Abstract
Description
Anti-tumor application of R406 and its derivatives in synergy with TRAIL Technical Field
[0001] The present invention relates to the field of medicine, and in particular, provides the anti-tumor use of drug R406 and its derivatives in synergistic effect with TRAIL. Background Art
[0002] In recent years, the incidence and mortality of tumors have been increasing, posing new challenges to tumor treatment. Numerous studies have shown that TNF-related apoptosis-inducing ligand (TRAIL) selectively induces apoptosis in tumor cells, with virtually no significant toxic side effects on normal tissue cells. In human cells, there are four types of TRAIL surface receptors: TRAIL-R1, TRAIL-R2, TRAIL-R3, and TRAIL-R4. TRAIL-R1 and TRAIL-R2 contain a death domain (DD) within their cells. Upon TRAIL stimulation, TRAIL-R1 and TRAIL-R2 can initiate apoptosis by recruiting the death domain-containing protein FADD. TRAIL-R3 and TRAIL-R4 are decoy receptors that do not contain a death domain within their cells, and therefore do not induce apoptosis upon TRAIL stimulation. Different TRAIL receptors are differentially expressed in normal and tumor cells. TRAIL-R1 and TRAIL-R2 are highly expressed in tumor cells, while TRAIL-R3 and TRAIL-R4 are highly expressed in normal cells. These characteristics allow TRAIL to specifically induce tumor cell death, making research on its anti-tumor properties highly sought after. Currently, TRAIL-based treatments have been used in numerous clinical trials, including studies investigating TRAIL in non-small cell lung cancer (NSCLC), B-cell non-Hodgkin's lymphoma (NHL), and colorectal cancer. Although TRAIL demonstrated high tolerability and safety in Phase I clinical trials, multiple clinical trials have shown that the efficacy of TRAIL alone for tumor treatment is very limited. Numerous preclinical studies have demonstrated that most tumor cells are insensitive to TRAIL alone, leading to a growing number of studies exploring the combination of TRAIL with other small molecules to enhance tumor cell sensitivity to TRAIL. Currently, research on TRAIL combination therapy has entered the clinical stage, and clinical results have shown that TRAIL combination therapy has a good effect on tumor suppression. Therefore, the development of new small molecule drugs that can synergize with TRAIL to achieve better anti-tumor effects can provide a good reference and hope for subsequent targeted tumor treatment.
[0003] R406 is a reported inhibitor of the tyrosine kinase Syk. R788, also known as fostamatinib, is the phosphate form of R406. As a prodrug molecule for R406, R788 can be rapidly converted to R406 in vivo. Currently, R788 has been approved by the FDA for the treatment of idiopathic thrombocytopenic purpura (ITP), but there are no studies reporting the combined use of R788 with TRAIL for the treatment of tumors. Therefore, there is no known application of R406 or its phosphate, R788, in combination with TRAIL for the treatment of tumors.
[0004] Therefore, there is an urgent need to develop small molecule drugs that can synergize with TRAIL to exert better anti-tumor function.

[0118] The synthesis of the compounds of the present invention all uses the corresponding aminopyridine derivatives as raw materials, and the reaction process is as follows:
[0119] To a 15 mL reaction flask, add the corresponding aminopyridine derivative (2.59 mmol), 5-fluoro-2,4-dichloropyrimidine (650 mg, 3.9 mmol), DIPEA (850 μL, 5.18 mmol), and DMF (5 mL). Heat to 90°C and stir overnight. After the reaction is complete, dilute with ice water, filter, and wash the filter cake with water and an organic mixed solvent (PE / DCM = 10 / 1) before drying to obtain the pyrimidine-substituted intermediate.
[0120] The intermediate (0.31 mmol) was dissolved in NMP (2 mL) with the corresponding primary amine (0.46 mmol) and 2M dioxane hydrochloride solution (15 μL, 0.03 mmol). The mixture was microwaved at 120°C for 1 h. After completion, the reaction solution was purified by reverse phase chromatography (H₂O / MeCN = 1:1) and HPLC to yield the title compound.
[0121] Among them, the preparation routes of various aminopyridine derivatives are as follows:
[0122] 1. Preparation of compound TXY-4-44
[0123] (1) Preparation of compound TXY-3-21:

[0189] 1.7 Experimental Results
[0190] 1.7.1 R406 directly binds to TAK1
[0191] Currently, recombinant protein expression of the TAK1 kinase domain is typically performed using insect cells. Since TAK1 only possesses kinase activity when fused to TAB1, TAK1-TAB1 was expressed using an insect cell expression system, as shown in Figure 2. This system was then used to study the binding of R406 and R788 to TAK1-TAB1. Protein thermal stability can be used to investigate changes in protein stability when binding to small molecules or other proteins. When small molecules bind to a protein, they stabilize its globular structure, causing changes in the protein's Tm value during thermal stability experiments. Therefore, changes in Tm can reflect the binding activity.
