[0001] A novel enantioselective crystal floatation process

[0002] The present invention relates to a process for the separation of enantiomeric crystals by enantioselective crystal floatation and to a process in which preferential crystallization is combined with enantioselective crystal floatation.

[0003] Compounds which are identical, non-superimposable mirror images of each other are referred to as enantiomers. They have the same physiochemical properties and only differ by the direction that they rotate polarized light. Although these molecules are identical in structure, the way they operate on a molecular level within a living organism is different due to their stereospecific interactions in a chiral environment. It is widely known that one enantiomer can have either a lower efficacy than its counter enantiomer or even result in undesired or harmful side effects. Regulatory bodies have placed rigorous screening protocols on chemicals that contain racemic mixtures. Therefore, access to enantiomerically pure compounds is highly desirable, particularly in industrial production schemes.

[0004] Current methods to obtain enantiomerically pure compounds make use of chiral synthesis, selectively targeting one enantiomer by asymmetric synthesis or using the chiral pool. Another approach, chiral resolution, builds on enantiomer separation of racemic mixtures via different techniques such as chiral membranes, chiral chromatography, enzyme-based kinetic separation or diastereomer salt separation.

[0005] These techniques have advantages and disadvantages unique to their processes but the major disadvantage prevalent throughout is that they are usually tailored to one compound. Especially in industrial settings it is desirable that the enantioselective process be highly adaptable to different compounds.

[0006] Preferential crystallization techniques have gained a lot of interest over the past decades as a simple, inexpensive, and adaptable option for enantiomeric separation. It is a process in which one enantiomer crystallizes out of solution via secondary nucleation, for instance after the solution has been seeded with crystals of the desired enantiomer (eutomer), while the undesired enantiomer (distomer) remains in solution. The process is kinetically driven so the crystals must be recovered before the distomer begins to crystallize (H. Lorenz et al., Chem. Eng. Process. Process Intensif, 2006, 45, 863-873).

[0007] Chiral compounds crystallize into three different crystal forms: racemic crystals, conglomerates of crystals, and pseudo racemates. 90-95% of chiral compounds crystallize as racemic crystals, where both enantiomers are incorporated into the same unit cell in equal ratio. 5-10% crystallize as conglomerate crystals, which is defined as a ‘physical mixture of pure enantiomeric crystals' and very few crystallize as solid solutions (F. Cascella et al., Chem. Eng. Technol., 2020, 43, 329-336; S. Srisanga et al., Cryst. Growth Des., 2010, 10, 1808-1812). In both, conglomerate and racemic crystal batch systems, the crystallization must be stopped before contamination by the distomer or the racemic crystal, respectively, because the separation at the end is accomplished via filtration, which does not discriminate between the crystals.

[0008] Binev et al. (Cryst. Growth Des., 2016, 16, 1409-1419) and Temmel et al. (Crystals, 2020, 10, 394) suggested to separate enantiomeric crystals spatially within a fluidized bed crystallizer. The seeded crystals of the eutomer are larger and tend to sink to the bottom of the fluidized bed, while the smaller, nucleated distomer crystals are swept to the top by the moving fluid. This separation method utilizes a difference in crystal mass. While enantiomer molecules exhibit the same thermophysical properties the arrangement these enantiomers take within the crystal lattice results in crystal surface properties that are unique for each enantiomeric crystal. Likewise, the enantiomers within the unit cell of a racemic crystal can interact in a different way as compared to any one of the enantiomeric crystals resulting in a changed arrangement of the crystal lattice, leading to different surface energies. Disparities in the surface properties of individual organic crystal faces has been discussed in literature (J. J. Segura et al., J. Am. Chem. Soc. 2009, 17853-17859; P. Klitou et al., Cryst. Growth Des., 2022, 22, 6103-6113).

[0009] The process of the present invention harnesses these differences and provides a novel, enantioselective crystal separation process by floatation, in particular a process in which crystals are formed in a preferential crystallization, whereas they are subsequently spatially separated in situ, i.e. as soon as the crystals have nucleated and selectively adsorb to the bubbles of a gas phase that runs through the crystallization vessel. The bubbles eventually form a stable foam phase containing crystals. This process offers a unique advantage over conventional preferential crystallization in that it no longer needs to be stopped before the distomer nucleates. This increases the total theoretical yield of the eutomer recovered.

[0010] In crystal floatation physical phases may be differentiated as follows: in cases where the process is conducted in a melt comprising the enantiomers, or in an auxiliary liquid, which does not have the capacity to dissolve the enantiomeric crystals in appreciable amounts in a technical setting, there is typically a liquid phase and a foam phase. In systems involving a solvent, there is the dissolved phase, the pulp phase and the foam phase. The dissolved phase comprises of the solute that is molecularly dissolved in the solvent. The pulp phase is defined as the total concentration of the solute in the vessel including the crystal mass and the dissolved phase. The foam phase is the result of a buildup of bubbles and contains crystals in suspended form. It may as well contain residual constituents of the pulp phase entrained in the process of separation.

