Abstract
Description
[Technical field]
[0001] The present invention relates to an efficient method for preparing a crystalline form of sodium 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetate trihydrate. [Background technology]
[0002] Cytomegalovirus (CMV) is a common opportunistic infection that causes significant morbidity and preventable mortality after solid organ transplantation and allogeneic hematopoietic stem cell transplantation.
[0003] (S)-(+)-{8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-trifluoromethyl)phenyl]-3,4-dihydroquinazolin-4-yl}acetic acid or 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetic acid, known as letermovir, was developed in WO 2004 / 096778 as an antiviral drug, particularly to combat infections caused by human cytomegalovirus (HCMV). Salts of 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetic acid are obtained in crystalline or amorphous form (WO 2013 / 127971 A1). In particular, some of the sodium or calcium salts of letermovir are obtained mainly in amorphous form. In the case of the sodium salt of letermovir, some crystalline sodium hydrate is also obtained.
[0004] However, as is evident from the independent claims, there remains a need for an effective method for preparing crystalline forms of sodium 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetate trihydrate. Summary of the Invention
[0005] In one embodiment, the present invention provides a process for the preparation of ... [ka] The present invention relates to crystalline sodium 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetate trihydrate. No alcohol, particularly methanol or ethanol, is involved in any step of the preparation of said crystalline sodium 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetate trihydrate. This results in a product that does not contain said alcohol.
[0006] In another aspect, the present invention provides a method for preparing a crystalline form of sodium 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetate trihydrate of formula (I), comprising the steps of: [ka] Follow these steps: Step 1) preparing a solution of 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetic acid in a mixture of (C1-C6) alkyl acetate and (C1-C6) dialkyl ether, the molar ratio of (C1-C6) alkyl acetate:(C1-C6) dialkyl ether being 1:1 to 1:3 at a concentration of 0.3M to 0.7M, and the temperature of the solution being preferably in the range of 30°C to 60°C; Step 2) adding 1.0 to 2.0 molar equivalents of aqueous sodium hydroxide to the solution resulting from step 1 relative to the 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetic acid content to obtain a mixture; Step 3) stirring the mixture resulting from step 2 at a temperature ranging from 30° C. to 60° C. for at least 30 minutes to obtain a suspension containing a solid compound; Step 4) separating the solid compound from the suspension resulting from step 3; and Step 5) Dry the solid compound resulting from step 4 at a temperature ranging from 30° C. to 60° C. for at least 1 hour to obtain crystalline sodium 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetate trihydrate. The present invention relates to a method comprising the steps of:
[0007] Prior art methods required separate preparation of the sodium salt of letermovir, which was then treated in an aqueous alcohol such as methanol or ethanol for one week, after which a crystalline solid was obtained and filtered off. In the present disclosure, it is demonstrated that the prior art crystalline solid is not letermovir sodium trihydrate, but a mixed alcohol and water solvate form of letermovir.
[0008] When methanol was used as the solvent, a crystalline form of 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetate sodium methanol monohydrate was formed, as shown in Figure 1. The X-ray structural data of the crystalline letermovir sodium methanol monohydrate is listed in Table 1.
[0009] When ethanol was used as the solvent, a crystalline form of 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetate sodium ethanol monohydrate was formed, as shown in Figure 2. The X-ray structural data of the crystalline letermovir sodium ethanol monohydrate is listed in Table 2.
[0010] The crystals obtained, i.e., Letermovir sodium methanol monohydrate and Letermovir sodium ethanol monohydrate, need to be dried at room temperature and ambient humidity for two weeks for conversion to crystalline 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetate sodium trihydrate as shown in FIG. 4.
[0011] Also, the use of water as a co-solvent does not result in the formation of letermovir sodium trihydrate. Thus, there are no successful methods in the prior art that use water as a co-solvent to produce letermovir sodium salt trihydrate.
