Abstract
Description
Technical Field
[0001] This invention belongs to the field of cosmetics, and in particular relates to a gene expression restorer, its application, and skin care and cosmetic products. Background Technology
[0002] Biological rhythms refer to the phenomenon that all life activities change in a certain temporal sequence and periodically. Normal human physiological functions are closely related to biological rhythms, which can be divided into monthly rhythms, weekly rhythms, and daily rhythms based on their duration. Daily rhythms include physiological activities such as sleep-wake cycles, blood pressure, and body temperature cyclical changes, with a cycle of approximately 24 hours.
[0003] The balance and stability of biological rhythms are essential for the healthy life cycle of the body, while disruption of the circadian rhythm can lead to various physiological and psychological abnormalities. With the development of science and technology and changes in people's lifestyles, more and more people are habitually staying up late. Frequently staying up late not only leads to decreased immunity, high blood pressure, and poor skin condition, but also disrupts the body's circadian rhythm, thus affecting both physical and mental health, and in severe cases, even causing disease.
[0004] Molecular mechanisms of circadian rhythms have been studied, revealing that Bmal1 and CLOCK proteins bind to form heterodimers, promoting the expression of genes such as Per (Per1-3) and Cry (Cry1 / 2). When Per and Cry accumulate to a certain level, they form heterodimers and inhibit Bmal1 / CLOCK activity, thereby reducing the transcription and translation of Per and Cry, forming a negative feedback loop with a cycle of approximately 24 hours. Further research has shown that a relatively stable expression level of circadian rhythm genes is essential for normal bodily function. For example, a significant decrease in CLOCK gene expression can weaken or even disrupt circadian rhythms, leading to a loss of circadian rhythms and a series of problems. Conversely, excessive release of CLOCK genes leads to p65 phosphorylation, further enhancing the NFκB signaling pathway and causing inflammation and other issues. Therefore, maintaining circadian rhythm gene homeostasis has become a key research focus.
[0005] Numerous studies have focused on exploring the efficacy of ingredients or compositions that influence the skin's circadian rhythm. For example, patent CN110448478A proposes a composition and its application in the preparation of skincare products that regulate the skin's circadian rhythm. This patent uses skin conditioning agent A containing glycoproteins and amino acids, skin conditioning agent B containing glutamine ethyl imidazole, and adenosine as raw materials to prepare a composition that regulates the skin's circadian rhythm. At the molecular mechanism level, this composition mainly studies how to improve a series of skin problems in people who stay up late by promoting cellular energy synthesis. It only briefly mentions that it can regulate the expression of circadian clock genes without conducting in-depth research on the expression of circadian rhythm genes. Further research has found that this composition has a certain promoting effect on the expression of circadian rhythm genes, but the promoting effect is limited, especially the promoting effect on the expression of CLOCK and Pers genes, which needs to be improved. Therefore, the inventors of the aforementioned patent filed a new patent application, CN116440035A. This new patent application adds Lespedeza bicolor extract as an enhancer to the composition of CN110448478A. Through the interaction between Lespedeza bicolor extract and the promoter, the expression of the CLOCK and Per1 genes can be efficiently promoted. However, this patent does not consider how to maintain the expression of rhythm genes at a relatively stable and normal level to help the body recover to normal levels. Summary of the Invention
[0006] The technical problem to be solved by this invention is to overcome the deficiencies and defects mentioned in the background art above, and to provide an application of glutamine-ethylindole in promoting the expression of circadian rhythm genes, a circadian rhythm gene expression restorer that can restore the expression of CLOCK and Per3 genes to normal and stable levels, as well as skin care products and cosmetics. To solve the above technical problem, the technical solution proposed by this invention is as follows:
[0007] Application of glutamine-ethylindole in promoting the expression of circadian rhythm genes.
[0008] In the above applications, the preferred biological rhythm genes are the CLOCK gene and the Per3 gene.
[0009] As a general technical concept, the present invention also provides a restorer of circadian rhythm gene expression, comprising glutamine ethyl indole.
