Description

Oral composition containing herbal ingredients as active ingredients and breath freshener containing the same

[0001] The present invention relates to an oral composition comprising a herbal ingredient as an active ingredient and a breath freshener comprising the same, and more specifically, to an oral composition comprising pine needles, Korean knotweed, and rhubarb as active ingredients and a breath freshener comprising the same.

[0002] Halitosis is a foul odor detected in the oral cavity, which interferes with social activities such as interpersonal relationships. While halitosis can be caused by a variety of factors, including gum disease, dental caries, and gastrointestinal diseases, the most common and significant causes are gum disease and tongue coating. The volatile sulfur compounds produced by oral bacteria in gum disease and tongue coating as they break down oral proteins are the primary contributor to bad breath. Therefore, eliminating the oral bacteria that produce these volatile sulfur compounds is the most effective way to eliminate bad breath.

[0003] Traditionally, mouthwashes and other mouthwashes have been used primarily to combat bad breath. However, these contain alcohol, which can lead to complex problems like dry mouth. Listrin, a representative mouthwash, is known to be highly effective in suppressing bad breath, but its alcohol content is known to be over 10%. Furthermore, conventional mouthwashes and other mouthwashes fail to address the root cause of bad breath, as they do not address the antibacterial activity against periodontal bacteria, the root cause of bad breath.

[0004] Accordingly, there is a growing need for oral hygiene products that are non-toxic to the human body as they do not contain chemical ingredients such as alcohol, and oral hygiene products that can suppress the growth of periodontal bacteria, the fundamental cause of bad breath.

[0057] Hereinafter, examples of the present invention will be described in detail, but it is obvious that the present invention is not limited to the following examples.

[0058]

[0059] Experimental Example 1. Analysis of the ability of 252 herbal ingredients to inhibit the growth of periodontal bacteria.

[0060] 252 herbal ingredients were distributed from the Korea Plant Extract Bank (currently the Natural Products Central Bank of the Korea Research Institute of Bioscience and Biotechnology, Cheongju, Republic of Korea), and the inhibitory activity against periodontal bacteria growth of the corresponding preparations was confirmed through absorbance measurement. The serial numbers, distribution numbers, and country / product names of the 252 herbal ingredients are listed in Table 1. All herbal ingredients used in the experiment were prepared using 95.0% ethyl alcohol, and the detailed extraction method is described in Experimental Example 5 below.

[0061]

[0062]

[0063]

[0064] Hereinafter, the absorbance measurement method is described in detail. First, GAM broth, a periodontal bacteria culture medium, was dispensed into each 96-well plate, and the same amount of periodontal bacteria (Porphyromonas gingivalis or Fusobacterium nucleatum) was applied, and then each of the 252 herbal medicines was added to prepare 252 samples. Next, the absorbance of the 252 samples before placing them in an anaerobic incubator was measured, and the absorbance after 24 hours of incubation in the incubator was measured to calculate the inhibition of periodontal bacteria growth. In order to correct for the difference in absorbance due to the herbal medicine, the absorbance of samples to which only the culture medium and each herbal medicine was applied without adding periodontal bacteria were also measured before and after incubation to establish a reference point.

[0065] The absorbance measured in the above Experimental Example 1 is shown graphically in Fig. 2. The absorbance of all 252 herbal preparations was measured, but only 32 herbal preparations listed in Table 2 below are shown in Fig. 2. The vertical axis of Fig. 2 represents the periodontal bacteria inhibition ability, and the degree of periodontal bacteria proliferation according to the presence or absence of herbal treatment was confirmed by measuring the absorbance, and the periodontal bacteria inhibition ability was calculated using the following Equation 1. More specifically, the lower the y value in Fig. 2, the better the periodontal bacteria inhibition ability.

[0070]

[0071]

[0072] As a result of Experimental Example 1, the periodontal bacteria inhibition ability against P. gingivalis was excellent in many herbal medicine preparations, including Hojanggeun (serial number 205), Rheum amurense (serial number 207), and Songjeol (serial number 217), but the periodontal bacteria inhibition ability was not excellent in tangerine kernel (serial number 61) and Yugeunpi (serial number 219).

[0073] In addition, the periodontal bacteria inhibition activity against F. nucleatum was excellent in many herbal medicine preparations, including Hojanggeun (serial no. 205) and Songjeol (serial no. 217), but the periodontal bacteria inhibition activity was not excellent in broiler chicken (serial no. 170) and rhubarb (serial no. 207).

[0074]

[0075] Experimental Example 2. Analysis of the antibacterial activity of 252 herbal ingredients against periodontal bacteria.

[0076] The antibacterial activity of 252 herbal ingredients against periodontal bacteria was confirmed using the disk diffusion method. The disk diffusion method is described in detail below. First, agar was added to GAM broth, a periodontal bacteria culture medium, and heated to sterilize. After sufficient cooling, periodontal bacteria were mixed and poured into culture dishes to prepare culture dishes containing a mixture of periodontal bacteria.

[0077] Next, an antibacterial activity measurement disk with a thickness of 0.7 mm and a diameter of 60 mm was placed on the culture dish, and 10 μl of the herbal ingredient at a concentration of 100 mg / ml was applied to the disk. After culturing in an anaerobic incubator for 24 hours, the antibacterial activity against periodontal bacteria was confirmed by observing the zone of transparency around the disk. Metronidazole, a representative antibiotic against anaerobic bacteria, was used as a positive control, and the relative antibacterial effect was calculated based on the effect of metronidazole as 100%.