[0192] R406 and R788 were incubated with hTAK1-TAB1 protein for 60 minutes respectively, and then the thermal stability experiment was started. The results are shown in Figure 3. From the Tm value and thermal stability fluorescence change spectrum of TAK1, it can be seen that R406 and R788 can increase the Tm value of the thermal stability of the hTAK1-TAB1 reaction by 4-6°C, which has a significant effect on thermal stability, indicating that R406 can directly bind to TAK1.
[0193] 1.7.2 R406 directly activates TAK1
[0194] Flag-RIPK1 and Flag-IKKβK44A plasmids were overexpressed in HEK293T cells, respectively. To minimize the background autophosphorylation of RIPK1, 10 μM Nec-1s was added during overexpression. 24 h later, RIPK1 and IKKβ were immunoprecipitated using Flag beads, respectively. Then, in vitro kinase reactions were performed with recombinant His-tagged TAK1-TAB1 protein. At the same time, 20 μM R406 and 1 μM 5Z7 were added, respectively. After incubation at 30°C for 60 min, the samples were boiled for Western detection, as shown in Figure 4.
[0195] The results showed that R406 promoted the phosphorylation of IKKβ and RIPK1-S321 in vitro, while 5Z7, as a control, inhibited their phosphorylation. As shown in Figure 5, R406 also promoted its own phosphorylation in vitro, as well as the phosphorylation of TAK1 peptide substrates. These data indicate that R406 can directly activate TAK1 in vitro.
[0196] 1.7.3 Design and Utilization of the R406 Photosensitive Affinity Probe
[0197] Based on the structure of R406, small molecules were modified, as shown in Table 1, and their structure-activity relationship for TAK1 was analyzed. Then, a cell survival assay was performed to detect whether the modified small molecules still had the activity of R406 in promoting TNFα-induced cell death, as shown in Table 2. Then, suitable structural sites were found to introduce photo-crosslinking tags without affecting the activity of R406, and photo-crosslinking probes targeting TAK1 were synthesized, as shown in Table 1, to assist in finding the interaction sites between R406 and TAK1, as shown in Table 2.
[0198] Table 1 R406 derivatives
[0199] Table 2 Activity data of various R406 derivatives ("+" represents that the compound promotes the death of MEFs cells by ≤10%, "++" represents that the compound promotes the death of MEFs cells by ≤20% but >10%, "+++" represents that the compound promotes the death of MEFs cells by ≥30%)
[0200] Table 3 Characterization data of some compounds of the present invention
[0201] The results showed that the small molecules based on the structural modification of R406 were sequentially subjected to cell survival assays, and it was finally found that S-8-56 was the most active small molecule. T-5-27 was a photocrosslinking probe derived from S-8-56. Cells were treated with small molecules, or small molecules and proteins were co-incubated, as shown in Figure 6. After photocrosslinking and click chemistry reactions, the biotin-labeled target protein could be co-precipitated and detected through the biotin-avidin system.
[0202] The diazirine group of T-5-27 can undergo a photochemical reaction under ultraviolet light of a certain wavelength, allowing this part of the group to be covalently cross-linked to the vicinity of the target protein binding site, while the alkyne group on the left can undergo a click reaction with the azide group of the biotin probe to enrich the protein labeled with the photocross-linking probe. The enriched protein can be subjected to mass spectrometry analysis after trypsin digestion. An attempt is made to find the approximate binding site of T-5-27 on TAK1 by increasing the molecular weight peptide information shown in the figure.
[0203] As shown in Figure 6, the photosensitized affinity probe T-5-27 can indeed enrich and isolate TAK1 from cell lysates, whereas S-8-56, a derivative of the R406 photostable control small molecule, fails to coprecipitate TAK1. Furthermore, in the presence of S-8-56, the cross-linking effect of the probe is weakened, demonstrating the effectiveness of the photocross-linking probe.
