Attorney Docket No.43820-0007P01 (SBP Ref.22-015-02PCT) Methods of Treatment Polycystic liver disease (PLD) is characterized by the development and growth of cholangiocyte-derived liver cysts. The pathogenesis of PLD involves defects in the primary cilium of the cholangiocyte that impair cilia function. Some patients are asymptomatic and require no therapeutic intervention, other patients suffer from a myriad of symptoms from the compressive effects of enlarged liver cysts, malnutrition, and liver decompensation. See, e.g., Masyuk, et al., (2017) 66, 4, pp.1197-1218. Provided herein are methods for treating a disease associated with TGR5 (e.g., PLD) by inhibiting the activity of TGR5. For example, provided herein are compounds that reduce or inhibit TGR5 function that are useful for treating a disease associated with TGR5. This reduction or inhibition of TGR5 function can be, for example, through allosteric binding that changes the conformation of the protein and reduces its affinity for one or more ligand(s), and / or through orthosteric binding that directly competes with one or more ligand(s) for binding to the receptor. The ability of test compounds to act as a TGR5 antagonist may be demonstrated by assays known in the art. The activity of the compounds and compositions provided herein as TGR5 antagonists can be assayed in vitro, in vivo, or in a cell line. In vitro assays include assays that determine activation of the protein and / or a change in its conformation. Potency of a TGR5 antagonist as provided herein can be determined by IC50value. A compound with a lower IC50value, as determined under substantially similar conditions, is a more potent TGR5 antagonist relative to a compound with a higher IC50value. FIG. 34 shows representative images of HUCCT1 cells that demonstrated that TGR5 agonist, TLCA, increased cell proliferation in a time-dependent manner. The inactive compound, SBI-155, did not affect cell proliferation. TGR5 antagonist, SBI-364, reduced HUCCT1 proliferation in a time-dependent manner. SBI-155 and SBI-364 were added in the presence of TLCA. TGR5 antagonist SBI-364 decreased levels of p-ERK (i.e., cAMP downstream target) in cholangiocarcinoma cell lines. FIG.35 shows representative immunofluorescence confocal microscopy images that demonstrate the increased levels of p-ERK (stained in red) in response to TGR5 agonist, TLCA. No changes in p-ERK was observed after treatment with inactive compound, SBI-155. In contrast, TGR5 antagonist, SBI-364, markedly decreased p-ERK levels. Cell were treated with all compounds for 30 min. SBI-155 and SBI-364 were added in the presence of TLCA. n = 5-6 for each treatment condition. TGR5 antagonist SBI-364 decreased the tumor growth in mouse model of cholangiocarcinoma. NOD SCID (Nonobese diabetic / severe combined immunodeficiency) male mice (n=5) were implanted with human tumors which express TGR5 (confirmed by western blot, data not shown). As shown in FIG.36, after 27 days of tumor implantation, mice were injected with SBI-319 (3 mg / kg) for 9 days. The graph shows that tumor size was reduced by 30% in response to treatment. Dotted blue line is a projection of a tumor growth in un- treated mice.