[0011] Accordingly, the present invention relates to a process (herein referred to as embodiment E1) for the separation of enantiomeric crystals comprising the steps of a) charging a vessel with a liquid composition comprising a chiral compound consisting of both enantiomers of one of the chiral compound's constitutional isomers; b) generating an interface between a gas phase and the liquid composition of step a) to produce bubbles that form a foam containing enantiomeric crystals and / or racemic crystals, whereas the foam is either enriched in one of the enantiomeric crystals or the foam is enriched in the racemic crystals; and c) separating the foam produced in step b) from the liquid composition; whereas, if the liquid composition is a melt of the chiral compound or if the liquid composition comprises a solvent and the chiral compound is partly or completely dissolved in the liquid composition, both enantiomers form enantiomeric and / or racemic crystals under the conditions as defined in step b); and whereas the crystals separated in step c), under the conditions as defined in step b), differ in overall surface energy as compared to the counter enantiomeric crystals or racemic crystals.

[0016] As mentioned, hydrophobicity of the crystal surface plays an important role in the differentiation of crystals by floatation. One parameter oftentimes referred to when measuring hydrophobicity (or the wetting capacity) of crystals, is the angle formed on contact of a gas bubble, in a liquid phase, with the solid phase, the so-called contact angle. The Pendent Drop method is used to calculate the contact angle. A liquid droplet is placed on the surface of the sample being analyzed and the angle that forms between these two phases in air is the contact angle. It is determined by the surface energy of a solid surface. Surfaces that have higher surface energies will interact more strongly with a liquid droplet resulting in a greater degree of wetting and lower contact angle. More hydrophobic samples have a lower surface energy therefore interact weakly and the contact angle is high. The surface energy is an intrinsic property of the crystalline sample that is under investigation. Contact angle measurement has been widely used as a descriptive parameter in floatation processes, however, it has several disadvantages, such as the size of the sample surface necessary to perform a measurement. Usually, a droplet needs to be placed on to the surface, which requires the use of relatively large crystals, which may be difficult or impossible to attain. Crystal agglomeration or growth on the surface may prevent the successful measurement as a completely flat surface is needed. Typically, this is overcome by tableting smaller crystals and performing the drop measurement. However, tablets will not have a rougher surface and will contain pores between the microscopic crystals which liquid may seep through.

[0017] An alternative method to measure surface energy is through Inverse Gas Chromatography (IGC, herein below also referred to, and specifically described as, the "IGC method”), which yields more realistic results (Khodakarami et al., Am. Chem. Soc., Omega, 2019, 4, 13319-13329). IGC measures both, average polar surface energy, which is the contribution to the sample surface energy from polar interactions, and the average dispersive surface energy, which is the contribution to the surface energy from non-polar forces such as London or van-der-Waals forces. IGC is a universally adaptable method to determine the overall surface energy (OSE) of the sample with relative ease. A detailed description of the methodology that was used for the determination of overall surface energy values when referred to with regard to the present invention is given below. The method provides information on the interaction across the bulk surface including all facet surface energy contributions. Hydrophobic interactions originate in the dispersive energy contribution. IGC experiments to determine non-polar surface energies have been shown to provide identical or comparable results to dynamic contact angle measurements (Dove et al., Int. J. Pharm., 1996, 138, 199-206).

[0018] The inventors of the present invention found that, for selective floatation to occur, the crystals to be separated must exhibit a difference in OSE, and whereas the OSE is measured by the IGC method depicted herein below.

This can be achieved through structural changes to the aerator, such as pore size, or by suitably choosing the agitation speed or type of agitation.

[0091] Inverse Gas Chromatography (IGC method) A Surface Energy Analyzer (obtained by company Surface Measurement Systems) was used to measure surface energy of dry solid particles. Particles were packed into a quartz column of length 25 cm and width 4 mm. The material mass inside the column is important and depends on the size and surface area of the particles. Brunauer- Emmett-Teller (BET) surface area analysis was performed, and targeted surface coverage was set to 0.05 (unit: amount adsorbed per monolayer capacity) for surface energy experiments. In these experiments the sample mass was between 400-500 mg. The column was conditioned for 120 minutes at 30°C and 0% relative humidity. The flowrate was 8 standard cubic centimeter per minute and the carrier gas was helium.

[0092] The Infinite Dilution IGC (ID-IGC) method was used to determine the surface energy of the solid samples (unit: mJ / m2). This method involved using very small amount of the probe molecules and passing them through the solid sample with a carrier gas. The retention time is defined as the time it takes probe molecules to pass through the sample and be detected by the FID detector with respect to a reference probe, in this case methane, which does not interact with the sample. Two groups of probes were used in this measurement, non-polar probes (n-hexane, n- heptane, n-octane, n-nonane and n-decane) and polar probes (ethyl acetate, dichloromethane). The non-polar probes were used to determine dispersive surface energy and the polar probes the polar surface energy. The retention times are fitted to a Shultz plot from which dispersive and polar contribution to the samples surface energy can be calculated. The average dispersive surface energy was taken from these experiments (reading as max dispersive surface energy) and added the average polar surface energy (reading as specific surface energy).

