Abstract
Description
Ozone-containing composition and method for producing the same
[0001] The present invention relates to an ozone-containing composition and a method for producing an ozone-containing composition.
[0002] Ozone (ozone gas) has long been known to have extremely strong bactericidal properties, which are due to the destruction of bacterial cell membranes or nucleic acids. Therefore, compared to common disinfectants and antibiotics, ozone is capable of inactivating (killing) all bacteria, including highly resistant spore-forming bacteria, and is characterized by not generating resistant bacteria.
[0003] The present inventors have independently developed disinfectants and germicides containing ozone, utilizing these characteristics of ozone, and have filed patent applications and obtained patent rights for them. For example, JP 2009-525974 A (Patent Document 1) discloses a disinfectant containing ozone, which is imparted with germicidal power by mixing ozone into liquid paraffin or Vaseline while stirring the mixture. Furthermore, JP 2013-234132 A (Patent Document 2) discloses an ozone-containing silicone oil imparted with germicidal power by stirring the silicone oil while blowing ozone into the silicone oil as bubbles, thereby incorporating ozone into the silicone oil as fine bubbles. Furthermore, Japanese Patent Laid-Open Publication No. 2015-071586 (Patent Document 3) discloses a volatile disinfectant that is obtained by stirring a volatile solvent containing volatile liquid paraffin, volatile silicone oil, or a mixture thereof as a base while blowing ozone gas into the volatile solvent, thereby incorporating ozone gas into the volatile solvent, and that when the disinfectant is applied to a specified object, it inactivates pathogens on the object and then volatilizes from the object.
[0004] Meanwhile, in addition to its disinfection and sterilization uses, the aforementioned characteristics of ozone are being utilized to develop pest control and insecticide methods and to decontaminate and decompose anticancer drugs used in medical settings. For example, Japanese Patent Laid-Open Publication No. 05-170610 (Patent Document 4) discloses a method for exterminating pests by ozone exposure. It claims that exposure to ozone at a concentration of 0.2 ppm or higher, or to such ozone in combination with trace amounts of nitrogen oxides, can exterminate household pests and nuisance pests such as cockroaches, mites, termites, fleas, lice, flies, and mosquitoes. It also claims that pest extermination can be carried out efficiently using a pest extermination device equipped with an ozone generation means and an ozone decomposition means.
[0005] Furthermore, Japanese Patent Laid-Open Publication No. 2019-208864 (Patent Document 5) discloses a sterilization or pest control system for sterilizing or pest-controlling fruits and vegetables by exposing the fruits and vegetables to a sterilizing or pest-control gas having a sterilizing or pest-control effect in a gas-filled space. This system includes a memory unit, a selection / designation receiving unit, a determination processing unit, and a control processing unit. The memory unit stores a combination of a sterilizing or pest-control gas concentration and an exposure time of the fruits and vegetables to the sterilizing or pest-control gas as a sterilization or pest-control condition for each type of fruit and vegetables. The selection / designation receiving unit receives a selection of the type of fruit and vegetables and a designation of the exposure time or gas concentration. The determination processing unit reads from the memory unit the sterilization or pest-control condition corresponding to the type of fruit and vegetables selected by the selection / designation receiving unit, and determines a gas concentration corresponding to the exposure time designated by the selection / designation receiving unit or an exposure time corresponding to the gas concentration designated by the selection / designation receiving unit. The control processing unit generates a sterilizing or pest control gas and controls the gas-filled space to maintain the gas concentration determined by the determination processing unit or to expose fruits and vegetables to the sterilizing or pest control gas for an exposure time determined by the determination processing unit. The sterilizing or pest control gas is ozone gas. This makes it possible to obtain a sufficient sterilizing or pest control effect on fruits and vegetables using a sterilizing or pest control gas that has a sterilizing or pest control effect on the fruits and vegetables at a gas concentration or exposure time that does not cause damage to the fruits and vegetables.
[0006] Furthermore, Patent Publication No. 2019-073996 (Patent Document 6) discloses a method for treating harmful substances in which ozone gas is applied to decompose or sterilize harmful substances in a humidified environment using an aqueous surfactant solution. The harmful substances in question are anticancer drugs, and the surfactants used include nonionic surfactants, anionic surfactants, cationic surfactants, and amphoteric surfactants. This method is said to be capable of decomposing harmful substances that are difficult to decompose using ozone gas alone, or of promoting the detoxification of harmful microorganisms.
[0007] Furthermore, Patent Publication No. 2014-208428 (Patent Document 7) discloses a method for decomposing an anticancer drug by using air containing ozone and humidified by a humidifying means to decompose the anticancer drug, which is said to be able to protect medical personnel from anticancer drugs that are dispersed outside (such as in a safety cabinet or dispensing room) during dispensing.
[0008] Japanese Patent Publication No. 2009-525974 Japanese Patent Publication No. 2013-234132 Japanese Patent Publication No. 2015-071586 Japanese Patent Publication No. 05-170610 Japanese Patent Publication No. 2019-208864 Republished No. 2019-073996 Republished No. 2014-208428
[0009] However, gaseous, liquid, and solid chemicals are used to kill and exterminate pests, but these chemicals usually have the problem of having adverse effects on the human body.In addition, since chemicals usually have residual properties, there is a problem that the chemicals remaining after application have an adverse effect on the external environment.Furthermore, even if ozone is sprayed on pests, there are cases where the pests do not die, and there is a problem that ozone alone is not effective in killing and exterminating pests.
