Description

[Technical field]

[0001] The present invention relates to compounds of formula (I) or pharma- ceutically acceptable salts or stereoisomers or tautomers thereof, useful as MALT-1 inhibitors and in the treatment of MALT-1 dependent diseases or disorders. The present invention also relates to processes for the preparation of compounds of formula (I), pharmaceutical compositions containing them, and their use as therapeutic agents for the treatment of diseases, particularly cancer and autoimmune diseases. [Background technology]

[0002] MALT-1 (mucosa-associated lymphoid tissue lymphoma translocation protein 1) is an intracellular signaling protein known from innate immune cells (natural killer cells NK, dendritic cells DC, and mast cells) and adaptive immune cells (T cells and B cells). In association with BCL10 and CARD11, MALT-1 functions as a scaffolding protein for activating the inhibitor of IκB kinase (IKK) complex. The function of MALT-1 is best known in the context of T cell receptor (TCR) signaling, where it mediates nuclear factor kappa-beta (NF-κB) signaling to control lymphocyte activation, survival, and differentiation (S. Hailfinger et al., Immunol. Rev., 232 (2009), pp. 334-347, M. Thome, Nat. Rev. Immunol., 8 (2008), pp. 495-500). In addition to its scaffolding function, MALT-1 is the only gene encoding a paracaspase belonging to the caspase (cysteine-aspartic acid protease) family of proteases that shows high homology to mammalian caspases and plant and fungal metacaspases (AGUren et al., Mol. Cell, 6 (2000), pp. 961-967). Paracaspase MALT-1 plays a key role in the activation of immune cells by certain subtypes of immune receptors, which induces nuclear translocation of NF-κB transcription factor complexes leading to activation of the NF-κB signaling pathway (M. Jaworski et al., Cell Mol. Life Science 2016, 73, 459-473). NF-κB target genes include cytokines and anti-apoptotic proteins that together trigger the receptor to promote the activation, proliferation and survival of activated immune cells, thereby enabling efficient generation of immune responses.

[0003] Studies of BCL10 and MALT1-deficient mice seem to suggest that they play an important role in the signaling cascade from antigen receptors to the transcription factor NF-kB. Furthermore, chromosomal translocations leading to overexpression of BCL10 and MALT-1 or generating the constitutively active fusion protein API2-MALT-1 appear to activate NF-kB in an uncontrolled and stimulus-independent manner. Inhibitors of the proteolytic activity of MALT-1 have been described to have activity in preclinical lymphoma models (M. Vincendeau et al. Int. J. Hematol. Oncol. 2013, 2, 409-417). Furthermore, several publications seem to suggest that MALT-1 and its proteolytic function play an important role in signaling cascades triggered by innate cell receptors such as the dectin receptor, and by G protein-coupled receptors in many cell types.

[0004] International Applications Nos. 2015181747, 2017081641, 2018020474, 2018119036, 2018141749, 2018165385, 2018226150, 2020010252, 2020111087, 2021000855 and 2020208222 report several small molecule compounds and their derivatives that can target MALT-1 and thereby inhibit its activity. However, there is a demand for the development of small molecule MALT-1 inhibitors that could attenuate NF-kB signaling in various proliferative and autoimmune diseases.
