[0162] The compounds of the present disclosure can be synthesized in part by following the steps outlined in Scheme 1-1a below, which contains different orders of assembling intermediates or compounds. Starting materials can be purchased or made by known procedures reported in the literature or as shown below.
[0163] Schema 1
[0164]
[0165] In step 1 of Scheme 1, protected piperazine-2-ethanol (compound (1)) is coupled with 4-bromo-2,5-difluoro-benzoic acid (compound (2)) in amide bond formation using a coupling agent (such as CDMT) and an organic base (such as NMM) in a solvent system (such as acetonitrile and water) and an inorganic base (such as K2CO3). R is a protecting group developed for amino groups, such as carbamate and amide. Such protecting groups are well known and understood in the art, such as carbamate protecting groups, including allyloxycarbonyl, fluorenylmethoxycarbonyl or benzyloxycarbonyl. A common and preferred protecting group can be Boc. Those skilled in the art will recognize that there are many methods and reagents for amide formation resulting from the reaction of carboxylic acids and amines. For example, the reaction of an amine compound with an appropriate carboxylic acid in the presence of a coupling agent in the presence or absence of an organic base such as DIPEA or TEA can provide compound (3). Other coupling reagents include carbodiimides, such as DCC, DIC, EDCI or carbonyldiimidazoles, such as CDI. Amide coupling additives such as HOBt and HOAt can also be used to enhance the reaction. In addition, uronium or phosphonium salts of non-nucleophilic anions such as HBTU, HATU, PyBOP and PyBrOP can be used instead of more traditional coupling reagents. Additives such as DMAP can be used to enhance the reaction. Alternatively, the acyl chloride of compound (2) can be used in the presence of a base such as TEA or pyridine to obtain compound (3).
[0166] In step 2, the intramolecular cyclization of compound (3) is accomplished using a suitable base such as potassium tert-butoxide, sodium tert-amyloxide, sodium tert-butoxide, sodium tert-amyloxide, DIPEA, TEA, DBU, sodium hydride in a solvent such as DMF to give compound 4. Other possible solvents may be NMP, DMAc, DMSO and THF. This intramolecular cyclization of compound (3) to compound (4) can be performed by slowly adding a solution of compound (3) to an excess of base to minimize impurities derived from the intermolecular reaction.
[0167] In step 3, compound (4) can be chlorinated with a chlorinating agent such as trichloroisocyanuric acid or NCS in a solvent such as acetonitrile or DMF under acidic conditions using an acid such as TFA to give compound (5).

[0349] Bioassay
[0350] The following assays demonstrate that exemplary compounds are inhibitors of KRas G12C and inhibit the growth of certain tumors in vitro and / or in vivo.
[0351] KRas G12C probe occupancy TR-FRET assay
[0352] The purpose of this assay is to measure the ability of the inhibitor to compete with the probe for binding to KRas G12C at codon 12 and covalently modify KRas G12C. Europium on an antibody (Eu-labeled anti-histidine tag antibody LanthaScreen (Eu Anti-His antibody)) bound to KRas G12C is combined with a fluorescent tracer 647 (Alexa Fluor 647) bound to KRas G12C via streptavidin and a biotinylated inhibitor ("KRas probe", see Preparation 223). TM ) generates a signal by time-resolved fluorescence transfer between the
