Tongren Wuji Baifeng Wan (a traditional Chinese medicine formula) has the effects of replenishing qi and blood, regulating menstruation and stopping leukorrhea. It is used for qi and blood deficiency, emaciation, soreness and weakness of the waist and knees, irregular menstruation, metrorrhagia and leukorrhea. Its main functions and indications are to replenish qi and blood, regulate menstruation and stop leukorrhea. It is often used for irregular menstruation, menstrual cramps, lower abdominal pain, weakness and fatigue, and soreness and weakness of the waist and legs caused by qi and blood deficiency. Based on the formulation analysis and clinical reports of the drug, this application is the first to conduct a pharmacodynamic study on the treatment of premature ovarian failure with Tongren Wuji Baifeng Wan. Through pharmacodynamic experimental methods, the therapeutic effect of Tongren Wuji Baifeng Wan on premature ovarian failure was clarified.

[0086] The present experimental example verifies the influence of Tongren Wujibaifeng Pill on premature ovarian failure based on the immune premature ovarian failure model mice.

[0087] (I) Preparation of immune premature ovarian failure model and administration method

[0088] BALB / c mice were selected, female, and after adaptive feeding, they were randomly divided into 12 blank groups according to the body weight, and the rest were model groups. 10 mg pZP3 powder was added to 10 mL of normal saline to prepare a solution, which was prepared into an immune reagent with Freund's complete adjuvant at a ratio of 1:1, and an immune enhancer reagent with Freund's incomplete adjuvant at a ratio of 1:1. The model group was injected with 0.15 mL of the immune reagent subcutaneously at the back of the foot and the abdomen of each mouse on day 0, and was given 0.15 mL of the immune enhancer reagent again to strengthen the immunity at the back of the foot and the abdomen subcutaneously on day 14 to establish an immune premature ovarian failure model. All mice were subjected to vaginal exfoliative cell smears for 10 consecutive days, and the change of the estrous cycle of the mice was observed under an optical microscope. The model was successfully prepared when the estrous cycle disorder occurred. On day 21 of the experiment, the successfully modeled mice were randomly divided into a model group, a estradiol valerate tablet group, a high-dose, a medium-dose and a low-dose Tongren Wujibaifeng Pill group according to the body weight, with 12 mice in each group. The mice in each administration group were respectively given corresponding drugs by gavage, and the mice in the blank and model groups were given the same volume of purified water by gavage. The administration was continuously given for 8 weeks, and the specific administration dose was shown in Table 1 in the experimental example 1.

Due to the influence of the physiological structure of the mouse vagina, the cotton swab head was relatively large during the vaginal smear process, which might stimulate the vaginal mucosa, affect the mood and vaginal exfoliative cells of the mice, and interfere with the judgment of the estrous cycle results, so the results were only for reference.

The high dose of Tong Ren Wuji Baifeng Pills could significantly reduce the serum LH level of the model mice and increase the AMH level. The medium and low doses of Tong Ren Wuji Baifeng Pills could significantly increase the serum E2 and AMH levels and decrease the LH level of the model mice, which had significant differences compared with the model group (p<0.05, p<0.01, respectively). The results suggested that Tong Ren Wuji Baifeng Pills had obvious regulatory effect on the estrogen and gonadotropin disorder caused by immune premature ovarian failure, and could increase the anti-Mullerian hormone, indicating that Tong Ren Wuji Baifeng Pills could improve the ovarian reserve function of the model mice.
