Description

SOLID FORMS OF MDM2 DEGRADERSCROSS REFERENCE TO RELATED APPLICATIONS

[0001] This application claims the benefit of priority to U.S. Provisional Appl. No. 63 / 491,677, filed March 22, 2023, the contents of which are herein incorporated by reference.TECHNICAL FIELD OF THE INVENTION

[0002] The present disclosure relates generally to various solid forms and compositions thereof useful for the modulation of murine double minute 2 (MDM2) protein via ubiquitination and / or degradation and uses of the same in the treatment various diseases.BACKGROUND OF THE INVENTION

[0003] Ubiquitin-Proteasome Pathway (UPP) is a critical pathway that regulates key regulator proteins and degrades misfolded or abnormal proteins. UPP is central to multiple cellular processes, and if defective or imbalanced, it leads to pathogenesis of a variety of diseases. The covalent attachment of ubiquitin to specific protein substrates is achieved through the action of E3 ubiquitin ligases. UPP plays a key role in the degradation of short-lived and regulatory proteins important in a variety of basic cellular processes, including regulation of the cell cycle, modulation of cell surface receptors and ion channels, and antigen presentation.

[0004] The murine double minute 2 (MDM2) oncoprotein is a key E3 ubiquitin ligase that degrades the tumor-suppressor p53. Reversible small molecule inhibitors (SMIs) of the MDM2 / p53 interaction have been developed to stabilize p53 and to induce apoptosis in wildtype p53 tumors. However, MDM2 SMIs induce a p53 / MDM2 feedback loop, resulting in upregulation of MDM2 protein levels and p53 pathway inhibition thus drastically limiting their biological activity and clinical application. MDM2 targeted protein degradation suppresses p53 -dependent MDM2 protein feedback upregulation and is therefore expected to lead to a superior response compared to MDM2 SMIs.

[0005] The present invention addresses the need for improved MDM2 degraders and provides additional advantages.SUMMARY OF THE INVENTION

[0006] It has now been found that novel solid forms of (3lR,4'S,5lR)-6"-chloro-4l-(3-chloro-2- fhiorophenyl)-N-((lR,4R)-4-(4-(l-(2,6-dioxopiperidin-3-yl)-3-methyl-2-oxo-2,3-dihydro-lH- benzo[d]imidazol-5-yl)piperidine-l-carbonyl)cyclohexyl)-2"-oxodispiro[cyclohexane-l,2l-pyrrolidine- 3',3"-indoline]-5'-carboxamide, as described in the present disclosure, and compositions thereof, are usefulmodulators of targeted ubiquitination of MDM2 protein and exhibit desirable characteristics for the same. In general, solid forms and pharmaceutically acceptable compositions thereof, are useful for treating or lessening the severity of a variety of diseases or disorders as described in detail herein.BRIEF DESCRIPTION OF THE DRAWINGS

The amount of a compound of the present invention in the composition will also depend upon the particular compound in the composition.Uses of Compounds and Pharmaceutically Acceptable Compositions

[0118] Compounds and compositions described herein are generally useful for the degradation and / or inhibition of MDM2 protein activity.

[0119] MDM2 protein that is degraded and / or inhibited by the compounds and compositions described herein and against which the methods described herein are useful include those of the mouse double minute 2 homolog (MDM2) protein or E3 ubiquitin-protein ligase MDM2 that is encoded by the MDM2 gene. MDM2 is an important negative regulator of the p53 tumor suppressor. The p53 tumor suppressor is a principal mediator of growth arrest, senescence, and apoptosis in response to a broad array of cellular damage. Rapid induction of high p53 protein levels by various stress types prevents inappropriate propagation of cells carrying potentially mutagenic, damaged DNA. p53 can kill cells via a dual transcription-dependent and transcription -independent function in the nucleus and at the mitochondria. Ithas been demonstrated that cellular p53 protein levels are the single most important determinant of its function. In normal unstressed cells, p53 is a ven' unstable protein with a half-life ranging from 5 to 30 min, which is present at very low cellular levels owing to continuous degradation largely mediated by MDM2. Conversely, a hallmark of many cellular stress pathways such as DNA damage, hypoxia, telomere shortening, and oncogene activation is the rapid stabilization of p53 via a block of its degradation. MDM2 has emerged as the principal cellular antagonist of p53 by limiting the p53 tumor suppressor function. Moll and Petrenko, Mol. Cancer Res. 2003, 1: 1001.

