Abstract
Description
[Background technology]
[0001] (CROSS REFERENCE TO RELATED APPLICATIONS) This application claims priority to International Application No. PCT / CN2022 / 073235, filed January 21, 2022, and U.S. Application No. 63 / 310,765, filed February 16, 2022, the contents of which are incorporated by reference in their entireties herein.
[0002] BACKGROUND OF THEINVENTION Rat sarcoma virus (RAS) is one of the most validated cancer targets. However, despite the clinical importance of targeting RAS, discovering potent inhibitors of RAS has been difficult. RAS proteins have long had a reputation of being "undruggable."
[0003] In human cells, three closely related RAS genes, termed HRAS, KRAS, and NRAS, encode four highly related protein isoforms (H-Ras, N-Ras, K-Ras4A, and K-Ras4B). These RAS proteins are guanosine triphosphate degrading enzymes (GTPases) that are involved in a wide range of important molecular and cellular activities, including proliferation, differentiation, and cell death (Front. Oncol., October 18, 2019). RAS proteins constantly cycle between an inactive guanosine diphosphate (GDP)-bound state and an active guanosine triphosphate (GTP)-bound state to relay cellular signals in response to extracellular stimuli. Activation of Ras proteins is also controlled by guanine nucleotide exchange factors (GEFs), which catalyze nucleotide exchange, and GTPase-activating proteins (GAPs), which aid in GTP hydrolysis. Upon activation, RAS directly interacts with and activates several downstream effector pathways, including mitogen-activated protein kinase (MAPK) and phosphatidylinositol 3-kinase (PI3K) pathways. However, mutations in RAS typically render it GAP-independent and "lock" RAS in an active GTP-bound state, inhibiting the guanine exchange cycle, thereby activating downstream signaling pathways leading to tumor cell proliferation (PNAS, March 4, 2014 111 (9) 3401-3406; Nat Rev. Drug Discov., 2020 Aug; 19(8): 533-552).
[0004] Mutations in RAS genes occur frequently in solid and hematological malignancies, with at least one isoform mutation present in approximately 20% of all tumors. Most oncogenic mutations in RAS isoforms are missense gain-of-function mutations at codons G12, G13, or Q61, although their frequency of such mutations varies among RAS isoforms (Genes 2021 Jun; 12(6): 899). Mutations at amino acids G12, G13, Q61, and A146 are found in a variety of human cancers, including lung, colon, and pancreatic cancers (Cox et al., Nat. Rev. Drug Discov., 2014, 13(1): 828-51). However, recent biochemical analyses have demonstrated that these mutant proteins still require nucleotide cycling for activation, based on their intrinsic GTPase activity and / or partial sensitivity to exogenous GTPases. Therefore, mutant RAS proteins are sensitive to inhibition of upstream factors such as SOS or SHP2, upstream signaling proteins required for RAS activation (Nat Rev Drug Discov. 2020 Aug; 19(8): 533-552).
[0005] Son of Sevenless (SOS) has two homologous genes, SOS1 and SOS2. SOS1 encodes a protein that is a guanine nucleotide exchange factor (GEF) for RAS proteins. The product of this gene plays a key role in the activation of RAS proteins by mediating the exchange of GDP for GTP. SOS2 is the homolog of SOS1 in mammalian cells. It also functions as a GEF for the activation of RAS family proteins. SOS1 and SOS2 are approximately 70% identical in amino acid sequence. SOS2 knockouts in mice are viable, whereas SOS1 knockouts are embryonic lethal. Using a tamoxifen-inducible SOS1 knockout mouse model to investigate the role of SOS1 and SOS2 in adult mice, we demonstrated that while SOS1 knockout mice are viable, SOS1 and SOS2 (SOS1 / 2) double knockouts are lethal, suggesting functional redundancy and that selective inhibition of SOS1 may have a sufficient therapeutic index for the treatment of SOS1-RAS activated diseases (Mol Cell Biol., 2013 Nov; 33(22): 4562-4578).