[0012] In the present invention, the following parameters are tested during development and optimization: - Solvent / anti-solvent ratio -Addition of antisolvent (rate and timing of addition) -API concentration -Addition of NaOH (directly or dissolved in water) -Water amount -Reaction time -Reaction temperature - Seeding (optional)
[0013] According to the above process of the present invention, crystalline sodium 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetate trihydrate of formula (I) is directly obtained by using water as a co-solvent.
[0014] The method of the present invention has the following technical advantages: Sodium-2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetate trihydrate can be prepared directly from letermovir base. -The entire process requires a relatively short reaction time (4 hours + 15 hours for drying). -The process directly gives letermovir sodium trihydrate (no other transition forms). -The process is reproducible and scalable.
[0015] In another aspect, the present invention relates to pharmaceutical compositions comprising said crystalline sodium 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetate trihydrate and methods for the preparation of said pharmaceutical compositions.

[0052] The crystalline 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetate sodium trihydrate may exist in various polymorphic forms. Polymorphs are different crystalline forms of the same compound that may have substantially different physicochemical properties, such as solubility, stability, and bioavailability. Evaluation of the polymorphism of a drug substance plays an important role in formulation studies, since polymorphism can affect drug behavior. For example, the rate of dissolution of a drug substance affects the bioavailability of the final product. And the solubility depends on the polymorphism of the drug substance. Different polymorphs may have different solubilities, and therefore the corresponding pharmaceutical formulations may have different bioavailabilities.
[0053] Various methods can be used to study polymorphism. Such methods include microscopy, IR spectroscopy, Raman spectroscopy, solid-state NMR, TGA, DSC, XRPD, PDF and other techniques. A combination of different techniques can be applied. In particular, PXRD is a powerful technique to study polymorphism. X-rays are reflected from a crystal only if the angle between the beam and the plane in the crystal satisfies the Bragg condition. There are an infinite number of possible planes in a crystal. Each molecular repeat gives a unique set of reflections and therefore produces a unique pattern that can be recorded as a spectrum.
[0054] However, conventional XRPD analysis only gives the average structure of the material, e.g. average positions, displacement parameters and occupancies, and cannot provide information about local disorder in the material. For this purpose, the pair distribution function (PDF) can be used, which gives the probability of finding an atom at a certain distance from a given atom. The PDF is a Fourier sine transform of the total scattering diffraction pattern and provides information about the average interatomic distance, the disorder or distortion of the structure, and the average coordination properties. Thus, the PDF can distinguish different solid forms of the same compound that cannot be distinguished by conventional PXRD analysis. In particular, various amorphous forms characterized by different degrees of disorder can be determined by PDF analysis (Boetker et al.Pharmaceutics 2012,4,93-103).

[0069] The term capsule as described herein refers to a specific container or enclosure made of, for example, methylcellulose, polyvinyl alcohol, or modified gelatin or starch to hold or contain a composition containing an active ingredient. Hard shell capsules are typically made of a mixture of relatively high gel strength gelatin derived from bone or pigskin. The capsule itself may contain small amounts of dyes, opacifiers, plasticizers and / or preservatives. Under tablet is understood a compressed or molded solid dosage form containing the active ingredient together with a suitable diluent. Tablets can be prepared by compression of mixtures or granulations obtained by wet granulation, dry granulation, or by compression as is well known to those skilled in the art.
[0070] Oral gel refers to the active ingredients dispersed or solubilized in a hydrophilic semi-solid matrix. Powder for dissolution refers to a powder mix containing the active ingredients and suitable diluents that can be suspended in, for example, water or juice.
[0071] A suitable diluent is a substance that usually constitutes the majority of the composition or dosage form. Suitable diluents include sugars such as lactose, sucrose, mannitol, and sorbitol, starches derived from wheat, corn, rice, and potato, and celluloses such as microcrystalline cellulose. The amount of diluent in the composition can range from about 5 to about 95% by weight, preferably about 25 to about 75% by weight, more preferably about 30 to about 60% by weight of the total composition.