[0078] The results of the above Experimental Example 2 are shown graphically in Fig. 3. As with Fig. 2, the absorbance of all 252 herbal preparations was measured, but Fig. 3 only shows the 32 herbal preparations listed in Table 2.

[0080]

[0081] Experimental Example 3. Cytotoxicity Analysis of 252 Herbal Ingredients

[0082] The cytotoxicity of 252 herbal ingredients was confirmed using the MTT assay. The method for evaluating cytotoxicity is described in detail below. 8,000 cells were seeded onto each culture plate, and after 16 hours, the herbal ingredients were treated and cultured for 24 hours. A tetrazolium dye was applied to the cells treated with the herbal ingredients, and the cells were cultured for an additional 4 hours. The supernatant was removed. The cells from which the supernatant was removed were treated with dimethyl sulfoxide to dissolve the formazan crystals produced by the chemical reaction. The absorbance at a wavelength of 570 nm was measured using a microplate spectrophotometer. Two cell lines were used to confirm toxicity: Ca9-22 cells, which correspond to gingival epithelial cells, and HaCaT cells, which are keratinized epithelial cells.

[0083] The results of the above Experimental Example 3 are shown graphically in Fig. 4. As with Figs. 2 and 3, the absorbance of all 252 herbal preparations was measured, but Fig. 4 only shows the 32 herbal preparations listed in Table 2.

[0085]

[0086] Experimental Example 4. Analysis of the Bad Breath Inhibition Effect of 32 Primary Selected Herbal Ingredients Used Single

[0087] Based on the results of Experimental Examples 1 to 3 above, 32 herbal ingredients with excellent periodontal bacteria inhibition and antibacterial activity and low cytotoxicity were first selected, and the 32 herbal ingredients are as described in Table 2 above. The halitosis inhibition activity of the 32 herbal ingredients when used alone was analyzed using P. gingivalis and F. nucleatum, which are the main periodontal bacteria that produce halitosis. To analyze the halitosis inhibition activity, the methyl mercaptan inhibition activity when the 32 herbal ingredients were used at a concentration of 200 ㎍ / ㎖ was analyzed as a percentage based on the amount of methyl mercaptan when the herbal ingredients were not treated. Briefly, periodontal bacteria were inoculated into the culture medium and cultured for 16 hours. The amount of methyl mercaptan was measured using a halitosis measuring device after 6 hours with or without applying the herbal ingredients. The inhibitory activity was expressed as a percentage of the amount of methyl mercaptan in the treated and untreated samples.

[0088] The results of the above Experimental Example 4 are shown graphically in Fig. 5. As with Figs. 2 to 4, the bad breath suppressing ability of all 252 herbal preparations was measured, but Fig. 5 only shows the 32 herbal preparations listed in Table 2.

The halitosis suppression activity against F. nucleatum was excellent in Jiyu (serial number 187) and Songjeol (serial number 217), and in particular, Songjeol's halitosis suppression activity against F. nucleatum was very excellent.

[0090]

[0091] Experimental Example 5. Analysis of the Bad Breath Control Effect of the Four Final Selected Herbal Ingredients Used Single

[0092] Based on the results of the above Experimental Example 4, four herbal ingredients (Pine needle, Pilbal, Hojanggeun, and Rhubarb) with excellent bad breath suppression properties were finally selected. The extraction method for each preparation was the same, but the initial application weight and final extract weight were different: 1) in the case of Pine needle (distribution number CA04-030), 31.12g was obtained by using 140g, 2) in the case of Pilbal (distribution number CA04-079), 7.54g was obtained by using 100g, 3) in the case of Hojanggeun (distribution number CA03-097), 5.85g was obtained by applying 80g, and 4) in the case of Rhubarb (distribution number CA04-008), 9.53g was obtained by using 74g.

[0093] The extraction method for each preparation is briefly described as follows. The shade-dried herbal medicine was ground into powder, mixed with 1 L of 95.0% ethyl alcohol, and then ultrasonic extraction was performed for 15 minutes at 40 Kz and 1500 W using an ultrasonic extractor (SDN-900H, SD-ULTRASONIC CO., LTD). The extraction was repeated 30 times (cycles) at room temperature. The extract (Qualitative Filter No. 100, HYUNDAI MICRO CO., LTD) was filtered and dried under reduced pressure. The bad breath inhibition ability of the four herbal ingredients mentioned above when used alone was analyzed in the same manner as in Experimental Example 4, and the herbal ingredients were used at a concentration of 20 μg / ml. The results of Experimental Example 5 are shown in a graph in Fig. 6.

[0094] As a result of Experimental Example 5, when the above-mentioned Songjeol, Pilbal, Hojanggeun, and Rhubarb were used alone, the bad breath suppression effect was not excellent. More specifically, when Songjeol was used alone, the bad breath suppression effect against P. gingivalis appeared to be somewhat excellent, but the degree was minimal, and when all ingredients were used alone, the bad breath suppression effect against F. nucleatum was not found.

[0095]

[0096] Experimental Example 6. Analysis of nitric oxide inhibition ability when the four selected herbal ingredients were used alone.

[0097] After selecting four herbal ingredients (Pine Tree, Pine Tree, Hojanggeun, and Rheum) with excellent halitosis suppressing properties, their anti-inflammatory effects were confirmed through nitric oxide suppression assay. First, Raw264.7 macrophages were cultured at 2×10 5 The cells were seeded in 24-well plates and cultured for 16 hours, and then treated with four herbal ingredients at a concentration of 50 μg / ml and cultured for an additional 24 hours.