[0010] Among the aforementioned recovery agents, glutamine and acetylglutamine are preferred.
[0011] Preferably, the above-mentioned restorative agent comprises the following components by mass percentage:
[0012] Glutamidoethylindole 0.2-1%;
[0013] Glutamine 0.01-1%;
[0014] Acetylglutamine 0.01-1%;
[0015] Stabilizer 1-1.5%;
[0016] The remainder is water.
[0017] The stabilizer is specifically p-hydroxyacetophenone and 1,2-hexanediol, with a mass ratio of 1:(1-2). More preferably, the mass ratio of p-hydroxyacetophenone and 1,2-hexanediol is 1:1.2, and their respective proportions in the stabilizer are 0.5% and 0.6%.
[0018] Among the above-mentioned recovery agents, the preferred biological rhythm genes are the CLOCK gene and the Per3 gene.
[0019] As a general technical concept, the present invention also provides the application of the above-mentioned circadian rhythm gene expression restorer in skin care products.
[0020] As a general technical concept, the present invention also provides a skin care product, the components of which include the aforementioned circadian rhythm gene expression restorer. The mass percentage of the restorer is 0.01-2%. When the mass percentage of the restorer is lower than the above percentage, the effect may be less obvious; when the mass percentage of the restorer exceeds the above percentage, it may burden the skin.
[0021] As a general technical concept, the present invention also provides the application of the above-mentioned circadian rhythm gene expression restorer in cosmetics.
[0022] As a general technical concept, the present invention also provides a cosmetic product whose components include the aforementioned circadian rhythm gene expression restorer. The mass percentage of the restorer is 0.01-2%. When the mass percentage of the restorer is lower than the aforementioned percentage, the effect may be less noticeable; when the mass percentage of the restorer exceeds the aforementioned percentage, it may burden the skin.
[0023] In the skin care and cosmetic products of the present invention, other components are not limited and can be conventional components in the prior art, such as conventional moisturizers, thickeners, emulsifiers, emollients, pH adjusters and other components.
[0024] Glutamyl ethyl indole, molecular formula C 15 H 19 Nitrogen oxides (N3O3) are commonly used as a dipeptide in cosmetics and skincare products. By protecting nerve cell lines, they can improve the skin's defense function, resisting damage from environmental stress, ultraviolet radiation, and oxidation, and alleviating discomfort such as dryness and itching. Its molecular structure is as follows:
[0025]
[0026] Generally, existing research on glutamine ethyl indole primarily focuses on its use as an additive in cosmetics, where it works in conjunction with other components, without delving into its promoting effect on circadian rhythm gene expression. Based on previous research, this invention demonstrates that glutamine ethyl indole promotes the expression of circadian rhythm genes, particularly the CLOCK and Per3 genes, and can help restore these genes to normal levels.
[0027] To further enhance the effect of glutamine-ethylindole on the restoration of expression of the CLOCK and Per3 genes, this invention also introduces two other components to synergistically work with glutamine-ethylindole, specifically glutamine and acetylglutamine, whose conventional applications in the prior art are as follows:
[0028] Glutamine, chemically known as 2-amino-5-carboxypentamide, is the amide of glutamic acid. Glutamine is an amino acid that encodes proteins and is also the most abundant free amino acid in muscles, accounting for 60% of the total free amino acids in the human body. Although glutamine is not an essential amino acid for mammals—it can be synthesized in the body from glutamic acid, valine, and isoleucine, and can also be obtained through food—it plays a crucial role in muscle strength, physical exertion, and the repair of damaged tissues. Especially when the body is unable to synthesize enough glutamine due to strenuous exercise, illness, or injury, supplementation is necessary. Today, glutamine is increasingly being promoted and used by athletes and fitness enthusiasts as an important nutritional supplement.