[0072] The term disintegrant refers to materials added to a composition to aid in the break-down (disintegration) and release of the pharma- ceutical active ingredients of a pharmaceutical product. Suitable disintegrants include starch, "cold water soluble" modified starches such as sodium carboxymethyl starch, natural and synthetic gums such as locust bean, karaya, guar, tragacanth and agar, cellulose derivatives such as methylcellulose and sodium carboxymethylcellulose, microcrystalline cellulose and crosslinked microcrystalline cellulose such as croscarmellose sodium, alginates such as alginic acid and sodium alginate, clays such as bentonite, and effervescent mixtures. The amount of disintegrant in the composition can range from about 2 to about 20% by weight of the composition, more preferably from about 5 to about 10% by weight.
[0073] Binders are substances that bind or "glue" powder particles together and make them cohesive by forming granules, thus acting as the "adhesive" in the formulation. Binders enhance the cohesive strength already present in the diluent or bulking agent. Suitable binders include sugars such as sucrose, starches derived from wheat, corn, rice, potato, natural gums such as acacia, gelatin and tragacanth, derivatives of seaweed such as alginic acid, sodium alginate and calcium ammonium alginate, cellulosic materials such as methylcellulose, sodium carboxymethylcellulose and hydroxypropylmethylcellulose, polyvinylpyrrolidone, and inorganic compounds such as magnesium aluminum silicate. The amount of binder in the composition can range from about 2 to about 20% by weight of the composition, preferably from about 3 to about 10% by weight, more preferably from about 3 to about 6% by weight.
[0074] Lubricants refer to a class of substances added to dosage forms to reduce friction and wear, thereby allowing tablet granules and the like to be removed from the mold or die after compression. Suitable lubricants include metal stearates, such as magnesium stearate, calcium stearate, or potassium stearate, stearic acid, high melting point waxes, and other water-soluble lubricants, such as sodium chloride, sodium benzoate, sodium acetate, sodium oleate, polyethylene glycol, and D,L-leucine. Lubricants must be present on the surface of the granules, so they are usually added at the very last step before compression. The amount of lubricant in the composition can range from about 0.2 to about 5% by weight of the composition, preferably from about 0.5 to about 2% by weight of the composition, and more preferably from about 0.3 to about 1.5% by weight of the composition.
[0075] Lubricants prevent the components of the pharmaceutical composition from sticking and improve the flow properties of the granules, resulting in a smooth and homogeneous flow. Suitable lubricants include silicon dioxide and talc. The amount of lubricant in the composition can range from about 0.1 to about 5% by weight, preferably from about 0.5 to about 2% by weight, of the final composition.
[0076] A colorant is an excipient that provides color to a composition or dosage form. Such excipients can include food grade dyes adsorbed onto a suitable adsorbent, such as clay or aluminum oxide. The amount of colorant can be about 0.1 to about 5% by weight of the composition, preferably about 0.1 to about 1% by weight.
[0077] The sodium letermovir trihydrates of the present invention exhibit an antiviral effect against representatives of the Herpesviridae group (herpes viruses), in particular against cytomegalovirus (CMV), in particular against human cytomegalovirus (HCMV). They are therefore suitable for use in methods for treating and preventing diseases, in particular viral infections, in particular the viruses described herein and infections caused by them. The term "viral infection" is understood to mean not only infections caused by viruses, but also diseases caused by viral infections.
[0078] In another aspect, the present invention relates to a pharmaceutical composition obtainable by the methods described herein.
[0079] In another preferred embodiment, said pharmaceutical composition is obtainable by said process further comprising a subsequent addition step of adding at least one pharma- ceutically acceptable carrier, excipient and / or diluent to crystalline sodium 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetate trihydrate.
[0080] In a preferred embodiment, the pharmaceutical composition comprises crystalline sodium 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetate trihydrate, The particle size of sodium [trifluoromethyl]phenyl-4H-quinazolin-4-yl]acetate trihydrate is in the range of 0.3 to 1000 μm, and the particle size distribution is preferably defined as d(0.1) 1 to 100 μm, d(0.5) 30 to 250 μm, and d(0.9) 100 to 800 μm, and more preferably, the particle size distribution is defined as d(0.1) 2 to 50 μm, d(0.5) 25 to 100 μm, and d(0.9) 200 to 600 μm.