[0010] The same is true for the decontamination and decomposition of anticancer drugs used in medical settings. While anticancer drugs have pharmacological effects on patients when administered appropriately, they can also pose health hazards to medical professionals, such as doctors, nurses, and pharmacists, who are not patients themselves. For example, inhalation exposure to vaporized anticancer drugs or percutaneous exposure to leaked anticancer drugs can cause health hazards to medical professionals, leading to acute symptoms such as gastrointestinal and circulatory symptoms, or the chronic toxicity (carcinogenicity, etc.) of anticancer drugs, which can lead to cancer. Therefore, in actual practice, medical professionals carefully dispense, administer, and dispose of anticancer drugs according to prescribed procedures, but these measures are often insufficient. Furthermore, typical treatment agents used to decontaminate and decompose anticancer drugs have the problem of remaining on the anticancer drug after application or contaminating the target. The technologies described in Patent Documents 1-7 are unable to solve the above-mentioned problems.
[0011] Therefore, the present invention has been made to solve the above-mentioned problems, and aims to provide an ozone-containing composition and a method for producing an ozone-containing composition that has insecticidal effects, pest repellent effects, and effects such as decontamination and decomposition of anticancer drugs, is safe for the human body, and allows for easy post-treatment after application.
[0012] The ozone-containing composition of the present invention contains a volatile oily solvent, a volatile aqueous solvent, and ozone. The volatile oily solvent has a concentration within the range of 1% to 99% by weight based on the total composition. The volatile aqueous solvent has a concentration within the range of 1% to 99% by weight based on the total composition. The volatile liquid paraffin is one or more volatile liquid paraffins selected from the group consisting of light isoparaffin, light liquid isoparaffin, and hydrogenated polyisobutene. The volatile silicone oil is a volatile silicone oil including cyclic dimethyl silicone oil. The volatile aqueous solvent is a volatile alcohol having a boiling point of 150°C or less and being both hydrophilic and lipophilic, a volatile ketone having a boiling point of 150°C or less and being both hydrophilic and lipophilic, or a mixture of the volatile alcohol and the volatile ketone. The volatile alcohol includes any of ethanol, isopropyl alcohol, and 1-pentanol. The volatile ketone includes any of methyl ethyl ketone and 2-hexanone. The ozone-containing composition according to the present invention is used for insecticide, pest repellent, or decontamination and decomposition of anticancer drugs.
[0013] The method for producing an ozone-containing composition according to the present invention comprises a mixing step and a stirring step. In the mixing step, a volatile oil-based solvent, a volatile aqueous solvent, and ozone are mixed in a predetermined mixing vessel. In the stirring step, the mixture in the mixing vessel is stirred with a predetermined stirrer.
[0014] According to the present invention, it is possible to obtain an insecticidal effect, a pest repellent effect, and an effect of decontaminating and decomposing anticancer drugs, and it is safe for the human body and to facilitate post-treatment after application.

[0016] Hereinafter, an embodiment of the present invention will be described with reference to the accompanying drawings to help understand the present invention. Note that the following embodiment is an example of the present invention, and is not intended to limit the technical scope of the present invention.
[0017] The present inventor has been conducting extensive research into disinfectants and sterilants containing ozone for many years. These disinfectants and sterilants basically involve dissolving ozone in an oily solvent, retaining the ozone in the oily solvent, and bringing the oily solvent into contact with bacteria, whereby the ozone in the oily solvent comes into contact with the bacteria and inactivates them.
[0018] Here, the inventor has attempted to apply such disinfectants and germicides to killing and repelling pests, but has found that pests are surprisingly hydrophilic, and that even if an oily solvent containing ozone is applied to pests, it is difficult to obtain the desired insecticidal or pest repellent effect. Here, insects such as pests do not have lungs, and breathe by directly exchanging oxygen and carbon dioxide through their trachea. Even if ozone is sprayed on pests, the ozone does not properly come into contact with the pests' trachea, so it does not interfere with the pests' breathing, and it has been found that killing or repelling pests using ozone alone is difficult.
[0019] The present inventors also attempted to decontaminate and decompose anticancer drugs, and found that, as described above, anticancer drugs also have hydrophilic properties, so that even if an oily solvent containing dissolved ozone is applied to the anticancer drug, it is difficult to obtain a sufficient effect of decontaminating and decomposing the anticancer drug.
[0020] Therefore, the inventors added a certain amount of an aqueous solvent miscible with the oily solvent and surprisingly discovered that the ozone in the oily solvent contacted the trachea of pests via the aqueous solvent, resulting in insecticidal and pest repellent effects. The same was true for decontamination and decomposition of anticancer drugs, where the ozone in the oily solvent contacted the anticancer drug via the aqueous solvent, resulting in decontamination and decomposition of the anticancer drug. As a result, the inventors have completed the present invention based on the examples described below.