[0097] The enantioselective crystal floatation experiment was performed in a 100 mL glass jacketed vessel equipped with a magnetic stirrer bar at 600 RPM. Argon bubbles (300-800 pm) were sparged into an undersaturated solution of mandelic acid at 35°C at a flowrate of 10 mL / minute. The solution was cooled to below the saturation limit of 20°C to 10°C at a rate of 0.5°C / minute and held to induce nucleation. The foam phase was mechanically collected, and enantiomeric excess analyzed via chiral HPLC.

[0098] In the experiment a 180 g / L aqueous solution (%ee = 50) was used. As the solution was cooled two nucleation events occurred, one for S-mandelic acid crystals and the other for RS-mandelic acid crystals. The S-mandelic acid crystals adsorbed to the bubble immediately and were taken into the foam phase. RS-mandelic acid crystals remained in the crystallizer and were not taken into the foam. The foam was analyzed via chiral HPLC yielding an %ee of 92.

[0099] The overall surface energy values determined for S-mandelic acid was 40 mJ / m2, for RS-mandelic acid 44 mJ / m2.

Claims

Claims1. A process for the separation of enantiomeric crystals comprising the steps of a) charging a vessel with a liquid composition comprising a chiral compound consisting of both enantiomers of one of the chiral compound's constitutional isomers; b) generating an interface between a gas phase and the liquid composition of step a) to produce bubbles that form a foam containing enantiomeric crystals and / or racemic crystals, whereas the foam is either enriched in one of the enantiomeric crystals or the foam is enriched in the racemic crystals; and c) separating the foam produced in step b) from the liquid composition; whereas, if the liquid composition is a melt of the chiral compound or if the liquid composition comprises a solvent and the chiral compound is partly or completely dissolved in the liquid composition, both enantiomers form enantiomeric and / or racemic crystals under the conditions as defined in step b); and whereas the crystals separated in step c), under the conditions as defined in step b), differ in overall surface energy as compared to the counter enantiomeric crystals or racemic crystals.

2. The process according to claim 1, wherein the overall surface energy is measured by the IGC method depicted in the description.

The process according to claim 1 or 2, wherein the liquid composition in step b) is a solution of the chiral compound in a solvent, under conditions allowing for crystallization to form a mixture of enantiomeric crystals and racemic crystals; and whereas the solution of the enantiomers is enriched with one of the enantiomers to the extent that the crystal formation of the enriched enantiomer is kinetically favored over the formation of the counter enantiomeric crystals.

4. The process according to claim 1 or 2, wherein the liquid composition in step b) is a solution of the chiral compound in a solvent, under conditions allowing for crystallization to form a conglomerate of enantiomeric crystals.

5. The process according to claim 3 or 4, wherein the liquid composition in step b) is a supersaturated solution of the enantiomers.

6. The process according to any one of claims 3 to 5, wherein the solvent is an organic compound or water, or mixtures of organic compounds and water.

7. A process for the separation of enantiomeric crystals comprising the steps ofa) charging a vessel with a liquid composition comprising an auxiliary liquid and enantiomeric crystals and / or racemic crystals of a chiral compound; b) generating an interface between a gas phase and the liquid composition of step a) to produce bubbles that form a foam containing enantiomeric crystals and / or racemic crystals, whereas the foam is either enriched in one of the enantiomeric crystals or the foam is enriched in the racemic crystals; and c) separating the foam produced in step b) from the liquid composition; whereas the crystals separated in step c), under the conditions as defined in step b), differ in overall surface energy as compared to the counter enantiomeric crystals or racemic crystals.

8. A process for the separation of enantiomeric crystals comprising the steps of d) charging a vessel with a liquid composition comprising a melt of the chiral compound; e) generating an interface between a gas phase and the liquid composition of step a) to produce bubbles that form a foam containing enantiomeric crystals and / or racemic crystals, whereas the foam is either enriched in one of the enantiomeric crystals or the foam is enriched in the racemic crystals; and f) separating the foam produced in step b) from the liquid composition; whereas the crystals separated in step c), under the conditions as defined in step b), differ in overall surface energy as compared to the counter enantiomeric crystals or racemic crystals.

9. The process according to claim 7 or 8, wherein the overall surface energy is measured by the IGC method depicted in the description.

10. The process according to any one of claims 1 to 9, wherein the chiral compound is organic in nature.

11. The process according to any one of claims 1 to 10, wherein the bubble size in diameter of the gas phase introduced in step b) is in the range from 0.1 mm to 10 mm.

12. The process according to any one of claims 1 to 11, wherein the gas phase is selected from the group consisting of air, nitrogen, noble gases, hydrogen, carbon monoxide, carbon dioxide, synthesis gas, methane, ammonia, or mixtures thereof.

13. The process according to any one of claims 1 to 12, wherein the temperature in step b) is in the range of from -40°C to 300°C.