[0006] SOS proteins bind to RAS not only through a catalytic binding site that promotes nucleotide exchange, but also through an allosteric site that binds to GTP-bound RAS family proteins that enhance the catalytic function of SOS (PNAS, November 7, 2006, 103 (45) 16692-16697). Binding to the allosteric site relieves steric blockage of the catalytic site and is therefore required for full activation of the catalytic site. Retention of the active conformation at the catalytic site after interaction with the allosteric site is maintained in isolation, as interactions of the key domains are strengthened in the active state. Selective pharmacological inhibition of the interaction between SOS1 and RAS is expected to prevent activation of RAS signaling via SOS1.
[0007] In the present invention, the applicant provides novel compounds that inhibit the interaction between SOS1 and RAS proteins, thereby preventing KRAS from recycling to its active GTP-bound form. These novel compounds provide therapeutic benefits for a wide range of cancers, particularly RAS protein-associated cancers.

[0068] As used herein, the term "heteroaryl" refers to a monocyclic or fused ring (i.e., rings sharing adjacent pairs of atoms) of 5 to 12 ring atoms containing 1, 2, 3, or 4 ring heteroatoms selected from N, O, or S, with the remaining ring atoms being C, and further having a fully conjugated pi-electron system. Examples of unsubstituted heteroaryl groups include, but are not limited to, pyrrole, furan, thiophene, imidazole, oxazole, thiazole, pyrazole, pyridine, pyrimidine, quinoline, isoquinoline, purine, triazole, tetrazole, triazine, carbazole, benzimidazole, benzoxazole, benzthiazole, indazole, and quinazoline. Heteroaryl groups can be substituted or unsubstituted. Examples of substituents are halo, alkyl, alkynyl, haloalkyl, cycloalkyl, heterocycloalkyl, hydroxyl, alkoxyl, amino, amine, cyano, alkenyl, cycloalkenyl, heterocycloalkenyl, aryl, heteroaryl, carbonyl, alkoxycarbonyl, aminocarbonyl, hydrocarboxyl, alkylthio, -D(deuterium), oxo, -C(O)-(CH 2 )n-OR, -(CH 2 ) n -S(O) 2 -R, -C(O)-R, -C(O)-NR, -(CH 2 ) n -OR, -NR-C(O)-(CH 2 ) n -, CD 3 Includes, but is not limited to, -O-, and -CN (where R is H or alkyl).
[0069] As used herein, the term "oxo" refers to an oxygen atom (i.e., =O) as a divalent substituent. When it is attached to a carbon atom it results in a carbonyl group, when it is attached to a sulfur atom it results in a sulfoxide or sulfone group, or when it is attached to a nitrogen atom it results in an N-oxide group. In some embodiments, heterocyclic groups can be optionally substituted with one or two oxo substituents.
[0070] The above defined groups may include prefixes and / or suffixes that form substituents commonly used and further understood in the art. As an example, the term "haloalkoxy" or "haloalkyloxy" refers to a haloalkyl group attached to the parent molecular moiety through an oxygen atom. The term "(haloalkyl)oxyalkyl" refers to an alkyl group substituted with one, two, or three (haloalkyl)oxy groups. As another example, the term "hydroxyalkamino" refers to an amino group substituted with one or two hydroxyalkyl groups.
[0071] As used herein, the term "stereoisomer" refers to an isomer of identical constitution that differs in the arrangement of atoms in space. Enantiomers and diastereomers are examples of stereoisomers. The term "enantiomer" refers to one of a pair of molecular species that are mirror images of each other and are not superimposable. The term "diastereomer" refers to a stereoisomer that is not a mirror image. The term "racemate" or "racemic mixture" refers to a composition composed of equimolar amounts of two enantiomeric species, which composition lacks optical activity. When a disclosed compound is named or depicted by structure without showing stereochemistry, it is understood that the name or structure includes all possible stereoisomers, geometric isomers, and combinations thereof, including essentially pure stereoisomers or geometric isomers.