[0029] The molecular mechanisms of glutamine's action include: protecting tissue metabolic function under stress, reducing insulin resistance, maintaining muscle mass, mitigating intestinal barrier dysfunction, anti-inflammatory and immunomodulatory effects, and protective functions against other organs. Studies have shown that glutamine helps increase the activity of superoxide dismutase and catalase, promoting the antioxidant capacity of substances. Furthermore, it can enhance the intestinal epithelial cell barrier function, increase the synthesis of intestinal toxins and bacterial invasion by colonic and small intestinal mucosal cells.
[0030] Glutamine is also widely used in cosmetics. It not only acts as a moisturizing factor for short-term skin hydration, but also promotes the expression of aquaporin AQP3 and caspase-14 to achieve deep hydration and barrier stabilization. Its molecular structure is as follows:
[0031]
[0032] Acetylglutamine, a neuropharmaceutical, was introduced in the early 1980s and has since been combined with acupuncture for the treatment of Alzheimer's disease. Acetylglutamine is an acetylated derivative of glutamine and belongs to the neuropeptide family. While similar in properties to glutamine, acetylglutamine is more stable in water, which is beneficial for its transport, metabolism, and biological effects. In recent years, acetylglutamine has been used as a nutritional support during the recovery process from high-intensity training (racing, swimming, etc.) to promote cell growth. Reports indicate that acetylglutamine promotes amino acid transport, enhances intracellular glutathione and DNA synthesis, and promotes cell mitosis.
[0033] The main functions of acetylglutamine in cosmetics include promoting keratinocyte renewal and enhancing skin's moisturizing ability. It can accelerate the metabolism of the stratum corneum, prevent clogged pores and blackheads caused by thickened stratum corneum, and promote skin cell regeneration, making the skin smoother and more delicate. Acetylglutamine can also increase skin's moisturizing ability, improve skin elasticity and softness, and prevent and improve dryness and roughness. Its molecular structure is as follows:
[0034]
[0035] The above analysis shows that glutamine and acetylglutamine have certain applications in cosmetics and skincare products, but neither has been found to restore the expression of circadian rhythm genes. Our further research indicates that using either glutamine alone or in combination to restore the expression of the CLOCK and Per3 genes is not very effective. However, when glutamine-ethyl indole is combined with glutamine and acetylglutamine, and the amounts of glutamine and acetylglutamine are controlled, glutamine-ethyl indole shows significantly better results in restoring the expression of the CLOCK and Per3 genes. The synergistic effect of glutamine-ethyl indole, glutamine, and acetylglutamine significantly restores the expression of the CLOCK and Per3 genes to normal and stable levels.
[0036] This invention provides an ideal solution for skin problems caused by lack of sleep by using glutamine ethyl indole or its combinations to restore and promote the expression of circadian rhythm genes CLOCK and Per3. The invention uses blue light to construct skin cells with disordered circadian rhythms, and after intervention with glutamine ethyl indole and its combinations, the expression of circadian rhythm genes in these skin cells can be restored to normal levels. Further research revealed that the circadian rhythm genes in skin cells treated with glutamine ethyl indole and its combinations were essentially at the same level as those in skin cells not exposed to blue light, indicating that glutamine ethyl indole and its combinations can restore the expression of circadian rhythm genes in skin cells under blue light exposure to normal levels. This invention also found that glutamine ethyl indole and its combinations can enhance the expression of circadian rhythm genes in cells with disordered rhythms, helping to restore normal circadian rhythm cycles, providing a new skincare approach for circadian rhythm-based cosmetics / skincare products.
[0037] This invention has the following characteristics:
[0038] Delving into Circadian Rhythm Genes: Many biorhythm skincare products achieve daytime protection and nighttime repair by using different products with different functions at different times. This invention addresses the root cause of circadian rhythm disorder by using glutamine ethyl indole and its compositions to restore the low expression of circadian rhythm genes in skin cells to normal levels, thereby ensuring normal skin function and maintaining the skin's biological rhythm.
[0039] Restoration and Normalization of Circadian Rhythm Genes: With the continuous development of functional skincare, many consumers believe that the higher the efficacy and data, the better, neglecting the skin's own needs. This invention, by using glutamine-ethyl indole and its composition in skin cells with disordered circadian rhythms, can restore their circadian rhythm genes to normal levels, without exceeding them by a significant margin. This ensures that the expression of circadian rhythm genes in the skin is at a healthy, normal level, thus helping the skin to return to a healthy state.