[0353] Inhibitors were tested in a dose response format from 10 mM stocks in 100% DMSO. Labycyte 555 was used for dilution and 100 nL / well containing 10-point 2.8-fold serial dilutions were transferred to the assay plate. Two copies of the assay plate were prepared to measure the potency of the inhibitor after 5 and 60 minutes of incubation with KRas G12C. His-tagged KRas G12C (20 nM) was added to the plate in assay buffer (20 mM Tris-HCl, pH 7.5, 0.01% TX-100 and 1 mM DTT). After 5 or 60 minutes of incubation, 1 μM KRas probe was added and allowed to covalently modify free KRas G12C for 1 hour. This was diluted 4-fold in a buffer containing EuAnti-His antibody and Streptavidin-Coated Tracer 647 (both from Life Technologies) to achieve KRas G12C (5 nM), Anti-Hi antibody (2 nM), KRas probe (300 nM), and Streptavidin-Coated Tracer 647 (500 nM). TM Fluorescence signals were read on a plate reader (excitation at 340 nM, tracer emission (em) at 665 nM, and antibody emission at 615 nM). The maximum control wells lacked inhibitor, while the minimum control wells lacked inhibitor and KRas G12C. The signal ratio (em at 665 / em at 615) was converted to percent inhibition using the following equation: % inhibition = 100 - [(test compound signal - median minimum signal) / (median maximum signal - median minimum signal) x 100]. The fluorescence signals were read on a plate reader using Genedata y = (A + ((BA) / (1 + ((C / x) ^ D)))) The percentage inhibition at each inhibitor concentration was fitted to a four-parameter nonlinear logistic equation to determine the IC 50 , where y = % inhibition, A = minimum asymptote, B = maximum asymptote, and C = relative IC 50 or the inhibitor concentration that produces 50% inhibition within the fitted range of the two asymptotes, and D = Hill slope.
[0354] Compounds within the scope of the present disclosure are evaluated in this assay essentially as described above. Exemplary compounds of the present disclosure evaluated in this assay exhibit KRas G12C inhibitor activity by competing with the probe for binding to KRas G12C at codon 12 and covalently modifying KRAS G12C as shown in Table 3.

[0358] Phospho-ERK in H358 cells
[0359] The purpose of this assay is to measure the ability of test compounds to inhibit the phosphorylation of p-ERK1 / 2, a downstream effector of KRas in human lung cancer cells H358 (ATCC CRL-5807). Ultra TM The p-ERK 1 / 2 (Thr202 / Tyr204) assay is a sandwich immunoassay for quantitative detection of phosphorylated-ERK1 / 2 (phosphorylated on Thr202 / Tyr204 in ERK1 or Thr185 / Tyr187 in ERK2) in cell lysates using AlphaTechnology (Perkin Elmer Cat# ALSU-PERK-A50K).
[0360] H358 cells were seeded at 40K cells / well in 100 μL of culture medium (RPMI 1640, GIBCO Cat#22400-071) containing 10% FBS (GIBCO Cat#:10082-147) in a 96-well plate (Costar#3596) and incubated overnight in a humidified chamber at 37°C, 5% CO2. The next morning, 10 μL of serially diluted (3-fold) test compounds (50 μM top concentration) and 10 μL of controls (maximum signal well: 5% DMSO, minimum signal well: 2 μM N-(3-{3-cyclopropyl-5-[(2-fluoro-4-iodophenyl)amino]-6,8-dimethyl-2,4,7-trioxo-3,4,6,7-tetrahydropyrido[4,3-D]pyrimidin-1(2H)-yl}phenyl)acetamide (trametinib, as a positive control)) were added to the cell plate and incubated for 2 hours at 37°C / 5% CO2 in a humidified tray. Lysis buffer containing a cocktail of protease and phosphatase inhibitors was prepared at ambient temperature. The culture medium was removed by inverting and shaking the cell plate in a sink and then blotting onto a paper towel. Lysis buffer was added to the cell plate (50 μL / well) and the plate was incubated on a shaker at ambient temperature for 10 minutes. For p-ERK detection, acceptor beads were diluted in buffer to a suspension mixture. Using a STARlet liquid handler, 5 μL of acceptor beads and 2 μL of cell lysate were transferred to a 384-well assay plate as a single-step in-tip dilution. The assay plate was sealed with foil and incubated at ambient temperature for 2 hours. Donor beads were diluted in buffer to a suspension mixture. Using a STARlet, 5 μL of donor beads were added to the assay plate, which was then sealed and wrapped with foil. The plate was incubated at ambient temperature in the dark for 2 hours. The plate was then incubated in an EnVision TM Assay plates were read on a plate reader (Perkin Elmer) using the luminescence program.