[0120] MDM2 is transcriptionally activated by p53 and MDM2, in turn, inhibits p53 activity by at least three mechanisms. Wu et al., Genes Dev. 1993, 7: 1126. First, MDM2 protein directly binds to the p53 transactivation domain and thereby inhibits p53-mediated transactivation. Second, MDM2 protein contains a nuclear export signal sequence, and upon binding to p53, induces the nuclear export of p53, preventing p53 from binding to the targeted DNAs. Third, MDM2 protein is an E3 ubiquitin ligase and upon binding to p53 is able to promote p53 degradation.

[0121] The activity of a compound utilized in this invention as a degrader and / or inhibitor of MDM2 protein or a mutant thereof, may be assayed in vitro, in vivo or in a cell line. In vitro assays include assays that determine inhibition of either the activity and / or the subsequent functional consequences of activated MDM2 protein, or a mutant thereof. Alternate in vitro assays quantitate the ability of the inhibitor to bind to a MDM2 protein. Inhibitor binding may be measured by radiolabeling the inhibitor prior to binding, isolating the inhibitor / MDM2 complex and determining the amount of radiolabel bound. Alternatively, inhibitor binding may be determined by running a competition experiment where new inhibitors are incubated with a MDM2 protein bound to known radioligands. Representative in vitro and in vivo assays useful in assaying a MDM2 inhibitor include those described and disclosed in, e.g., Zhange et al., “Fluorescence polarization assay and inhibitor design for MDM2 / p53 interaction” Anal. Biochem. 2004, 333(1): 138; Herman et al., “Discovery of Mdm2-MdmX E3 Ligase Inhibitors Using a Cell-Based Ubiquitination Assay” Cancer Discovery. 2011, 1(4): 312. Detailed conditions for assaying a compound utilized in this invention as a degrader and / or inhibitor of MDM2 proteins, or a mutant thereof, are set forth in the Examples below.

[0122] As used herein, the terms “treatment,” “treat,” and “treating” refer to reversing, alleviating, delaying the onset of, or inhibiting the progress of a disease or disorder, or one or more symptoms thereof, as described herein. The exact amount required will vary from subject to subject, depending on the species, age, and general condition of the subject, the severity of the infection, the particular agent, its mode of administration, and the like. Compounds of the invention are preferably formulated in dosage unit form for ease of administration and uniformity of dosage. The expression "dosage unit form" as used herein refers to a physically discrete unit of agent appropriate for the patient to be treated. It will be understood, however, that the total daily usage of the compounds and compositions of the present invention will be decided by the attending physician within the scope of sound medical judgment. The specific effective dose level for any particular patient or organism will depend upon a variety of factors including the disorder being treated and the severity of the disorder; the activity of the specific compound employed; the specific composition employed; the age, body weight, general health, sex and diet of the patient; the time of administration, route of administration, and rate of excretion of the specific compound employed; the duration of the treatment; drugs used in combination or coincidental with the specific compound employed, and like factors well known in the medical arts. The term “patient”, as used herein, means an animal, preferably a mammal, and most preferably a human.

[0243] Pharmaceutically acceptable compositions of this invention can be administered to humans and other animals orally, rectally, parenterally, intracistemally, intravaginally, intraperitoneally, topically (as by powders, ointments, or drops), bucally, as an oral or nasal spray, or the like, depending on the severity of the infection being treated. In certain embodiments, the compounds of the invention may be administered orally or parenterally at dosage levels of about 0.01 mg / kg to about 50 mg / kg and preferably from about 1 mg / kg to about 25 mg / kg, of subject body weight per day, one or more times a day, to obtain the desired therapeutic effect.

[0244] Liquid dosage forms for oral administration include, but are not limited to, pharmaceutically acceptable emulsions, microemulsions, solutions, suspensions, syrups and elixirs. In addition to the active compounds, the liquid dosage forms may contain inert diluents commonly used in the art such as, for example, water or other solvents, solubilizing agents and emulsifiers such as ethyl alcohol, isopropyl alcohol, ethyl carbonate, ethyl acetate, benzyl alcohol, benzyl benzoate, propylene glycol, 1,3-butylene glycol, dimethylformamide, oils (in particular, cottonseed, groundnut, com, germ, olive, castor, and sesame oils), glycerol, tetrahydrofurfuryl alcohol, polyethylene glycols and fatty acid esters of sorbitan, and mixtures thereof. Besides inert diluents, the oral compositions can also include adjuvants such as wetting agents, emulsifying and suspending agents, sweetening, flavoring, and perfuming agents.

