There remains a need for more efficacious cell-based therapies that demonstrate enhanced in vivo activity, persistence and extended treatment duration compared to traditional cell-based therapies.SUMMARY
[0005] In one aspect, provided herein is a polynucleotide comprising: (a) a nucleic acid sequence encoding a chimeric antigen receptor (CAR); and (b) a nucleic acid sequence encoding a CD70 shRNA, wherein the CD70 shRNA comprises the sequence set forth in SEQ ID NO: 49.