[0361] Convert the signal to percent inhibition using the following equation:
[0362] % Inhibition = 100 - [(Test compound signal - median minimum signal) / (median maximum signal - median minimum signal) x 100]. The maximum signal is the control well with no inhibitor. The minimum signal is the control well containing the reference inhibitor sufficient to completely inhibit activity. y = (A + ((BA) / (1 + ((C / x) ^ D)))) The percentage inhibition at each inhibitor concentration was fitted to a four-parameter nonlinear logistic equation to determine the IC 50 , where y = % inhibition, A = minimum asymptote, B = maximum asymptote, and C = relative IC 50or the inhibitor concentration that produces 50% inhibition within the fitted range of the two asymptotes, and D = Hill slope.

[0366] H358 Cell Active RAS GTPase ELISA
[0367] The purpose of this assay is to measure the ability of the test compound to inhibit constitutive RAS GTPase activity in human lung cancer cells H358 (ATCC CRL-5807). The RAS GTPase ELISA kit (Active Motif Cat#52097) contains a 96-well plate pre-coated with glutathione to capture the GST-Raf-RBD protein supplied with the kit. Activated RAS (GTP-bound) in the cell extract specifically binds to Raf-RBD. The bound RAS is detected with a primary antibody that recognizes human KRas. The secondary antibody conjugated to HRP recognizes the primary antibody, and the developing solution provides a chemiluminescent readout.
[0368] H358 cells were seeded at 80,000 cells / well in 90 μL serum-free medium (RPMI 1640, Gibco) and incubated overnight at 37°C / 5% CO2. The next morning, 10 μL of serially diluted (3-fold) test compounds (500 μM highest concentration) and 10 μL of controls (maximum signal well: 5% DMSO, minimum signal well: 500 μM 1-[4-[6-chloro-8-fluoro-7-(3-hydroxy-1-naphthyl)quinazolin-4-yl]piperazin-1-yl]prop-2-en-1-one, WO2015054572, as inhibitor) were added to the cell plate and incubated at 37°C / 5% CO2 for 2 hours. Complete lysis / binding buffer containing protease inhibitor cocktail and GST-Raf-RBD was prepared and stored on ice. One hour before the cell plate incubation was completed, 50 μL of GST-Raf-RBD was diluted in lysis / binding buffer and the buffer was added to the ELISA assay plate, which was incubated at 4 ° C for 1 hour under gentle shaking. After 2 hours, the cells were washed with 100 μL of ice-cold PBS and lysed with 100 μL of lysis / binding buffer. The cell plate was vibrated for 10 minutes at ambient temperature. The cell plate was then centrifuged at 1500 rpm for 10 minutes at ambient temperature. During this period, 1X washing buffer was prepared at ambient temperature and then used to wash (3x100 μL) GST-Raf-RBD coated assay plates. After washing, 50 μL of cell lysate was added to the GST-Raf-RBD coated assay plate and incubated for 1 hour at ambient temperature under gentle shaking. During this incubation period, 1X antibody binding buffer was prepared and allowed to reach ambient temperature. The assay plate was washed 3x100 μL with 1X Wash Buffer, then 50 μL of primary antibody (diluted 1:500 in 1X Antibody Binding Buffer) was added. The plate was incubated for 1 hour at ambient temperature. The assay plate was washed 3x100 μL with 1X Wash Buffer, then 50 μL of secondary antibody (diluted 1:5000 in 1X Antibody Binding Buffer) was added and incubated for 1 hour at ambient temperature. The assay plate was washed 4x100 μL with 1X Wash Buffer, then 50 μL of Chemiluminescent Working Solution was added at ambient temperature. The plate was then incubated for 1 hour at EnVision TM Assay plates were read on a plate reader (Perkin Elmer) using the luminescence program.
