[0003] Amyloid beta precursor protein (APP) is an integral membrane protein expressed in many tissues and concentrated in the synapses of neurons. APP functions as a cell surface receptor and has been implicated as a regulator of synapse formation, neural plasticity, antimicrobial activity, and iron export. It is coded for by the APP gene and regulated by substrate presentation. APP is best known as the precursor molecule whose proteolysis generates amyloid beta, a polypeptide containing 37 to 49 amino acid residues, where the amyloid fibrillar form is the primary component of amyloid plaques found in the brains of Alzheimer’s disease (AD) , early onset familial AD (EOFAD) , sporadic or late onset AD patients, and cerebral amyloid angiopathy (CAA) patients, and AD-related psychiatric disorders.
[0004] Moreover, APP plays a critical role in synaptic formation and repair. Its expression is upregulated during neuronal differentiation and after neuronal injury. APP may also play a role in cell signaling, long-term potentiation, and cell adhesion. Furthermore, molecules in the cell bodies of neurons must be conveyed outward to the distal synapses via fast anterograde transport. APP plays a role in mediating the interaaction between cargo and kinesin and thus facilitates this transport.

[0006] The present disclosure provides compositions targeting APP and methods of reducing APP expression for treatment of subjects having a disease, disorder, or symptom associated with APP expression level. In some embodiments, the expression level is elevated. In some embodiments, the expression level of APP may not be elevated but suppression of its expression is of benefit.

[0045] The present disclosure provides isolated oligonucleotides (oligonucleotide (s) ) that form a double stranded region, preferably small interfering RNAs (siRNAs) , that can decrease APP mRNA expression, in turn leading to a decrease in the degree of APP protein expression in target cells. The oligonucleotides disclosed herein can have therapeutic applications in regulating the expression of APP for the prevention and / or treatment of a disease or disorder involving APP such as, but not limited to, neurological disorders (e.g., Alzheimer’s disease, dementia, EOFAD, sporadic AD, late onset AD, AD-related psychiatric disorders, Parkinson’s disease, Huntington’s disease, stroke, multiple sclerosis, epilepsy, brain tumors, ataxia, aneurysm, encephalitis, amyotrophic lateral sclerosis, Bell’s palsy, Guillain-Barre syndrome, etc. ) , cardiovascular diseases (e.g., cerebral amyloid angiopathy (CAA) , peripheral artery disease, , stroke) , cancer and malignancies (including metastasis and tumor growth) , and any other disease or disorder that is characterized by increased levels of APP. B6N-Apptm1.1 (APP) Mdk / J, JAX#038920) . The humanized App transgenic mice are dosed subcutaneously at 3 mg / kg and liver samples are collected for APP mRNA silencing analysis.