[0081] The parameter d(0.1) refers to the mesh size of a single notional sieve that will pass 10% of the sum of all particles in the sample. Thus, d(0.1)=2-100 μm means that the upper limit of the particle size range that defines 10% of the smallest particles in the sample is 2 μm-100 μm. Thus, 10% of all particles have a particle size equal to or less than d(0.1), which means that those particles have a maximum particle size between 2 μm and 100 μm.
[0082] The parameter d(0.5) thus refers to the mesh size of a single notional sieve that will pass 50% of the sum of all particles in the sample. Thus, d(0.5)=30-250 μm means that the upper limit of the particle size range that defines the notional half of the sample that contains smaller particles is 30 μm-250 μm. Thus, 50% of the sum of all particles have a particle size equal to or less than d(0.5), which means that in this case those particles have a maximum particle size between 30 μm and 250 μm.
[0083] The parameter d(0.9) thus refers to the mesh size of a single notional sieve that will allow 90% of the sum of all particles in the sample to pass, i.e., only 10% of the sample will be retained. Thus, d(0.9)=100-800 μm means that the lower limit of the particle size range that defines 10% of the largest particles in the sample is 100 μm-800 μm. Thus, 90% of all particles have a particle size equal to or less than d(0.9), which means that in this case, those particles have a maximum particle size between 100 μm and 800 μm.
[0084] Preferably, the antiviral agent used in the pharmaceutical composition of the present invention is effective against viruses of the Herpesviridae family, particularly cytomegalovirus (CMV), especially human cytomegalovirus (HCMV).
[0085] Preferably, the pharmaceutical composition further comprises a pharma- ceutically acceptable carrier, excipient and / or diluent.
[0086] The pharmaceutical compositions are preferably administered orally or intravenously.
[0087] The sodium 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetate trihydrate of the present invention exhibits an antiviral effect against representatives of the Herpesviridae group (herpes viruses), in particular against cytomegalovirus (CMV), and in particular against human cytomegalovirus (HCMV). The present invention is therefore suitable for use in methods for treating and preventing diseases, in particular viral infections, in particular the viruses described herein and infections caused by them. The term "viral infection" is understood to mean not only infections caused by viruses, but also diseases caused by viral infections.
[0088] Thus, another aspect of the present invention relates to a pharmaceutical composition comprising crystalline sodium 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetate trihydrate for use in a method for the treatment and / or prevention of diseases caused by and / or associated with cytomegalovirus, in particular human cytomegalovirus.
[0089] Furthermore, the present invention relates to a pharmaceutical composition comprising crystalline sodium 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetate trihydrate for the preparation of a medicinal product for the treatment and / or prevention of a disease, in particular a viral infection, preferably a human cytomegalovirus (HCMV) infection or an infection with another member of the Herpesviridae group.
[0090] Furthermore, the present invention provides a method for treating and / or preventing a disease associated with and / or caused by cytomegalovirus (CMV), in particular human cytomegalovirus (HCMV), or an infection by another member of the Herpesviridae group, comprising administering to said subject a pharmaceutical composition comprising a therapeutically effective amount of crystalline 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetate sodium trihydrate.
[0091] The term "effective amount" means an amount of a compound that, when administered to a subject in need of such treatment, is sufficient to: (i) Treating or preventing a particular disease, condition, or disorder; (ii) reducing, ameliorating, or eliminating one or more symptoms of a particular disease, condition, or disorder; or (iii) preventing or delaying the onset of one or more symptoms of a particular disease, condition or disorder described herein;
[0092] The amount of sodium 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetate trihydrate will vary depending on factors such as the particular compound, the disease state and its severity, the identity of the subject requiring treatment (e.g., weight), and the like, but can be routinely determined by one of ordinary skill in the art.