[0245] Injectable preparations, for example, sterile injectable aqueous or oleaginous suspensions may be formulated according to the known art using suitable dispersing or wetting agents and suspending agents. The sterile injectable preparation may also be a sterile injectable solution, suspension or emulsion in a nontoxic parenterally acceptable diluent or solvent, for example, as a solution in 1,3 -butanediol. Among the acceptable vehicles and solvents that may be employed are water, Ringer's solution, U.S.P. and isotonic sodium chloride solution. In addition, sterile, fixed oils are conventionally employed as a solvent or suspending medium. For this purpose any bland fixed oil can be employed including synthetic mono- or diglycerides. [0247] In order to prolong the effect of a compound of the present invention, it is often desirable to slow the absorption of the compound from subcutaneous or intramuscular injection. This may be accomplished by the use of a liquid suspension of crystalline or amorphous material with poor water solubility. The rate of absorption of the compound then depends upon its rate of dissolution that, in turn, may depend upon crystal size and crystalline form. Alternatively, delayed absorption of a parenterally administered compound form is accomplished by dissolving or suspending the compound in an oil vehicle. Injectable depot forms are made by forming microencapsule matrices of the compound in biodegradablepolymers such as polylactide-polyglycolide. Depending upon the ratio of compound to polymer and the nature of the particular polymer employed, the rate of compound release can be controlled. Examples of other biodegradable polymers include poly(orthoesters) and poly(anhydrides). Depot injectable formulations are also prepared by entrapping the compound in liposomes or microemulsions that are compatible with body tissues.

Upon antigen binding, such CARs link to endogenous signaling pathways in the effector cell and generate activating signals similar to those initiated by the TCR complex.

[0348] For example, in some embodiments the CAR-T cell is one of those described in U.S. Patent 8,906,682 (June; hereby incorporated by reference in its entirety), which discloses CAR-T cells engineered to comprise an extracellular domain having an antigen binding domain (such as a domain that binds to CD 19), fused to an intracellular signaling domain of the T cell antigen receptor complex zeta chain (such as CD3 zeta). When expressed in the T cell, the CAR is able to redirect antigen recognition based on the antigen binding specificity. In the case of CD 19, the antigen is expressed on malignant B cells. Over 200 clinical trials are currently in progress employing CAR-T in a wide range of indications. [https: / / clinicaltrials.gov / ct2 / results ?term=chimeric+antigen+receptors&pg=l].

[0357] In some embodiments, an immuno-oncology agent is an immune checkpoint inhibitor as described herein.

[0358] The term “checkpoint inhibitor” as used herein relates to agents useful in preventing cancer cells from avoiding the immune system of the patient. One of the major mechanisms of anti -tumor immunity subversion is known as “T-cell exhaustion,” which results from chronic exposure to antigens that has led to up-regulation of inhibitory receptors. These inhibitory receptors serve as immune checkpoints in order to prevent uncontrolled immune reactions.

[0359] PD-1 and co-inhibitory receptors such as cytotoxic T-lymphocyte antigen 4 (CTLA-4, B and T Lymphocyte Attenuator (BTLA; CD272), T cell Immunoglobulin and Mucin domain-3 (Tim-3), Lymphocyte Activation Gene-3 (Lag-3; CD223), and others are often referred to as a checkpoint regulators. They act as molecular “gatekeepers” that allow extracellular information to dictate whether cell cycle progression and other intracellular signaling processes should proceed.

[0360] In some embodiments, an immune checkpoint inhibitor is an antibody to PD-1. PD-1 binds to the programmed cell death 1 receptor (PD-1) to prevent the receptor from binding to the inhibitory ligand PDL-1, thus overriding the ability of tumors to suppress the host anti -tumor immune response.