[0040] Compared with the prior art, the advantages of the present invention are as follows:
[0041] 1. The circadian rhythm gene expression restorer of the present invention includes glutamine ethyl indole, which can restore the expression of CLOCK gene and Per3 gene to normal and stable levels, help restore the normal circadian rhythm cycle, and provide a new skin care idea for circadian rhythm cosmetics / skin care products.
[0042] 2. The biological rhythm gene expression restorer of the present invention has extremely high biocompatibility, will not cause additional irritation to the skin, has a high safety factor, and is applicable to a wide range of scenarios. It has broad application potential and can be widely used in skin care products, cosmetics and other fields. Attached Figure Description

[0050] An agent for restoring the expression of biological rhythm genes includes glutamine ethyl indole, as detailed in Table 1 below.
[0051] Table 1: Ratio of accelerators in the Examples and Comparative Examples
[0052]
[0053] In the table above, the parts represent the mass percentages. For example, in Example 1, the restoring agent includes 0.2% glutamine-ethyl indole by mass, with the remainder being water and stabilizers. The stabilizers comprise 1.1% by mass (0.5% p-hydroxyacetophenone and 0.6% 1,2-hexanediol), with the balance being water. The following examples and comparative examples may include stabilizers with the same mass percentages as described above.
[0054] The circadian rhythm gene expression restorers in the examples and comparative examples can be prepared using the following method: A measured amount of p-hydroxyacetophenone and 1,2-hexanediol are added to a measured amount of ultrapure water. The mixture is heated and stirred until dissolved and homogeneous, then cooled. When the temperature drops to 45°C, measured amounts of glutamine-ethylindole, glutamine, and acetylglutamine are added, and water is added to make up the difference. After thorough stirring until homogeneous, the mixture is filtered to obtain the restorer. If a component is missing in the examples or comparative examples, it can be omitted during preparation. In comparative examples 4 and 5, indoleacetic acid is used instead of glutamine-ethylindole.
[0055] The aforementioned glutamine-ethyl indole, glutamine, acetylglutamine, and indoleacetic acid can all be obtained from existing commercially available products, such as those from the Titan Technology Exploration Platform or cosmetic raw material suppliers with relevant raw material production and sales qualifications.

[0072] 3) Test Results
[0073] CLOCK gene recovery rate calculation formula: C% = (C x -C B ) / (C0-C B );
[0074] Where C represents the CLOCK gene recovery rate;
[0075] C x This represents the average gene expression value in the efficacy test group after cell experiments;
[0076] C B The average gene expression value of the positive control group (Blue Light, 45 min) after the cell experiment is represented.

[0078] The data was organized to obtain Table 2 below and Figure 1 As shown.
[0079] Table 2: CLOCK gene recovery rate in the examples and comparative examples
[0080]
[0081] from Figure 1 As shown in Table 2, the CLCOK gene restoration agent provided by this invention restores the expression level of rhythmic genes in skin cells to a certain extent, gradually restoring the gene expression level that had decreased after blue light irradiation. This indicates that glutamine ethyl indole and its composition can restore the expression of rhythmic genes in skin cells after blue light irradiation. In particular, in Examples 5-7, the restoration rate of the CLCOK gene exceeds 100%, indicating that it can completely restore the expression of rhythmic genes and help the skin restore normal expression of rhythmic genes even under blue light conditions, while not exceeding the normal value by a large margin. Even if the amount of effective components in the restoration agent is increased, the restoration rate of the CLCOK gene will not increase indefinitely, but will remain at a relatively normal and stable level, which is beneficial for restoring the normal rhythm cycle.
[0082] Comparison of comparative examples and embodiments also shows that the components of the composition of the present invention have a synergistic effect. When some components are missing, the efficacy of the composition is significantly reduced. Furthermore, by optimizing the dosage ratio and proportion of the effective components of the present invention, the expression of the CLOCK gene can be restored to a normal and stable level, which helps the body to recover.