[0103] Comparative Example 01: Monosodium salt of 2-[(4S)-8-fluoro-2-[4-(3-methoxy-phenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetic acid (Corresponding to Example 1 of U.S. Patent Application Publication No. 2015 / 0038514(A1)) 333.1 g of 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetic acid are dissolved in 1300 mL of a mixture of ethanol and diisopropyl ether (1:1) in a 2000 mL three-neck flask. 21.9 g (546.84 mmol) of NaOH are added as a solid to the solution. The mixture is heated to an internal temperature of 50° C. for 25 minutes, which gives a clear orange solution. The solution obtained is stirred at this temperature for 3 hours, after 1 hour a thin suspension is already formed. The reaction mixture is then cooled to an internal temperature of 20° C. for 10 hours at a cooling rate of 3° C. / h and then stirred at this temperature for a further 5 hours. The total volume of the reaction mixture is reduced under vacuum to about 750 mL and the suspension thus obtained is stirred at 20° C. for 2 hours. Then, 250 mL of diisopropyl ether is added to the reaction mixture obtained over a period of 10 minutes, and the mixture is stirred for another 2 hours. The crystalline product obtained is sucked off with an aspirator, washed twice with 250 mL of diisopropyl ether each time, and dried in a vacuum drying cabinet at 20° C. and 160 mbar for 20 hours. The crystalline solid thus obtained is then dried in an IR dryer at 90° C. for 10 minutes, and then in a vacuum drying cabinet at 60° C. for another 16 hours. In this way, a total of 274.4 g (86% theoretical yield) of the desired crystalline sodium salt is obtained.

The X-ray structural data of the crystalline letermovir sodium methanol monohydrate is listed in Table 1.
[0107] When ethanol was used as a solvent, a crystal of 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetate sodium ethanol monohydrate was obtained, as shown in Figure 2. The X-ray structural data of the crystalline letermovir sodium ethanol monohydrate is listed in Table 2.
[0108] The asymmetric unit contains one anion molecule, one sodium cation, one water molecule, and one ethanol molecule. The water and ethanol molecules coordinated to the sodium cation are disordered at two positions (ratio 60:40). The compound crystallizes in the chiral space group R3, but a determination of the absolute structure could not be made. The structure obtained by using ethanol is isostructural with that obtained using methanol. The structure is of high quality with an R1 value of 4.75%.

In light of previous results, NaOH was added as a concentrated aqueous solution and anti-solvent addition was controlled.
[0112] As for the poor solvent, diisopropyl ether was selected as the main poor solvent in the reaction carried out.
[0113] Solvents: As evidenced by Comparative Example 02, which was performed according to the prior art (WO 2013 / 127971), letermovir sodium forms a significant number of solvates, especially with alcohols such as methanol and ethanol. Therefore, the present development excluded the use of alcohols and other solvents that may promote the formation of solvates other than sodium trihydrate.
[0114] Other parameters During development and optimization the following parameters were varied: - Solvent / anti-solvent ratio -Addition of antisolvent (rate and timing of addition) -API concentration -Addition of NaOH (directly or dissolved in water) -Water amount -Reaction time -Reaction temperature - Seeding
[0115] An overview of the experiments carried out can be seen in Table 3. The results suggest that a reduction in the amount of water appears to have a positive effect on the outcome of the reaction. The rate of anti-solvent addition does not appear to have a significant effect on the procedure. The yields obtained were generally average to low. Diisopropyl ether proved to be the most reliable anti-solvent, but all solvents gave comparable results.