[0021] That is, the ozone-containing composition of the present invention contains a volatile oily solvent, a volatile aqueous solvent, and ozone. The volatile oily solvent has a concentration within the range of 1% by weight to 99% by weight based on the total composition. The volatile aqueous solvent has a concentration within the range of 1% by weight to 99% by weight based on the total composition. The volatile oily solvent is a lipophilic volatile liquid paraffin having a boiling point of 170°C, a lipophilic volatile silicone oil having a boiling point within the range of 170°C to 200°C, or a mixture of a volatile liquid paraffin and a volatile silicone oil. The volatile liquid paraffin is one or more volatile liquid paraffins selected from the group consisting of light isoparaffin, light liquid isoparaffin, and hydrogenated polyisobutene. The volatile silicone oil is a volatile silicone oil including cyclic dimethylsilicone oil. The volatile aqueous solvent is a volatile alcohol having a boiling point of 150°C or less and being both hydrophilic and lipophilic, a volatile ketone having a boiling point of 150°C or less and being both hydrophilic and lipophilic, or a mixture of a volatile alcohol and a volatile ketone.
[0022] Here, volatile liquid paraffin and volatile silicone oil are lipophilic and stable against ozone (do not oxidize). Furthermore, the boiling point of volatile liquid paraffin is 170°C, and the boiling point of volatile silicone oil is in the range of 170°C to 200°C, so both volatile liquid paraffin and volatile silicone oil will volatilize and dry if left exposed to the atmosphere for a while. The SP value of volatile liquid paraffin {(cal / cm 3 ) 1/2} is in the range of 6.5 to 7.5, and the SP value of the volatile silicone oil {(cal / cm 3 ) 1/2} is in the range of 6.5 to 9.5, and the SP value of the volatile liquid paraffin and the SP value of the volatile silicone oil overlap partially, so that the volatile liquid paraffin is mixed uniformly with the volatile silicone oil without reacting with it. 3 ) 1/2 is (evaporation energy / molar volume) 1/2 SP is a parameter that reflects the cohesive strength of a substance, and solvents with similar or overlapping SP values have good compatibility. The SP values are estimated by the Fedors method. In the present invention, ozone is dissolved in such a volatile oily solvent.
[0023] On the other hand, the volatile alcohols and volatile ketones of the volatile aqueous solvents are hydrophilic and lipophilic. Here, the volatile alcohols refer to alcohols with a boiling point of 150°C or less, and the volatile ketones refer to ketones with a boiling point of 150°C or less. The SP value of the volatile alcohols (cal / cm 3 ) 1/2} is in the range of 10.0 to 13.8, and the SP value of the volatile ketone {(cal / cm 3 ) 1/2} is in the range of 8.0 to 10.0. In other words, the SP values of the volatile alcohol and the volatile ketone partially overlap, and also partially overlap with the SP values of the volatile liquid paraffin and the volatile silicone oil. The volatile alcohol is uniformly mixed with the volatile ketone without reacting with it, and the volatile alcohol and the volatile ketone are uniformly mixed with the volatile liquid paraffin and the volatile silicone oil without reacting with it. Furthermore, the volatile alcohol and the volatile ketone have lower reactivity with ozone than carboxylic acids and fatty acids. Therefore, in the present invention, by mixing a volatile aqueous solvent with a volatile oily solvent and ozone, the overall mixture is imparted with hydrophilic properties, which increases the accessibility of the ozone to the trachea of pests and the anticancer drug, thereby enabling the ozone to effectively contact the anticancer drug and pests, thereby achieving insecticidal effects, pest repellent effects, and decontamination / decomposition effects of the anticancer drug. Furthermore, since the boiling point of the volatile aqueous solvent is lower than that of the volatile oily solvent, the volatile aqueous solvent volatilizes first, and then the volatile oily solvent containing ozone volatilizes, making it possible for the ozone to properly come into contact with the trachea of pests and the anticancer agent.
[0024] Furthermore, the volatile oil-based solvents, volatile liquid paraffin and volatile silicone oil, are used in cosmetics, food additives, etc., and are harmless to the human body even if inhaled by a user. Ozone is approved as a food additive by the U.S. Food and Drug Administration (FDA), and at low concentrations, is harmless to the human body even if inhaled by a user. Furthermore, the volatile aqueous solvents, volatile alcohol and volatile ketone, are unlikely to affect the human body, ensuring overall safety. Therefore, the present invention can exert insecticidal effects, pest repellent effects, decontamination and decomposition of anticancer drugs, and other effects without harming the health of users who use them.
[0025] Here, the volatile oily solvent does not react with ozone, and when the volatile aqueous solvent is mixed with the volatile oily solvent, the volatile oily solvent hardly reacts with ozone. Therefore, for example, if the present ozone-containing composition is stored in a sealed container, it can be stored for a long period of time.
[0026] In this way, the ozone-containing composition of the present invention can suitably obtain insecticidal effects, pest repellent effects, decontamination and decomposition of anticancer drugs, etc. in the fields of hygiene, medicine, agriculture, dairy farming, etc. In addition, the composition can also be applied to other uses and fields that require decomposition of hydrophilic objects by ozone.
[0027] Here, the type of ozone is not particularly limited, and examples thereof include a mixed gas of ozone and oxygen, a single gas of ozone only, etc. The mixture ratio of ozone to oxygen in the mixed gas is, for example, in the range of 50% by weight to 2% by weight of ozone and in the range of 50% by weight to 98% by weight of oxygen.
[0028] Furthermore, there are no particular limitations on the concentration of ozone in the present ozone-containing composition. For example, in terms of ensuring the effectiveness of insecticidal effects, pest repellent effects, and decontamination / decomposition of anticancer drugs, as well as safety for the human body, the concentration is preferably in the range of 0.1 ppm to 5000.0 ppm, more preferably 0.1 ppm to 2500.0 ppm, and even more preferably 0.1 ppm to 100.0 ppm, relative to the present ozone-containing composition. This makes it possible to adjust the concentration of ozone in the atmosphere that volatilizes together with the volatile solvent, thereby ensuring safety for the human body. Note that the ozone concentration in the gas and the ozone concentration in the composition (solvent) of the present invention are completely different.
