Description

[Technical field]

[0001] CROSS-REFERENCE TO RELATED APPLICATIONS This application claims priority to U.S. Provisional Patent Application No. 62 / 832,133, filed April 10, 2019, and U.S. Provisional Patent Application No. 62 / 836,659, filed April 20, 2019, the disclosures of each of which are incorporated by reference in their entireties herein.

[0002] The present disclosure generally relates to novel inhibitors of phosphatidylinositol 3-kinase (PI3K). More specifically, the present invention further relates to the preparation of the disclosed PI3K gamma inhibitor analogs and their use in pharmaceutical compositions for the treatment of various diseases, conditions and disorders associated with PI3K gamma activity. [Background technology]

[0003] Class I phosphoinositide 3-kinases (PI3Ks) regulate phosphatidylinositol 4,5-bisphosphate (PIP2) phosphorylation. PI3K converts PIP2 to the scaffold-binding element phosphatidylinositol (3,4,5)-trisphosphate (PIP3). PIP3 plays an important regulatory role in cell survival, signal transduction, control of membrane trafficking, and other functions. (Di Paolo, G. et al. Nature 2006, 443, 651; Parker, PJ et al. Biochem. Soc. Trans. 2004, 32, 893; Hawkins, PT et al. Biochem. Soc. Trans. 2006, 34, 647; Schaeffer, EM et al. Curr. Opin. Immunol. 2000, 12, 282). Its dysregulation leads to various disease states such as cancer, inflammatory and autoimmune disorders.

[0004] Class I PI3K consists of four kinases that are further divided into two subclasses. Class 1A PI3K consists of three closely related kinases, PI3Kα, β, and δ, that exist as heterodimers consisting of a catalytic subunit (p110α, β, or δ) and one of several regulatory subunits. They generally respond to signaling through receptor tyrosine kinases (RTKs). The PI3Kγ single class 1B isoform responds primarily to G protein-coupled receptors (GPCRs) and is composed of a p110γ catalytic subunit and one of two distinct regulatory subunits. PI3Kα and PI3Kβ are ubiquitously expressed across a wide variety of tissue and organ types. The expression pattern of PI3Kδ is restricted to the spleen, thymus, and peripheral blood leukocytes (Knight, Z. et al. Cell 2006, 125, 733). PI3Kγ is found primarily in white blood cells, but also in skeletal muscle, liver, pancreas, and heart (Cantly, C. Science 2002, 1655).

[0005] Recently, Schmid showed that p110γ can be activated by receptor tyrosine kinases and TLR / IL1R in myeloid cells. PI3kγ serves as a single convergence point to promote tumor inflammation and progression. In animal models, treatment of mice with selective PI3Kγ inhibitors inhibited the catalytic activity and adhesion of myeloid cell p110γ to VCAM-1, due to effects on the tumor microenvironment instead of direct inhibition of tumor cells. (Schmid,MC et al. Cancer Cell 19,715-727,2011,PLoS ONE 8,e60226,2013). Furthermore, inhibition of PI3Kγ enhanced proinflammatory cytokines and reduced expression of immunosuppressive factors in tumors and TAMs. Kaneda and De Henau concluded that PI3Kγ controls the TAM switch between immunosuppression and immunostimulation. (Kaneda,MMet al.Nature 539,437-442,2016;De Henau,O.et al.Nature 539,443-447,2016).

[0006] Camps et al. described that treatment with the selective PI3Kγ inhibitor AS-60485023 suppressed the progression of joint inflammation and damage in two different mouse models of rheumatoid arthritis (Camps M, et al., Nat. Med. 2005, 11, 936-943).

[0007] Based on studies of cellular levels and efficacy observed in various disease models, PI3Kγ inhibitors could potentially be used to treat a variety of diseases such as inflammation, metabolism and cancer (Cushing, TD, et al, J. Med. Chem. 2012, 55, 8559-8581; Ruckle, T. et al, Nat. Rev. Drug Discovery 2006, 5, 903-918; Stark, AK et al, Curr. Opin. Pharmacol. 2015, 23, 82-91).