[0361] In one aspect, the checkpoint inhibitor is a biologic therapeutic or a small molecule. In another aspect, the checkpoint inhibitor is a monoclonal antibody, a humanized antibody, a fully human antibody, a fusion protein or a combination thereof. In a further aspect, the checkpoint inhibitor inhibits a checkpoint protein selected from CTLA-4, PDL1, PDL2, PD1, B7-H3, B7-H4, BTLA, HVEM, TIM3, GAL9, LAG3, VISTA, KIR, 2B4, CD 160, CGEN- 15049, CHK 1, CHK2, A2aR, B-7 family ligands or a combination thereof. In an additional aspect, the checkpoint inhibitor interacts with a ligand of a checkpoint protein selected from CTLA-4, PDL1, PDL2, PD1, B7-H3, B7-H4, BTLA, HVEM, TIM3, GAL9, LAG3, VISTA, KIR, 2B4, CD160, CGEN-15049, CHK 1, CHK2, A2aR, B-7 family ligands or a combination thereof. In an aspect, the checkpoint inhibitor is an immunostimulatory agent, a T cell growth factor, an interleukin, an antibody, a vaccine or a combination thereof. In a further aspect, the interleukin is IL-7 or IL-15. In a specific aspect, the interleukin is glycosylated IL-7. In an additional aspect, the vaccine is a dendritic cell (DC) vaccine.

[0362] Checkpoint inhibitors include any agent that blocks or inhibits in a statistically significant manner, the inhibitory pathways of the immune system. Such inhibitors may include small molecule inhibitors or may include antibodies, or antigen binding fragments thereof, that bind to and block or inhibit immune checkpoint receptors or antibodies that bind to and block or inhibit immune checkpoint receptor ligands. [0387] Solubility of amorphous compound 1 was measured in 10 solvents at RT and in nine solvents at 50 °C using the gravimetric method. Approximately 20 mg solid was added to a 2 mL vial followed by 0.6 mL of solvent. Slurries were stirred at a constant temperature overnight. After stirring overnight, the slurries were centrifuged, and supernatants were recovered for gravimetric solubility measurement. Supernatant solutions were evaporated to dryness at 35 °C under a mild flow of nitrogen for 30 min, then placed at 50 °C under vacuum overnight before final weighing. Results are summarized in Table 3. Solubility assessment by the addition method was also done with dioxane, THF, THF:ethanol (EtOH, 95:5 vol. and 8:2 vol.), and DMSO.Table 3. Solubility of Compound 1 at RT and 50 °C.Hyphen indicates that solubility was not assessed.aSolubility was done by addition.Evaporative Crystallization

[0388] Approximately 500 pL of supernatant from the slurry experiments was recovered for evaporative crystallization. The solutions were evaporated to dryness at 35 °C under a mild flow of nitrogen for 30 min, then placed at 50 °C under vacuum overnight. If sufficient solids were available after evaporation and drying, they were analyzed by X-ray powder diffraction (XRPD). The results are summarized in Table 4.The undersaturated crystallizations were from the experiments at RT, while the saturated crystallizations were from the experiments at 50 °C. Hyphen indicates no data were collected.Short-Term Slurries

[0389] Slurry experiments were completed in 10 solvents at RT and in nine solvents at 50 °C during the initial screening. After removing the supernatant for solubility measurement, fresh solvent (500 pL) was added to each vial and stirred at a constant temperature overnight. If an immobile slurry was generated, additional solvent was added to generate a stirrable slurry. After stirring overnight, the solids were filtered and recovered for XRPD analysis. Results are summarized in Table 5.Table 5. XRPD patterns of Solids Obtained from Short-Term Slurry Experiments.Hyphen indicates no data were collected.Vapor Diffusion onto Amorphous Solid

[0390] Vapor diffusion experiments onto amorphous solid were set up using a wide range of solvents. Approximately 10 mg of amorphous compound 1 was added to 4 mL vials. Each vial was then placed inside a 20 mL scintillation vial containing approximately 3 mL of the corresponding solvent. The results are summarized in Table 6. The unchanged solids were dissolved in THF and used for antisolvent vapor diffusion crystallization.Table 6. Summary of Results from Vapor Diffusion onto Amorphous Solid.Hyphen indicates no data were collected.Antisolvent Vapor Diffusion

[0391] Unchanged solids from vapor diffusion onto amorphous solid experiments were dissolved in THF and used for antisolvent vapor diffusion crystallization. Each 4 mL vial was then placed inside a 20 mL scintillation vial containing approximately 3 mL of the corresponding solvent. Results are summarized in Table 7.Table 7- Summary of Results from Antisolvent Vapor Diffusion.Hyphen indicates no data were collected.Antisolvent Crystallization