[0029] Furthermore, there are no particular limitations on the type of volatile liquid paraffin used as the volatile oily solvent, and examples thereof include light isoparaffins such as isododecane and isohexadecane, light liquid isoparaffin (a mixture of hydrocarbons having side chains obtained by copolymerizing isobutene and n-butene and then hydrogenating the copolymer), and hydrogenated polyisobutene (a branched aliphatic hydrogen oxide, a synthetic liquid oil primarily composed of isoparaffin). There are no particular limitations on the type of volatile silicone oil used as the volatile oily solvent, and examples thereof include cyclic dimethyl silicone oil. There are no particular limitations on the composition of the volatile liquid paraffin, and it may be either a single type of volatile liquid paraffin or a mixture of two or more types of volatile liquid paraffins. There are no particular limitations on the composition of the volatile silicone oil, and it may be either a single type of volatile silicone oil or a mixture of two or more types of volatile silicone oils. The terms volatile liquid paraffin and volatile silicone oil used herein encompass both of these meanings.
[0030] Furthermore, the type of volatile alcohol in the volatile aqueous solvent is not particularly limited, and includes alcohols with a boiling point of about 150° C. or less. Examples of volatile alcohols include methanol (boiling point about 64° C.), ethanol (boiling point about 78° C.), isopropyl alcohol (boiling point about 80° C.), tert-butyl alcohol (boiling point about 82° C.), allyl alcohol (boiling point about 97° C.), 1-propanol (boiling point about 97° C.), 1-propyl alcohol (boiling point about 97° C.), 2-butanol (boiling point about 100° C.), 2-methyl-2-butanol (boiling point about 102° C.), 3-pentanol (boiling point about 116° C.), and 1-butanol (boiling point about 118° C.). Examples of such alcohols include 2-methoxyethanol (boiling point of approximately 124°C), 2-chloroethanol (boiling point of approximately 128°C), 3-methyl-1-butanol (boiling point of approximately 131°C), isoamyl alcohol (boiling point of approximately 131°C), isopentyl alcohol (boiling point of approximately 131°C), propylene glycol monoethyl ether (boiling point of approximately 133°C), 2-ethoxyethanol (boiling point of approximately 135°C), 1-pentanol (boiling point of approximately 138°C), and propylene glycol monopropyl ether (boiling point of approximately 150°C).
[0031] Furthermore, the type of volatile ketone in the volatile aqueous solvent is not particularly limited, and includes ketones having a boiling point of about 150° C. or less. Examples of volatile ketones include acetone (2-propanone) (boiling point: about 56° C.), methyl ethyl ketone (2-butanone) (boiling point: about 80° C.), methyl vinyl ketone (3-buten-2-one) (boiling point: about 81° C.), methyl isopropyl ketone (3-methyl-2-butanone) (boiling point: about 93° C.), diethyl ketone (3-pentanone) (boiling point: about 101° C.), methyl n-propyl ketone (2-pentanone) (boiling point: about 102° C.), methyl n-butyl ketone (2-hexanone) (boiling point: about 128° C.), acetylacetone (boiling point: about 141° C.), di-n-propyl ketone (boiling point: about 144° C.), and ethyl n-butyl ketone (boiling point: about 148° C.).
[0032] The concentration of the volatile oily solvent is not particularly limited as long as it is within the range of 1% by weight to 99% by weight of the total composition, but is more preferably within the range of 10% by weight to 90% by weight, and even more preferably within the range of 10% by weight to 70% by weight of the total composition, which increases the amount of ozone dissolved in the volatile oily solvent and enables the effects of ozone to be obtained.
[0033] The concentration of the volatile aqueous solvent is not particularly limited as long as it is within the range of 1% by weight to 99% by weight of the total composition, but is more preferably within the range of 10% by weight to 90% by weight of the total composition, and even more preferably within the range of 30% by weight to 90% by weight of the total composition. This allows the volatile aqueous solvent to promote volatility, making it possible to obtain an ozone-containing composition with excellent volatility.
[0034] Furthermore, there are no particular limitations on the mixing weight ratio (-) of the volatile oily solvent to the volatile aqueous solvent. Here, when the mixing weight ratio of the volatile oily solvent to the volatile aqueous solvent (volatile oily solvent / volatile aqueous solvent) is close to 99.00, the amount of volatile oily solvent is greater than the amount of volatile aqueous solvent, resulting in a greater amount of solvent that dissolves ozone, resulting in an ozone-containing composition with high oxidizing power by ozone. On the other hand, when the mixing weight ratio of the volatile oily solvent to the volatile aqueous solvent is close to 0.01, the amount of volatile oily solvent is less than the amount of volatile aqueous solvent, resulting in an ozone-containing composition with excellent volatility. When insecticidal effects, pest repellent effects, decontamination / decomposition effects of anticancer drugs, etc. are expected, the mixing weight ratio (-) of the volatile oily solvent to the volatile aqueous solvent is preferably, for example, within the range of 0.01 to 99.00, more preferably within the range of 0.4 to 15.0, and even more preferably within the range of 0.4 to 10.0. This allows the amount of ozone dissolved in the volatile oily solvent to be increased, thereby achieving the effects of ozone.