[0072] As used herein, the term "tautomer" or "tautomeric form" refers to structural isomers of different energies that are interconvertible via a low energy barrier. When tautomerization can occur (e.g., in solution), a chemical equilibrium of tautomers can be reached. "Optional" or "optionally" means that the subsequently described event or circumstance may occur, but does not necessarily occur, and the description includes cases where the event or circumstance occurs and cases where it does not occur. For example, "a heterocyclyl group optionally substituted with an alkyl group" means that the alkyl may be present, but does not necessarily have to be present, and the description includes situations where the heterocyclyl group is substituted with an alkyl group and situations where the heterocyclyl group is not substituted with an alkyl group.
[0073] As used herein, the phrase "optionally substituted" means that the substitution is optional, and thus includes both unsubstituted and substituted atoms and moieties. A "substituted" atom or moiety indicates that any hydrogen of the specified atom or moiety can be replaced with one selected from the specified substituents, provided that the normal valence of the specified atom or moiety is not exceeded and the substitution produces a stable compound. For example, when a methyl group is optionally substituted, three hydrogen atoms on the carbon atom can be replaced with a substituent.
[0074] As used herein, the term "pharmaceutical acceptable" means suitable for use in humans or other mammals.
[0075] The term "pharmaceutical acceptable salt" used herein refers to salts with known cations or anions, and can be applied to the use in the field.Suitable salts with bases, for example, salts formed from compounds of formula (I) containing carboxyl groups, include salts of alkali metals (such as sodium, potassium), alkaline earth metals (such as calcium, magnesium), ammonium, and amines.Suitable salts with acid radical additives, for example, salts formed from compounds of formula (I) containing amino, include salts formed with inorganic acids such as hydrochloride, sulfate, sulfite, phosphate, hydrogen phosphate, and nitrate, and salts formed with organic acids such as acetic acid, malic acid, tartaric acid, citric acid, lactic acid, salicylic acid, oxalic acid, etc.
[0076] Furthermore, if a compound is obtained as an acid addition salt, the free alkali can be obtained by alkalizing a solution of the acid salt. In contrast, if the product is a free alkali, the addition salt can be prepared by dissolving the free alkali in a suitable organic solvent and treating the solution with an acid, which is consistent with the conventional process of preparing an acid addition salt from an alkaline compound. Those skilled in the art will recognize the various synthetic methods that can be used to prepare acceptable addition salts without undue experimentation.
[0077] In addition, when the compound of the present invention has a carboxyl group, the compound can be converted to the corresponding alcohol (e.g., C 1-6 By reacting the carboxylic acid with an alcohol) a pharma- ceutically acceptable ester can be obtained by a conventional method (for example, a condensation reaction between a carboxylic acid and an alcohol).
[0078] As used herein, "pharmaceutical composition" refers to a mixture of one or more of the compounds described herein, or their pharmaceutically acceptable salts or prodrugs, and other chemical components, such as pharmaceutically acceptable excipients. The purpose of a pharmaceutical composition is to facilitate the administration of a compound to an organism.
[0079] As used herein, the term " pharmaceutically acceptable excipient " refers to the inert substance added to pharmaceutical compositions to further facilitate the administration of compound. The examples of excipient include, but are not limited to, calcium carbonate, calcium phosphate, various sugars and starches, cellulose derivatives, gelatin, vegetable oil and polyethylene glycol.

[0146] The following examples were synthesized using the methods described above, except for example 174, which used the same method as example 159. JPEG2025504495000082.jpg159170
[0147] Biological Examples Example A. Protein-Protein Interaction (PPI) Assays This example demonstrates that exemplary compounds of the invention inhibit the interaction of SOS1 and KRAS proteins.
[0148] The ability of compounds of formula (I) to inhibit the interaction of SOS1 and KRAS was measured using the SOS1-KRAS Binding Assay Kit (CAT # 63ADK000CB16PEG, Cisbio). This assay utilizes HTRF (homogeneous time-resolved fluorescence) technology to allow simple and rapid characterization of compounds and antibody blockers in a high-throughput format. The interaction between Tag1-SOS1 and Tag2-KRAS-G12C is detected using anti-Tag1-Terbium antibody (HTRF donor) and anti-Tag2-XL665 antibody (HTRF acceptor). When the binding of SOS1 and KRAS-G12C brings the donor and acceptor antibodies into close proximity, excitation of the donor antibody induces fluorescence resonance energy transfer (FRET) to the acceptor antibody, which then emits light specifically at 665 nm. This specific signal is directly proportional to the extent of KRAS-G12C / SOS1 interaction.