[0392] Antisolvent crystallization was completed in various solvent systems. First, 20 mg of amorphous compound 1 was dissolved in solvent. Antisolvent cry stallization was then carried out using the direct addition method. For direct antisolvent addition, more than three times the volume of solvent was used as antisolvent and was added dropwise in four equal portions over 1 h. Solutions / slurries were mixed during antisolvent addition. Results are summarized in Table 8.Table 8. Summary of Solids Obtained from Antisolvent Crystallization Experiments.L.C. = low -crystalline. Hyphen indicates that solids were not collected, indicates the sample was seeded with the wet cake of Form A.

[0393] One new pattern, Form A, was obtained from DMSO / ACN, but was unstable to drying at both 50 °C (-29 in Hg) for 3 h and RT (no vacuum) overnight, converting into an amorphous solid. HPLC analysis showed 98.44 area % purity for the Pattern A wet cake. To improve the crystallinity of Form A, the sample was heated up to 50 °C for 1 h while stirring at 400 rpm, then cooled down to RT and stirred for another hour. XRPD showed that the temperature cycling improved the crystallinity of Form A (FIG.l).Slow Evaporative Crystallization

[0394] Approximately 10 mg of amorphous compound 1 was weighed into a 2 mL vial and 0.75 mL solvent was added. The slurry was stirred at RT at 500 rpm overnight. The supernatant was separated using a centrifuge at 4400 rpm for 10 min. It was then transferred into a new 2 mL HPLC vial and the cap was poked with needles to evaporate the solvent at RT without stirring. Results are summarized in Table 9.Table 9. XRPD Patterns of Solids Obtained from Slow Evaporative Crystallization.Thermal Treatment

[0395] Thermal treatment experiments were performed with amorphous compound 1. The first experiment employed a ramp from 30-237 °C and was cooled back to 30 °C at 10 °C / min. The second experiment employed a ramp from 30-210 °C and was cooled back to 30 °C at 10 °C / min. The materials recovered from the pans were plated for XRPD analysis. Y ellow and white amorphous solids were observed for the first and second experiments, respectively.Slurry and Seeding Experiment Using Pattern A

[0396] Approximately 10 mg of amorphous compound 1 was weighed into a 2 mL vial and 0.5 mL of solvent was added. After 5 min of stirring (400 rpm) at RT and 50 °C, the samples were seeded with the Form A. The samples were filtered after 15 min for XRPD. Results are summarized in Table 10 and an XRPD pattern overlay of Form A and Form B + A is shown in FIG 2.Table 10. Summary of Slurry and Seeding Experiments in ACN.L.C. = low -crystalline, indicates the sample was seeded with the wet cake of Form A.Reslurry of Form A

[0397] Approximately 27 mg of Form A was filtered and transferred to a 2 mL vial. EtOH (20 vol.) was added, followed by stirring at RT and 400 rpm for 4 days. This resulted in an off-white slurry . The sample was filtered for XRPD analysis. Amorphous solid (white powder) was obtained from reslurry in EtOH, but the filtration yield was very low for NMR analysis. Therefore, approximately 30 mg of Form A was filtered first and washed with EtOH. It was then dried at 50 °C under vacuum (-29 in Hg) for 3 h beforesampling for XRPD and NMR. Results are summarized in Table 11. Results showed that DMSO content decreases after more EtOH used for washing Form A.Table 11. Summary of the Washing Experiment of Form A.Slow-Cooling Crystallization

[0398] Slow-cooling crystallization was conducted in a range of solvents / solvent mixtures using Crystall6. Experiments were earned out from 55 °C to 5 °C at a rate of -2.5 °C per hour. For all experiments, approximately 15 mg of amorphous compound 1 was weighed into a 2 mL vial. Solvent was then added incrementally at 50 °C until dissolution. In all cases, solids were completely dissolved before cooling. Solutions obtained from slow-cooling crystallization were used for seeding experiments with a crystallographic template. Samples were sonicated for 1 h followed by stirring at RT for 2 h. Results are summarized in Table 12.Table 12. Summary of XRPD Patterns of Solids Obtained from Slow-Cooling Crystallization.CT = crystallographic template. Hyphen indicates no data were collected. Conclusion