[0083] ②Per3 gene immunofluorescence assay
[0084] 1) Test Background
[0085] The study simulated a decrease in Per3 gene expression in a human body after exposure to blue light, and then used a recovery agent to restore gene expression in order to analyze the effect of the recovery agent on the Per3 gene.
[0086] 2) Testing Methods
[0087] A. Cell seeding: Adjust the density of the fibroblast (HFF-1) suspension to 1.5 × 10⁻⁶. 4 After inoculating each well with 100 μL of the solution, the cells were seeded into 96-well plates and incubated at 37°C in a 5% CO2 incubator for 24 h.
[0088] B. Experimental Groups: A blank control group, a positive control group (blue light irradiation, without the addition of the recovery agent), and an efficacy test group (blue light irradiation, with the addition of the recovery agent) were set up.
[0089] C. Model Construction: Blue light was used for model construction, specifically irradiated at a radiance of 90 ppm and a height of 3 cm for 0, 0.5, 1, and 1.5 hours. Afterward, 100 μL / well of complete culture was added, and the cells were incubated at 37°C with 5% CO2 for 24 hours for static repair. Immunofluorescence staining was then performed to determine the success of model construction.
[0090] D. Raw material efficacy testing: First, blue light modeling was used to construct a cell model irradiated by blue light. The efficacy test group was added with the recovery agent to be tested (corresponding to the recovery agents in Examples 1-7 and Comparative Examples 1-5, respectively). The blank control and positive control group were added with an equal amount of complete culture medium, 100ul / well, and placed in a 37℃, 5% CO2 incubator for static repair for 24h.
[0091] E. Immunofluorescence staining of cells: After sample collection, cell fixation, permeabilization, first wash, blocking, primary antibody, secondary antibody, second wash, and image acquisition, fluorescent images of different samples are obtained.
[0092] F. Image Processing: The fluorescence images were analyzed using ImageJ's average fluorescence intensity.
[0093] G. Statistical analysis: GraphPad Prism 8.0 software was used for data statistical analysis and graph plotting. Quantitative data are expressed as x±s. One-way ANOVA analysis was used to analyze differences between groups, and P<0.05 was considered statistically significant.
[0094] 3) Test Results
[0095] Formula for calculating Per3 gene recovery rate: P% = (P x -P B ) / (P0-P B );
[0096] Where P represents the Per3 gene recovery rate;
[0097] P x This represents the average gene expression value in the efficacy test group after cell experiments;
[0098] P B The average gene expression value of the positive control group (Blue Light, 45 min) after the cell experiment is represented.
[0099] P0 represents the average gene expression value of cells in the blank control group.
[0100] The data was organized to obtain Table 3 below. Figure 2 As shown.
[0101] Table 3: Per3 gene recovery rate in the examples and comparative examples
[0102]
[0103] from Figure 1As shown in Table 2, the Per3 gene recovery agent provided by this invention restores the expression level of rhythmic genes in skin cells to a certain extent, gradually restoring the gene expression level that had decreased after blue light irradiation. This indicates that glutamine ethyl indole and its composition can restore the expression of rhythmic genes in skin cells after blue light irradiation. In particular, in Examples 5-7, the recovery rate of the Per3 gene exceeds 100%, indicating that it can completely restore the expression of rhythmic genes and help the skin restore normal expression of rhythmic genes even under blue light conditions, while not exceeding the normal value by a large margin. Even if the amount of effective components in the recovery agent is increased, the recovery rate of the Per3 gene will not increase indefinitely, but will remain at a relatively normal and stable level, which is beneficial for restoring the normal rhythm cycle.
[0104] Comparison of comparative examples and embodiments also shows that the components of the composition of the present invention have a synergistic effect. When some components are missing, the efficacy of the composition is significantly reduced. Furthermore, by optimizing the dosage ratio and proportion of the effective components of the present invention, the expression of the Per3 gene can be restored to a normal and stable level, which helps the body to recover.