[0035] In addition to the volatile oil-based solvent, volatile aqueous solvent, and ozone described above, the ozone-containing composition of the present invention may contain additives, such as oxidizing agents, reducing agents, alkaline agents, acidic agents, fragrances, solvents, dyes, stabilizers, pH adjusters, UV inhibitors, and antioxidants, as long as the additives do not impair the effects of the present invention.
[0036] The type of additive is not particularly limited, but examples thereof include oxidizing agents such as hydrogen peroxide, manganese dioxide, and nitric acid; reducing agents such as iron (II) ions, lithium aluminum hydride, sulfite, oxalic acid, and formic acid; alkaline agents such as sodium hydroxide; acidic agents such as hydrochloric acid; non-volatile liquid paraffin, non-volatile silicone oil, cyclomethylhydrogen silicone oil, hydrocarbons, isoparaffins, mineral spirits, acyclic hydrocarbons, cyclic hydrocarbons, halogenated hydrocarbons, ethers, esters, and carboxylic acids. Examples of acyclic hydrocarbons include pentane, hexane, heptane, octane, nonane, decane, undecane, and dodecane. Examples of cyclic hydrocarbons include cyclopentane, cyclohexane, cycloheptane, and cyclooctane. Examples of halogenated hydrocarbons include methylene chloride, ethyl chloride, chloroform, carbon tetrachloride, and trichloroethylene. Examples of ethers include dimethyl ether, diethyl ether, butyl ether, and tetrahydrofuran (THF). Examples of the ester include methyl acetate, ethyl acetate, n-butyl acetate, γ-butyrolactone, etc. Examples of the carboxylic acid include acetic acid, etc. Also included are solvents containing a double bond such as isophorone and oleic acid.
[0037] Here, from the viewpoint of enhancing the oxidizing power of ozone, examples of additives include alkaline agents such as hydrogen peroxide and sodium hydroxide. The concentration of the additive is set within a range that does not impair the effects of the present invention, and is, for example, preferably within the range of 0.1 wt % to 10.0 wt % of the total composition, more preferably within the range of 1.0 wt % to 5.0 wt % of the total composition, and even more preferably within the range of 1.0 wt % to 3.0 wt % of the total composition. Hydrogen peroxide alone is particularly preferred as the additive, but an alkaline agent alone may also be used, or a mixture of hydrogen peroxide and an alkaline agent may also be used. The presence of an additive containing hydrogen peroxide can be expected to strengthen and sustain the oxidizing power of ozone.
[0038] Furthermore, there are no particular limitations on the manner of use of the ozone-containing composition of the present invention. Here, since both the volatile oil-based solvent and the volatile aqueous solvent have high fluidity, the ozone-containing composition may be directly applied to, sprayed on, or immersed in the target object. Alternatively, the ozone-containing composition may be impregnated into a cloth such as gauze and then applied to the target object. Furthermore, the ozone-containing composition may be filled into an aerosol container (spray container) together with a high-pressure gas (compressed gas, liquefied gas) and used as an aerosol product. Alternatively, a diluted version of the ozone-containing composition may be used.
[0039] Furthermore, there is no particular limitation on the type of pest that can be targeted by the ozone-containing composition of the present invention, and it can be applied to all pests, such as mosquitoes, mites, fleas, lice, flies, bees, horseflies, cockroaches, ants, spiders, and stink bugs.
[0040] Furthermore, the type of anticancer drug that can be treated with the ozone-containing composition of the present invention is not particularly limited, and the composition can be applied to all types of anticancer drugs. Examples of anticancer drugs include alkylating agents, antimetabolites, platinum compounds, topoisomerase inhibitors, microtubule inhibitors, and antibiotics. Examples of alkylating agents include nitrogen mustards such as cyclophosphamide, ifosfamide, busulfan, and melphalan, and nitrosoureas such as nimustine, ranimustine, carmustine, and streptozocin. Examples of antimetabolites include pyrimidine antagonists such as 5-fluorouracil, gemcitabine, cytarabine, and tegafur; purine antagonists such as 6-mercaptopurine, fludarabine, nelarabine, and pentostatin; and folate antagonists such as methotrexate and pemetrix sodium. Examples of platinum compounds include cisplatin, carboplatin, miriplatin, oxaliplatin, and nedaplatin. Examples of topoisomerase inhibitors include etoposide, irinotecan, nogitecan, and sobuzoxane. Examples of microtubule inhibitors include taxanes such as paclitaxel and docetaxel, vin alkaloids such as vincristine, vinblastine, and vindesi, and eribulin such as eribulin mesylate. Examples of antibiotics include anthracyclines such as doxorubicin, mitomycin, and daunomycin, actinomycin D, and bleomycin.
[0041] Next, an apparatus and method for producing an ozone-containing composition according to the present invention will be described. As shown in FIG. 1 , an apparatus 10 for producing an ozone-containing composition according to the present invention includes a generator 11, a mixing tank 12, a supply unit 13, and an agitator 14. The generator 11 generates ozone. The mixing tank 12 contains volatile solvents 12a, including both a volatile oil-based solvent and a volatile aqueous solvent. The supply unit 13 blows ozone 12b into the volatile solvent 12a in the mixing tank 12. The agitator 14 agitates the volatile solvent 12a while blowing in the ozone 12b, thereby incorporating the ozone 12b into the volatile solvent 12a.
