Abstract

The present invention relates to the technical field of bacterial detection, and in particular to a high-throughput detection method for Salmonella pullorum in chickens. The method comprises the following steps: inoculating Salmonella from chickens into a TSB culture medium; after cultivation and collection of the bacterial suspension, inactivating the bacterial suspension, centrifuging to remove the supernatant, staining with a crystal violet ethanol solution, and adjusting the concentration of Salmonella to 5×107 CFU / mL to 5×109 CFU / mL, thereby preparing a Salmonella antigen solution. The serum to be tested is diluted and added to a multi-well plate, followed by drying and fixation; after blocking with a PBS blocking solution, the Salmonella antigen solution is added for incubation, followed by washing, and the results are determined by visual observation or by measurement using a microplate reader. The present invention enables high-throughput, low-cost, and rapid detection of pullorum disease in chickens, while overcoming the problem of insufficient accuracy in traditional manual interpretation, and provides a technical method for the identification and diagnosis of pullorum disease in chickens.