[0008] PI3K gamma selective inhibitors have been disclosed in recent years. IPI-549 is in clinical trials as a monotherapy and combination cancer immunotherapy with the checkpoint PD-1 inhibitor nivolumab (Evans, CA et al, ACS Med. Chem. Lett. 2016, 7, 862-867). Pemberton N et al reported that selective PI3K gamma inhibition resulted in dose-dependent inhibition of LPS-induced airway neutrophilia in rats (Pemberton, N. et al, Journal of Medicinal Chemistry 2018, 61, 5435-5441). The engineering of a series of azaisoindolinones by Come, JH et al. provided CNS-permeable and orally bioavailable PI3K gamma inhibitors that were effective in a mouse EAE model and demonstrated that PI3K gamma inhibition has potential for the treatment of multiple sclerosis (Come, JHet al, Journal of Medicinal Chemistry 2018, 61, 5245-5256).

[0009] PI3K gamma inhibitors with related structures but different binding modes have been disclosed in recent years. For example, WO2015048318 discloses (R)-6-(1-(2,2-difluoroethyl)-1H-pyrazol-4-yl)-4,7,7-trimethyl-2-(5-(2,2,2-trifluoro-1-hydroxyethyl)pifudin-3-yl)-6,7-dihydro-5H-pyrrolo[3,4-b]pyridin-5-one as a selective inhibitor of PI3K gamma. WO2011087776 disclosed isoindolinone inhibitors of phosphatidylinositol 3-kinase. WO2017153527 disclosed novel inhibitors of phosphatidylinositol 3-kinase gamma. The discovery of highly isoform-selective, orally bioavailable phosphoinositide 3-kinase (PI3K) gamma inhibitors and the design and synthesis of a novel series of 3-kinase gamma (PI3Kγ) inhibitors have been reported (Journal of Medicinal Chemistry 2018, 61, 5435-5441 and Journal of Medicinal Chemistry 2018, 61, 5245-5256).

[0010] However, these reported PI3K gamma inhibitors have less than optimal potency, selectivity and pharmacokinetic properties. Thus, there is a need in the art to develop new PI3K gamma inhibitors with improved potency, selectivity and pharmacokinetic properties. Summary of the Invention

In some embodiments, when a mixture of stereoisomers is separated by HPLC, the resulting individual stereoisomers or mixtures can be assigned sequential labels (e.g., first enantiomer, and second enantiomer), which order should be understood to imply the order of the isomers eluted from the HPLC column. The absolute stereochemistry of the first enantiomer and the second enantiomer can be obtained by known methods.

[0194] Biological Examples Activity testing was performed in the examples below using methods described herein and well known in the art.

[0195] Compound characterization This example compares the biological activity of compounds of formula (I), including (Ia), (Ib), (Ic), and (Id).

[0196] The enzymatic activity of different PI3K isoforms was measured to compare the inhibitory potency and selectivity of the compounds provided herein with the selectivity for PI3K isoforms, particularly PI3K gamma over delta.Solubility was also measured to access the drug potential of the test compounds in terms of physicochemical properties.

[0197] Each of these biological experiments is described below.

[0198] Enzyme activity of PI3K isoforms: The enzymatic activity of class I PI3K isoforms in the presence of the compounds in Table 1 was measured using the ADP-Glo luminescent assay for PI3Kα, PI3Kβ, PI3Kδ and PI3Kγ with an ATP concentration at 25 μM. IPI-549 was used as the reference compound. Compounds are tested in duplicate from 3-fold dilutions for 10 doses of 1 or 10 μM.

[0199] Compound preparation: Compounds are diluted in 100% DMSO to 100x the final desired highest inhibitor concentration in the reaction. 100 μl of this compound dilution is transferred to wells of a 96-well plate and serially diluted in DMSO to a total of 10 concentrations.