[0042] Here, the method by which the generator 11 generates the ozone 12b can be, for example, a silent discharge method, an electrolysis method, a photoreaction method, a high-frequency electrolysis method, a solid polymer electrolysis method, etc. Also, an ozone cylinder containing the ozone 12b may be used as the generator 11 as is.
[0043] The supply unit 13 may blow in the ozone 12b by, for example, arranging the end 12d of a long supply pipe 12c directly on the bottom surface of the volatile solvent 12a, as shown in FIG. 1, or by attaching an air diffuser 12e made of a porous material that releases the enclosed gas as fine bubbles to the end 12d.
[0044] The method of stirring the volatile solvent 12a in the mixing tank 12 by the stirrer 14 may be, for example, by rotating a predetermined stirring blade 14a at a predetermined rotation speed or by rotating a stirrer using magnetic force, as in a magnetic stirrer, as shown in Fig. 1. Alternatively, the ozone 12b may be stirred and atomized by vibrating a predetermined vibrator placed in the volatile solvent 12a or by irradiating the volatile solvent 12a with ultrasonic waves using a predetermined ultrasonic generator.
[0045] Furthermore, since the concentration of ozone 12b in the volatile solvent 12a is proportional to the blowing time of the ozone 12b, the target concentration of ozone 12b (target concentration) can usually be obtained by adjusting the blowing time (sec).
[0046] The method for producing an ozone-containing composition according to the present invention uses the above-described production apparatus 10 and includes a mixing step of mixing a volatile oily solvent, a volatile aqueous solvent, and ozone in a mixing tank 12, and a stirring step of stirring the mixture in the mixing tank 12 with a predetermined stirrer 14. This allows the production of the ozone-containing composition of the present invention.
[0047] In addition to the above, the method for producing an ozone-containing composition according to the present invention may also be modified in such a way that, for example, ozone is incorporated into a volatile oily solvent in advance, and the volatile oily solvent containing the ozone and the volatile aqueous solvent are placed in the mixing tank 12 as a mixture in the mixing step, thereby mixing the volatile oily solvent, the volatile aqueous solvent, and the ozone. Alternatively, for example, ozone may be incorporated into a volatile aqueous solvent in advance, and the volatile aqueous solvent containing the ozone and the volatile oily solvent are placed in the mixing tank 12 as a mixture in the mixing step, thereby mixing the volatile oily solvent, the volatile aqueous solvent, and the ozone. Furthermore, for example, the mixing step may involve placing the volatile oily solvent and ozone in the mixing tank 12 as a mixture and mixing them, and then adding the volatile aqueous solvent and mixing. There is no particular limitation on the order in which the volatile oily solvent, the volatile aqueous solvent, and the ozone are mixed.
[0048] Examples and comparative examples of the present invention will be specifically described below, but the application of the present invention is not limited to these examples.
[0049] (Production of Ozone-Containing Composition) An ozone-containing composition according to the present invention was produced using the production apparatus shown in FIG. 1. As shown in the tables of FIGS. 4 to 8, the components were mixed to produce ozone-containing compositions of Examples 1 to 41 and Comparative Examples 1 and 2. The values in the tables indicate concentrations (wt %). The concentration of ozone, among the components, was adjusted to fall within the range of 0.1 ppm to 5000.0 ppm by adjusting the blowing time. When the ozone concentration is marked "◯", it means a low concentration of, for example, 0.1 ppm to 100.0 ppm, and when the ozone concentration is marked "◎", it means a high concentration of, for example, 100.0 ppm to 5000.0 ppm.
[0050] (Evaluation Method) Next, the performance of the produced ozone-containing compositions was evaluated. The performance evaluation items were (1) mosquito killing test, (2) acaricidal test, (3) anti-mite test, (4) paclitaxel decontamination test, (5) 5-fluorouracil decontamination test, (6) volatility test, and (7) cockroach killing test.
[0051] (1) Mosquito Insecticidal Test A glass cylinder with an inner diameter of 20 cm and a length of 43 cm was placed on its side, one opening blocked with nylon mesh cloth, and the other opening blocked with nylon mesh cloth with sleeves. A predetermined number of test pest mosquitoes (approximately 60) were placed inside the cylinder, and the ozone-containing composition was sprayed directly from the nylon mesh cloth with sleeves toward the inside for 5 seconds. Five minutes and 60 minutes after spraying, the mosquitoes were transferred from the cylinder to a glass petri dish, and the number of dead mosquitoes was counted. The quotient obtained by dividing the number of dead mosquitoes by the number of introduced mosquitoes was calculated as the mosquito mortality rate (%) and ranked according to the following criteria. A rating of "good" or "great" indicates a passing product. Figure 2 shows a front, right side, and left side photographs taken immediately after spraying, showing an example of the results of the mosquito insecticidal test in Example 1.
[0052] <Criteria> ◎: When the mosquito mortality rate is 75% to 100% ○: When the mosquito mortality rate is 50% to 75% △: When the mosquito mortality rate is 25% to 50% ×: When the mosquito mortality rate is 0% to 25%
[0053] (2) Miticidal activity test: 0.5 mL of the liquid ozone-containing composition was evenly dropped onto a filter paper 10 cm long and 5 cm wide, and the filter paper was folded in half to make a size of 5 cm x 5 cm. A predetermined number (several dozen) of live mites (test pests) were placed into the folded part, and the three sides other than the fold were completely sealed with clips. After 24 hours, the clips were removed, the live mites were observed under a microscope, and the number of dead live mites was counted. The quotient obtained by dividing the number of dead mites by the number of introduced mites was calculated as the mosquito mortality rate (%), and the product was ranked according to the following criteria. Evaluations of "good" and "good" indicate that the product is acceptable.