[0116] Based on the experimental results, which depended on the methodological variables, the most important reaction parameters were selected for a series of optimization experiments. [Table 3]
[0117] 2. Further optimization From the results of the development experiments, the following important factors were outlined: -API concentration - Solvent / anti-solvent ratio -Addition of antisolvent -Water amount
[0118] The optimization aimed to obtain a robust methodology that could be easily scaled up. Therefore, experiments were performed on a 10g-20g scale using EasyMax™. The results can be seen in Table 4. The experiments performed with EasyMax™ revealed some issues that were not observed on a smaller scale. [Table 4]
[0119] The following parameters were optimized: - A more concentrated NaOH solution was used (1 g / mL in water). NaOH was added in excess (1.5 molar equivalents). -A more concentrated API solution was used (approximately 0.33 g / mL). -The stirring speed was medium to high (poor stirring can lead to the formation of oil, resin and two-phase systems). -Diisopropyl ether was added at various times and rates with no visible improvement in the results. The reaction was streamlined by dissolving the API in a solvent / anti-solvent mixture prior to the addition of -NaOH. - Optionally, the initial solution and base were seeded with 0.5 wt% letermovir sodium trihydrate crystal seeds to better induce letermovir sodium trihydrate precipitation. Under optimized reaction conditions, isopropyl acetate was found to be the most suitable solvent, the use of which leads to highly crystalline letermovir sodium trihydrate in quantitative yield.
[0120] Based on the optimized parameters, a general procedure for the synthesis of letermovir sodium trihydrate is outlined.
[0121] 2-[(4S)-8-Fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetic acid (letermovir) is dissolved in a 1:2 mixture of isopropyl acetate and diisopropyl ether at 50 °C (final API concentration: 0.33 g / mL). Then, 1.5 molar equivalents of NaOH (1 g / mL aqueous solution) are added, the solution is seeded with letermovir sodium trihydrate A (1-0.5% w / w) and stirred at 50 °C for 4 h. The suspension is then gradually cooled (50 °C to 20 °C in 1 h). The reactor contents are filtered under vacuum, washed three times with diisopropyl ether and dried at 50 °C for 15 h under reduced pressure (2-3 hPa). A white dense powder (letermovir sodium trihydrate) is obtained.
[0122] The technical characteristics of Letermovir Sodium Ethanol Monohydrate (Water) and Letermovir Sodium Trihydrate are summarized below: [Table 5]
[0123] Example 02. Preparation of sodium 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetate trihydrate on a 500 g scale 499 g of 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetic acid are dissolved in a 1:2 (0.75 L / 1.5 L) mixture of isopropyl acetate and diisopropyl ether at 50 °C, followed by the addition of about 1.5 equivalents of NaOH (aqueous, 1 g / mL, 50 mL) and seeding the solution with sodium 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetate trihydrate (0.5-1% wt / wt with respect to base). The suspension is stirred for 4 h at 50 °C (stirring speed: 150 rpm, glass anchor shaft). A large amount of white solid precipitates within the first hour. The reactor is gradually cooled (50°C to 20°C in 1 hour) and the contents are filtered under vacuum and washed three times with diisopropyl ether. The white solid obtained is dried at 50°C under reduced pressure (2-3 mBar) for 15 hours.
[0124] 510 g of sodium 2-[(4S)-8-fluoro-2-[4-(3-methoxyphenyl)piperazin-1-yl]-3-[2-methoxy-5-(trifluoromethyl)phenyl]-4H-quinazolin-4-yl]acetate trihydrate are obtained.
[0125] Example 03. Preparation of single crystals of letermovir sodium trihydrate Crystals were obtained by partial evaporation of a concentrated solution of letermovir sodium in an isopropyl acetate / diisopropyl ether mixture.
[0126] The asymmetric unit contains one molecule of anionic letermovir, a sodium cation, and three water molecules. The composition of the asymmetric unit is shown in Figure 5 (disordered atoms are omitted). The water molecules are disordered in two positions with ratios of 77:23, 67:33, and 63:37, respectively. Also, the CF3 groups are disordered in two directions with a ratio of 63:37. The trihydrate crystallizes in the highly symmetric trigonal space group R3 and forms a trimeric complex in which the three letermovir anions form a channel in which the sodium cation and water molecules are contained (Figure 6). A negatively charged carboxylate salt is located in the channel and interacts with the sodium cation.