[0200] Kinase Reaction: Prepare a solution of PI3K in 1x kinase buffer at twice the final concentration of each reagent in the assay. Control wells contain 1x kinase buffer without enzyme. Prepare substrate solutions of PIP2 substrate and ATP in 1x kinase reaction buffer at twice the final concentration. Add concentrated substrate solution to each well to final concentrations. Cover assay plate, swirl to mix and incubate at room temperature for 1 hour. Add 5μl ADP-Glo Reagent to each well to stop the reaction. Mix briefly by swirl quickly and shake slowly on a shaker for 120 minutes. Add 10ul kinase detection reagent to each well, shake for 1 minute and equilibrate for 30 minutes before reading on a luminescence plate reader.

[0201] Data Analysis: RLU values are converted to percent inhibition values as follows: Percent Inhibition = (Max-Sample RLU) / (Max-Min) * 100. "Min" - no enzyme control; "Max" - DMSO control Data is fitted with XLFit excel add-in version 5.4.0.8 to obtain IC50 values. Y = Bottom + (Top-Bottom) / (1 + (IC50 / X)^HillSlope). For compounds of the present disclosure, as exemplified in the Examples, IC50 values are listed in Tables 2 and 3. 50 The values were shown.

[0202] Kinetic solubility test: Stock solutions of test compounds and control compound progesterone were prepared at a concentration of 10 mM in DMSO. 15 μL of stock solution (10 mM) was placed in the appropriate 96-well rack in sequence. 485 μL of PBS pH 7.4 was added to each vial of the capless solubility sample plate. The assay was performed in singlets. One stir stick was added to each vial and sealed using a molded PTFE / silicone plug. The solubility sample plate was then shaken at 25° C. and 1100 rpm for 2 hours. After the completion of the 2 hours, the samples from the solubility sample plate were transferred to a filter plate. All samples were filtered using a vacuum manifold. A 5 μL aliquot was taken from the filtrate followed by the addition of 495 μL of a mixture of H2O and acetonitrile (1:1) containing the internal standard. A fixed percentage of ultrapure water was used to dilute the diluent according to the peak shape. The dilution factor was varied according to the solubility value and LC-MS signal response.

[0203] Preparation of 3 μM Standards (STD): From the 10 mM DMSO STD plate, 6 μL was transferred to the remaining blank plate, then 194 μL of DMSO was added to the plate to have a STD concentration of 300 μM. From the 300 μM DMSO STD plate, 5 μL was transferred to the remaining blank plate, then 495 μL of a mixture of H2O and acetonitrile (1:1) containing internal standards was added to the plate to have a final STD concentration of 3 μM. A fixed proportion of ultrapure water was used to dilute the diluent according to the peak shape. The concentration of the standard samples was varied according to the LC-MS signal response.

[0204] Sample analysis procedure: The plate was placed in a well plate autosampler. Samples were evaluated by LC-MS / MS analysis.

[0205] Data Analysis: All calculations were performed using Microsoft Excel.

[0206] The filtrate was analyzed and quantified against standards of known concentration using LC coupled with mass spectral peak identification and quantification. Solubility values of the test and control compounds were calculated as follows: JPEG0007644023000127.jpg27141 Any values for compounds not within the specified limits were rejected and the experiment was repeated. Solubility values are shown in Table 2.