[0054] <Criteria> ◎: When the mite mortality rate is 75% to 100% ○: When the mite mortality rate is 50% to 75% △: When the mite mortality rate is 25% to 50% ×: When the mite mortality rate is 0% to 25%
[0055] (3) Antimite Test: A 40 mm diameter filter paper onto which a liquid ozone-containing composition had been dropped was placed in a petri dish, and 50 mg of a mite rearing medium (approximately 250 mites) for the test pest was spread in the center of the filter paper to a diameter of approximately 3 cm. Additionally, a mite rearing medium was placed in an empty petri dish adjacent to the petri dish. The petri dish was immersed in saturated saline, maintained at 25°C, and left for 24 hours. The mite migration status was then observed. The mite repellency rate (%) was calculated by dividing the diameter of the mites (mite rearing medium) after migration by the diameter of the mites before migration (approximately 3 cm), and the result was ranked according to the following criteria. Evaluations of "good" and "great" indicate acceptable products. Figure 3 is a photograph showing an example of the antimite test results for Reference Example 1 and Example 1. Reference Example 1 is a control, a mixture of a volatile oily solvent and a volatile aqueous solvent that does not contain ozone. As shown in Figure 3, in the anti-mite test, the mites in Reference Example 1 were shown as circles with a diameter of about 3 cm, which was almost the same as the diameter of the mites before migration. On the other hand, the mites in Example 1 were shown as circles with a diameter of less than 1 cm, indicating that the ozone-containing liquid composition, which has a mite-repellent effect, reduced the diameter of the mites.
[0056] <Criteria> ◎: When the mite repellency rate is 0% to 25% ○: When the mite repellency rate is 25% to 50% △: When the mite repellency rate is 50% to 75% ×: When the mite repellency rate is 75% to over 100%
[0057] (4) Paclitaxel Decontamination Test: 10 μL of paclitaxel was dropped onto a stainless steel pad, which was then wiped once with a cleaning wipe soaked in 15 mL of water. The pad was then sprayed with an ozone-containing composition liquid or wiped once with a wipe soaked in the ozone-containing composition. 250 μL of extraction solution was then dropped onto the pad, and the solution was wiped twice with filter paper. The wiped filter paper was then placed into a test tube, and the residual paclitaxel concentration (ng / mL) was calculated using a liquid chromatograph mass spectrometer (LC-MS / MS) and ranked according to the following criteria: A rating of "good" or "great" indicates a passable product.
[0058] <Criteria> ◎: When the paclitaxel concentration after residue is 0.00ng / mL to 0.30ng / mL ○: When the paclitaxel concentration after residue is 0.30ng / mL to 0.60ng / mL △: When the paclitaxel concentration after residue is 0.60ng / mL to 0.90ng / mL ×: When the paclitaxel concentration after residue is 0.90ng / mL or more
[0059] (5) 5-fluorouracil decontamination test. (4) Paclitaxel decontamination test. 10 μL of 5-fluorouracil (5-FU) was dropped onto a stainless steel pad, which was then wiped once with a cleaning wipe soaked in 15 mL of water. An ozone-containing composition liquid was sprayed onto the pad, or the pad was wiped once with a wipe soaked in the ozone-containing composition. 250 μL of extraction solution was then dropped onto the pad, and the solution was wiped twice with filter paper. The wiped filter paper was then placed into a test tube, and the residual 5-FU concentration (ng / mL) was calculated using a liquid chromatograph mass spectrometer (LC-MS / MS) and ranked according to the following criteria. Evaluations of "good" and "great" indicate acceptable products.
[0060] <Criteria> ◎: When the residual 5-FU concentration is 0.00ng / mL to 0.30ng / mL ○: When the residual 5-FU concentration is 0.30ng / mL to 0.60ng / mL △: When the residual 5-FU concentration is 0.60ng / mL to 0.90ng / mL ×: When the residual 5-FU concentration is 0.90ng / mL or higher
[0061] (6) Volatility Test A predetermined amount (about 1 mL, the amount equivalent to one spray) of the ozone-containing composition liquid was dropped onto a glass plate, and then the plate was kept at 25°C and left to stand. 10 minutes after the dropping, the state of drying of the ozone-containing composition liquid was observed. The weight of the ozone-containing composition liquid after 10 minutes was divided by the amount dropped to calculate the dryness rate (%), and the results were ranked according to the following criteria. Evaluations of "good" and "good" indicate that the product is acceptable.
[0062] <Standards> ◎: When the drying rate is 0% to 25% ○: When the drying rate is 25% to 50% △: When the drying rate is 50% to 80% ×: When the drying rate is 80% to 100%
[0063] (7) Cockroach Insecticidal Test: A semi-transparent container measuring approximately 13 cm in diameter, 12 cm in height, and 1.4 L in volume, containing three cockroaches, was used as one treatment area. Next, an aerosol product was prepared by filling an aerosol container with an ozone-containing composition together with high-pressure gas (LPG, liquefied petroleum gas). The aerosol product was sprayed for 3 seconds from a height of approximately 20 cm from the opening of the cockroach container. The cockroaches in the cockroach container were then visually inspected to see if they remained motionless for approximately 30 seconds or if they remained motionless even after shaking the container. If the cockroaches remained motionless, they were considered dead. The time from spraying to the time the cockroaches were confirmed dead was calculated and ranked according to the following criteria. A rating of "good" or "great" indicates a passing product.