[0399] In Example 1, two crystalline patterns, Form A and Form B + A, were obtained. Form A was obtained from direct antisolvent crystallization in DMSO / ACN, and Form B + A was formed from slurry and seeding experiments in DMSO:ACN (1:4 and 1:5 vol.). Forms A and B + A were both unstable on drying at RT overnight and converted into amorphous solids. Microscopy images on the slurry of Form A showed it was rod-like materials. Form A was reproduced many times and it was washed with EtOH to remove residual solvent (DMSO) from the sample. NMR results showed that the residual DMSO contentwas 1.11 wt%. after washing the wet cake with five vol. of EtOH for 10 times. HPLC showed 98.44 area % purity for Form A.Example 2. Salt Screening

[0400] Experiments were set up for the salt screening process in 2 mL vials containing 6.3 mm stir bars. A stock solution of compound 1 was prepared in THF with a concentration of 41.52 mg / mL. Stock solutions of counter ions were prepared in EtOH. The equivalent of 25 mg freeform solution was added to each vial, followed by the appropriate counter ion stock solution. The vials were left to stir at RT for 1 h, after which they were uncapped and the solvent was evaporated using a mild flow of nitrogen. The vials were then placed under active vacuum (—29 inHg) at RT for 2 days to dry thoroughly . Approximately 300 pL of screening solvent was added to each vial, and the samples were stirred (400 rpm) at RT overnight. Vials that demonstrated significant precipitation or gumming were vortexed and sonicated often to ensure thorough mixing.

[0401] XRPD analysis was done in three stages. First, XRPD analysis of the wet cake was completed for all samples where solids were observed. Unique solids were then left on XRPD plates and dried under vacuum at RT (—29 in Hg) for 3 h before they were analyzed by XRPD. Finally, solids were exposed to > 95 % RH overnight and XRPD on the resulting solids was carried out. All XRPD patterns were compared to counter ion XRPD patterns and known freeform patterns. 2-Methyltetrahydrofuran (2-MeTHF), methyl isobutyl ketone (MIBK), and EtOH were chosen for the first round of solvents. If no crystalline salt was formed, the solvents were evaporated as before, and a second round of solvents (acetone, THF, DMSO:ACN (1:5 vol.)) was added. The third round of screening was completed using DCM, methyl acetate (MeOAc), and dioxane. In this round, solvent was added to each vial followed by stirring (400 rpm) at 45 °C (RT for DCM samples) for 1 h. Samples were cooled down to RT at 10 °C / h and stirred for 3 days. A summary of the XRPD data from the salt screening is shown Table 13.Table 13. Summary of the Salt ScreenAm. = amorphous. Hyphen indicates no data were collected.aGum manipulation included adding 100 pL of solvent to the vial and sonicated for 2 h, followed by 1 h of stirring at RT and 400 rpm. If no slurry formed, the previous step was repeated three times.

[0402] No crystalline salts were formed from the salt screening experiments using 14 counter ions and nine screening solvents. However, the purity of some isolated amorphous salts, including HC1, ethanedisulfonic acid (0.5 eq.), 2-naphthalenesulfonic acid, and maleic acid were found to have higher purity than compound 1 free base. Due to the amorphous nature, select salts were analyzed by NMR to confirm if the isolated amorphous solids had formed salts as shown in Table 14.Table 14. Summary of NMR data on select salts.

[0403] A summary of the compound 1 salts formed are shown in Table 15.Table 15. Salts of Compound 1Conclusion

[0404] In Example 2, no crystalline salts were formed. Purity analysis by HPLC indicated that HBr, sulfuric acid, nitric acid, methanesulfonic acid, and pyruvic acid were not preferred for salt formation, showing an HPLC chemical purity of less than 90 area %. On the other hand, HC1, benzenesulfonic acid, ethanedisulfonic acid (0.5 eq.), TFA, oxalic acid, 4-bromobenzenesulfonic acid, 2-naphthalenesulfonic acid, and maleic acid formed amorphous salts, with an HPLC chemical of over 97 area %. Although the benzene sulfonic acid, ethanedisulfonic acid, and bromobenzenesulfonic acid salts exhibited relatively high residual solvent (1.95, 1.25, and 2.95 wt. %, MeOAc, respectively), the MeOAc peak could have been overestimated in the NMR peak integration as it was combined with the compound 1 peaks.
