A method for the synthesis of deflazacort

Abstract

The application discloses a synthesis method of deflazacort and belongs to the technical field of medicine preparation. According to the method, compound 2 (pregn-21-hydroxy-1, 4, 9(11), 16-tetraene-3, 20-diketone-21-acetate (3TR)) is used as a starting material, and deflazacort is obtained through four steps of epoxidation, double hydroxylation, cyclization and bromohydroxy in sequence and finally through debromination treatment. Compared with the prior art, the method has the advantages of mild reaction conditions, economic environmental protection, simple production operation, a short synthesis route, high product yield and the like, and can significantly reduce the production cost of the product. Moreover, the solvent used in the synthesis process can be recycled and utilized, and is very beneficial to industrialized scale production.
