the replication of complete infectious viruses. The chosen cell systems (HeLa, A549, A549-ACE2 cells) are relevant for the envisaged infectious models and have been validated for the expression of the Axl molecule in flow cytometry. Infectious models used:

[0148] Virus Model Strain Cells ZIKV mCherry Reporter BeH-8 HeLa DENV Luciferase reporter DENV2 HeLa WNV Luciferase reporter Kunjin (KUNV) HeLa CHIKV Luciferase reporter LROPY-1 HeLa SARS-CoV-2 Viral isolate BetaCoV / France / IDF0371 / 202 A549-ACE2, VeroE6, primary bronchial epithelium 1- Antiviral activity against the Zika virus

[0149] The assays are performed by pre-incubating 2.5 × 10⁴ HeLa cell line (cervical carcinoma; ATCC# CCL-2) cultured in 96-well plates with increasing concentrations of compounds for 1 h at 37°C. The cells are cultured in a 5% CO₂ atmosphere in Dulbecco's modified Eagle's medium (DMEM) containing 10% calf serum and 1% penicillin / streptomycin. The cells are then exposed to the ZIKV BeH8 strain (MOI = 0.1) for 1 h in the presence of the compound.

[0150] The viral inoculum is then removed and the cells are maintained in the presence of inhibitors for 24 h. Cell infection is determined by quantification of viral RNA detected by qRT-PCR using the Luna Universal One-Step RT-PCR kit and the primers 5'-CCGCTGCCCAACACAAG and 5'-CCACTAACGTTCTTTTGCAGACAT using the Luna Universal One-Step RT-PCR kit. The values (DCT) are normalized to the quantification of GAPDH mRNA. The values represented are the means of triplicates + standard deviation.

[0151] Control conditions were achieved by incubating the cells in the presence of DMSO, the solvent used for solubilization of the tested compounds. The volumes of DMSO used under these conditions correspond to the volumes provided by the compounds under the test conditions.

[0152] A parallel treatment, under the same experimental conditions, was carried out with R428 / Bemcentinib, Axl inhibitors in clinical trial phase developed by the company Bergenbio.

[0153] The results obtained with compounds 1, 2, 3, 10 and 11 are illustrated in Figure 3 The IC50 values of compounds 1, 10 and R428 / Bemcetinib were determined using Graphpad Prism software.

[0154] Under these experimental conditions, compounds 1, 2, 3, 10 and 11 used at a concentration greater than or equal to 150 nM prevent infection of HeLa cells by the ZIKV BeH-8 strain. The in vitro efficacy of these compounds is at least equivalent to that of R428 / Bemcentinib described in the literature for its efficacy against the ZIKV virus at concentrations of 1 to 3 µM. 2- Antiviral activity against the Dengue virus

[0155] The assays are performed by preincubating 4×10 4< HeLa cells (human cervical adenocarcinoma; ATCC# CCL-2) grown in 96-well plates with increasing concentrations of compounds for 1 h. The cells are grown in a 5% CO2 atmosphere in Dulbecco's modified Eagle's medium (DMEM) comprising 10% calf serum and 1% Penicilin / streptomycin. The cells are then exposed to the DENV subtype 2 strain (DENV2) expressing a nanoluciferase gene upstream of the capsid protein coding sequence, in the presence of the compounds, for 1 h. The viral inoculum is then removed and the cells are maintained in the presence of the inhibitors for 3 days.

[0156] Cells were lysed using Passive Lysis Buffer reagent (Promega). Viral infection was detected by quantifying nanoluciferase activity in the cell lysate in the presence of Genofax A reagent (Yelen) using an Infinite F200PRO fluorometer (Tecan). Values were normalized to the total protein content of the cell lysate, determined using the BCA Protein Assay kit (Pierce) by measuring absorbance at 562 nm. Values shown are triplicate means plus standard deviation. Control conditions were identical to those described previously.

[0157] The results obtained with compounds 10, 11 and 14 are illustrated in Figure 4 .

[0158] Under these experimental conditions, compounds 10, 11 and 14 used at a concentration greater than or equal to 500 nM prevent infection of HeLa cells by the DENV2 subtype. 3- Antiviral activity against WNV virus Kunjin strain

[0159] The assays are performed by preincubating 4×10 4< HeLa cell line (ATCC# CCL-2) cultured in 96-well plates with increasing concentrations of compounds for 1 h. The cells are cultured in a 5% CO2 atmosphere in Dulbecco's modified Eagle's medium (DMEM) comprising 10% calf serum and 1% Penicillin / streptomycin. The cells are then exposed to WNV virus, Kunjin strain expressing a nanoluciferase gene upstream of the sequence coding for the capsid protein, in the presence of the compounds, for 1 h. The viral inoculum is then removed and the cells are maintained in the presence of the inhibitors for 24 h.