[0204] Next, the probe was photocrosslinked to TAK1 protein in vitro. The probe was linked to the biotin via a click chemistry reaction. The probe-crosslinked TAK1 protein was then precipitated using streptavidin agarose beads. The TAK1 protein was then digested with trypsin, and peptides containing the probe and biotin were eluted for protein mass spectrometry analysis to identify the interaction sites between R406 or its derivatives and TAK1. As shown in Figure 7, if the peptide is linked to the probe and biotin, it will have an increased mass shift of 1000.4865 Da. The amino acids in the peptide that are linked to the probe and biotin can be identified through the secondary mass spectrometry spectrum. Finally, mass spectrometry analysis identified the peptide "35-EIEVEEVGR-44" as having a mass increase of 1000.4865 Da. This indicates that E6 in the peptide, or glutamic acid at position 40 of TAK1, is covalently coupled to the probe.

[0218] 1.4 Experimental Results
[0219] 1.4.1 R406 / R788 promotes TNFα-induced RIPK1-dependent programmed cell death in tumor cells
[0220] In human leukemia FADD-deficient Jurkat cells, after pretreatment with R406 (3 μM), R788 (3 μM) or Nec-1s (10 μM) for 30 minutes, TNFα (20 ng / ml) was added to induce cell necrosis.
[0221] As shown in Figure 8, R406 and R788 significantly promoted TNFα-induced cell necrosis with prolonged TNFα treatment, and this promotion was protected by Nec-1s. Furthermore, R406 and R788 significantly enhanced the phosphorylation of RIPK1 and MLKL with increasing TNFα treatment time, and this phosphorylation was protected by Nec-1s. This suggests that R406 can promote TNFα-induced RIPK1-dependent programmed necrosis.
[0222] Human colon cancer cells HT-29 and human lung adenocarcinoma cells NCI-H1975 were pretreated with R406 (3 μM) and Nec-1s (10 μM) for 30 minutes, and then TNFα (20 ng / ml) and caspase inhibitor zVAD.fmk (20 μM) were added to induce cell necrosis. The SYTOX Green level of the cells was measured every 7 hours to assess cell death.
[0223] As shown in Figures 9-12, R406 significantly promoted TNFα / zVAD.fmk-induced cell necrosis with prolonged TNFα and zVAD.fmk treatment, and this promotion was protected by Nec-1s. Furthermore, R406 significantly enhanced the phosphorylation of RIPK1 and MLKL with increasing TNFα / zVAD.fmk treatment time, and this phosphorylation was protected by Nec-1s. This suggests that R406 can promote TNFα-induced RIPK1-dependent programmed necrosis.
[0224] Subsequently, human lung cancer cells NCI-H1299 were pretreated with R406 (3 μM) and Nec-1s (10 μM) for 30 minutes, and then TNFα (20 ng / ml) was added. Similarly, the cell SYTOX Green level was measured every 7 hours to assess cell death.
[0225] As shown in Figure 11, R406 significantly promoted TNFα-induced cell death with prolonged TNFα treatment, and this promotion was protected by Nec-1s. Furthermore, R406 significantly enhanced RIPK1 phosphorylation and PARP1 cleavage with increasing TNFα treatment time, and these phosphorylation and cleavage were protected by Nec-1s. This suggests that R406 can promote TNFα-induced RIPK1-dependent cell apoptosis.
[0226] 1.4.2 R406 / R788 promotes TRAIL-induced apoptosis
[0227] E0771 cells were treated with different concentrations of TRAIL to induce apoptosis and 1 μM, 3 μM, and 5 μM R406, respectively. Cellular ATP levels were detected 24 hours later, and cell viability was evaluated using the CellTiter-Glo assay.
[0228] As shown in Figure 12, when TRAIL alone treated E0771 cells, its IC 50 was 34.21 nM. With the addition of R406, the IC 50 It showed a gradually decreasing trend, and the higher the concentration of R406, the higher the IC of TRAIL 50 The lower.
[0229] In order to establish an animal model with ZsGreen-LUC-expressing E0771, it is first necessary to construct a stable cell line with the luciferase gene, and this cell line has unchanged sensitivity to the compound. Therefore, the experiment shown in Figure 13 was performed.
[0230] The experimental results showed that the modified ZsGreen-LUC-expressing E0771 cells with the strongest luciferase activity showed no difference in sensitivity to the compound compared to E0771-BLANK cells. Treatment with either the compound or TRAIL alone promoted 20%-30% cell death (12 hours). The combination of the compound and TRAIL promoted 70%-80% cell death (12 hours) in both cell types. This promotion effect was significantly different. In addition, the cell death promoted by compound R788 under TRAIL-induced conditions was inhibited by Nec-1s, but the cell death promoted by TRAIL itself was not inhibited by Nec-1s.