[0213] Pharmacokinetics in mice Compound 34 dosing solution was prepared at 2 mg / mL in 5% DMSO / 40% PEG400 / 55% water. Dosing solution was administered to male CD-1 mice via oral gavage (PO) at 20 mg / kg. Blood and brain tissue samples were collected at 0.5, 2, and 8 hours after dosing. Blood samples were then centrifuged for 5 minutes in a refrigerated centrifuge at 4° C. Brain samples after blood removal were homogenized in water. Plasma and brain homogenate samples were then analyzed using LC / MS / MS to determine the concentration of Compound 34. The results are listed in Table 6. Compound 34 showed good exposure in mouse brain tissue and good blood-brain penetration. JPEG0007644023000134.jpg41170

[0214] Antitumor Efficacy of Compounds Described Herein in Syngeneic Mouse Tumor Models Compound 34 dosing solution was prepared at 2 mg / mL in 5% DMSO / 40% PEG400 / 55% water. BALB / c mice were subcutaneously inoculated in the fourth mammary pad with 4T1 cells for tumor development. The day after tumor cell inoculation, mice were assigned to two groups using stratified randomization with 10 mice per group based on body weight and inoculation order. Treatment of mice by oral gavage with either vehicle or Compound 34 at 100 mg / kg was initiated on the day of randomization. Tumor size was measured three times a week during treatment. Tumor volume was calculated as length x width. 2 The tumor growth was calculated by the formula: ##EQU1## As shown in FIG. 1, treatment with compound 34 significantly inhibited tumor growth.

[0215] Efficacy study of compound 34 against MOG35-55-induced EAE in C57BL / 6 mice Objective: Experimental autoimmune encephalomyelitis (EAE) is the most common animal model of human multiple sclerosis to evaluate the efficacy of therapeutic strategies. The aim of this study was to evaluate the efficacy of compound 34 in a myelin oligodendrocyte glycoprotein (MOG)-induced EAE mouse model.

[0216] Experimental procedure: Animal model: Mix 10 mL of complete Freund's adjuvant (CFA) with 50 mg of Mycobacterium tuberculosis H37Ra to a final concentration of 6 mg / mL. Synthetic peptide MOG 35-55 was dissolved in saline to a final concentration of 4 mg / ml. Using a high-speed homogenizer (30,000 rpm, IKA Equipment Shanghai Co., Ltd.), an equal volume of mixed CFA and MOG was 35-55 The solution is emulsified on ice for 1 hour.

[0217] Forty female C57BL / 6 mice, 6–8 weeks of age, were randomly assigned into four groups as follows: JPEG0007644023000135.jpg59170

[0218] Mice were inoculated with 0.1 mL of MOG at three sites on the shaved back: one along the midline of the back between the shoulders, and one on each side of the midline. 35-55 The emulsion was injected subcutaneously. Bordetella pertussis toxin (PTX) (400 ng in 200 μL of PBS) was injected intraperitoneally on day 0 and 48 hours after immunization.

[0219] Treatment: Mice in groups 3 and 4 were administered compound 34 according to the designated dosing regimen. Mice in group 2 were administered the positive compound FTY-720 as a reference. Mice in group 1 served as vehicle control. Dosing began on day 10 of disease onset and continued for 14 days.

[0220] Assessment: Body weights were measured before immunization. Body weights were measured daily from day 8 until the end of the study. EAE clinical scores were recorded before immunization. Scores were recorded daily from day 8 until the end of the study. Scoring system for clinical assessment of EAE: JPEG0007644023000136.jpg54164 a Limp tail: complete tail relaxation and no curling at the tip of the tail when the mouse is picked up. b Hind limb weakness: observed as a wobbly gait, the objective sign is the mouse's hind limbs dropping through the top of the wire cage while walking. c Partial hindlimb paralysis: Mice can no longer maintain a standing position or walk using their hindlimb, but can still move one or both limbs to some extent. d Complete paralysis of the hind limbs: Complete loss of movement of the hind limbs. The mouse only drags its forelimbs. Data Analysis: Clinical scores at baseline, day 8, and during treatment were analyzed using GraphPad Prism software.

[0221] result As shown in FIG. 2, treatment with compound 34 at 20 mg / kg and 80 mg / kg BID significantly improved clinical symptoms in EAE-induced mice, with efficacy comparable to that of fingolimod (FTY-720) at 1 mg / kg.
