[0003] The renal outer medullary potassium channel (ROMK) is a weak inward rectifying K+ channel with a key role in renal K+ recycling and secretion (Ho et al., Nature, 1993, 362, 31-38; Shuck et al., The Journal of Biological Chemistry, 1994, 269(39), 24261-24270; Lee and Hebert, American Journal of Physiology -Renal Physiology, 1995, 268(6), Fl 124-F1131 ; Lu et al., The Journal of Biological Chemistry, 2002, 277, 37881-37887; and Hebert et al., Physiological Reviews, 2005, 85:319-371). In the thick ascending limb (TAL) of a nephron, ROMK channel activity provides the K+ gradient necessary for Na and Cl reabsorption by the Na+-K+-2C1- (NKCC2) co-transporter. In the distal convoluted tubule (DCT) and cortical collecting duct (CCD), ROMK channels form the major secretory pathway for K+ and as a result, play an important role in K+ homeostasis under physiological conditions (Welling and Ho, American Journal of Physiology -Renal Physiology, 2009, 297(4): F849-F863).

[0004] Inhibition of ROMK channel activity can result in natriuresis, diuresis, and reduced blood pressure. Therefore, ROMK inhibition may offer a novel mechanism of blood pressure regulation and diuresis in patients suffering from hypertension, congestive heart failure or any other edematous disease conditions. Inhibition of ROMK may produce diuretic efficacy that is equivalent to or better than currently available loop diuretics and with potentially lower incidence of hypokalemia. [0005] In one aspect, provided herein is an oligonucleotide for inhibiting expression of ROMK, wherein the oligonucleotide comprises an antisense strand comprising at least 14 contiguous nucleotides substantially complementary to a sequence of nucleotides encoding ROMK (e.g., sequence of an RNA produced from KCNJ1), with no more than 4 mismatched nucleotides.

The P3000 enhancer reagent master mix (P3) was prepared by adding 100 ng / well of plasmid DNA and P3000 enhancer reagent at 0.2 pL / well to 5 pL / well of OptiMEM media. Subsequently, L3 and P3 master mixes were combined 1 : 1 to make the L3P3 master mix and incubated at room temperature for 25 minutes. During the incubation for L3 and P3, a master mix of Lipofectamine RNAiMax (ThermoFisher, 13778150) at 0.05 pL / well and OptiMEM 30pL / well was created. 30 pL of RNAiMax master mix was added to each diluted siRNA well for a total of 60 pL per preparation well and allowed to incubate at room temperature for 15 minutes.

Then, Firefly luciferase activity was measured by adding DUAL-GLO® Luciferase Reagent kit (Promega, E1960) containing Luciferase Assay Reagent II (LARII) and STOP & GLO® (S&G) reagents to each well. A Synergy Hl multimode plate reader (Agilent, 8041000N) equipped with dual injectors was used to dispense LARII and S&G, capture and read luminescence. 100 pL of LARII reagent was dispensed per well and the luminescence was captured. Following LARII reagent dispense, 100 pL of S&G was dispensed per well and the second luminescent readout was captured. The DUAL-GLO® STOP & GLO® Reagent works by quenching the firefly luciferase signal and sustaining luminescence for the Renilla luciferase reaction. siRNA activity was determined by normalizing the Renilla ROMK signal to the Firefly (control) signal within each well. The magnitude of siRNA activity was then assessed relative to cells that were transfected with the same vector but were not treated with siRNA or were treated with a nontargeting siRNA.

91. The oligonucleotide of any one of claims 9-85, further comprising a phosphate or phosphate mimic at the 5’-end of the antisense strand.

92. The oligonucleotide of claim 91, wherein the phosphate mimic is a 5 ’-vinyl phosphonate (VP).

93. A pharmaceutical composition comprising the oligonucleotide of any one of claims 1-92.

94. The pharmaceutical composition of claim 93, further comprising a targeting moiety.

95. The pharmaceutical composition of claim 94, wherein the targeting moiety is a targeting ligand.Attorney Docket No. 63578-71660196. The pharmaceutical composition of claim 95, wherein the targeting ligand is a small molecule-based, sugar-based, fatty acid-based, protein-based, or nucleic acid-based targeting ligand.

97. The pharmaceutical composition of claim 96, wherein the protein-based targeting ligand is an antibody, nanobody, affibody, peptibody, or a peptide.

98. A method for inhibiting ROMK expression in a subject, the method comprising administering an effective amount of the oligonucleotide of any one of claims 1-92 or the pharmaceutical composition of any one of claims 93-97 to the subject.

99. The method of claim 98, wherein the subject is a human.

100. The method of claim 98 or 99, wherein the reduction of ROMK mRNA or protein expression levels is measured in a population of kidney cells derived from the subject.

101. The method of any one of claims 98-100, wherein the reduction of ROMK mRNA or protein expression levels is measured in tissues derived from the subject.

102. The method of any one of claims 98-101, further comprising administering to the subject an additional agent or a therapy suitable for treatment or prevention of an ROMK related disorder.