[0231] LLC cells were treated with different concentrations of TRAIL to induce apoptosis and 1 μM, 3 μM, and 5 μM R406, respectively. Cell ATP levels were detected 24 h later, and cell viability was evaluated using the CellTiter-Glo assay.
[0232] As shown in Figure 14, when TRAIL alone treated E0771 cells, its IC 50 is 37.77nM. With the addition of R406, the IC 50 It showed a gradually decreasing trend, and the higher the concentration of R406, the higher the IC of TRAIL 50 The lower.
[0233] As shown in the Western Blot results of Figures 12 and 16 , in LLC and E0771 cells, R406 can promote TRAIL-induced PARP1 cleavage and RIPK1-S166 phosphorylation, thereby promoting RIPK1 activation.
[0234] These results indicate that R406 / R788 can promote TRAIL-induced apoptosis in E0771 and LLC cells.

[0300] 1.8 Experimental Results
[0301] 1.8.1 R788 enhances the anti-tumor effect of TRAIL in the mouse E0771 model (statistical analysis)
[0302] The constructed ZsGreen-LUC-expressing E0771 tumor cells were implanted into C57BL / 6 female mice. On days 11 and 15, respectively, the mice were injected orally with D-Luciferin (150 mg / kg). The light intensity changes were detected by a small animal in vivo imaging device to monitor the drug effect.
[0303] As shown in Figure 15, the tumor size of the mice in the R788 alone treatment group and the TRAIL alone treatment group was reduced compared with the control group. After the combination of TRAIL and R788, the tumors of the mice were not only significantly reduced compared with the control group, but also significantly reduced compared with the tumors of the mice in the R788 alone and TRAIL alone groups.
[0304] Tumor growth curves and tumor size were statistically analyzed, as shown in Figure 16. A total of 32 6-8 week old C57BL / 6 female mice successfully inoculated with ZsGreen-LUC-expressing E0771 tumor cells were used in the experiment. Eight mice were used in each experiment, for a total of four experimental groups. Tumor volume and body weight were recorded daily, starting from the first day of treatment. Eleven days after treatment, tumors were weighed ex vivo, and the statistical results were compared and analyzed.
[0305] Compared with the control group, the tumor growth rate of the two groups of mice treated with R788 alone and TRAIL alone was slowed down. When the drug was stopped, the tumor volume was about 1500mm 3 When the control group stopped taking the drug, the tumor volume was about 2300mm 3 The tumor volume of mice treated with R788 and TRAIL combined was significantly slower than that of the control group and the R788 and TRAIL alone groups. When the drug was stopped, the tumor volume of the mice treated with the combined drug was approximately 700 mm 3 .
[0306] The statistical results of in vitro weighing of ZsGreen-LUC-expressing E0771 tumors are shown in FIG17 . R788 and TRAIL administered alone had certain anti-tumor effects, but the combined use of R788 and TRAIL had a better effect.
[0307] 1.8.2 R788 Enhances the Antitumor Effect of TRAIL in the E0771 Mouse Model (Tissue Section Analysis)
[0308] The cleavage of Caspase-3 represents the occurrence of cell apoptosis. The larger the cleavage area, the more intense the apoptosis. As shown in Figure 18, after the combination of R788 and TRAIL, the immunohistochemical staining area of cleaved Caspase-3 in mouse ZsGreen-LUC-expressing E0771 tumor tissue sections was the largest, indicating that cell apoptosis is most obvious at this time.
[0309] This proves that R788 has an anti-tumor effect by promoting TRAIL-induced cell apoptosis, thereby slowing down tumor growth.
[0310] Ki-67 is a marker of cell proliferation. The darker the Ki-67, the more vigorous the cell proliferation. As shown in Figure 19, Vehicle has the darkest color, and the ZsGreen-LUC-expressing E0771 tumor cells proliferate the most vigorously. Compared with the control group, Ki-67 levels in the TRAIL alone and R788 alone treatments were weaker, and the tumor cell proliferation ability decreased. The contrast was more obvious in the TRAIL+R788 group. Compared with the control group, Ki-67 showed significant changes and became significantly weaker, and the tumor cell proliferation ability was the worst.
[0311] CD31 is a marker for endothelial cells. The darker the CD31 immunohistochemical staining, the faster the endothelial cells grow, the greater the microvessel density, and the richer the nutrition supplied to tumor cells, which will accelerate the growth of tumor cells.
