Abstract
Description
A KOR antagonist and its medical application Technical Field
[0001] The present invention relates to a compound of general formula (I) or its stereoisomers, tautomers, racemates, deuterated compounds, solvates, prodrugs, metabolites, pharmaceutically acceptable salts or cocrystals, as well as intermediates and preparation methods, and use of the compound in preparing drugs for treating diseases associated with antagonizing KOR. Background Art
[0002] Opioid receptors are a collective term for a group of G protein-coupled receptors. The kappa-opioid receptor (KOR) is a member of the opioid receptor family. It binds to the opioid peptide dynorphin, its primary endogenous ligand. Kappa-opioid receptors are widely distributed in the brain, spinal cord, and pain-sensing neurons, participating in important physiological processes such as pain perception, neuronal secretion, emotional behavior, and cognition. When receiving sustained neural signals, dynorphin acts on kappa-opioid receptors, and agonizing them can cause negative emotions such as anxiety and depression.
[0003] Research indicates that blocking the effects of dynorphin with κ-opioid receptor antagonists has therapeutic benefits for patients with mood disorders, anxiety disorders, and substance use disorders. Currently available κ-opioid receptor antagonists suffer from delayed onset of action, lasting several hours to days, and persistent antagonism at the minimum effective dose for several weeks. Furthermore, they have low blood-brain barrier penetration. While progress has been made in the development of highly selective and potent κ-opioid receptor antagonists, the discovery of safer, shorter-acting κ-opioid receptor antagonists with improved pharmacokinetics remains clinically valuable.

[0461] 4.CYP450 enzyme inhibition test
[0462] The purpose of this study was to evaluate the effects of test substances on the activities of five cytochrome P450 (CYP) isoforms (CYP1A2, CYP2C9, CYP2C19, CYP2D6, and CYP3A4) in human liver microsomes using an in vitro assay system. Specific probe substrates for each CYP450 isoform were incubated with human liver microsomes and varying concentrations of the test substances. The reaction was initiated by the addition of reduced nicotinamide adenine dinucleotide phosphate (NADPH). After the reaction, the samples were processed and metabolites generated from the specific substrates were quantitatively detected by liquid chromatography-tandem mass spectrometry (LC-MS / MS). Changes in CYP enzyme activity were measured, and IC50 values were calculated to evaluate the inhibitory potential of the test substances against each CYP enzyme isoform.
[0463] Conclusion: The inhibitory activities of the compounds of the present invention, such as compound 7, compound 8, and compound 10, against CYP1A2, CYP2C9, CYP2C19, CYP2D6, and CYP3A4 are all greater than 20 μM.
[0464] 5. hERG potassium channel effect test
[0465] Experimental platform: electrophysiology manual patch clamp system
[0466] Cell line: Chinese hamster ovary (CHO) cell line stably expressing hERG potassium channel
[0467] Experimental methods: CHO (Chinese Hamster Ovary) cells stably expressing hERG potassium channels were used to record hERG potassium channel currents using the whole-cell patch clamp technique at room temperature. Glass microelectrodes were pulled from glass electrode blanks (BF150-86-10, Sutter) using a puller. The tip resistance after perfusing the electrode liquid was about 2-5 MΩ. The glass microelectrode was inserted into the amplifier probe to connect to the patch clamp amplifier. The clamping voltage and data recording were controlled and recorded by a computer using pClamp 10 software, with a sampling frequency of 10 kHz and a filter frequency of 2 kHz. After obtaining the whole-cell recording, the cell was clamped at -80 mV to induce the hERG potassium current (I hERG ) was administered with a 2-second depolarization step from -80 mV to +20 mV, followed by repolarization to -50 mV for 1 second before returning to -80 mV. This voltage stimulus was administered every 10 seconds, and administration began after confirming that the hERG potassium current was stable (for at least 1 minute). Compounds were administered for at least 1 minute at each test concentration, and at least two cells (n ≥ 2) were tested for each concentration.
[0468] Data processing: Data analysis was performed using pClamp 10, GraphPad Prism 5, and Excel software. The degree of inhibition of hERG potassium current (peak hERG tail current induced at -50 mV) by different compound concentrations was calculated using the following formula:
[0469] Inhibition%=[1-(I / Io)]×100%
[0470] Wherein, Inhibition% represents the inhibition percentage of the compound on hERG potassium current, and I and Io represent the amplitude of hERG potassium current before and after drug addition, respectively.
[0471] Compound IC 50 Calculated using GraphPad Prism 5 software by fitting the following equation:
[0472] Y=Bottom+(Top-Bottom) / (1+10^((LogIC 50 -X)*HillSlope))
[0473] Where X is the Log value of the test sample concentration, Y is the inhibition percentage at the corresponding concentration, and Bottom and Top are the minimum and maximum inhibition percentages, respectively.
[0474] Conclusion: The compounds of the present invention have poor inhibitory activity on hERG.
[0475] 6. Liver Microsome Stability Test
[0476] In this study, human liver microsomes were used as an in vitro model to evaluate the metabolic stability of the test substances.
[0477] At 37°C, 1 μM of the test substance was incubated with microsomal proteins and coenzyme NADPH. The reaction was terminated by adding ice-cold acetonitrile containing an internal standard after a certain time (5, 10, 20, 30, 60 min). The concentration of the test substance in the sample was detected by LC-MS / MS. The T value was calculated based on the ln value of the drug residual rate in the incubation system and the incubation time. 1 / 2 , and further calculated the liver microsomal intrinsic clearance CL int(Liver) .
[0478] Table 6. Human liver microsome stability results

[0488] 8. Mouse forced swimming test
[0489] Male C57BL / 6J mice, 6-8 weeks old and weighing approximately 20-25 g, were purchased from Zhejiang Weitonglihua Laboratory Animal Technology Co., Ltd. After acclimation for 7 days, their weights were recorded and the mice were randomly divided into groups. A cylindrical swimming tub was filled with water (23 ± 1°C) at a height of two-thirds of the tub's volume. The mice were placed in the tub and allowed to swim for approximately 10 minutes. The mice were then removed, dried, and returned to their cages for formal testing the following day. The next day, the tub was filled with water (23 ± 1°C). The forced swimming apparatus (Cyons, model ABOS-01) and a camera (Cyons, model ABOS-01) were positioned and connected to a computer with Vistrack software (Shanghai Xinruan Information Technology Co., Ltd.). The mice were removed from their home cages and administered the test substance (or test substance) or intraperitoneally (or ketamine, the positive control). The mice were tested for swimming at designated time points after drug administration. Before the mouse swimming test, the experimenter soothed the animal for about 1 minute, then put the animal into the water, and the operator immediately left the video shooting range. The software recorded the mouse swimming video for 6 minutes. After the test, the mouse was taken out and thoroughly dried and put back into the cage. The above process was repeated until all mice were tested. After the experiment, all animals were euthanized. Finally, the data analyst evaluated the mouse's immobility behavior through the video (the immobility state is defined as passive floating, with no other movement except the movements necessary to keep the head above water), and analyzed the length of time the mouse was immobile from the 2nd minute to the 6th minute of the test phase. The longer the mouse was immobile, the more severe the depression. All raw data were entered into Excel, and statistical analysis was performed using GraphPad Prism 10.0.
[0490] Conclusion: The compounds of the present invention, such as the compounds in the examples, have good antidepressant effects.