[0149] The assay was performed according to the manufacturer's instructions. Compounds at an initial concentration of 10 mM in DMSO were first serially diluted in a 1:3 ratio using assay buffer on a 384-well plate to create a total of nine concentrations with 20 μL of each deposition. Blank buffer was included as a negative control. The final compound solution was a buffer solution containing 1% DMSO. Then, each well of the 384-well plate was incubated for 15 min with 2 μL of compound solution, 4 μL each of tagged SOS1 and KRAS(G12C) protein stocks, and a solution of 10 μM GTP. A mixture of 10 μL of anti-Tag1X665 and anti-Tag1-Terbium antibodies was introduced into each individual well. After 2 h of incubation at room temperature, fluorescence readings were measured using a PelkinElmer multiplate reader VICTOR Nivo.
[0150] Excitation was performed in the range of 245-395 nm, emission from the donor (emission 1) was detected at (657.5-672.5) nm, and emission from the acceptor (emission 2) was detected at (606.5-623.5) nm. The HTRF ratio was calculated using the following formula: (emission 1 @665 nm / emission 2 @620 nm) x 10000. Thus, compounds that inhibit the KRAS-G12C / SOS1 interaction cause a decrease in the HTRF signal ratio. The IC was then calculated using Graph pad Prism software. 50 The values were calculated, and the results are shown in Tables 1 and 2.
[0151] Example B: Phosphorylated ERK (p-ERK) Assay This example demonstrates that representative compounds of the present invention inhibit KRas-mediated GTP nucleotide exchange mediated by SOS1, thereby inhibiting KRas activity and thereby inhibiting the generation of the downstream effector pERK.
[0152] Hela cells (23,000 / w) were seeded in black clear flat-bottom 96-well cell culture plates (Coming, #3904) and cultured overnight at 37°C. On day 2, cells were dosed with compounds of formula (I) at a starting concentration of 10 μM and serially diluted 3-fold for a total of 9 concentrations. Cells were incubated with compounds dissolved in DMSO for 1 h at 37°C. Cells were immediately fixed by adding 100 μL of 8% formaldehyde to all wells in a fume hood and plates were incubated for 1 h at room temperature. Formaldehyde was removed from the plates and the cells were washed 5 times with 200 μL of Triton Washing Solution (0.1% Triton in PBS) for 5 min each time to permeabilize them. Cells were then blocked with 150 μL of Odyssey Blocking Buffer (LI-COR Biosciences #927-50010) at room temperature on a shaker. The blocking buffer was discarded and 50 μL of primary antibodies pERK (cell signaling Technology #4370; rabbit, 1:500) and β-actin (AF5001; mouse, 1:2000) diluted in Odyssey blocking buffer was added. The plate was incubated overnight at 4 °C on a shaker.
[0153] On day 3, the primary antibody solution was removed. Each plate was washed 5 times with 200 μL of 1xPBST (PBS + 0.1% Tween 20) and incubated with 50 μL of secondary antibody. Anti-rabbit (LI-COR Biosciences #926-32211) and anti-mouse (LI-COR Biosciences #68070) were diluted 1:1000 in Odyssey blocking buffer and agitated on a shaker at room temperature for 1 hour (protected from light). The secondary antibody solution was removed and each plate was washed 5 times with PBST. Any remaining liquid was discarded and the plates were photographed using a Licor Odyssey machine with the focal length set to 3.5 mm and both 800 nm and 700 nm filters, according to the manufacturer's instructions. The β-actin normalized scan value for each well was divided by the average of the vehicle wells to calculate the % of p-ERK inhibition. IC was then calculated using Graph pad Prism software. 50 The p-ERK IC values were calculated. The results are shown in Tables 1 and 2. In general, the p-ERK IC 50 Data is PPI IC 50 Matches the data.