[0312] As shown in Figure 20, Vehicle had the darkest immunohistochemical staining and the fastest endothelial cell proliferation rate, while CD31 was weaker in the R788 alone and TRAIL alone treatment groups compared with the control group, and the endothelial cell proliferation ability decreased. The contrast was more obvious in the TRAIL+R788 group. Compared with the control group, CD31 showed significant changes and became significantly weaker. The endothelial cell proliferation ability was the worst, the blood vessels were the least, the nutrition supplied to the tumor cells was the poorest, and the tumor cells grew the slowest.
[0313] 1.8.3 R788 enhances the anti-tumor effect of TRAIL in the mouse LLC model (statistical analysis)
[0314] As shown in Figure 21, 17 days after LLC was subcutaneously inoculated into C57BL / 6 mice, tumors were removed from the four drug-treated groups and compared as shown in the figure. Tumor size in the R788-only group was not significantly different from that in the control group, while tumors in the TRAIL-treated group were slightly smaller than those in the control group. Combination therapy with TRAIL and R788 resulted in significantly smaller tumors not only compared to the control group but also compared to those in the TRAIL-only group. Tumor growth curves and tumor sizes were statistically analyzed.
[0315] As shown in Figure 22, a total of 24 6-8 week old C57BL / 6 male mice successfully inoculated with LLC tumor cells were used in the experiment. Six mice were used in each experiment, resulting in four experimental groups. Tumor volume and body weight were recorded daily starting from the first day of treatment. After 14 days of treatment, the tumors were weighed ex vivo, and the statistical results were compared and analyzed.
[0316] There was no significant difference in tumor growth rate between the R788 alone group and the control group. After 13 days of inoculation, both groups showed exponential growth rate, and when the drug was stopped, the tumor volume was nearly 1000 mm 3 Fourteen days after the injection, the group of mice that received intraperitoneal injection of TRAIL alone showed a slower tumor growth rate compared to the control group. When the drug was stopped, the tumor volume was approximately 600 mm. 3 The tumor volume of the mice treated with R788 and TRAIL was significantly slower than that of the control group and the TRAIL-only group. When the drug was stopped, the tumor volume of the combined treatment group was approximately 300 mm 3 .
[0317] The statistical results of LLC tumor in vitro weighing are shown in Figure 22. R788 alone had no effect, TRAIL had a certain anti-tumor effect, but the combination of R788 and TRAIL had a better effect.
[0318] 1.8.4 R788 enhances the antitumor effect of TRAIL in the mouse LLC model (tissue section analysis)
[0319] As shown in Figure 23, tumor tissue sections from mice treated with R788 alone showed virtually no damage or necrosis compared to the control group. Tissue sections from mice treated with intraperitoneal TRAIL showed some areas of damage and necrosis, but the areas of cell death were smaller. However, combined administration of R788 and TRAIL significantly expanded the areas of tissue damage and cell necrosis, accompanied by extensive hemorrhages. This suggests that R788 can promote TRAIL-induced cell death in LLC tumor tissues in mice. Immunohistochemical staining for Ki-67, CD31, and cleaved Caspase-3 demonstrates that R788 indeed exerts its anti-tumor effect by promoting TRAIL-induced apoptosis, thereby slowing tumor growth.
[0320] 1.8.5 R788 promotes TRAIL-induced activation of RIPK1 and caspase (in vivo)
[0321] As shown in Figure 24, 1 / 2 / 3 represent the numbers of mice treated with drugs in each group. Each group consisted of 6 mice, and tumor tissue samples from 3 of these mice were randomly selected for Western Blot analysis. The results showed that neither R788 alone nor the control group activated RIPK1 or caspases. However, in mice injected intraperitoneally with TRAIL alone, phosphorylation of RIPK1 at S166 was slightly activated, accompanied by caspase-3 and PARP1 cleavage. However, when R788 and TRAIL were combined, phosphorylation of RIPK1 at S166 was significantly enhanced, and caspase-3 and PARP1 cleavage was even more pronounced.
[0322] This indicates that R788 can be well absorbed in vivo and can promote TRAIL-induced RIPK1 and caspase activation in LLC and E0771 tumor tissues.
[0323] 2 Discussions
[0324] 2.1 The anti-tumor effect of R406 phosphate R788 was evaluated by using it in mice to determine whether it could promote the apoptosis of tumor cells mediated by other death receptor family members, such as TRAIL. Existing experimental data showed that the compound had the most significant effect in two mouse lung cancer cells, LLC and breast cancer E0771. In addition, in the mouse tumor models of these two cells, whether through biological characteristics observation, HE staining, or immunohistochemistry, it was demonstrated that R788 could promote the anti-tumor effect of TRAIL.