[0160] Cells were lysed using Passive Lysis Buffer reagent (Promega). Viral infection was detected by quantifying nanoluciferase activity in the cell lysate in the presence of Genofax A reagent (Yelen) using an Infinite F200PRO fluorometer (Tecan). The cells are grown in a 5% CO2 atmosphere in Dulbecco's modified Eagle's medium (DMEM) containing 10% calf serum and 1% penicillin / streptomycin. The cells are then exposed to the CHIKV virus, strain LR-OPY1, expressing either a luciferase gene or a GFP coding sequence upstream of the nsP3 protein coding sequence. The cells are incubated for 1 h in the presence of the compounds.

[0164] The viral inoculum is then removed and the cells are maintained in the presence of inhibitors for 24 h. Viral infection is measured after cell lysis using the Passive lysis Buffer reagent (Promega), either by direct quantification of GFP fluorescence or by quantification of luciferase activity in the cell lysate carried out in the presence of the Genofax A reagent (Yelen) and using an Infinite F200PRO fluorimeter (Tecan). The values are normalized to the amount of total protein contained in the cell lysate and determined using the BCA Protein Assay kit (Pierce) by measuring the absorbance at 562 nm. In both protocols, the values are normalized to the amount of total protein contained in the cell lysate and determined using the BCA Protein Assay kit (Pierce). The values represented are the means of triplicates + standard deviation. The control conditions are identical to those described previously.

[0165] The results obtained for compound 10 are illustrated in Figure 6 .

[0166] Under these experimental conditions, compound 10 used at a concentration greater than or equal to 100 nM prevents infection of HeLa cells by the Chikungunya virus.

[0174] The IC 50 of compounds 1, 2 and 3 were determined on VeroE6 and A549-ACE2 cells using Graphpad Prism software.

[0175] The results obtained are illustrated in Figure 9 and in the following table: IC 50 Compound 1 2 3 A549-ACE2 19 nM 57 nM 38 nM Vero E6 77 nM 289 nM ND

[0176] Compounds 1, 2, 3, 10 and 14 exhibit anti-SARS-CoV2 activity in vitro. b- Evaluation of antiviral activities in primary human bronchial epithelium model

[0177] Primary bronchial epithelial infection tests were performed using the MucilAir™ Human airway epithelial cells (Epithelix) model cultured in appropriate medium (HAE medium, Epithelix). Compounds are added to the basal compartment of the culture and preincubated for 1 hour at 37°C. Viral inoculum (MOI = 0.01) is then added to the apical compartment at the same time as the compounds. After 2 hours at 37°C, the viral inoculum is removed, replaced with medium containing the test compound and the culture is maintained at 37°C for 24 hours. Cell infection is determined by quantification of viral RNA detected by qRT-PCR using the Luna Universal One-Step RT-PCR kit. The values (DCT) are normalized to GAPDH mRNA.

[0178] The results obtained for compound 1 are illustrated in Figure 10 .

[0179] These evaluations indicate that compound 1 inhibits SARS-CoV2 infection of the primary bronchial epithelium.7- Preventive and curative activity of compound #1 against SARS-CoV2 coronavirus The inhibitory activity of compound 1 on SARS-CoV2 infection was evaluated in vitro in the context of preventive (Pre-Tt) or curative (Post-Tt) treatment of the A549-ACE2 line produced by transfection of the ACE2 receptor in the A549 line (lung epithelial carcinoma; ATCC# CCL-185) a), and the green monkey line VeroE6 (immortalized kidney epithelium, ATCC#CRL-1586).

[0180] Infection tests are performed on 4×10 4< cells cultured in 96-well plates, in a 5% CO2 atmosphere in Dulbecco's modified Eagle's medium (DMEM) comprising 2% calf serum and 1% penicillin / streptomycin. The cells are exposed to the SARS-CoV2 virus, strain BetaCoV / France / IDF0371 / 2020 (MOI = 0.01). The compounds are added to the cells either 1h before infection and maintained for 24h; or added to the cells 1h before infection and washed off 4h after viral challenge; or added to the cells 4h after exposure to the virus and maintained for 24h (Post-Tt). Residual infection is quantified by amplification of viral RNA by qRT-PCR using the Luna Universal One-Step RT-PCR kit. (DCT) values are normalized to GAPDH mRNA.

[0181] Control conditions were achieved by incubating cells in the presence of DMSO (SARS-CoV-2), the solvent used to solubilize the tested compounds. The volumes of DMSO used in these conditions correspond to the volumes provided by the compounds in the test conditions.

[0182] The results obtained are illustrated in Figure 11 .

[0183] These evaluations indicate that the preventive use of the compounds inhibits SARS-CoV2 infection in human A549-ACE2 cells and monkey VeroE6 cells. Administration of the compounds after viral challenge reduces viral multiplication in A549-ACE2 cells, but not in the VeroE6 cell line, which is characterized by a defect in the production of type I interferons.