[0064] <Criteria> ◎: Death occurred within 5 minutes ○: Death occurred between 5 and 10 minutes △: Death occurred between 10 and 30 minutes ×: Death occurred within 30 minutes, but the victim did not die
[0065] (Evaluation Results) When the evaluation results were confirmed, as shown in Figure 4, Comparative Example 1 contained a volatile oily solvent and ozone, but did not contain a volatile aqueous solvent, and therefore the ozone did not properly come into contact with the trachea of pests or the anticancer drug, and the evaluation results for (1) mosquito insecticidal test, (2) acaricidal test, (3) anti-acaricidal test, (4) paclitaxel decontamination test, and (5) 5-fluorouracil decontamination test were all "△". Here, Comparative Example 1 had a sufficient effect compared to the effect of general insecticides, but could not be evaluated as having a sufficient effect according to the standards of the present invention, and therefore was deemed to have failed as a product. In addition, Comparative Example 2 contains a volatile aqueous solvent and ozone, but does not contain a volatile oily solvent, so the ozone itself does not dissolve in the liquid, and the evaluation results of (1) mosquito insecticidal test, (2) acaricidal test, (3) anti-acaricidal test, (4) paclitaxel decontamination test, and (5) 5-fluorouracil decontamination test were all "X", and the product was unsuccessful.
[0066] On the other hand, in Examples 1 to 9, when the concentrations of the volatile oily solvent and the volatile aqueous solvent were within a specific concentration range, the evaluation results of (1) mosquito insecticidal test, (2) acaricidal test, (3) anti-mite test, (4) paclitaxel decontamination test, and (5) 5-fluorouracil decontamination test were "good" or "great". (6) The evaluation result of the volatility test was "good" or "great", and the product passed the test.
[0067] Next, as shown in FIG. 5, in Examples 10 to 18, even when the type of volatile oily solvent was any one of light isoparaffin, light liquid isoparaffin, hydrogenated isobutene, cyclic dimethyl silicone oil, or a mixture thereof, the evaluation results of (1) mosquito insecticidal test, (2) acaricidal test, (3) anti-acaricidal test, (4) paclitaxel decontamination test, and (5) 5-fluorouracil decontamination test were "◎", and the evaluation result of (6) volatility test was "◯", and the product was acceptable.
[0068] Furthermore, as shown in FIG. 6, in Examples 19 to 29, even when the type of volatile aqueous solvent was any of volatile alcohols (isopropanol, ethanol, 1-pentanol), volatile ketones (acetone, methyl ethyl ketone, 2-hexanone), or mixtures thereof, the evaluation results of (1) mosquito insecticidal test, (2) acaricidal test, (3) anti-acaricidal test, (4) paclitaxel decontamination test, and (5) 5-fluorouracil decontamination test were "◎", and the evaluation result of (4) volatility test was "◯", indicating that the product passed.
[0069] Furthermore, as shown in FIG. 7 , in Examples 30 to 36, even when a volatile oily solvent, a volatile aqueous solvent, and ozone were contained, and further, any of hydrogen peroxide, an alkaline agent such as sodium hydroxide, an acidic agent such as hydrochloric acid, a fragrance, and a colorant was added as an additive, the evaluation results of (1) mosquito killing test, (2) acaricidal test, (3) anti-mite test, (4) paclitaxel decontamination test, and (5) 5-fluorouracil decontamination test were "◎", and the evaluation result of (4) volatility test was "◯", and the product was acceptable.
[0070] Furthermore, as shown in Figure 8, in Examples 37 to 41, despite the low concentration of the volatile oily solvent and the low concentration of ozone, by adding hydrogen peroxide or sodium hydroxide as an additive, the evaluation results of (1) mosquito insecticidal test, (2) acaricidal test, (3) anti-mite test, (4) paclitaxel decontamination test, (5) 5-fluorouracil decontamination test, and (6) volatility test were "Excellent", indicating that the product passed. Hydrogen peroxide was particularly effective. This is thought to be because the oxidizing power of ozone was strengthened by adding an additive containing hydrogen peroxide.
[0071] Here, when the evaluation results of the (7) cockroach insecticidal test were examined, the aerosol product containing only LPG in Reference Example 2 did not kill cockroaches, resulting in an evaluation result of "×". Furthermore, the aerosol product containing the ozone-free composition (volatile oily solvent and volatile aqueous solvent) and LPG in Reference Example 3 took more than 10 minutes to kill cockroaches, resulting in an evaluation result of "△". Figure 8 shows a photograph showing an example of the cockroach insecticidal test in Example 1. On the other hand, the aerosol product containing the ozone-containing composition in Example 1 killed cockroaches immediately, resulting in an evaluation result of "◎", indicating that the product passed the evaluation.
[0072] As described above, the ozone-containing composition and method for producing an ozone-containing composition according to the present invention are useful not only in the fields of insecticide and pest repellent and the decontamination and decomposition of anticancer drugs, but also in the fields of hygiene, medicine, agriculture, dairy farming, etc., and are effective as ozone-containing compositions and methods for producing ozone-containing compositions that have insecticidal effects, pest repellent effects, decontamination and decomposition of anticancer drugs, etc., are safe for the human body, and allow for easy post-treatment after application.
